Paranodal structural abnormalities in rat CNS myelin developing in vivo in the presence of implanted O1 hybridoma cells.

O1 hybridoma cells, which produce a monoclonal IgM antigalactocerebroside, were implanted into the spinal cords of immature and mature rats and the cords examined 5-24 days later. Study of the younger group, in which myelin was developing at the time of implantation, revealed examples of abnormal myelin sheaths in which the repeat period was markedly increased. The paranodal regions of these abnormal sheaths were superficially normal in configuration; i.e. myelin lamellae terminated one by one as 'terminal loops' that indented the axolemma and formed normal axoglial junctions displaying periodic 'transverse bands'. Neighbouring terminal loops are normally joined by tight junctions that block passage of tracers from the paranodal periaxonal space into the compact myelin, as seen after implantation of a control hybridoma. In the abnormal sheaths that developed after O1 implantation, in contrast, terminal loops were usually widely separated from each other. As a result, multiple pathways from the paranodal periaxonal space into the myelin sheath remained patent, forming potential routes for shunting nodal action currents. This subtle abnormality could thus compromise conduction, even though the sheaths might appear to be normally myelinated at the histological level. Equivalent abnormalities in human neurological diseases, including multiple sclerosis and paraproteinemic neuropathies, could underlie functional loss in the absence of frank demyelination.

Serum biochemical values and mineral element contents of tissues in yaks.

Serum biochemical values and the wet weight to dry weight ratios of tissues were determined in yaks in Shandan county of Gansu province. The liver, kidney, heart and muscle contents of seven elements in yaks were also analysed. Most serum biochemical values were similar to those of cattle, camels and sheep, but the calcium concentration was considerably above the normal range for other ruminants. The liver contained the highest concentrations of copper, manganese and cobalt and the kidney of selenium, iron and calcium.

Object classification for human and ideal observers.

We describe a novel approach, based on ideal observer analysis, for measuring the ability of human observers to use image information for 3D object perception. We compute the statistical efficiency of subjects relative to an ideal observer for a 3D object classification task. After training to 11 different views of a randomly shaped thick wire object, subjects were asked which of a pair of noisy views of the object best matched the learned object. Efficiency relative to the actual information in the stimuli can be as high as 20%. Increases in object regularity (e.g. symmetry) lead to increases in the efficiency with which novel views of an object could be classified. Furthermore, such increases in regularity also lead to decreases in the effect of viewpoint on classification efficiency. Human statistical efficiencies relative to a 2D ideal observer exceeded 100%, thereby excluding all models which are sub-optimal relative to the 2D ideal.

Development of early bovine embryos in co-culture with KSOM and taurine, superoxide dismutase or insulin.

Three experiments, utilizing 2578 embryos, were designed to test the effects of media, taurine, Superoxide dismutase and insulin on the development of embryos produced by in vitro maturation and in vitro fertilization (IVM/IVF). Embryos showing at least 1 cleavage during culture for 40 to 44 h after IVM/IVF were selected for further culture under various conditions for 6 d at 39 degrees C in 5% C0(2):95% air. A Buffalo rat liver (BRL) cell co-culture was used in all 3 experiments. Experiment 1 was a 3 x 2 factorial arrangement with KSOM (a high potassium simplex optimization-derived medium containing only 12 ingredients), Menezo B(2) and TCM-199 media with or without 7 mM taurine. Blastocyst production in the 3 media, respectively, was 48, 36 and 29% (P<0.05). Addition of 7 mM taurine increased the percentage of blastocysts from 34 to 42 (P<0.05). In Experiment 2, Superoxide dismutase (SOD) did not improve blastocyst development (P>0.05). In Experiment 3, insulin (75 ng/ml) added to KSOM resulted in 46% morulae plus blastocysts compared with 35% for the control (P<0.05). These results indicate that the co-culture of embryos in KSOM with taurine or insulin added is superior to commonly used complex media for efficient production of blastocysts following IVM/IVF.

Study on the antimutagenic effect of pine needle extract.

The micronucleus test and sister-chromatic exchange (SCE) test were used to research the antimutagenic effect of pine needle extract. The results showed that the mutagenic effect of cyclophosphamide (CP) was inhibited by the pine needle extract. The micronucleus frequencies (MNF) of mouse bone marrow and human lymphocytes from peripheral blood were decreased with the effect of the extract (the dose was 2000 mg/kg or 5 mg/ml); the frequency of SCE in human lymphocytes was also reduced significantly, which indicated that the MNF and the SCE frequencies were negatively correlated with the dose of pine needle extract (r = -0.9782, -0.9587, -0.9765, respectively). This suggested that the pine needle extract was an effective antimutagen and it is important to choose the proper doses of pine needle extract for antitumor effect.

Mutagenic activity of dopamine after nitrosation.

Dopamine hydrochloride is reported to be a new mutagen precursor in this study. After treatment with nitrite under acidic conditions, dopamine hydrochloride showed direct-acting mutagenicity on Salmonella typhimurium TA100, TA98 and Escherichia coli WP2uvra. The addition of S9 mix did not affect the mutagenicity of nitrosated dopamine significantly in these three strains. Meanwhile, a comparison of the mutagenicity of nitrosated dopamine with nitrosated tyramine was carried out.

Morphine-induced prolactin release precedes a down-regulation of prolactin receptors in the male rat choroid plexus and hypothalamus.

In previous studies we provided evidence for changes in prolactin (PRL) receptor levels in the male rat brain after continuously infusing morphine using subcutaneously implanted miniosmotic pumps. In this work we have studied the binding of PRL in the male rat brain following morphine administration by both subcutaneous (s.c.) and intracerebroventricular (i.c.v.) injections. The binding in the choroid plexus and the hypothalamus was measured using iodinated ovine PRL (oPRL) as a radiolabel. The results indicated that the density of the PRL-binding sites in the hypothalamus and the choroid plexus were significantly decreased 4 h and 24 h after s.c. injections, and also 30 min and 4 h after i.c.v. injections. However, no decrease in PRL-binding was observed 15 min after i.c.v. injection of morphine. The plasma levels of PRL were measured by radioimmunoassay (RIA) and were found to be significantly increased after 30 min and 4 h in all treated animals. Following i.c.v. injection a significant increase in plasma PRL was observed after just 15 min. It was suggested that the down-regulation in PRL binding to some extent at least resulted from receptor overstimulation caused by the morphine-induced elevation in the concentrations of the endogenous hormone.

The changing availability of residential aged care in Australia.

Australian aged care services have undergone a series of substantial reforms in recent years under the rubric of the Aged Care Reform Strategy. Overall, there has been a progressive refinement of the targeting of available services on those most in need, defined in terms of both disability levels and financial resources. A key component of this process has been a deliberate reduction in the relative emphasis accorded to nursing home care within the aged care system. This has been accompanied by increases in the resources directed toward less intensive forms of residential care (hostels) and community based services. This paper explores the actual consequences of these intentional policy changes in terms of the availability of nursing home and hostel care, and the changing characteristics of nursing home residents. The results suggest that a more substantial reduction has occurred in the availability of nursing home care than has hitherto been suggested, with consequent decreases in the proportion of aged persons in nursing homes. The effect has been particularly marked amongst women and the very old. These findings raise policy questions about the appropriateness of current and planned future levels of provision.

Mesial temporal sclerosis: pathogenesis and significance.

Mesial temporal sclerosis (MTS) is a common pathologic finding in patients with temporal lobe epilepsy. Rarely MTS can be detected in children during the first decade of life, but is not commonly found until adolescence. Although the etiology of MTS remains controversial, there is now a considerable amount of evidence demonstrating that MTS is both a result and a cause of seizures. Clinical studies suggest that prolonged seizures or complicated febrile seizures may result in MTS. A variety of epileptogenic agents administered to adult animals have resulted in MTS and spontaneous recurrent seizures. The mechanism of the lesions is due to excessive excitability secondary to release of excitatory amino acids, primarily glutamate. Glutamate, acting at a number of subreceptors on the postsynaptic membrane, leads to prolonged depolarization of neurons and results in the entry of cytotoxic amounts of calcium. Interestingly, the same agents that produce MTS in adult animals do not produce MTS in immature animals. Clinical and experimental evidence suggests that although prolonged seizures or complicated febrile seizures can place a child at risk for MTS, a period of time is required for the lesions to develop fully.

Two-dimensional crystallization of avidin on biotinylated lipid monolayers.

Two-dimensional crystals of avidin were obtained on mixed lipid monolayers containing biotinylated lipids (N-biotinyl-dipalmitoyl-L-alpha-phosphatidyl ethanolamine and dioleoyl phosphatidyl choline) by specific interaction. Image analysis of electron micrographs of these crystals revealed p2 symmetry with the unit cell parameters a = 66 +/- 2 A, b = 68 +/- 1 A, and gamma = 121 +/- 4 degrees. The projection map showed, at a resolution of about 27 A, that the four subunits within one avidin molecule are separated into two parts. Comparison between avidin and streptavidin reveals that avidin molecule binds to the lipid monolayer in an orientation similar to that of streptavidin.

Simultaneous determination of guanine, uric acid, hypoxanthine and xanthine in human plasma by reversed-phase high-performance liquid chromatography with amperometric detection.

A reversed-phase high-performance liquid chromatographic method with amperometric detection is described for the separation and quantification of uric acid, guanine, hypoxanthine and xanthine. The isocratic separation of a standard mixture of the compounds was achieved in 5 min on a Spherisorb 5 C18 reversed-phase column, with a mobile phase of NaH2PO4 (300 mmol dm-3, pH 3.0)-methanol-acetonitrile-tetrahydrofuran (97.8 + 0.5 + 1.5 + 0.2). Uric acid, guanine, hypoxanthine and xanthine were completely separated, with detection limits in the range 2-20 pmol per injection. The effect of pH and the composition of the mobile phase on the separation are described. The hydrodynamic voltammograms of these compounds were recorded at a glassy carbon electrode. The linear range of the calibration graph for each compound was: uric acid, 1-5000 mumol dm-3; guanine, 0.5-2000 mumol dm-3; hypoxanthine, 0.1-500 mumol dm-3 and xanthine, 0.5-5000 mumol dm-3. The within- and between-day precision was good. The uric acid and hypoxanthine content in human plasma was measured using the proposed method. Good recoveries of uric acid (97.9-103%), hypoxanthine (98.0-99.2%), guanine (96.0-98.3%) and xanthine (96.0-102%) were obtained from human plasma. The results of electrochemical detection were in good agreement with those of UV detection.

The effects of augmented information on motor learning: a multidimensional assessment.

Subjects performed trials of a two-hand coordination task involving the manipulation of a small, steel ball through a maze. During the acquisition phase, control subjects received no augmented information while subjects in two experimental groups were provided with visual aids or a cognitive strategy. One day later, subjects performed under control conditions on familiar (relearning phase) and unfamiliar (transfer phase) sections of the maze. A 3 x 3 (Group x Phase) repeated measures multivariate analysis of variance revealed significant phase differences for all dependent measures but no significant group effects. Chi-square analysis of ratings of cue usage as well as postexperimental interviews suggested, however, that practice conditions differentially influenced the search strategies of subjects. Implications of the findings for contemporary theories of motor learning are discussed.

Radiation-sensitive mutants of Arabidopsis thaliana.

Five Arabidopsis mutants have been isolated on the basis of hypersensitivity of leaf tissue to UV light. For each mutant, the UV-hypersensitive phenotype (uvh) was inherited as a single recessive Mendelian trait. In addition, each uvh mutant represented a separate complementation group. Three of the mutations producing the UV hypersensitive phenotype have been mapped relative to either genetic markers or physical microsatellite polymorphisms. Locus UVH1 is linked to nga76 on chromosome 5, UVH3 to GL1 on chromosome three, and UVH6 to nga59 on chromosome 1. Each uvh mutant has a characteristic pattern of sensitivity based on UV sensitivity of leaf tissue, UV sensitivity of root tissue, and ionizing radiation sensitivity of seeds. On the basis of these patterns, possible molecular defects in these mutants are discussed.

Virus burden in long-term survivors of human immunodeficiency virus (HIV) infection is a determinant of anti-HIV CD8+ lymphocyte activity.

Persons infected with human immunodeficiency virus (HIV) for > 8 years were studied to delineate virologic and immunologic attributes of long-term survival. Whereas those with 300-700 CD4+ cells/microL often had circulating cytotoxic T lymphocytes (CTL) against HIV antigens, those with > 1000 CD4+ cells/microL did not. The subjects with > 1000 CD4+ cells/microL had low virus burden, low levels of Gag-specific CTL precursors, and minimal CD8+ cell activation. Overall, elevated levels of CD8+ cells, CD38 antigen expression on CD8+ cells, and anti-HIV functions were correlated with increased virus burden, provirus load, and HIV plasma RNA levels. A factor that suppressed HIV replication was spontaneously secreted from CD8+ cells of most subjects but not from those with high CD4+ cell counts. CD8+ cell activities, therefore, may reflect chronic viral stimulation of the immune system. Long-term survivors with high levels of CD4+ cells maintained control of viral replication but lacked the CD8+ cell activities.

Effects of amino acids on the development of in-vitro matured/in-vitro fertilization bovine embryos in a simple protein-free medium.

The effects of commercially available amino acids, referred to as Eagle's non-essential amino acids (NEAA) and essential amino acids (EAA), on the development of bovine embryos produced by in-vitro maturation and in-vitro fertilization (IVF) in a simple modified protein-free KSOM medium containing polyvinyl alcohol (PVA) were studied. Embryos produced by IVF were cultured in an atmosphere of 5% O2, 5% CO2 and 90% N2 at 39 degrees C for the first 40-44 h in modified KSOM. Then embryos with four or more cells were cultured in modified KSOM-PVA with different amino acids for another 6 days in experiments 1 and 2. In experiment 3, to study embryo hatching and degeneration further as criteria of medium suitability, the culture time was 8 days. In experiment 1, more blastocysts were produced (P < 0.05) in the medium with 0.5 x NEAA (37%) than with 0.25 or 0.5 x EAA (both 15%). In experiment 2, when 0.0, 0.5 and 1.0 x concentrations of NEAA were combined with 0.0 and 0.5 x concentrations of EAA, the combination 1.0 x NEAA with 0.5 x EAA resulted in the most blastocysts (39%), with 11% hatching (P < 0.05). Without NEAA, 1.0 x EAA resulted in 6% blastocysts (P < 0.05). In experiment 3, KSOM-PVA with 1.0 x NEAA plus 0.5 x EAA resulted in 41% blastocysts, similar to the 40% blastocysts in KSOM-bovine serum albumin (BSA) with the same amino acids added. When culture was continued for 2 more days, more embryos hatched in these two treatments. These results indicate that the combination of 1.0 x NEAA with 0.5 x EAA promoted blastocyst formation and hatching, but EAA alone was detrimental. With PVA successfully replacing BSA, a completely defined medium resulted, which was equal or superior to complex medium, and is potentially useful for the culture of embryos from other species, such as humans.
