Growth of Serpulina (Treponema) hyodysenteriae under iron-restricted conditions.

Reference strains of Serpulina hyodysenteriae expressed at least three iron-regulated proteins with apparent molecular masses of > 200, 134, and 109 kDa when grown under iron-restricted conditions. Cells of S. hyodysenteriae grown under these conditions also showed increased outer membrane bleb formation when examined by electron microscopy after negative staining. S. hyodysenteriae did not use the 2 most common types of siderophore, namely catechol and hydroxamate. Western blotting with serum from a pig experimentally infected with S. hyodysenteriae B204 indicated that the 109-kDa major iron-regulated protein was expressed in vivo and was conserved among all strains tested.

Protective effect of radix Salviae Miltiorrhizae Composita on cerebral ischemia.

The protective effects of Radix Salviae Miltiorrhizae Composita (RSMC) on cerebral ischemia in gerbils produced by unilateral ligation of left common carotid artery and on the development of ischemic cerebral edema were studied. It was found that the administration of RSMC resulted in a very significant decrease in neurological deficits and the time for exhibiting first signs of neurological deficit delayed compared with untreated animals. Although the values of specific gravity of fresh cerebral tissue on the ligated side of common carotid artery in untreated animals (1.0406 +/- 0.0039 SD) and in RSMC-treated animals (1.0453 +/- 0.0009) were decreased significantly compared to that (1.0468 +/- 0.0003) of sham-operated animals, the specific gravity of cerebral tissue in RSMC-treated animals was significantly heavier than that in untreated animals (t = 5.5022, P < 0.001), i.e. the administration of RSMC decreased the severity of cerebral edema. A significant negative correlation was found between stroke index and specific gravity, and it suggests that one of the protective mechanisms of RSMC might be related to its ameliorating cerebral edema.

[Electromyographic study of human extrinsic laryngeal muscles].

Seven cases of human extrinsic laryngeal muscles were studied to determine the electrical activities during phonation, coughing, swallowing, fixation of thorax and respiration. The activities of these muscles all fired during phonation which were synchronous with discharge of the thyroarytenoid muscle (TAM) and increased with increasing volume of voice. They also fired during cough reflex and sphincteric action of larynx, and were basically synchronous with the TAM. They were weak during normal respiration. Although stronger electrical activities were recorded during inspiration in dyspnea, they were weaker than discharge during phonation, cough reflex and sphincteric action of larynx.

[The DNA damage by photodynamic effects of hematoporphyrin derivatives (HPD)].

Photodynamic effects of HPD has been used for cancer therapy (PDT) successfuly. The biological mechanism of the PDT was studied by using DNA as a target in this work. After treatment of phage M13 DNA with HPD + light the molecule of the DNA kept intact in TE buffer. But the smear bands by puting DNA in 0.1 mol/L NaOH at 90 degrees C and separating with electrophoresis denoted the degradation of DNA at the alkali-labile sites of phosphodiester bond in which the groups of the bases were photooxydized. The primer extension experiment showed the aggregation of DNA template by photooxydation. The DNA sequencing results showed that some of the wrong sequences appeared both at the site of A,G,C,T bases. The mouse Ehrlich ascetes carcinoma cell DNA was treated as above then degraded by Bgl II and BamH I respectively. It was found that the restriction fragments became larger than the controls. It also meant the alteration of the bases of DNA by the photooxydation. So the four bases of A,G,C,T of DNA were altered by photodynamic effects of HPD.

Anti-rhodopsin antibodies in sera from patients with normal-pressure glaucoma.

PURPOSE: To explore further the potential role autoimmunity may play in the pathogenesis of normal-pressure glaucoma (NPG) in some patients, the authors examined the sera of patients with NPG for the presence of antibodies directed toward retinal antigens. METHODS: Using patient sera, immunoblotting was performed on subcellular fractions of retina, purified bovine rhodopsin, and immunoaffinity-purified recombinant human rhodopsin. A chemiluminescence-enzyme-linked immunosorbent assay (ELISA) to detect anti-rhodopsin antibodies was developed and used. RESULTS: A patient with NPG was found to have a high titer of immunoglobulin M-lambda antibody against a 40-kd retina-specific glycoprotein antigen subsequently identified as rhodopsin. ELISA analysis conducted on sera from 28 patients with NPG and 26 patients with primary open-angle glaucoma (POAG) revealed highly significant differences in anti-rhodopsin antibody activity between these groups (P < 0.0002, Mann-Whitney test). For example, the majority of patients with NPG (19/28; 68%) had anti-rhodopsin antibody activity higher than the highest value obtained from among 26 age-matched patients with POAG. CONCLUSIONS: An elevated anti-rhodopsin antibody count is related to NPG. This may indicate that there is an autoimmune component in the optic neuropathy in these patients. The specific role of these autoantibodies, if any, in the pathogenesis of the disease remains to be determined.

p10 genes of Bombyx mori nuclear polyhedrosis virus and Autographa californica multiple nuclear polyhedrosis virus.

EcoR I-P fragment has been cloned from Autographa californica multiple nuclear polyhedrosis virus (AcMNPV) genomic DNA and used as a probe. 0.5-kb and 1.1-kb fragments including p10 gene from Bombyx mori nuclear polyhedrosis virus (BmNPV) have been hybridized. The p10 ORF was located in the EcoR I-R fragment. Initiation codon ATG of p10 from BmNPV has been mutated by PCR, and the ATG region became a Bgl II site. A novel transfer vector pBmAcPV-1 has been constructed using both the p10 5'-flanking region whose initiation codon ATG has been mutated with BmNPV and the p10 3'-flanking region of AcMNPV. The vector can recombine with not only AcMNPV DNA to express foreign gene in Sf cells, but also BmNPV DNA to express foreign gene in Bm cells. CAT gene was expressed at high level in Bm cells under the control of the mutated p10 promoter of BmNPV.

HBV surface antigen proteins with deletions in the preS region.

Four pairs of HBV surface antigen genes, in which the preS region was partially deleted, were constructed by the polymerase chain reaction (PCR). The comparison of the levels of the expression in mammalian cells of these genes and the ones constructed before, and the properties of these gene products showed that the missing of a part of the preS region did not affect the overall spatial structure of the S region and the surface localization of the preS region. The removal of the preS1 retention sequence (a. a. 2-19) alleviated significantly the shelter of the major antigenic determinants in the S region by the preS sequence. It was found that the long preS region seriously impaired the secretion of the surface antigen proteins from mammalian cells. In addition to the previously reported preS1 retention sequence, the preS1 sequence (a.a. 48-65) may also inhibit the secretion of the surface antigen proteins, whereas the preS2 region exerts no major influence on the retention of the large surface antigen protein. One of the expressed surface antigen proteins, in which the preS1 sequence (a.a. 21-47) and the S region were directly fused, deserves further study and may be developed into a new HBV vaccine which contains the preS1 binding site for hepatocyte receptors due to its stability, fine secretability and strong preS1 antigenicity.

Diagnosis and surgical treatment of bronchogenic cysts.

Between 1974 and 1993, 22 patients with bronchogenic cysts were operated on in our hospital; there were 14 men and 8 women, ranging in age from 11 to 62 years. The cyst locations were mediastinal in 13 (59.1%) and intrapulmonary in 9 (40.9%). There were symptoms (chest pain and recurrent bronchiolitis) in 20 patients (91%). The preoperative complications included infection in the lung and in the cyst and dysphagia due to esophageal compression. Chest pain was the main symptom in mediastinal cyst and recurrent infection of lung in intrapulmonary cyst. Plain chest radiograms showed that a round shadow, occasional air-fluid levels, and peripheral calcification may be found in cysts. An operation is the best treatment for cysts. All cysts were completely excised. No postoperative complications, late complications, or recurrence developed in our patients.

[A comparison study of various anterior capsulectomies].

The incidences of anterior capsular radial tears occurring in different capsulectomies performed on rabbit eyes under standardized conditions were compared. The eyes were randomly divided into 4 technique groups: (1) can opener, (2) capsulopuncture, (3) linear capsulotomy and (4) continuous circular capsulorhexis (CCC). The results demonstrated that the CCC technique has more advantages in comparison with other three groups. With the nuclear expression technique used in this study, no radial tears occurred with the CCC technique, whereas radial tears occurred in 100% of cases treated with other three techniques.

Effect of massotherapy on the in vivo free radical metabolism in patients with prolapse of lumbar intervertebral disc and cervical spondylopathy.

The endogenous free radical scavenger superoxide dismutase (SOD) and blood catalase (CAT) in 2 groups of patients with prolapse of lumbar intervertebral disc and cervical spondylopathy were lower than that of the healthy control group, while the -SH reflecting the metabolic disturbance of free radical was higher. After massotherapy, blood SOD and CAT were increased, while lipid peroxide (LPO), -SH in urine were decreased, demonstrating that there are distinct parallel relationships existing in the changes of these enzymes in blood and urine.

[Efficacy of praziquantel combined with albendazole in the treatment of clonorchiasis].

This paper reported on the clinical efficacy of praziquantel-albendazole combination in the treatment of clonorchiasis in Guangxi. Two hundred and two cases were divided into three groups. The first group comprised 66 cases who were treated with praziquantel at 180 mg/kg bwt for three days. The second group consisted of 62 cases who were treated with albendazole at 90 mg/kg bwt for three days. In the third group, 74 cases were treated with 90 mg/kg bwt praziquantel plus 45 mg/kg albendazole for three days. The results showed that the negative conversion rates were 98.5%, 61.3% and 87.8% in groups one, two and three, respectively, one month after treatment. In some cases who continued to be egg positive, the reduction rates in faecal egg counts were 99.7%, 65.0% and 97.3%, respectively. Although the cure rate in group three was not as high as in group one, the side effects were more mild and transient and the cost of treatment could be reduced by 60% as compared with group one. In cases with light and moderate infections, the praziquantel-albendazole combination proved more effective. In addition, the praziquantel-albendazole combination was also highly effective for treating those cases who were infected with Ascaris, Trichuris or hookworm.

[Fecundify after treatment of tubal pregnancy].

UNLABELLED: This study is a follow-up analysis of 103 patients who desired to preserve their fertile ability after treatment of tubal pregnancy. Among the 103 cases, 58 were treated by unilateral salpingectomy, 15 by salpingostomy and 30 by drug conservative therapy. RESULTS: 67 cases had intrauterine pregnancy and 9 cases repeated ectopic pregnancy. The intrauterine pregnancy rate of the surgical group (71.2%) was higher than that of the nonsurgical group (50.0%). And the pregnancy rate of salpingostomy group (86.7%) was higher than that of salpingectomy group (67.2%). No repeated ectopic pregnancy occurred in the salpingostomy group. After three months of treatment, 45 cases were randomly sampled for tubal patency test. Bilateral tubal patency rate in salpingostomy group was 93.3% and in drug treatment group was 20.0%. These results indicated that tubal patency is an essential factor for normal pregnancy. Prevention of post-operative adhesion and treatment of pelvic inflammatory disease are important for increasing intrauterine pregnancy rate and decreasing repeated ectopic pregnancy rate.

Duck hepatitis B virus model for screening of antiviral agents from medicinal herbs.

The effects of the extracts of 20 Chinese medicinal herbs and an antiviral drug foscarnet on duck hepatitis B virus (DHBV) endogenous DNA polymerase (DNAp) activity were compared. The extracts of P. urinaria showed a dose-dependent inhibition on DHBV DNAp. And those of other herbs showed little inhibition effect. Primary duck hepatocyte (PDH) cultures were used for evaluating effects of the extract of P. urinaria, foscarnet and acyclovir (ACV) on DHBV, and all the drugs or the extracts showed inhibition of DHBV DNA replication. Furthermore, in vivo trials were carried out. Peking ducks infected with LJ-76 strain of DHBV were treated with the extract of P. urinaria or ACV and compared with placebo treated control ducks. The treatment results in the loss or reduction of circulating viral DHBV DNA and DHBsAg.

[Clinical observation and therapeutic mechanism of blocking agent of substance P nerves in the treatment of perennial allergic rhinitis].

This paper reported the clinical effects and therapeutic mechanism of blocking agent of substance P(SP) nerves on prerennial allergic rhinitis. We applied capsaicin (CAP) in the treatment of 50 cases of perennial allergic rhinitis once a week and 4 times as a therapeutic course. By using highly specific and highly sensitive SP radioimmunoassay, the SP contents in nasal secretions were determined before and after CAP therapy. The results showed that clinical symptoms of allergic rhinitis were obviously relieved and SP content in the nasal secretions was remarkably reduced from 29.444 +/- 14.280pmol/L before CAP therapy to 16.848 +/- 10.622 pmol/L after therapy (P < 0.001). Of the 50 cases, 35 cases (70.0%) showed the best effects, 12 cases (24.0%) better effects and 3 cases (6.0%) no effects. The total effective rate was 94.0%. The study indicated that therapeutic mechanism of CAP on perennial allergic rhinitis is related to the reducing of SP and the blocking of axonal reflex of the SP nerves.
