Epitope mapping of anti-rhodopsin antibodies from patients with normal pressure glaucoma.

PURPOSE: The presence of anti-rhodopsin antibodies in patients with normal pressure glaucoma (NPG) has been previously demonstrated with western blot analysis and enzyme-linked immunosorbent assay. To learn more about the characteristics, origin, and possible significance of these antibodies, the epitopic specificity of the anti-rhodopsin antibodies was examined in four NPG patients. METHODS: Antibodies in patient sera were assayed by western blot analysis against purified bovine rhodopsin. Peptides derived from particular segments of the rhodopsin sequence were tested for activity in competing for rhodopsin-antibody binding. RESULTS: Of a series of nine peptides that constitute most of the hydrophilic regions of rhodopsin, only one, consisting of the C-terminal 25 amino acids, prevented binding of the patient antibodies to rhodopsin. Higher resolution mapping using a set of dodecamers of overlapping sequences from the C-terminal region demonstrated that antibody binding is completely dependent on the last two amino acids. Removing the C-terminal alanine alone, or amidating the C terminus carboxyl group, also eliminated antibody binding. CONCLUSIONS: Because four of four patient antibodies examined exhibited the identical epitopic specificity, it is likely that a common mechanism underlies their generation. This may indicate that molecular mimicry has occurred, because several pathogens contain similar C-terminal sequences. Although they may serve as diagnostic markers, and provide evidence that there is an autoimmune component in some patients with glaucoma, the role, if any, that these antibodies play in the pathogenesis of the disease remains unknown.

Computational differences between asymmetrical and symmetrical networks.

Symmetrically connected recurrent networks have recently been used as models of a host of neural computations. However, biological neural networks have asymmetrical connections, at the very least because of the separation between excitatory and inhibitory neurons in the brain. We study characteristic differences between asymmetrical networks and their symmetrical counterparts in cases for which they act as selective amplifiers for particular classes of input patterns. We show that the dramatically different dynamical behaviours to which they have access, often make the asymmetrical networks computationally superior. We illustrate our results in networks that selectively amplify oriented bars and smooth contours in visual inputs.

Visual segmentation by contextual influences via intra-cortical interactions in the primary visual cortex.

Stimuli outside classical receptive fields have been shown to exert a significant influence over the activities of neurons in the primary visual cortex. We propose that contextual influences are used for pre-attentive visual segmentation. The difference between contextual influences near and far from region boundaries makes neural activities near region boundaries higher than elsewhere, making boundaries more salient for perceptual pop-out. The cortex thus computes global region boundaries by detecting the breakdown of homogeneity or translation invariance in the input, using local intra-cortical interactions mediated by the horizontal connections. This proposal is implemented in a biologically based model of V1, and demonstrated using examples of texture segmentation and figure-ground segregation. The model is also the first that performs texture or region segmentation in exactly the same neural circuit that solves the dual problem of the enhancement of contours, as is suggested by experimental observations. The computational framework in this model is simpler than previous approaches, making it implementable by V1 mechanisms, though higher-level visual mechanisms are needed to refine its output. However, it easily handles a class of segmentation problems that are known to be tricky. Its behaviour is compared with psycho-physical and physiological data on segmentation, contour enhancement, contextual influences and other phenomena such as asymmetry in visual search.

Expression of an enzymatically active polymerase of human hepatitis B virus in an coupled transcription-translation system.

Genome replication of hepadnavirus proceeds by reverse transcription from a viral pregenomic RNA template by a virally encoded polymerase that possesses protein-priming, reverse transcriptase, DNA polymerase, and RNase H activities. Characterization of this enzyme has been hampered by failure to purify an active enzyme from virions and difficulties in expressing an active polymerase in heterologous systems. In this study, we constructed human hepatitis B virus polymerase cDNA under the control of a phage T7 promoter and expressed it in a rabbit reticulocyte lysate-coupled transcription-translation system. In vitro site-directed mutagenesis confirmed that the recombinant polymerase cDNA produced three products: a full-length protein (approximately 94 kDa), an internally initiated protein (approximately 81 kDa), and an N-terminal protein (approximately 40 kDa). The in vitro expressed polymerase possessed protein priming activity, as demonstrated by 32P-dGTP-labeling of the full size polymerase and the N-terminal portion of the molecule in an in vitro priming assay. The polymerase also exhibited polymerization activity, as detected in an in vitro polymerase assay by incorporation of radionucleotides into acid-precipitable polynucleotides and by synthesis of human hepatitis B virus (HBV) specific DNA with product lengths between 100 and 500 nucleotides. In addition, the polymerase was found to use an RNA sequence bearing HBV DR1/epsilon stem-loop motif as a template for DNA synthesis. Both the protein-priming and the reverse transcription activities of this recombinant polymerase are dependent on the RNA fragment containing the HBV DR1/epsilon stem-loop sequence known to be required for the polymerase activities. The in vitro systems used in this study will be applicable to further functional and biochemical studies of this enzyme.

Extraneural organ involvement in human rabies.

Human rabies is a fatal encephalomyelitis. After the development of the central nervous system infection, there is centrifugal spread of the rabies virus to extraneural (systemic) organs. With histochemical staining and localization of rabies virus antigen (RVA) with immunoperoxidase staining, we have examined tissue sections of organs from 14 postmortem pediatric and adult cases of human rabies acquired in Mexico and the People's Republic of China. RVA was found in nerve plexuses in multiple organs, including the gastrointestinal tract. RVA was observed in muscle fibers of the heart, tongue, and larynx. RVA frequently was observed in the adrenal medulla with an associated inflammatory reaction. Minor salivary glands of the tongue contained RVA and major salivary glands showed RVA in plexuses, but not in either acini or ducts. Epithelial cells of the tongue and taste buds were occasionally infected. RVA was observed in hair follicles of the skin and rarely in pancreatic islets. The infection of extraneural organs was sometimes, but not always, associated with an inflammatory reaction. These findings indicate that centrifugal spread of rabies virus to extraneural organs occurs frequently in human rabies.

[Effects of acetylcholine on inositol 1,4,5-triphosphate formation in cervix uteri myometrial cells of human].

AIM: To detect inositol 1,4,5-triphosphate (IP3) formation of pregnant and nonpregnant human myometrial cells induced by acetylcholine (ACh). METHODS: [3H] IP3 competitive protein binding assay. RESULTS: Basal levels of IP3 in pregnant and nonpregnant human myometrial cells were (82 +/- 9) and (96 +/- 10) nmol.g-1 (protein), respectively (n = 6). Incubated with ACh 50 mumol.L-1 for 5 min, IP3 reached the peak levels (109 +/- 11) and (122 +/- 15) nmol.g-1 (protein), respectively (n = 6), but difference of the increments of IP3 in pregnant and nonpregnant women was not significant. The increased IP3 induced by ACh was inhibited by atropine (Atr) 1 mumol.L-1. CONCLUSION: The basal IP3 level in pregnant cervix myometrial cells was higher than that in nonpregnant women. ACh increased the IP3 formation.

Dauricine suppressed CsCl-induced early afterdepolarizations and triggered arrhythmias in rabbit heart in vivo.

AIM: To study the effect of dauricine on CsCl-induced early afterdepolarizations (EAD) and ventricular arrhythmias in rabbits. METHODS: Monophasic action potentials (MAP) of the left ventricle of the rabbit heart in situ were recorded with MAP recording technique. CsCl 1-2 mmol.kg-1 i.v. was used to induce EAD and ventricular arrhythmias. RESULTS: CsCl resulted in decrease of MAP amplitude (MAPA, P < 0.05) and prolongation of MAP duration at 90% repolarization (MAPD90, P < 0.01), QRS, and R-R duration (P < 0.05) compared with those before CsCl in the dauricine and control group. CsCl injection induced EAD that appeared within about 30 s and disappeared 5-15 min thereafter. EAD always preceded ventricular arrhythmias including ventricular premature beats and paroxysmal ventricular tachycardia. The EAD amplitude (EADA) in the dauricine group (26% +/- 9% of MAPA) was smaller than that in the control group (52% +/- 5% of MAPA, P < 0.05) and the incidence of arrhythmias in dauricine group (28%) was lower than that in control group (80%, P < 0.05). CONCLUSION: Dauricine exerted an antagonistic effect on EAD and suppressed triggered ventricular arrhythmias by decreasing EADA.

[Risk factors for initiation of drug use among young males in Longchuan, Yunnan].

OBJECTIVE: To identify the risk factors for drug use among young males in Longchuan, Yunnan. METHODS: A non-concurrent cohort study was carried out based on a cross-sectional survey. Demographic, behavioral, and drug-using related information were collected using an anonymous questionnaire. The non-concurrent cohort included the period January 1, 1991 to August 1, 1994. Risk factors were assessed by univariate and multivariate analysis. RESULTS: The annual incidence of drug use increased between 1991 and 1993. Multivariate analysis identified the following risk factors for drug use: being born to Jingpo ethnic group (OR = 1.8, 95% C.I. 1.2-2.5), being divorced/ widowed/separated (OR = 8.9, 95% C.I. 1.8-43.3), smoking cigarettes (OR = 2.4,95% C.I. 1,6-3.8), having had pre-/extra-marital sex (OR = 1.5, 95% C.I. 1.1-2.2), having been encouraged by friends to try drugs (OR = 8.8, 95% C.I. 6.1-12.9) and having a family member who used drugs in 1991 (OR = 1.5, 95% C.I. 1.0-2.3). More than six years of education was a protective factor for drug use (OR = 0.6, 95% C.I. 0.4-0.98). The population attributable fractions were 70.8% for being encouraged by friends or others to try drugs, 50% for smoking cigarettes and 24% for being Jingpo ethnic group. CONCLUSION: Results suggested that community based programs to change attitudes towards smoking cigarettes and drug use could dramatically reduce the incidence of new drug users in Longchuan County. We, therefore, recommend a community-based intervention program targeting adolescent.

[Prevalence study of congenital heart disease in children aged 0-2 in Zhejiang Province].

OBJECTIVE: To find out the prevalence rate of Congenital Heart Disease (CHD) among infants and toddlers. METHODS: Heart auscultation and echocardiography examination on children aged 0-2 were examined in 13 cities (or counties) in Zhejing province. RESULTS: Findings showed that the prevalence rate in children aged 0-2 was 3.72/1000. The prevalence rates of CHD were quite different among age groups with the highest (5.54/1000) in age group 0, followed by 3.36/1000 in age group 1 and lowest (2.66/1000) in age group 2. No significant difference of prevalence rates was found between different sex. Ventricular septal defects (59.4%) was noticed as the most common lesion. CONCLUSION: The evidence indicated that CHD is one of the most important problems of public health in China. Preventing its occurrence by conducting CHD surveillance and its etiologic research will have great significance for enhancing the qualities of life of children.

[Study on seroprevalence of Helicobacter pylori infection among upper digestive tract cancer patients and their kindreds].

OBJECTIVE: In order to analyse the association between Hp infection and the risk of upper-digestive tract cancer. METHODS: In Huaian and Pizhou cities, Jiangsu province, Hp IgG quantitative-enzyme-immunoassay methods was used to identify IgG to H.pylori in the serum of 312 cases of upper-digestive tract cancer patients and their kindreds. RESULTS: (1) The level of IgG to H. pylori and the IgG positive rate (50.0%) in gastric cancer patients were both higher than that of cardia and esophageal cancer patients (P > 0.05, chi 2 test) but no significant differences were observed between cardia and esophageal cancer patients, 3 types of cancer patients and their kindreds; (2) the overall positive rates of both patients and kindreds in gastric cancer families (27.1%) were significantly higher than that of cardia or esophageal cancer families (P < 0.05) but no significant differences were observed between cardia and esophageal groups. CONCLUSION: H.pylori infection was not thought to be correlated with the development of gastric cancer, although higher clustering of Hp infection in families afflicted with gastric cancer was noticed.

hOGG1 Ser326Cys polymorphism and lung cancer susceptibility.

The human homologue of the yeast OGG1 gene, hOGG1, has been cloned, and its genetic structure has been determined. Several polymorphisms in the hOGG1 gene were detected in the Japanese populations, and among them, the Ser-Cys polymorphism at codon 326 has been shown to have a functional difference in complementation of mutant Escherichia coli that is defective in the repair of 8-hydroxyguanine. Activity in the repair of 8-hydroxyguanine is greater in hOGG1-Ser326 protein than in hOGG1(326) protein. Because many environmental carcinogens produce 8-hydroxyguanine residue and mismatching to this modified base potentially causes oncogenic mutations, the capacity to repair these lesions can be involved in cancer susceptibility in human beings. We, therefore, examined allele distributions of the Ser326Cys polymorphism in a case-control study of male lung cancer in Okinawa. The analyses based on 241 cases and 197 hospital controls disclosed the following findings. (a) Those with the Cys/Cys genotype were at an increased risk of squamous cell carcinoma and nonadenocarcinoma compared to those with the Ser/Cys and those with the Ser/Ser genotypes combined. The odds ratios adjusted for age and smoking history were 3.01 (95% confidence interval, 1.33-6.83) and 2.18 (95% confidence interval, 1.05-4.54), respectively. (b) The odds ratios for other histological subtypes of lung cancer or those in total were not significant. Those for Cys/Cys or Ser/Cys genotype against Ser/Ser did not reach statistical significance in any cell type. (c) The distributions of this polymorphism varied for different populations (Chinese, Japanese, Micronesians, Melanesians, Hungarians, and Australian Caucasians), with much less prevalence of Cys allele in the latter three populations. Although our sample size was limited, these results indicate that the Ser326Cys variant may be related to squamous cell lung cancer susceptibility. The Cys/Cys genotype appears to be more susceptible to squamous cell carcinoma, although the risk is less than that previously reported to be associated with the CYP1A1 gene. Further studies are needed to assess the importance of the interpopulation variation to cancer susceptibility.

Interaction of tumor-infiltrating lymphocyte from oral squamous cell carcinoma with FN enhances its adhesion and cytotoxicity.

OBJECTIVE: To investigate the influence of fibronectin (FN) on cytotoxicity and adhesion of tumor-infiltrating lymphocytes (TIL) from oral squamous cell carcinoma (OSCC). MATERIALS AND METHODS: TIL cells were isolated from surgical specimens of 9 patients with OSCC. Specific cytotoxicity for autologous tumor cells were tested by 3H-TdR 4 hours release, and specific TNF-alpha release was detected using enzyme-linked immunosorbent assay. Expression of intercellular adhesion molecule-1 (ICAM-1) and lymphocyte function-associated antigen-1 (LFA-1) on TIL cells were analyzed on FACScan following incubation with FITC-conjugated antibody. Cell adhesion ability was evaluated through detection by optical density (OD) values of bound cells and unbound cells. RESULTS: TIL-specific cytotoxic activity for autologous tumor was significantly enhanced, ranging from 17.7% to 67.6% by incubation on FN (P < 0.005). Cell adhesion was increased ranging from 42.5% to 62.7% by coculturing with FN (P < 0.005). The interaction of TIL with FN increased expression of ICAM-1 and LFA-1 on TIL cells as well. But specific TNF-alpha release was not obviously changed by FN (P > 0.5). CONCLUSION: The enhanced TIL activity by FN might be mediated by increased binding of TIL to targets through adhesion strengthening. This culture system would be useful to improve the TIL generation for cancer therapy.

[Changes and significance of natural killer cell, IL-2, IL-6 and TNF alpha of heroin addicts after detoxification].

In order to find out the activity of NKC and production of IL-2 in the peripheral blood mononuclear cells and the level of IL-6 and TNF alpha in the peripheral blood serum, we investigated 51 herion addicts who were in the course of detoxification. MTT colorimetric assay and ELISA assay methods were adopted. The results showed that, in the herion addicts, NK activity was 30.11 +/- 5.2%, IL-2 activity was 8.06 +/- 1.66 IU/ml, IL-6 level was 61.17 +/- 12.07 pg/ml, and TNF alpha level was 91.83 +/- 19.19 pg/ml, whereas in the healthy controls, the results were 44.89 +/- 4.75%, 15.91 +/- 3.83 IU/ml, 22.18 +/- 9.31 pg/ml and 30.55 +/- 11.94 pg/ml respectively. When the herion addicts were subjected to detoxication, their NKC and IL-2 activities, IL-6 and TNF alpha levels gradually restored to normal, and were correlated with the time of their detoxification (NKC: r = 0.626, P < 0.001; IL-2: r = 0.684, P < 0.001; IL-6: r = -0.791, P < 0.001; TNF alpha: r = -0.703, P < 0.001).

[Endothelial cell damage induced in vitro by subeschar tissue fluid].

OBJECTIVE: To assess the effect of subeschar tissue fluid (STF) on the function and structure of endothelial cell in vitro. METHODS: Human umbilical vein endothelial cells (HUVEC) were incubated with STF obtained from patients in the early postburn stage. Then the morphological changes in EC were observed, and activity of EC and contents of LDH, 6-keto-PGF1 alpha in media were determined. RESULTS: The activity of LDH and the content of 6-keto-PGF1 alpha were increased gradually after HUVEC incubated with STF, and were higher than that measured after HUVEC incubated with healthy volunteer's sera. Furthermore, activity of HUVEC decreased and they became deformed, showing shrinkage of the cell body with enlargement of intercellular space. In addition, small vacuoles appeared in cytoplasm with karyopyoknosis after 24 h. CONCLUSION: STF can damage endothelial cell and might play an important role in the pathophysiological process of burn injury.
