Viral oncolysate in the management of malignant melanoma. I. Preparation of the oncolysate and measurement of immunologic responses.

Primary explants of human malignant melanoma were utilized in the preparation of oncolysates by Newcastle disease virus. The concentrated lysate, administered parenterally, was employed in an effort to augment antitumor immunologic responses in patients with metastatic melanoma. Observed cellular changes suggested a benefit, but humoral antibody measurements were not impressive.

Viral oncolysate in the management of malignant melanoma. II. Clinical studies.

Melanoma cell oncolysate, prepared with Newcastle disease virus, was administered as an immunostimulant to 13 patients with metastatic melanoma. The oncolysate was well tolerated. Six treated patients evidenced a decrease in the size of skin nodules or diseased lymph nodes. Visceral lesions were not favorably influenced to any marked degree. One case of fulminating disease showed a change to slow progression and survived a year longer than was otherwise expected. Another patient, whose melanoma could not be controlled by surgery or chemotherapy, has been in complete remission for 2 years. It appears that viral oncolysate might be particularly helpful to patients with early disease.

Cytochemical and immunologic of women treated for squamous cell carcinoma of the cervix.

A total of 126 individuals were tested for circulating T lymphocyte levels: 10 patients with stage I-III squamous cell carcinoma of the cervix before treatment; 65 women previously treated with radiation for stage I and II squamous cell carcinoma of the cervix; and 51 healthy age-matched controls. Percentages of aneuploid cells and DNA content in vaginal or cervical smears were determined in 94 patients. All patients with squamous cell carcinoma of the cervix had lower ratios and levels of circulating T lymphocytes than healthy controls. Cytologic and cytochemical DNA studies of vaginal and cervical smears revealed that these individuals had high percentages of aneuploid cells in cervical smears as well as high DNA values. Patients with no evidence of dysplasia had increased circulating T lymphocyte levels compared to pretreatment values, a lower number of aneuploid cells, and mean DNA values close to diploid cells. Based on cytologic and quantitative DNA studies of vaginal and cervical smears, postirradiation dysplasia was diagnosed in 17 of 65 women previously treated by radiation for squamous cell carcinoma of the cervix. No difference in the levels of circulating T lymphocytes between women with postirradiation dysplasia and women without this mucosal disorder and no evidence of cancer was found.

Immunocompetence of patients with bronchogenic carcinoma.

Patients with lung carcinoma were tested for number of circulating thymus-derived (T-)lymphocytes and lymphocyte cytotoxicity before treatment and following operation or radiation therapy or a combination of the two. The proportion of circulating T-lymphocytes in healthy individuals was 75%, and the T-lymphocyte level was 1, 460 per cubic millimeter. All patients with lung cancer had a significantly decreased percentage and absolute number of T-cells before any treatment. Patients tested six to eight weeks following pneumonectomy or lobectomy showed an increased proportion of circulating T-cells in comparison to pretreatment values. Functional activity of immune lymphocytes as shown by cytotoxicity assay also improved in comparison to pretreatment values. The potential value of tests for immunocompetence is discussed.

Cytochemical response of kidney, liver and nervous system of fluoride ions in drinking water.

Morphological and cytochemical studies on the squirrel monkey have been made after maintaining the sujects on pure distilled water and fluoridated distilled water for 18 months with the objective of determining the effect of fluoride on the activity of some hydrolytic and oxidative enzymes in the kidney, liver and nervous system. Daily water intake by individual animals was measured over the final 10 months of the animal's exposure to 0,1 and 5 ppm fluoride. Water consumption was considerably higher in the animals on higher fluoride intake. Whereas the nervous system remained totally unaffected by this experimental procedure, the liver showed a slightly enhanced acivity of Krebs citric acid cycle enzymes. The kidneys, however, showed significant cytochemical changes, especially in the animals on 5 ppm fluoride intake in their drinking water. In these animals, the glomeruli showed an increase in the activity of acid phosphatase and the enzymes belonging to the citric acid cycle and the pentose shunt, whereas lactate dehydrogenase, a resentative of the anaerobic glyoclytic pathway, remained unchanged or only slightly changed. These observations suggest that fluoride in concentrations as low as 5 ppm interferes to some extent with the intracellular metabolism of the excretory system.

T-lymphocyte levels in the peripheral blood of patients with cancer of the head and neck.

Percentage and levels of circulating T-lymphocytes were studied in forty-six patients with squamous cell carcinoma of the head and neck before and/or after treatment and compared with control values obtained in fifty-one healthy volunteers. Mean percentages of circulating T-cells and mean T-cell levels in cancer patients prior to treatment were significantly lower than control values. After surgery or radiation therapy the mean percentage of T-cells remained lower in comparison with that of the controls whereas T-cell levels in patients evaluated after surgical treatment increased in comparison with pretreatment values and were not significantly different from those of the controls.

Short term effects of ionizing radiation on the recurrent or metastatic skin cancer--a quantitative DNA study.

A total of 72 smears from two cases of metastatic squamous cell carcinoma of the head and neck during radiation therapy were studied for DNA content. The percentage of aneuploid tumor cells in the range of lower ploidy was high in both cases and remained high throughout the treatment. The correlation between the percentage of aneuploid cells in metastatic squamous cell carcinoma and radiocurability of these lesions has been discussed.

Cytospectrophotometric analysis of DNA content in the invasive cancer of the vervix.

Fourteen cases of invasive, well differentiated squamous cell carcinoma were studied by means of spectrophotometry for relative DNA content in the tumor cells. Comprehensive review of literature on the subject is given. No correlation of relative DNA value in the tumor cells and clinical stage of the malignancy was found. Lack of the correlation between percentage of aneuploid cells and clinical stage of the tumor was observed. Based on the results of this paper and the literature on the subject, the interrelationship of the proportion of aneuploid cells with low arbitrary DNA value and radiosensitivity of the cervical lesion is suggested. The value of quantitative DNA study for treatment planning is emphasized.

Quantitative cytochemical studies of the RNA in the spinal cord of gamma-irradiated mice.

Quantitative analysis of RNA content was carried out in the oligodendroglia apposed to motor neurons and oligodendrocytes located far from the perikaryons in the spinal cord of mice subjected to 630 rd of whole body gamma irradiation. Decrease of relative RNA content in the glial cells apposed to the neuronal body in the irradiated animals 30 min to one hour after irradiation and the increase of RNA content in these cells two to eight hours after radiation was found. Glia located far from the perikaryons of motor neurons reacted with increase of RNA content 30 min after irradiation and its sharp decrease one hour later. This difference in the reaction of glia to whole body irradiation may be due to different metabolic patterns between glia linked to neurons in neuroglial unit and glia located far from perikaryons.

T-lymphocyte levels in the peripheral blood of gamma irradiated rhesus monkeys.

Circulating Thymus derived (T) lymphocytes were studied in Rhesus monkeys exposed to single whole body gamma radiation with doses of 400, 500 and 600 Rad. Spontaneous lymphocyte rosette assay (E-rosettes) was used to evaluate the level of T-lymphocytes. A decrease in total leukocytes, lymphocytes and T-lymphocytes levels was observed beginning from the 5th day after irradiation and continued for several weeks after radiation.
