Linkage of the genes controlling natural killer cell activity to HLA-B.

OBJECTIVE: To study whether there is a relationship between HLA-B alleles and low natural killer(NK) cell activity in Chinese. METHODS: The subjects were 34 unrelated healthy individuals (10 males and 24 females), aged 25-60 years, and 31 individuals (17 males and 14 females) from 11 families, aged 22-70 years, previously genotyped for HLA class I, II, complotypes and haplotypes that were assigned based on family studies. The unrelated individuals were typed for HLA-A, -B antigen by a microcytotoxicity assay and all the subjects were tested for NK cell activity by lactate dehydrogenase (LDH) enzyme-release assay. RESULTS: Statistical analysis showed that 30 lysis unit(LU) was the cutoff value of low and high NK reactors. The frequency of low reactor individuals homozygous for HLA-B was significantly higher than that for those for HLA-DR(P< 0. 05). The associated genes included HLA-B13, B40, B46, B7, B57, defined as NKB1 genes, and HLA-B44 as NKB2 gene. Family studies provide further evidence that the genes controlling low NK cell activity were located in the MHC region and it was a recessive trait. CONCLUSION: There were recessive genes associated with HLA-B controlling low NK cell activity in Chinese. The NKB alleles were partly different from those found in Caucasian.

[Remodeling and biomechanical properties of thoracic aorta in spontaneously hypertensive rats].

Morphometry and microstructure of thoracic aorta of spontaneously hypertensive rats (SHR) at different periods before and after hypertension were studied quantitatively by histological method and computer image analysis. We also observed the changes of opening angle in the zero-stress state and the relationship between pressure and diameter in SHR during established hypertension. The results showed that with blood pressure chronically increasing, there was a significant increase in morphometric parameters and microstructure parameters of the thoracic aorta in SHR. The zero-stress state of opening angle in SHR during established hypertension increased significantly. These reults suggest that aortic structural remodeling in hypertension such as thickness of vascular wall and disproportional increase of collagen may lead to decrease in distensibility of the aorta in SHR. The structural remodeling and changes of mechanical properties in aorta may contribute to the important pathophysiology of hypertension-induced complications.

[Assimilation efficiency of pacific oyster Crassortrea gigas].

The effects of body size, food density and its quality on assimilation efficiency of pacific oyster (Crassostrea gigas) were studied under artificial conditions in laboratory. The experimental results show that the filtration and ingestion rates of pacific oyster increased with its body size, which tallied with power function. The assimilation efficiency of pacific oyster was not significantly correlated with its body size, but correlated positively with the organic matter content of food and negatively with food density.

[Landscape structure of Longhua area in Shenzhen City during fast urbanization process--structure and heterogeneity analysis of urban construction area].

Remotely sensed mapping information of multi-period of time and methods of landscape pattern analysis were used to study the structure and spatial distribution characteristics of two types of land use, built-up areas and new developing areas, during the fast urbanization process in Longhua area, Shenzhen City. The results indicated that from 1988 to 1996, the patch number and average size in the two types of land use increased significantly, but the fragmentation degree decreased. The increase of constructed land use could be divided into two patterns, continued expanding of big residential patches and continuous appearing of new isolated small patches, and most of the new developing area was constructed from 1990 to 1994. Under the impacts of topography and traffic, the built-up areas showed macroscopic spatial distribution with decreasing lacunarity values. Distribution difference of constructed patches was very obvious in small and moderate scales. The new developing patches, affected by the conditions as the built-up areas, were also characteristic of macroscopic heterogeneity, but the expression scale of heterogeneity was larger than the built-up areas. The fast expanding of the constructed land use produced some very negative ecological problems, which should urgently be resolved by adjusting the land use polices.

Cloning of Chinese obese cDNA and its expression in E. coli.

OBJECTIVE: To obtain the sequence of Chinese obese (OB) cDNA and establish a method of leptin production in China. METHODS: Han Chinese OB cDNA fragment was obtained by reverse transcriptase-polymerase chain reaction (RT-PCR) with total RNA extracted from human adipocytes and was inserted into the expressing vector pBV220. Then the constructed recombinant plasmid pBV220-OB was transformed to E. coli DH5 alpha for leptin expression. The recombinant expressing system was confirmed by restriction endonuclease digestion, DNA sequencing and protein expression. E. coli cells were lysed by high-pressure homogenization. After cell membrane was extracted, the inclusion bodies were mainly renatured and purified primarily by precipitation with ammonium sulfate and gel chromatography through a Sephadex G75 column. The activity of recombinant leptin was determined by its influence on the satiety and weight gain of mice. RESULTS: Analysis of DNA sequence showed that Han Chinese OB cDNA included the glutamine codon at 49. The amount of recombinant leptin expressed in E. coli accounted for 31%-47% of total cellular proteins. From 1 L of fermentative bacteria about 40 mg of pure recombinant human leptin was isolated with a purity of being above 95%. The recombinant human leptin could reduce food intake and inhibit weight gains in mice. CONCLUSION: The glutamine codon at 49 is not missing in Chinese OB gene. The biologically active human leptin can be obtained by a relatively simple method of recombinant DNA technology.

[Pathologic diagnosis of usual interstitial pneumonia].

OBJECTIVE: To discuss pathologic differential diagnosis between UIP and the other types of IPF. METHOD: The slides (routine and histochemical stain) of 4 autopsy cases of UIP were reviewed. RESULTS: The most significant characteristic of UIP is that the interstial changes show temporal heterogeneity and variable distribution. The earlier stage of UIP shows an interstitial infiltrate of chronic inflammation cells and the progressive stage appears significant interstitial fibrosis. The honeycomb change and muscular cirrhosis are the characteristics of the end stage. CONCLUSION: For clinical purpose, the distinction of UIP and the other types of IPF is of great significance because of marked differences in the prognosis and therapy.

[Intracranial hemangiopericytoma 32 cases reports].

OBJECTIVE: To further elucidate the clinical feature, pathogenesis, treatment and prognosis of intracranial hemangiopericytomas (HPC). METHODS: Thirty-two cases of HPC treated in our department were analyzed retropectively. RESULTS: Grade I surgery, complet tumor removal with excision of involved dural and bone (7 cases); Grade II surgery, complet tumor removal with aggressively cauterization of dural and/or bone (13 cases); Grade III surgery, complet tumor removal but no satisfactory attempt to deal with involved dural or bone (7 cases); Grade IV surgery, tumor residual (7 cases). 13 cases were recurrent. CONCLUSION: The prognosis of intracranial hemangiopericytomas is dismal. The recurrent rates of HPC are very high. Microneurosurgery and adjuvant radiotherapy are main treatment. It is very important that counterchecking at regular intervals.

[Detection of K-ras gene mutations in DNA extracted from the plasma of patients with pancreatic cancer].

OBJECTIVES: To detect mutations of the K-ras codon 12 in DNA extracted from the plasma of patients with pancreatic cancer, and to explore the possibility of using this method in early diagnosis of pancreatic cancer. METHODS: Plasma DNA was isolated from the blood of 22 patients with pancreatic cancer and from 20 normal controls. K-ras codon 12 mutations were detected by mutant enriched polymerase chain reaction-restriction fragment length polymorphism technique and subsequent product sequencing. The relation of K-ras mutations in plasma to clinical features in pancreatic cancer patients was analyzed. RESULTS: Seventeen (77.3%) of 22 patients with pancreatic cancer had a codon 12 K-ras mutation in their plasma DNA. In two patients, the PCR products were sequenced and the mutations were confirmed. The occurrence of K-ras mutations in the plasma DNA was not related to tumor location, tumor size, and TNM stage. No K-ras mutation was detected in the plasma specimen of any of the normal controls. CONCLUSIONS: K-ras mutations are frequently found in the plasma DNA of patients with pancreatic cancer. Analysis of K-ras mutation in the plasma DNA may be useful in the early detection of pancreatic cancer.

[Experimental study on herpes simplex virus-1 functional gene latency in cornea].

OBJECTIVE: To investigate the possible existence of herpes simplex virus-1 (HSV-1) functional gene latency in cornea. METHODS: A herpes simplex keratitis (HSK) model was established in New Zealand White (NZW) rabbit corneas, latently infected corneas were transplanted to one eye of each naive NZW rabbit. One week after the operation, the corneal buttons were removed and divided into three portions. One portion was processed for HSV-1 antigen detection, another was for HVS-1 latency associated transcript (LAT), thymidine kinase (TK) and DNA polymerase gene detections by polymerase chain reaction (PCR), and the last portion after tissue culture for 3 weeks, it was co-cultured with rabbit kidney cells (RK) for one week, then the expression of HSV-1 antigen in the RK cells and this portion of corneal button was detected. RESULT: In the 15 corneal samples negative for HSV-1 antigen, LAT gene was detected in 8 samples, TK gene in 10 and DNA polymerase gene in 6 samples. HSV-1 antigen was detected in 3 samples of RK cells and 6 cultured corneal samples. CONCLUSION: HSV-1 functional genes are retained in latently infected cornea.

[An epidemiology study on limb injuries in infants aged 0 - 14 years old in Tengzhou area during 1994 - 1998].

OBJECTIVE: To find out epidemiological features on limb injuries in infants. METHODS: Stratified random sampling was carried out to investigate limb injuries in children aged 0 - 14 years old in four villages and townships, Tengzhou city Shandong province from 1994 to 1998. RESULTS: Results showed that the total rate of limb injuried was 2.96% with a ratio of male/female 3.20:1. The injury on arms was higher than on legs. The first three leading rates of injuries were falling, hurt by sharp stuff and traffic accidents. The highest rate of injury took place from May to October during the year. The rate of limb injuried seemed to be increasing with a disabled rate of 115.97/100,000. CONCLUSION: Measures should be taken to reduce the rate of injuries, according to the epidemiological findings.

[Analysis on the intervention effect of HIV/AIDS cases].

OBJECTIVE: To study the effect of a comprehensive intervention strategy on the HIV infected individuals and their families. METHODS: Eleven HIV positive persons and their families were included to a comprehensive intervention strategy including routine surveillance, safer sex education and improving the status of hygiene. The outcome was compared with 2 fully developed AIDS cases who were not included to the intervention strategy. RESULTS: Of those HIV positive persons, that subjected to the intervention strategy, none had developed to AIDS, with only 1 transmitted HIV to his spouse among the 6 married individuals. All of their families were stable. Of those that not had been subjected, 2 individuals developed to AIDS and transmission in family was confirmed. CONCLUSION: The comprehensive intervention strategy on HIV/AIDS was effective which could the possibility of HIV in-family transmission and enhance the quality of life and the stability of their families.

[Study on the needle biopsies of 100 prostatic adenocarcinoma].

OBJECTIVE: To study the morphological diagnostic criteria of prostatic adenocarcinoma in needle biopsy. METHODS: 250 cases of prostatic needle biopsy including 100 cases of adenocarcinoma were observed retrospectively. The diagnostic significance of 20 parameters of morphological criteria including the architectural changes, cytologic features, invasion and some additional histologic features (intraepithelial neoplasm, intraluminal crystelloid body, intraluminal basophilic mucin secretion, collagen nodule and coagulative necrosis) were analyzed. RESULTS: All 20 parameters of morphological criteria had practical significance in pathologic diagnosis, the most significant ones being architectural changes, prominent nucleoli, perineural and stromal invasion. CONCLUSION: The diagnosis of prostatic adenocarcinoma is based on a constellation of cytologic and architectural features.

[The development of immune-mediated aplastic anemia in mice can be blocked by cimetidine].

OBJECTIVE: To investigate the effect of H(2) receptor antagonist cimetidine on the development of immune-mediated aplastic anemia (AA) in mice. METHODS: The immune-mediated aplastic anemia mice model was induced by irradiation and half-matched allogeneic lymphocytes transfusion (ALT). The mice were divided into 3 groups: the irradiation alone control group, the irradiation + ALT group (AA group) and the irradiation + ALT + cimetidine group. In cimetidine group, each mouse was intraperitoneally injected cimetidine at different doses once a day for 8 days. On day 14, the bone marrow histology was examined. RESULTS: In 0.5 mg, 1.0 mg and 2.0 mg x (10g)(-1) x d(-1) cimetidine groups, the percentages of hematopoietic tissue volume in bone marrow were (46.58 +/- 20.41)%, (51.88 +/- 24.94)% and (59.12 +/- 32.48)%, respectively, and all much higher than that in AA group [(19.99 +/- 10.98)%, P < 0.01], but were not different from that in irradiated control group [(53.81 +/- 31.72)%, P > 0.05]. CONCLUSION: The development of immune-mediated aplastic anemia in irradiated mice by half-matched allogeneic lymphocytes transfusion was blocked by H(2) receptor antagonist cimetidine.

[Antitumor activity of the lysates prepared from anti-CD3 antibody activated killer cells].

This study was directed at the antitumor activity of the lysates prepared from CD3McAb activated killer cells. We separated the peripheral blood monocytes(PBMC) from normal adults. The PBMC were induced by monoclonal antibody specific for CD3(CD3McAb) and activated by rIL-2. The CD3McAb-activated killer cells (CD3AK) were smashed by ultrasonic wave, along with frozen and thawed three times, then the lysates were obtained by centrifugation. The lysates were tested for antitumor activity in vivo and in vitro. The results revealed that the inhibition rate of the lysates that acted on mice solid tumor hepatoma 22(H22) was 68.20% and the killing activity of the lysates on K562 and Raji were 83.32%, and 66.83% respectively. The results of this experiment suggested that the lysates prepared from CD3McAb-activated killer cells is one of probable agents that might apply to biotherapy for tumors.

[Zero-stress state of renal artery in spontaneously hypertensive rats].

By observing the opening angle of the renal artery, the zero-stress state of the renal artery was studied in spontaneously hypertensive rats (SHR) prior to and posterior to their hypertension, and the role of endogenous angiotensin II (Ang II) and endothelin (ET) on the zero-stress state was studied by giving SHR Losartan(AT1 receptor antagonist of Ang II) and BMS-182874 (an A type receptor antagonist of ET) respectively. The opening angle of the renal artery in SHR with established hypertension was much greater than that in wistar-Kyoto rats(WKY) (114.2 +/- 42.2 vs 70.2 +/- 30.4, P < 0.01). The opening angles of the renal artery in SHR treated with Losartan and BMS-182874 were much smaller than those in control SHR. The opening angles of the renal artery in Losartan-treated and BMS-182874-treated SHR were 65.9 +/- 32.7 and 66.6 +/- 41.54 respectively. The results suggest that non-unequal growth exists in the renal arterial wall in SHR with established hypertension and the residual stress and strain in SHR are greater than those in age-matched WKY. Endogenous Ang II and ET may play a role in the changes of zero-stress state in SHR with hypertension.

[Detection of bacterial DNA in blood with three pairs of primers].

OBJECTIVE: To develop a stable and sensitive method to detect gut-derived bacterial DNA present in the blood. METHODS: Blood culture and PCR were performed simultaneously when temperature of patients (n = 23) was higher than 38.5 degrees C after laparotomy. PCR was performed after DNA extraction, target genes being beta-lactosidase gene of Escherichia coli(E. coli), glutamine synthase gene of Bacteroides Fragilitas, and 16SrRNA gene of most pathogenic bacteria. DNA of standard strains of E. coli and Bacteroides Fragilitas served as positive controls, blank control as negative controls, and healthy volunteers (n = 20) served as normal controls. We performed PCR twice with part of the samples (n = 20) in order to test the repetition of this method. RESULTS: The repetition rate of PCR was 95%. Three of blood cultures were positive (13.0%, 3/23), the corresponding positive ratio of PCR was 43.5% (10/23) which was higher than that of the blood culture (P = 0.016). CONCLUSION: PCR is more sensitive than blood culture in detection of bacteremia.

[Principle of evidence-based medicine and its application in clinical trial practice for glutamine and gut permeability].

Evidence-based medicine (EBM) was developed from the progress of traditional clinical model trial patterns. EBM was based on well-designed, randomized, double blind, controlled, multiple center trails in order to assess end pont parameters, prognosis parameters (as: mortality, morbidity of complications and hospital stay etc.), quality of life and cost/effect ratio. It has been accepted in developed countries and could be the guidelines for Chinese clinical trials in the future. This article summarized the principle of EBM and attached with a clinical trial sample about effects of glutamine dipeptide on gut barrier function, post-operative complications and the hospital stay.

[Ototoxicity of gentamicin assessed by the vestibular evoked potentials].

OBJECTIVE: To assess ototoxicity of gentamicin by the vestibular evoked potentials (VsEPs) and auditory brainstem response (ABR). METHODS: 40 guinea pigs were divided randomly into 4 groups, 10 animals of group A were injected gentamicin intramuscularly 60 mg/(kg.d). 10 of group B were injected Ns intramuscularly 0.5 ml/d as control. 10 of group C were given gentamicin intratympanic 5 mg/d(0.1 ml). 10 of group D were given Ns intratympanic 0.1 ml/d as control. ABR and VsEPs were measured prior to and following the administration. RESULTS: After treatment group A 21 days, 40 days, group C 5 days, 10 days, ABR and VsEPs showed significant changes, corresponding to histopathological examination of the temporal bones. CONCLUSION: VsEPs used in assessing ototoxicity of gentamicin just as ABR.
