Helicobacter pylori serology using specimens collected on filter paper.

A simple method for the collection of blood specimens on filter paper for serology of Helicobacter pylori was devised. Blood was freshly applied to small Whatman 3 mm filter paper and allowed to dry. Serological activity, as measured by reactivity to H pylori antigens (AMRAD, Australia), was retained after storage for one month at both room temperature and at 4 degrees C. This technique eliminates formal venepuncture, separation of sera, and arduous packaging of specimens to be sent to the laboratory.

The changes of fluorescein angiography and visual electrophysiology in ocular posterior aging degeneration syndrome (OPADS).

Ocular posterior aging degeneration syndrome (OPADS) is related to age excepting inflammation, trauma, tumor, congenital abnormalities. Clinically, it can be divided into several types: degeneration, pre-retinal membrane, hole and mixed types. Changes can be seen in the choroidoretina with fluorescein angiography. In the degeneration type, there are changes of chroid-blood vessels, pigment epithelium and neovasculization. The manifestation in pre-retinal membrane type are small vascular tortuosity, expansion or fluorescein leadage, retinal pigment epithelium window defects. The hole type shows lack of retinal capillaries and retinal pigment epithelium window defects. Visual electrophysiological tests by various intensities and colors show the abnormal ERG. The bio-electricity active changes indicated the correlation among several ocular posterior aging degeneration types.

[Anti-inflammatory activities and effect of rhizoma Alismatis on immune system].

10 and 20 g/kg ig of Rhizoma Alismatis (RA) markedly decrease the clearance rate of charcoal particles in mice, but have no significant effect on the weight of immune organs, or on the content of serum antibody hemolysin and immunoglobulin G and the delayed footpad edema induced by sheep red blood cell. The contact dermatitis of mouse pinnae immunized with dinitrochlorobenzene is inhibited by RA when it is given before challenge. In addition, RA 20g/kg suppresses the swelling of mouse rinnae induced by xylene and the proliferation of granuloma induced by cotton-pellet in rats. However, the content of vitamin C in the adrenal of rats is not clearly affected by RA.

[Statistical analysis of age-related macular degeneration (AMD) in the differentiation of symptoms and signs].

Ninety-eight individuals according with entry criterion were allowed to participate in the study. Their clinical symptoms and signs were classified to seven symptoms by means of epidemiology. The results indicated syndrome of vital energy in patient group had more significant than controlled group, the others had not. In the syndrome of deficiency of vital energy, the frequency that the corpalent tongue or margin and tip of the tongue look jagged and the pulse lost its vitality emerged in the patient group surpassed significantly the controlled group. So, we initially considered that traditional Chinese medicine syndrome of AMD patient is mainly the deficiency of vital energy. The happening and development of AMD is probably related to the physique of deficiency of vital energy, it is a key point of determination of treatment with traditional chinese medicine based on differentiation of symptoms and signs.

[The role of nucleus tractus solitarii (NTS) in acupuncture inhibition of visceral-somatic reflex (VSR)].

Experiments were carried out on 164 rabbits. The transmission of abdominal vagal impulses and the mechanism of inhibitory effect by acupuncture systematically were investigated. 1) Electrical stimulation (single pulse of 20 V, 0.5 ms) of abdominal vagus nerve (AVN) produced two kinds of potential on cervical vagus nerve: the fast wave (10.4-24.4 M/sec) and the slow wave (0.9-1.7 M/sec), the later was more stable. 2) In dorsal medulla oblongata the 'M' shaped evoked potentials with long duration (100 ms or so) were recorded. The results suggest that afferents from AVN are mainly transmitted through the small fibres and multi-synaptic connections in Medulla. 3) Out of 301 response units, 281 units (93.6%) concentrated in NTS and adjacent regions. This result was also confirmed by injection of Horseradish Peroxidase (HRP) in the trunk of AVN. 4) On 24 awake rabbits the VSR were elicited by strong stimulation of AVN. Electroacupuncture (EA) on "Zusanli" points inhibited VSR, an effect lasted over 20 min. This inhibitory effect of EA was reversed partly by Naloxone (0.5 mg/kg iv) and was attenuated after lesion (DC 0.5 mA, 1 min.) of NTS. In conclusion, NTS plays an important role in suppression of VSR by EA (P less than 0.01).

Effect of thyroid hormone on epidermal growth factor-like immunoreactivity and growth velocity in cynomolgus monkeys.

To examine the potential role of epidermal growth factor (EGF) in mediating the effects of thyroid hormone on linear growth, we measured serum EGF levels by RIA in cynomolgus monkeys before and during methimazole-induced hypothyroidism, and after 9 weeks of T4 replacement at different doses. Ten castrated prepubertal monkeys were rendered hypothyroid by methimazole (0.0125% in drinking water for 12 weeks). Methimazole was continued, and T4 was then administered for 9-week intervals. Six weeks elapsed between successive T4 doses. The sequence of different T4 doses for each animal was random. Serum EGF level was measured at baseline and at the end of each treatment period with a newly developed RIA using a polyclonal antiserum against human recombinant EGF. Serum EGF level correlated significantly with the level of serum thyroxine but not with serum triiodothyronine, over the thyroxine dosage range of 1-4 micrograms/kg/day (r = 0.41, p less than 0.005). Lower-leg growth rate correlated significantly with serum EGF level over this same thyroxine dosage range (r = 0.41, p less than 0.005). These data are consistent with the hypothesis that EGF may mediate some of the effects of thyroid hormone on skeletal growth.

Effect of clonazepam on brain development of mice.

Twenty-seven Kun-Ming white mice were divided randomly into three groups of 9 animals. One of these groups served as a control group. Clonazepam (CZP), 0.1 or 0.5 mg/kg/day, was administered by oral gavage for 22 days. The water-maze test was performed before giving CZP and 10 and 22 days later. The step-down test was performed on the 1st and the 2nd day after discontinuing CZP treatment to determine toxic effects on brain development and learning-memory function. Body weights and brain weight were also recorded. Brain development and learning-memory function, evaluated by water-maze and step-down tests, were not impaired by CZP. Although there was a trend toward lower weights in the higher-dose group, CZP treatment did not significantly affect either body weight or total or regional brain weight. The results of this study indicate that 4 weeks of treatment with CZP does not impair brain development and learning-memory function in baby mice.

Biphasic response of rat tibial growth to thyroxine administration.

To evaluate the dose-response relationship between thyroxine and tibial growth, 60 male rats age 21 days were rendered hypothyroid by administration of methimazole in the drinking water. Twenty-one days later, the hypothyroid rats were randomly divided into 5 groups which received 0, 2, 8, 32, or 64 micrograms.kg-1.day-1 of T4 im for 21 days. All animals were sacrificed at age 64 days. Rat tibia were removed for measurement of epiphyseal growth plate width and longitudinal growth rate. Serum T4 and IGF-I levels were determined by RIA. Methimazole therapy significantly decreased serum T4, IGF-I, epiphyseal growth plate width, and longitudinal growth rate compared to controls. Epiphyseal growth plate width gradually increased when T4 was administered at doses from 2 to 32 micrograms.kg-1.day-1 (271 +/- 14, 311 +/- 15 and 324 +/- 11 microns), and subsequently decreased when T4 was given at a dose of 64 micrograms.kg-1.day-1 (267 +/- 8 microns). A similar profile was observed for longitudinal growth rate and IGF-I. We conclude that rat tibial growth has a biphasic response to exogenous T4 administration, and that the effects of T4 on tibial growth may be mediated through IGF-I secretion.

[Free amino acid and synthesis of polyamines in Plasmodium-berghei-infected RBC].

The contents of free amino acids and the activity of ornithine decarboxylase in infected RBC of chloroquine-sensitive strain of Plasmodium berghei(CS) were similar to those in chloroquine-resistant strain (CR). After treatment with chloroquine (10mg/kg im) no effect on the formation of free amino acids was found after 20th, but chloroquine (5mg/kg im) inhibited the enzymatic activity of the CS RBC by 79.6% whereas that of the CR by 55.7%. The contents of spermidine in CS and CR infected RBC were found to be 139 +/- 27 and 528 +/- 140 nmol/10(9) infected respectively. Cycloleucine inhibited the formation of spermidine by 44% in CS and 57% in CR groups, while co-administration of methionine (100mg/kg) with cycloleucine (80mg/kg) could reverse the inhibitory effect and raise the contents of spermidine to 294 +/- 56 and 657 +/- 129 nmol/10(9) infected RBC in CS and CR groups respectively. This indicates that methionine had significant effect on the formation of spermidine in CS strain, but there was still a remarkable difference, when compared with CR strain. It is suggested that the activities of S-adenosyl-methionine synthetase and other enzymes involved in polyamine formation might be the causal factors.

[Antiarrhythmic effect of Oenanthe javanica (Bl.) DC. injection].

The antiarrhythmic effect of Shuiqin (Oenanthe javanica) injection was studied in rats. An injection of 3 ml/kg iv could significantly antagonize the arrhythmias induced by aconitine and BaCl2, and decrease the rates of ventricular fibrillation and death induced by CaCl2. The results suggest that Shuqin injection has a significant antiarrhythmic effect on experimental rats.

A preliminary report on screening for congenital hypothyroidism in newborns with time-resolved fluorescence immunoassay (TR-FIA) of TSH.

From November 1986 to September 1987, 27,253 newborns were screened for congenital hypothyroidism (CH) with Time-Resolved Fluoroimmunoassay (TR-FIA) for TSH concentrations in specimens of dried blood spots on filter paper. Specimens were obtained in 12 hospitals by heel pricking of the babies 48-72 hours after birth. The specimens with TSH concentration higher than 20 mu u/ml were retested, and the babies were called back for confirmation of CH by TSH, T3, T4, Tg serum radioimmunoassay etc. and bone-age radiography if the repeated tests were positive. In the present series, 337 specimens were retested (1.23%). Altogether 25 babies were recalled (0.09%) and CH was confirmed in 5 cases, giving a screening incidence of 1:5 450. In addition, one case failed to respond to the recall because of a wrong address. TR-FIA or DELFIA (Pharmacia-LKB) is non-radioactive and does no harm to man and the environment, and is thus feasible in ordinary laboratories. The method is sensitive and the results are reproducible, with a precision of 1.4 mu u/ml. Being rapid in terms of measurement time, it is suitable for mass screening.

[Studies on the variability of incubation period of vivax malaria following mosquito biting].

From September 1985 to May 1987, 30 people were experimentally infected by Anopheles sinensis infected with Plasmodium vivax isolated from northwestern part of Hunan Province. The findings displayed in graphs demonstrate that the incubation period was variably depended on the number of infective mosquitoes used in the experiment. The short incubation period of 15.8 +/- 1.9d was frequently observed in 6 persons bitten by 10 infected mosquitoes. However, when one mosquito was allowed to feed on each of 24 volunteers, the incubation period was either short (17.3 +/- 1.2d) or prolonged (284.7 +/- 37.7d) in the ratio of 1 to 1. Based on these data it is suggested that tertian malaria of long incubation period must exist everywhere in China. In addition, the rational classification of vivax malaria and the ratio between long and short incubation periods in nature are also discussed in detail.

[Experimental studies on the incubation period and relapse patterns in vivax malaria in west Hunan].

The paper reports on some biological characteristics of Plasmodium vivax isolated from Cili County of Hunan Province. 27 volunteers inoculated with sporozoites were observed for 1.5 to 2 years. Treatment with chloroquine 1.5g base for 3 days was initiated on the first day of the primary attack and of each relapse. The data indicate that the first latent period in 13 cases developing malaria after short incubation period is very long, being 243.4 (191-297) days; however it is always short, being 55.3 (51-80) days in 14 patients with long incubation period. Definite relationship existing between incubation period and relapse patterns in vivax malaria is thus demonstrated.
