A clinical study of yi zhi capsules in prevention of vascular dementia.

The raised score in the revised Hasegawa dementia scales (HDS) demonstrated that the effect of Yi Zhi Capsules (YZC) in treating loss of intellectual function after cerebrovascular diseases was significantly better than that of the western drug Piracetam [symbol: see text] (P < 0.01). The morbidity of vascular dementia was much lower in the treatment group than that of the control group (P < 0.05) after a one-year follow-up; and the blood lipid level and some indexes in rheological examinations were also improved significantly (P < 0.05, or < 0.01). The data indicate that YZC is a relatively good preparation for prevention of vascular dementia.

[Clinical study on morphological characteristics of placenta in severe pregnancy induced hypertension].

OBJECTIVE: To study morphological characteristics of placenta in severe pregnancy induced hypertension (SPIH) and its relationship to pregnant outcome. METHODS: Morphological changes were observed by light microscopy; Blood biochemical analyses were used to predispose pregnant outcome. RESULTS: Difference in pathological changes of placental bed between normal term pregnancy (NTP) and SPIH groups was significant: the placental weight, proliferation of cytotrophoblasts, numbers of the placental villi with syncytial knots, thickness of basal lamina, fibrinoid necrosis and deposition of matrix, stromal edema and fibrosis of villi, the vascular numbers of villi, stasis; lack of physiologic changes in decidual spiral arteries. Clinical examination showed that the rate of anemia, thrombocytopenia, blood concentration, hypoproteinemia, ascites, eye ground artery spasm in SPIH were higher in NIT. CONCLUSIONS: Pathological changes of placenta play important roles in development of SPIH, and the pathological changes are paralleled with the severity of the disease.

Flow injection analysis of fluoride: optimization of experimental conditions and non-linear calibration using artificial neural networks.

This paper deals with the application of artificial neural networks (ANNs) to two common problems in spectroscopy: optimization of experimental conditions and non-linear calibration of the result, with particular reference to the determination of fluoride by flow injection analysis (FIA). The FIA system was based on the formation of a blue ternary complex between zirconium(IV), p-methyldibromoarsenazo and F- with the maximum absorption wavelength at 635 nm. First, optimization in terms of sensitivity and sampling rate was carried out by using jointly a central composite design and ANNs, and a neural network with a 3-7-1 structure was confirmed to be able to provide the maximum performance. Second, the relationship between the concentration of fluoride and its absorbance was modeled by ANNs. In this process, cross-validation and leave-k-out were used. The results showed that good prediction was attained in the 1-4-1 neural net. The trained networks proved to be very powerful in both applications. The proposed method was successfully applied to the determination of free fluoride in tea and toothpaste with recoveries between 96 and 101%.

[Recognition of multiple information of human auricular points by linear model].

The three variables of the electric characteristic of human auricular points can be regarded as observed targets that reflect the physiologic or pathologic changes. Medical principle design follows the theory of the channels and collaterals and the viscera-state doctrine of traditional Chinese medicine and that of the pattern recognition. The linear integration model can be established by drawing the characteristic variables of the correlation groups of the auricular points and applying the multi-variable normalization. With this model, the recognition of the three variations of the auricular points can be of use for differential diagnosis. In this paper, the test to identify the cases of Upper-Digestive tract achieves good result. This research, based on the integration of modern science with traditional Chinese medicine, provides a method for building the specialist system of the auricular point diagnosis, and probably has some prospect in clinical application.

[Research progress on PGPR/AMF interactions].

As one of the rhizospheric microorganisms PGPR(Plant Growth Promoting Rhizobacteria) and AMF(Arbuscular Mycorrhizal Fungi) play an important role in promoting plant growth. It is of significance to further study and elucidate the interactions between them to utilize and regulate the interactions among rhizospheric microorganisms, and promote and protecte plant growth. Many research results show that on one hand, there exists synergism between PGPR and AMF. AMF can transfer PGPR or act as a media in the process of spread of PGPR along roots, where PGPR create many beneficial conditions for the infection of AMF. Both of them can indirectly enhance the other side's colonization or infection ability through their own promoting role on plant growth. On the other hand, they compete with each other for nutrients and niches, and probably produce some secondary metabolites which cause detrimental effects on the other. However, whether these interactions are synergistic or competitive depends upon the AM fungal or PGPR species involved. So far, the research work is extensive, even in molecular level in some aspects, but not systematic and deep. It is believed however, with the development of techniques in molecular biology and the increasing application of advanced testing methods, the new breakthroughs will be gained in the study and understanding on the interactions.

[Study on apoptosis and its related genes in pregnancy induced hypertension].

OBJECTIVE: To investigate apoptosis and its related genes expression in serious pregnancy induced hypertension (SPIH). METHODS: 40 cases of normal term pregnancy(NTP) and 40 cases of SPIH were enrolled in this study. Apoptosis was detected using DNA in situ terminal deoxynucleotidyl transferas (TDT)-mediated deoxyuridine triphosphate (dUTP)-biotin nick ending labeling (TUNEL); bax and bcl-2 were detected by immunohistochemical staining (SP). RESULTS: In NTP group apoptosis index(AI) of cytotrophoblasts(CT) and syncytiophoblasts(ST) in placenta was (1.1 +/- 0.9)% and (41.8 +/- 1.5)%; bax positive rate was (1.0 +/- 0.9)% and (28.9 +/- 9.7)%; bcl-2 was (2.2 +/- 0.8)% and (22.9 +/- 0.7)% respectively; bax/bcl-2 was 0.7-1.7. In SPIH group, AI was (4.3 +/- 1.2)% and (45.3% +/- 1.4)%; bax was (2.2 +/- 0.8)% and (42.5 +/- 11.7)%; bcl-2 was (3.2 +/- 0.8)% and (23.3 +/- 7.8)%; bax/bcl-2 was 1.0-3.2. The difference between SPIH and NTP was significant respectively (P < 0.01); bax/bcl-2 was balance in NTP, while the balance was broken in SPIH. CONCLUSIONS: Apoptosis and its related genes expression are simultaneous. The balance and imbalance of apoptosis and bax/bcl-2 affect the structure and the function of placenta, and then may further affect the outcome of pregnancy.

[The effect of progesterone on proliferation and apoptosis in ovarian cancer cell].

OBJECTIVE: To investigate the regulatory effect of progesterone on proliferation and apoptosis in ovarian cancer cell line HO8910 in vitro. METHODS: Ovarian cancer cell line HO8910 originated from human ovarian serous cystadenocarcinoma was cultured in vitro. Two groups were set up: study group (progesterone in different concentrations) and control group without progesterone. Cell proliferation was measured by 3-(4, 5-dimethylthiazol-z-yl)-2,5-dipheny tetrazolium blue (MTT) colorimetric assay. Cell cycle and apoptotic percentage were detected by flow cytometry, morphological changes of apoptotic cells were observed by light and electron microscopy, and apoptotic cells were quantitatively determined by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL). In addition, expression of intracellular bcl-2 protein was analyzed by flow cytometric indirect immunofluorescent technique. RESULTS: Progesterone of 1 x 10(-7)-1 x 10(-5) mol/L inhibited HO8910 cell growth significantly in a dose-dependent manner (P < 0.01). After treatment with progesterone, the enhanced G0/G1 arrest was accompanied with the enhanced apoptotic peak and percentage, as well apoptotic cells were found more than those in control group (P < 0.05). By light and electron microscopy, there were many morphological characteristics of apoptosis including compaction and margination of nuclear chromatin, nuclear fragments, and apoptotic bodies. Analysis on expression of intracellular bcl-2 protein showed that progesterone could down-regulate bcl-2 protein and at concentration of 1 x 10(-5) mol/L it could almost block bcl-2 expression. CONCLUSIONS: It is suggested in the present study that progesterone can inhibit the proliferation of epithelial ovarian cancer cells in vitro and there is an accordant dose-response relationship. Its anticancer effect seems to be due to induction of apoptosis which maybe a result of down-regulation of the anti-apoptotic protein bcl-2.

[Factor analysis on the intelligence of patients with schizophrenia].

The authors made a factor analysis on the intelligence of patients with schizophrenia. 119 patients with schizophrenia were measured by Wechsler Adult Intelligence Scale-Revised China (WAIS-RC). The results revealed the average full intelligence quotient (IQ) of schizophrenia was 79.70, the verbal intelligence quotient(VIQ) 84.43, and the performance intelligence quotient(PIQ) 74.10. Multiple regression showed the regression coefficient of educational level for IQ was 8.6721. The regression coefficient of the course of disease for IQ was negative 0.1519. Factor analysis showed that the cumulative percent of three-factor solution of schizophrenia was 73.5. In conclusion, the IQ of most patients with schizophrenia was lower than norm, VIQ > PIQ(P = 0.0000). Educational level had greater influence on IQ. The longer the course of disease was, the worse the intelligence came to be. Factor analysis indicated that three-factor solution of schizophrenia was similar to that of norm, but each subtest value was lower than norm.

[Studies on biosorption of Pd2+ by bacteria].

The strain R08, showed relatively strong ability of adsorbing Pd2+, was screened from different source of bacterial strains. The strain R08 was identified as Bacillus licheniformis. The optimum pH value of Pd2+ biosorption by R08 dead biomass is 3.5. The biosorption is a rapid and non-temperature dependent process. The biomass and Pd2+ initial concentration in solution affected the biosorption process. The bosorptive capacity could reach 224.8 mg/g under the conditions of Pd2+ 200 mg/L, 0.4 g biomass/L, pH3.5 and 30 degrees C for 45 min. TEM observation showed that R08 biomass is able to adsorb and reduce Pd2+ ions to Pd0 particles. IR spectrum analysis indicated that -COO- and HPO(4)2- groups on the cell walls may involve in process of adsorbing Pd2+.

[Effects of phytohormones on growth and content of depsides in Salvia miltiorrhiza suspension cells].

This paper deals with the effects of 2,4-D, BA and GA3 on the growth and content of two depsides (rosmarinic acid and lithospermic acid B) in suspension cells of Salvia miltiorrhiza. The results showed cell growth and rosmarinic acid content reached the maximum on the 12th day and lithospermic acid B content on the 16th day after incubating cells in the subculture medium MS + 2, 4-D 1 mg/L + KT 0.1 mg/L. This cell line was a growth-product-associated. With the same concentration (1 mg/L), 2, 4-D stimulated the cell growth but prohibited the formation of lithospermic acid B; GA3 inhabited the cell growth but stimulated the formation of two depsides; The effects of BA is between 2, 4-D and GA3. The concentration optimum of phytohormones tested displayed 3 mg/L for BA and 1 mg/L for GA3. For the optimum time of adding BA and GA3 was in med-term (the 8th day) and early term (at the beginning) of culture period respectively. The synergiatic function of BA and GA3 on the depside formation was also showed that adding GA3 (1 mg/L) was favour for the formation of lithospermic acid B of the suspension cell cultured in the medium containing BA 3 mg/L. The suitable time of adding GA3 was the 6the day after incubating cells in the medium of MS + BA 3 mg/L.

[The influence of external stimulation on content and quality of volatile oil in Lignun Santali albi].

The authors analyzed the quality of Ligmum Santali Albi formed by the external stimulation of hormone and windburn by GC-MS-DS. The results showed that the content of volatile oil is 2.34% in the heart wood formed in 10 years tree age of Santalum album (SA) after 2 years stimulation continuously with a definite concentration of hormone, which is near to the 25 years tree age of SA in the same place. The GC-MS analysis showed that the content of santalol and other chemical components in volatile oil are similar to the 25 years tree age of SA. It is indicated that a definite concentration of hormone stimulated the SA may shorten the formation of the heart wood. The heart wood can be also formed by the broken branches after 2 years windburn, but its content of volatile oil is only 1/2 of the heart wood formed by hormone stimulation.

[Study on the photoinduced-color changing about novel azobenzocrown ether derivatives 16ABC-OH and 16ABC-NH2 in water solution and Langmuir-Blodgett film].

Two kinds of azobenzocrown ether derivation 16ABC-OH and 16ABC-NH2 have been synthesized, and their photoinduced color changing phenomena has been investigated in water solution, the reaction of 16ABC-OH is the photochemical Cis-trans-isomerization, and it is a first-order reaction, but the reaction of 16ABC-NH2 photochemical Cis-trans-isomeriation is very complex; and it is not a first-order reaction. In Langmuir-Blodgett film of 16ABC-OH, there is not molecular self-assembly phenomenon, the part of Cis-16ABC-OH become to iso-16ABC-NH2, but in the film, 16ABC-NH2 form H-Polymer by self-assembly, and the photochromic Cis-trans-isomerization is forbidded.

The MinC component of the division site selection system in Escherichia coli interacts with FtsZ to prevent polymerization.

Positioning of the Z ring at the midcell site in Escherichia coli is assured by the min system, which masks polar sites through topological regulation of MinC, an inhibitor of division. To study how MinC inhibits division, we have generated a MalE-MinC fusion that retains full biological activity. We find that MalE-MinC interacts with FtsZ and prevents polymerization without inhibiting FtsZ's GTPase activity. MalE-MinC19 has reduced ability to inhibit division, reduced affinity for FtsZ, and reduced ability to inhibit FtsZ polymerization. These results, along with MinC localization, suggest that MinC rapidly oscillates between the poles of the cell to destabilize FtsZ filaments that have formed before they mature into polar Z rings.

ATP-Binding Cassette Transporters↗

Phosphate ion partially relieves the cooperativity of effector binding in D-3-phosphoglycerate dehydrogenase without altering the cooperativity of inhibition.

The binding of L-serine to phosphoglycerate dehydrogenase from E. coli displays elements of both positive and negative cooperativity. In addition, the inhibition of enzymatic activity by L-serine is also cooperative with Hill coefficients greater than 1. However, phosphate buffer significantly reduces the cooperative effects in serine binding without affecting the cooperativity of inhibition of activity. The maximal degree of inhibition and fluorescence quenching in Tris buffer occurs when an average of two serine binding sites out of four are occupied. This value increases to three out of the four sites at maximal levels of inhibition and quenching in phosphate buffer. The increase from two to three sites appears to be due to the ability of phosphate to reduce the site to site cooperative effects and render each ligand binding site less dependent on each other. The correlation between the level of inhibition and the fractional site occupancy indicates that in Tris buffer, one serine is bound to each interface at maximal effect. In the presence of phosphate, the order of binding appears to change so that both sites at one interface fill before the first site at the opposite interface is occupied. In each case, there is a good correlation between serine binding, conformational change at the regulatory site interfaces, and inhibition of enzyme activity. The observation that phosphate does not appear to have a similar effect on the cooperativity of inhibition of enzymatic activity suggests that there are two distinct cooperative pathways at work: one path between the four serine binding sites, and one path between the serine binding sites and the active sites.
