Functional comparison of the three isoforms of the Na+/Ca2+ exchanger (NCX1, NCX2, NCX3).

Three distinct mammalian Na+/Ca2+ exchangers have been cloned: NCX1, NCX2, and NCX3. We have undertaken a detailed functional comparison of these three exchangers. Each exchanger was stably expressed at high levels in the plasma membranes of BHK cells. Na+/Ca2+ exchange activity was assessed using three different complementary techniques: Na+ gradient-dependent 45Ca2+ uptake into intact cells, Na+ gradient-dependent 45Ca2+ uptake into membrane vesicles isolated from the transfected cells, and exchange currents measured using giant patches of excised cell membrane. Apparent affinities for the transported ions Na+ and Ca2+ were markedly similar for the three exchangers at both membrane surfaces. Likewise, generally similar responses to changes in pH, chymotrypsin treatment, and application of various inhibitors were obtained. Depletion of cellular ATP inhibited NCX1 and NCX2 but did not affect the activity of NCX3. Exchange activities of NCX1 and NCX3 were modestly increased by agents that activate protein kinases A and C. All exchangers were regulated by intracellular Ca2+. NCX1-induced exchange currents were especially large in excised patches and, like the native myocardial exchanger, were stimulated by ATP. Results may be influenced by our choice of expression system and specific splice variants, but, overall, the three exchangers appear to have very similar properties.

Cystic fibrosis transmembrane conductance regulator activation by cAMP-independent mechanisms.

Recent studies have demonstrated that several compounds with diverse structures can activate wild-type cystic fibrosis transmembrane conductance regulator (CFTR) by non-receptor-mediated mechanisms. Some of these compounds have been shown to enhance cAMP-dependent activation of DeltaF508-CFTR. This study was undertaken to compare the mechanisms by which genistein, IBMX, milrinone, 8-cyclopentyl-1, 3-dipropylxanthine (CPX), the benzimidazolone NS004, and calyculin A increase CFTR activity. Our studies demonstrate that, in transfected NIH-3T3 cells, maximal enhancements of forskolin-dependent DeltaF508-CFTR activity are greatest with genistein, IBMX, and NS004. Milrinone, genistein, CPX, NS004, and calyculin A do not increase cellular cAMP. Because forskolin and calyculin A increase in vivo phosphorylation of cAMP binding response element (CREB), the inability of milrinone, genistein, CPX, and NS004 to increase CREB phosphorylation suggests that they do not stimulate protein kinase A or inhibit phosphatase activity. Our data suggest that the mechanisms by which genistein and NS004 activate CFTR differ. We also demonstrate that, in NIH-3T3 cells, IBMX-dependent enhancement of cAMP-dependent CFTR activity is not due to an increase in cellular cAMP and may involve a mechanism like that of genistein.

Semaphorins III and IV repel hippocampal axons via two distinct receptors.

The semaphorins are the largest family of repulsive axon guidance molecules. Secreted semaphorins bind neuropilin receptors and repel sensory, sympathetic and motor axons. Here we show that CA1, CA3 and dentate gyrus axons from E15-E17 mouse embryo explants are selectively repelled by entorhinal cortex and neocortex. The secreted semaphorins Sema III and Sema IV and their receptors Neuropilin-1 and -2 are expressed in the hippocampal formation during appropriate stages. Sema III and Sema IV strongly repel CA1, CA3 and dentate gyrus axons; entorhinal axons are only repelled by Sema III. An antibody against Neuropilin-1 blocks the repulsive action of Sema III and the entorhinal cortex, but has no effect on Sema IV-induced repulsion. Thus, chemorepulsion plays a role in axon guidance in the hippocampus, secreted semaphorins are likely to be responsible for this action, and the same axons can be repelled by two distinct semaphorins via two different receptors.

[Changes of carbohydrate metabolism in normal pregnancy and its relationship with placental lactogen concentrations].

OBJECTIVE: To observe the changes of glucose metabolism and its relationship with human placental lactogen (HPL) in normal pregnant women. METHOD: 94 normal pregnant women had their serum HPL concentration tested and had undergone a 75 g oral glucose tolerance tests (OGTT). Both serum insulin and glucose levels at different time intervals were determined, and the areas under curve (IAUC, GAUC) were caculated, also insulin resistance (IAI) was evaluated. RESULTS: GAUC, IAUC, IAI and HPL are all significantly increased with advance of gestation (P < 0.05). Multiple correlation coefficient study demonstrated that HPL is significantly positively correlated with GAUC, IAUC and IAI(P < 0.001). But when IAUC and IAI were controlled, the relationship between HPL and GAUC was not found. CONCLUSION: There are hyperinsulinemia and insulin resistance during pregnancy, and they are related to HPL. HPL is one of the factors that contributing to carbohydrate metabolism changes during pregnancy.

[Chemosensitivity test for 170 human breast carcinoma samples].

We tested the in vitro sensitivity of 170 primary breast carcinoma samples to the anti-cancer drugs conventionally used in clinical chemotherapy. It was found that: 1. The high sensitivity rate (HSR) of breast carcinoma cells to 5-fluorouracil (5-Fu), mitomycin (MMC), cisplatin (DDP), adriamycin(ADM), methotrexate(MTX), vincristin(VCR), vepesid(VP-16), taxol, and vindesine(VDS) were 57.1%, 49.4%, 48.3%, 35.8%, 21.8%, 21.2%, 10%, 18.2% and 20.9%, respectively. The increase of HSR of cancer cells to 5-Fu, MMC, DDP, taxol was more significant than that of the other drugs tested (P < 0.05). 2. No correlation was shown between drug sensitivity and factors, i.e. pathological type, clinical type, expression of estrogen receptor (ER) and epithelium growth factor receptor(EGFR) for breast carcinoma (P > 0.05). 3. Cyclosporin A can increase part of the sensitivity of ADM-resistant breast carcinoma cells to ADM.

[Short-and long-term therapeutic effects of brachytherapy on intracavitary residual tumor in 563 nasopharyngeal carcinoma (NPC) patients].

OBJECTIVE: To evaluates the effect of brachytherapy on intracavitary residual tumor in patients with NPC after external radiation therapy. METHODS: Five hundred fifty three NPC patients with residual tumor in the nasopharynx were treated with large dose brachytherapy (20-25 Gy). The treatment results were retrospectively evaluated. RESULTS: Within 3 months after brachytherapy, the overall regression rate of the residual tumor was 90.4%. Upon long-term follow-up, the 1-, 3-, and 5-year survival rate was 89.6%, 67.0% and 55.4%, respectively. The major complications of the after-loading treatment were nasal obstruction (16.2%), necrosis of the naso-pharynx (0.9%) and perforation of soft palate in 2 patients. CONCLUSION: Brachytherapy is effective in the treatment of residual NPC especially in patients in early stage. It may not be helpful for stage IV patients.

[Effect of zinc deficiency on apoptosis of spermatogenic cells of rat tostis].

OBJECTIVE: To study the changes of testis apoptosis in zinc deficient rats will promote the understanding of the molecular mechanism of zinc deficiency in the development and function of testis. METHODS: 16 Wistar rats were divided randomly into zinc control group (ZC) and zinc deficiency group (ZD). The serum and testis zinc contents were measured with atomic absorse method; the apoptosis of spermatogenic cells was studied with in situ nick translation (ISNT) technique. RESULTS: Under zinc deficient status, the zinc contents of the serum and testis were obviously decreased (P < 0.05). The apoptosis number of spermatogenic cells was significantly increased (P < 0.01). CONCLUSION: The adequate amount of zinc is essential to the development of testis, whereas zinc deficiency can harmfully affect it. This effect is perhaps carried out in different ways, but the increasing apoptosis numbers of spermatogenic cells might be one of molecular miechanisms of the effect of zinc defficiency on testis development.

[TGF-beta 1 antisense gene transfer into ito cells and suppressed extracellular matrix production].

OBJECTIVE: To investigate possible role of antisense TGF-beta 1 RNA in the regulation of TGF-beta 1 and ECM production in Ito cells. METHOD: A human transforming growth factor-beta 1(TGF-beta 1) cDNA (1467 bp) was inserted in reverse orientation into the retroviral vector, constructed retroviral vector pLATSN of antisense RNA for TGF-beta 1. A higher-titer, recombinant retroviral vector carried antisense RNA for TGF-beta 1 produced in PA317 packaging cells has been introduced into human Ito cells lines LI90. After selection with G418, resistant colonies were obtained. RESULTS: Stable integration of retrovirus in infectants was shown the presence of antisense RNA was detected by RT-PCR. The expression of TGF-beta 1 protein and the production of extracellular matrix such as FN, Co1A1 were markedly decreased in the antisense TGF-beta 1 transfected cultured cells by ELISA, immunohistochemistry and in situ hybridization. CONCLUSION: Antisense RNA of TGF-beta 1 can be successfully used to inhibit Ito cells activated, endogenous TGF-beta 1 mRNA and extracellular matrix produced, and may provide a basis for the development of anti-fibrosis gene therapy.

[Estimation of cytosolic free calcium within synaptosomes with fluorescene indicator Fura-2].

Intracellular free calcium [Ca2+]i concentration has become a focus of attention for investigators in the field of brain edema. It is the purpose of the present study that the changes of the concentration of the free calcium within cortical neurons were investigated on the model of brain edema induced by freezing injury. [Ca2+]i was determined in calcium fluorescent indicator Fura-2/AM loaded neuronal synaptosomes by using mono-wave length spectrofluorophotometer. The method for preparing synaptosomes, and the method and mechanism of concentration of cytosolic free calcium using fluorescent indicator Fura-2/AM were studied.

[Orthogonal design-direct analysis for PCR optimization].

Polymerase chain reaction (PCR) optimization is difficult but very important. It is hard to gain the optimal condition due to the confusion of too many factors influencing the experiment. We report a new method, orthogonal design-direct analysis, which has been confirmed to be scientific and easy to follow by many times of PCR.

[A clinical analysis of 64 cases of pemphigus].

OBJECTIVE: Reasonable therapeutic dosage regimen for pemphigus are analyzed to find the factors related to prognosis. METHODS: A retrospective study is made on 64 cases of pemphigus who were admitted to our hospital between 1986-1996. RESULTS: Sixty-two of 64 cases with pemphigus were treated with systemic administration of corticosteroids. The average dosage of prednisone for controlling the diseases were 0.958 mg.kg-1.d-1 for mild-type of pemphigus (12 cases) and 1.145 mg.kg-1.d-1 for severe-type (50 cases) respectively. Furthermore, 26 of 62 cases were treated with corticosteroids only, and the average dosage of prednisone for controlling the diseases was 1.072 mg.kg-1.d-1 while in 36 cases treated with corticosteroids combined with immunosuppressors, the average dosage of prednisone was 1.178 mg.kg-1.d-1. The rate of side-effects and complications were 15.4% in patients with corticosteroids alone and 36.1% in patients with combined therapy. Six of eight patients who died were diagnosed pemphigus vulgaris and seven with a course of disease less than 3 years. CONCLUSIONS: We recommend a moderate to high dosage of corticosteroids be the first choice to control pemphigus. Those who were effective with steroids are not recommended to use additional immunosuppressors. Patients with pemphigus vulgaris usually have a poor prognosis while those who could survived more than 4 years usually have a better outcome.

[Identification and study on effect of new stress protein from Neisseria gonorrhoeae].

OBJECTIVE: To investigate the influence of changing environmental growth conditions on the virulence of Neisseria gonorrhoeae. METHODS: The gonococci were subjected to 1 mmol/L hydrogen peroxide (sublethol concentration) for one hour. RESULTS: A novel gonococcal stress protein with molecular weight of 62,000 was identified by radioactive labeling and sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Gonococci exposed to 1.0 mmol/L hydrogen peroxide made it significantly more resistant to higher concentrations of hydrogen peroxide and to neutrophils than control organisms (P < 0.01). CONCLUSIONS: These results suggest that the 62,000 stress protein might increase the virulence of N. gonorrhoeae and might be an adapted regular protein produced by N. gonorrhoeae survived within human neutrophils.

[Effect of Shenmai injection on morphological structure of kidney and peritoneum in rats with 5/6 nephrectomy].

OBJECTIVE: To study the protective effect of Shenmai injection (SMI) on kidney and peritoneum in rats with 5/6 nephrectomy. METHODS: By using cell morphological quantification, light microscopy and scanning electron microscopy, the morphological structure changes of peritoneum and kidney in rats with 5/6 nephrectomy caused chronic renal failure model after the intramuscular or intraperitoneal injection of SMI were observed. RESULTS: The SMI injection could relieve glomerular sclerosis, promote the recovery and proliferation of injured peritoneal mesothelial cell. The effect of the SMI intraperitoneal injection is better than that intramuscular of injection. CONCLUSION: The SMI has the protective effect on kidney and peritoneum in rats with 5/6 nephrectomy.

[Analysis and toxicological evaluation of hazardous gases in sealed cabin].

82 volatile organic compounds (VOCs) of eight organic sorts and 3 target inorganic compounds in a sealed cabin that simulating the flying spaceship were identified and quantified for 5 d, the law of hazardous gas concentration variation was discussed, and the atmosphere toxicology was evaluated preliminarily. It provides a basis for detecting gas compounds and evaluating the atmosphere toxicology in the spaceship.

[The timing and mode of surgery in patients with severe acute pancreatitis].

OBJECTIVE: To investigate the timing and mode of surgical management in patients with severe acute pancreatitis (SAP). METHOD: The timing, mode of operation and mortality were reviewed retrospectively in 50 SAP cases undergoing laparotomy in our hospital from 1985 to 1994. RESULT: The overall mortality was 14% and 85.7% of the mortality falls on the age between 40 to 60 years, with no difference in etiology and sex. The operative mortality reduced gradually in patients with longer duration of illness (P > 0.05). The operative mortality of 31.8% in patients undergoing laparotomy within 12 h after hospitalization was significantly higher (P < 0.05) than in these operated on later. When the number of injured extra-pancreatic organs increased, the mortality also increased significantly (chi(2) = 5.99, P < 0.05). The related mortality with shock, pancreatic necrosis and multiorgan failure (MOF) was 31.8%, 31.6% and 28%, respectively (P < 0.05). The main procedures of surgical management were pancreatic debridement and/or drainage, and combined surgical measures. CONCLUSION: The laparotomy timing is an important factor influencing mortality as well as the severity of pancreatic necrosis, sepsis, and MOF. Delayed laparotomy and 24 h preoperative management is recommended. Surgery adopted should be simple, efficient in drainage and debridement.

[Long-term results of combined splenorenal shunt and porta-azygos devascularization in patients with portal hypertension].

OBJECTIVE: To investigate the long-term effects of combined splenorenal shunt and porta-azygos devascularization in patients with portal hypertension. METHOD: We analysed retrospectively the follow-up results of 140 portal hypertensive patients undergoing the combined therapy in our hospital from April 1978 to April 1997. Changes of portal hemodynamics were studied by Doppler Flowmeter and DSA pre- and-postoperatively and direct FPP measurement intraoperatively. RESULT: The operative mortality was 3.6%. No rebleeding was noted in a short period and rebleeding rate was 8.3% in a long term follow-up. The rate of postoperative hepatic encephalopathy was 5.0%. The long-term survival rate of 5-, 10- and 15-years was 83.6%, 64.5% and 54.5%, respectively. After operation FPP and PVP decreased significantly (P < 0.01) and FPP maintained at the level of 3.2 +/- 0.4 kPa. CONCLUSION: The combined procedure is a rational and practical procedure for the treatment of patients with portal hypertension.
