The role of hydroxyl radical as a messenger in the activation of nuclear transcription factor NF-kappaB.

Although it is generally believed that reactive oxygen species activate NF-kappaB, a primary oxidative stress-responsive transcription factor, it is unclear which one among these species causes NF-kappaB activation. Our hypothesis is that hydroxyl radical (*OH) functions as a messenger for the activation of NF-kappaB. Jurkat cells, macrophages and JB6 cells were used to test this hypothesis. Cr(VI), silica and ZnO were used as sources of *OH radicals. None of these *OH generating systems involves exogenous H2O2. Cr(VI) expressed enhanced activity in induction of NF-kappaB in Jurkat cells. This activation of NF-kappaB was decreased by a metal chelator, diethylene triaminepentaacetic acid or a H2O2 scavenger, catalase, but was increased by superoxide dismutase. Mn(II), which reacts with Cr(IV) to inhibit this metal ion-mediated *OH generation, decreased the NF-kappaB activation. Sodium formate, an *OH radical scavenger, also inhibited the NF-kappaB activation. Electron spin resonance measurements show that Cr(VI) was reduced by Jurket cells to Cr(IV) and Cr(V). During the reduction process, molecular oxygen was reduced to O2 and then to H2O2, which reacted with Cr(IV) and Cr(V) to generate *OH radical. The *OH generation correlated with the Cr(VI)-induced NF-kappaB activation. Similarly, silica caused NF-kappaB activation in macrophages via the *OH radical-mediated reaction. This radical was generated via metal mediated reaction from H2O2, which was generated by the reduction of molecular oxygen via O2- as an intermediate during the silica-stimulated 'respirable burst'. Silica particles did not cause *OH generation either in Jurket or in JB6 cells and thus did not cause any observable NF-kappaB activation in these cells. ZnO induced NF-kappaB activation in JB6 cells through the generation of *OH resulting from light irradiation of ZnO which was measured by electron spin resonance. The results thus show that *OH radical functions as a messenger for NF-kappaB activation. Antioxidants, which scavenge *OH radical or its precursors, inhibit NF-kappaB activation. Metal chelators, which make metal ions incapable of generating *OH from H2O2, inhibit activation of this transcription factor.

Antioxidant properties of aspirin: characterization of the ability of aspirin to inhibit silica-induced lipid peroxidation, DNA damage, NF-kappaB activation, and TNF-alpha production.

Electron spin resonance (ESR) was used to investigate the reaction of aspirin toward reactive oxygen species, such as hydroxyl radicals (*OH), superoxide radicals (O2-) and H2O2. The Fenton reaction (Fe(II) + H2O2 ---> FE(III) + *OH + OR) was used as a source of *OH radicals. The results show that aspirin is an efficient *OH radical scavenger with a reaction rate constant of k = 3.6 x 10(10) M(-1) sec(-1), which is faster than several well established antioxidants, such as ascorbate, glutathione and cysteine. However, aspirin is not a good scavenger for O2- or H2O2. Through its antioxidant property, aspirin exhibited a protective effect against silica-induced lipid peroxidation and DNA strand breakage. Aspirin also inhibited the activation of nuclear transcription factor-kappaB induced by silica, lipopolysaccharide or the transition metal, Fe(II), as demonstrated by electrophoretic mobility shift assay. The results show that aspirin functions as an antioxidant via its ability to scavenge *OH radicals. This antioxidant property may explain some of its various physiological and pharmacological actions.

Rat-tail replantation model.

The authors evaluated the reliability of a rat-tail replantation model Eleven male adult Sprague-Dawley rats were involved. The tails were completely amputated 3 to 6 cm distal to the base. They were then replanted in the following order: fixation of the coccygeal vertebrae; suturing of tendons; anastomosis of the artery; anastomosis of the two dominant veins (or one of them); and skin closure. Tails were inspected daily for 14 postoperative days for color, capillary refill, skin consistency and wound condition. Subsequently, ultrasound Doppler inspection and angiography were carried out to observe the patency of the vessels. All replanted tails were pink, viable, and had normal capillary refill and appearance. Complete survival was achieved. Doppler flow imaging and angiography demonstrated patency of the vessels. This rat-tail replantation is a reliable and reproducible experimental model of microvascular anastomosis. Crucial steps in the procedure are presented.

Resveratrol suppresses cell transformation and induces apoptosis through a p53-dependent pathway.

Resveratrol, a plant constituent enriched in the skin of grapes, is one of the most promising agents for the prevention of cancer. However, the mechanism of the anti-carcinogenic activity of resveratrol is not well understood. Here we offer a possible explanation of its anti-cancer effect. Resveratrol suppresses tumor promoter-induced cell transformation and markedly induces apoptosis, transactivation of p53 activity and expression of p53 protein in the same cell line and at the same dosage. Also, resveratrol-induced apoptosis occurs only in cells expressing wild-type p53 (p53+/+), but not in p53-deficient (p53-/-) cells, while there is no difference in apoptosis induction between normal lymphoblasts and sphingomyelinase-deficient cell lines. These results demonstrate for the first time that resveratrol induces apoptosis through activation of p53 activity, suggesting that its anti-tumor activity may occur through the induction of apoptosis.

Membrane type 1-matrix metalloproteinase (MT1-MMP) and MMP-2 immunolocalization in human prostate: change in cellular localization associated with high-grade prostatic intraepithelial neoplasia.

Membrane type 1-matrix metalloproteinase (MT1-MMP) is a known activator of latent MMP-2 (pro-MMP-2), and increased MMP-2 expression has been associated with tumor aggressiveness in prostate cancer. However, expression of MT1-MMP in human prostate tissue has not been described. We investigated the expression and immunolocalization of MT1-MMP and MMP-2 in the epithelial components of benign prostate epithelium, high-grade prostatic intraepithelial neoplasia (HGPIN), and prostate cancer. Tissue sections from the peripheral zone of 50 prostates (radical prostatectomy specimens) were chosen based on their containing benign glands, HGPIN, and prostate cancer glands. All 50 sections were immunostained for MT1-MMP and MMP-2 and were evaluated for staining pattern, uniformity, and intensity. Western blotting and gelatin zymography were done to confirm expression of MT1-MMP and activity of MMP-2, respectively. Comparisons were made between benign epithelium, HGPIN, and cancer. In benign glands, basal cells (BCs) uniformly stained intensely for MT1-MMP, whereas secretory cells (SCs) were rarely positive (P < 0.0001). Conversely in HGPIN, SCs showed consistent cytoplasmic staining (P < 0.0001). In cancer cells, staining was heterogeneous and varied from no staining to very intense staining in select glands. MMP-2 in normal tissue stained both BCs and the apical region of SCs, whereas in HGPIN, staining was observed in the SC in a predominantly cytoplasmic pattern. Similar to MT1-MMP, staining in cancer tissue for MMP-2 was heterogeneous; however, there was a significant association between the pattern of MMP-2 and MT1-MMP staining within the epithelial components of the cancer glands in individual specimens (P < 0.001). Finally, MMP-2 and MT1-MMP were confirmed to be expressed in the prostate tissues by gelatin zymography and Western blotting. In conclusion, we found that consistent changes in localization and intracellular distribution of MMP-2 and MT1-MMP were associated with the transition from benign prostate epithelium to HGPIN, suggesting that regulation of these enzymes is altered during the earliest stages of prostate cancer.

Intercellular adhesion molecule-1 and accumulation of eosinophils in nasal polyp tissue.

OBJECTIVE: To investigate the relationship between intercellular adhesion molecule-1 (ICAM-1) and the accumulation of eosinophils in nasal polyp tissue to better understand the mechanism of airway eosinophilic inflammation. METHODS: The expression of ICAM-1 and its natural ligand, lymphocyte function-associated antigen-1 (LFA-1), in normal nasal mucosa from 6 controls and in nasal polyp tissue from 19 patients with nasal polyposis were determined with immunohistochemistry. With dual immunohistochemistry and May-Griünwald Giemsa stain (MGG), the expression of LFA-1 and infiltrating eosinophils in nasal polyp tissue was observed. RESULTS: The expression of ICAM-1 and LFA-1 was stronger in the nasal polyp tissue than in normal nasal mucosa. There was a positive relationship between the infiltration of eosinophils and the expression of LFA-1 on eosinophils. CONCLUSION: Accumulation of eosinophils in nasal polyp tissue is associated with the counter-effect between adhesion molecules and its ligand on eosinophils.

[WT1 gene expression in leukemia patients and its correlation with prognosis and multidrug resistance].

OBJECTIVE: To evaluate the value of expression of WT1 gene in predicting the prognosis of leukemia patients, and explore the relationship between WT1 gene expression and multidrug resistance and cell apoptosis. METHODS: Expressions of WT1, MRP and mdr1 were measured in 68 leukemia patients by RT-PCR method. Expression of bcl-2 was measured in 32 AML patients by immunofluorescence flow cytometry. RESULTS: Expression of WT1 was revealed in 36 of 68 leukemia patients and none of 23 normal controls. Complete remission rate (59.46%) was lower in WT1 positive patients than that (87.10%, P = 0.011) in WT1 negative patients. The rate of MRP expression was also higher in patients with WT1 expression (58.33%), than in those without WT1 expression (32.26%, P = 0.033). Thirty-two AML patients were divided into high-, intermediate-, and low-risk groups according to the expression of WT1 and bcl-2. CR rates were significantly different among these 3 groups (33.33% for high-, 47.37% for intermediate-, and 100% for low-risk group, P < 0.05). CONCLUSION: The expression of WT1 can predict the treatment outcome and the prognosis for leukemia patients. Expression of WT1 is the most important risk factor, and the coexpression of WT1 and bcl-2 predicts poor prognosis of AML patients.

Effects of rhidosin on expression of fibroblast growth factor on olfactory bulb in rats and its relation with senile hyposmia.

OBJECTIVES: To determine the expression of fibroblast growth factor (FGF) on olfactory bulb in rats and its relation with senile hyposmia, and to discuss the effects of rhidosin on expression of FGF on olfactory bulb in senile rats and its significance. METHODS: Four groups of rats, i.e., young group, senile group, rhidosin group, and physiological saline control group were used in this study. After the removal of the heads of these rats, olfactory mucosa and olfactory bulbs were immediately fixed with neutral formalin, followed with paraffin-embedding, serial sectioning, immunohistochemical staining and light microscopic observation. RESULTS: The positive expression rate of FGF on olfactory bulb in young group was significantly higher than that in senile group (P < 0.01). The positive expression rate of FGF on olfactory bulb in rhidosin group was significantly higher than that in physiological saline control group (P < 0.01). No positive expression of FGF on olfactory mucosa was observed in these four groups. CONCLUSION: These results suggest that the expression of FGF on olfactory bulb gradually decline with aging. The decrease of expression of FGF on olfactory bulb may play a key role in the occurrence of senile hyposmia. Rhidosin can increase the expression of FGF on olfactory bulbs in senile rats and this may be one of the mechanisms by which rhidosin promotes the growth of olfactory cells and inhibits aging effects on the olfactory system.

[The relationship between surgical staging, pathologic grading, operative type and postoperative recurrence in giant cell tumor of bone].

Sixty-eight surgical procedures of 58 patients with Giant Cell Tumor of Bone(GCT) were followed up over two years. The recurrence rate of GCT was Stage 1: 0 in 5, Stage 2: 15.4% in 39 and Stage 3: 37.5% in 24. It was significantly higher in Stage 3 than in Stage 2 (P < 0.05). The recurrence rate of GCT in Stage 2 was 30.8% in 13 intralesional curettage, and 0 in 17 wide or radical resection(P < 0.05). We conclude that the surgical staging of GCT has important value in predicting postoperative recurrence, in accurately evaluating the operative efforts and in guiding treatment, and that the operative type is a significant factor effecting on the recurrence of GCT.

[Electrophysiological criteria for selection of operative procedure in the treatment of late incomplete obstetrical palsy].

OBJECTIVE: Using electrophysiological method to evaluate the severity of incomplete obstetric paralysis brachial plexus injuries, and establish an electrophysiological criteria for selection of operative procedure in the treatment of cases of late stage. METHODS: Neurolysis was performed in 16 patients, and during the operation, the neuroma at the upper trunk was discovered. The electrophysiological study was carried out before and during operation to evaluate the conducting function of neuroma. In the follow-up the operative result was analysed by Mallet test. RESULTS: The preoperative study showed that the compound muscular active potential (CMAP) amplitude of supraspinatus deltoid and biceps were decreased more than 60% in comparison with that of the healthy side. After external neurolysis of the neuroma, by stimulation of the proximal and distal ends of the neuroma the average decrease of CMAP amplitude of the above three muscles was 37.45% +/- 20.97%, 47.85% +/- 26.23%, 47.05% +/- 21.23%, respectively and no significant improvement was observed in the shoulder and elbow function in all of the 16 cases. CONCLUSION: When the preoperative electrophysiological study found that the CMAP amplitude decreased more than 60% in comparison with that of the healthy side, transposition of nerve or nerve grafting was indicated.

[Cell infiltration in nasal polyps and it's pathological significance].

OBJECTIVE: To study the relation between inflammatory cells infiltration in nasal polyp tissues and it's pathogenesis. METHOD: Nasal mucosa from 10 normal individual and 4 patients of nasal polyps were studied with immunohistochemical technique and histopathologic examination. RESULT: The number of eosinophils, CD68 positive cells and monocytes was significantly higher (P < 0.05) in nasal polyps tissues than in the normal controls. CONCLUSION: Infiltration of inflammatory effector cells such as eosinphils and chronic inflammatory changes of the nasal mucosa might play a role in nasal polyps sprouting.

[Study of the relation between MMP2, MMP9 and nasopharyngeal carcinoma].

OBJECTIVE: To evaluate the role of matrix metalloproteinase (MMP) in the invasion and metastasis of nasopharyngeal carcinoma (NPC). METHOD: Twenty-six cases of NPC were studied by immunohistochmical method using monocolonal antibody to MMP2 and MMP9. RESULT: It was showed that there were significantly correlation between MMP2 or MMP9 and the invasion and metastasis of NPC. The positive rate of MMP2 and MMP9 were higher in NPC with cervical lymph node involvement than that of no cervical lymph node involvement. And they were lower in well differentiated NPC than poorly differentiated NPC. CONCLUSION: The results indicate that MMP2 and MMP9 may play a critical role in assisting carcinoma cell to degrade basement membrane to invade surrounding tissues and to form metastatic colonies in lymph nodes.

[Studies on human nasal cycle with acoustic rhinometry].

OBJECTIVE: The aim of this article was to study the classifications and characteristics of nasal cycle of adult and children and the influence of pathological state of nasal mucosa on nasal cycle with acoustic rhinometry. METHOD: We examined the nasal cycle of 16 healthy adults, 11 healthy children and 14 patients with bilateral nasal obstruction. Acoustic rhinometry was performed every 30 minutes (20 minutes in children) over 4-8.5 hours to determine minimum cross-sectional area of nasal cavity (MCA) and nasal cavity volume (NV) which were used as indices of nasal patency. RESULT: Nasal cycles were observed in all subjects. Classical nasal cycle was most commonly observed in healthy adults and patients with nasal diseases. In healthy children, in concert nasal cycle was the most common type. The distribution of the healthy adults and patients with nasal diseases between the cycle categories was not statistically significant different (P > 0.05), the mean periodicity of the cycle was no significantly different between these two groups (P > 0.05). The mean alteration amplitude of nasal cycle of healthy adults was significantly longer than that of patients with nasal diseases (P < 0.01). The distribution of the healthy children and the healthy adults between the cycle categories was significantly different (P < 0.01), the mean periodicity of the cycle of healthy children was distinctively shorter than that of healthy adults (P < 0.01). CONCLUSION: The above results illustrated that nasal cycle exists as central control and local modulation. In healthy children, in concert nasal cycle was most commonly observed and this may be related with the immature condition of their autonomic nervous system.

[Preparation of immobilized penicillin V acylase from Fusarium oxysporum and its properties].

Extracellular penicillin V acylase from Fusarium oxysporum FP941 was partially purified by means of adsorption on, and elution from gamma-alumina and (HN4)2SO4 fractional precipitation. The enzyme was immobilized on acrylic polymer support linkaged with covalent bond. The activity of wet immobilized enzyme was 217 IU/g, The activity yield of Immobilized enzyme was 53%. Optimum pH and temperature for immobilized enzyme action wer 8.0 and 55 degrees C, respectively. The enzyme was stable below 50 degrees C and pH 4-11. The activity of the immobilized enzyme remained 90% after it was reused 25 times.

[Preliminary study on NS5 region gene sequencing and variants of hepatitis G virus in China].

OBJECTIVE: To study the genomic specificity and variation of Hepatitis G virus(HGV) in China. METHODS: The 141nt cDNA fragments locating at nonstructural region 5 (NS5) of HGV were obtained from the sera of 16 HGV infected patients by RT-nested-PCR using NS5 primers, then purified and directly sequenced by dideoxynucleotide chain termination method. RESULTS: The sequence diversity near the N-terminus of the region between nt 7,820 and 7,960 was more marked than that near the C-terminus. Compared with the HGV-US isolate, the nt alterations of NS5 in Chinese isolates usually took place at nt 7,824, 7,892, 7,914, 7,917, 7,935 and in region nt 7,845-7,860, the rate of variability at one nt position was 0-89.5%. The nucleotide homology was 83.6%-95.7% (89.6% in average) and the homology of deduced amino acid sequence was 83.0-100%(91.2% in average). CONCLUSION: The nucleotide and deduced amino acid sequences of NS5 of Chinese HGV are relatively conserved but are obviously different from the HGV and GBV-C reported in USA in this study.

CONCLUSION: Rhidosin can increase the expression of FGF on olfactory bulbs in senile rats and these may be one of the mechanisms by which rhidosin exerts its anti-aging effects on the olfactory system.

[Expression of estradiol receptors in mast cells of the upper airway].

OBJECTIVE: To examine the expression of estrogen receptors in mast cells, CD4+, CD8+ and CD68+ cells in human nasal polyps. METHODS: Immunohistochemical and dual immunostaining methods were used with mouse monoclonal antibodies against human estrogen receptors, a range of human immune cell types included CD4+ (helper T-cells), CD8+ (suppressor/cytotoxic T-cells), CD68+ (Macrophages) cells and mast cells. RESULTS: Coincident with the known features of human nasal polyps, all the immune/inflammatory cell types examined were clearly detected with corresponding monoclonal antibodies. The cells positive for estrogen receptor were morphologically similar to the mast cells but distinct from CD4+, CD8+ or CD68+ cells. The cells stained for estrogen receptors by dual immunostaining coincided exactly with cells labeled as mast cells but differed from CD4+, CD8+ or CD68+ cells. CONCLUSION: Estrogen receptors express only in mast cells, but not in CD4+, CD8+ or CD68+ cells in the human upper airway.

[Sodium channels in the apical membrane of human nasal epithelial cells].

OBJECTIVE: To study the physiological properties of sodium channels in the apical membrane of human nasal epithelial cells. METHODS: Serum-free cultured nasal epithelial cells of human inferior turbinate from obstructive sleep apnea syndrome patients on collagen gel-coated membranes at an air-liquid interface were studied by patch clamp technique. RESULTS: In cell-attached patches, a typical single channels current was recorded, their conductance was 21.09 pS, the reversal potential was -50.96, and 77.78% of them < -40 mV. Permeability ratio PNa/PK > 5.80. In the presence of 10(-4) mmol/L amiliride in the pipette, the incidence of sodium channels decreased from 26.67% to 5.13%. It revealed that a population of channels were inhibited by amiloride. Ca2+ did not influence the incidence of sodium channels(P > 0.05), there were no obviously association between voltage and the open probability of the channels. CONCLUSION: On Na+ channels in cell attached patches of human nasal epithelial cells, most of channels are amiloride-sensitive and selective Na+ over K+; The channels were not activated by extracellular Ca2+, but the open probability of them were voltage-independence.
