DNA methylation contributes to expression of the human neurotensin/neuromedin N gene.

The gut and liver share a common embryological origin. The gene encoding the gut hormone neurotensin/neuromedin N (NT/N) is expressed in the adult small bowel, and NT/N is transiently expressed in the fetal liver, suppressed in the adult liver, and reexpressed in certain liver cancers. In our present study, we found that the NT/N gene was expressed at high levels in the human hepatoma cell line Hep 3B but was not expressed in Hep G2 cells. To further determine the mechanisms regulating NT/N expression, we performed Southern blotting and gene cloning techniques. Neither alteration nor mutation of the NT/N gene was responsible for this differential NT/N expression pattern. Human NT/N promoter constructs were transfected into either Hep 3B or Hep G2. Both cell lines supported NT/N transcription, indicating that the absence of NT/N expression in Hep G2 cells was due to mechanisms other than the absence of positive transcription factors. The role of DNA methylation was next assessed. Methylation of NT/N promoter constructs in vitro resulted in a 67-fold reduction in promoter activity, whereas treatment with the demethylating agent 5-azacytidine induced NT/N expression in Hep G2 cells, thus suggesting that DNA methylation plays a role in the expression of the gut endocrine gene NT/N. Defining the mechanisms regulating NT/N expression in these hepatic-derived cell lines will provide not only a better understanding of cell-specific and developmental regulation of a gut endocrine gene but also possible insight into liver cell lineage patterns and the derivation of certain hepatocellular cancers.

Inhibition of ultraviolet C irradiation-induced AP-1 activity by aspirin is through inhibition of JNKs but not erks or P38 MAP kinase.

The exposure of mammalian cells to ultraviolet (UV) irradiation leads to the activation of transcription factors, such as AP-1 and NFkB. We demonstrate that aspirin, a promising cancer chemopreventative agent, inhibited UVC-induced AP-1 activity in JB6 cells. In JB6 cells, UVC stimulated Erks, JNKs and P38 kinase activities; aspirin only inhibited activation of JNKs, but not the other MAP kinases. Since the transcription factor AP-1 is important for the process of tumor promotion, the inhibitory effect of aspirin on AP-1 activation suggests that it can be used as a chemopreventative agent against skin cancer.

Vanadium induces AP-1- and NFkappB-dependent transcription activity.

Vanadate has been reported to be involved in the causation of cancer. In this study, we found that both AP-1 and NFkappaB activities were increased after treatment with sodium vanadate in JB6 cells. Maximum induction of AP-1 and NFkappaB appeared at 48 to 72 h. Phosphorylations of Erks and p38 kinases were markedly increased at 100 microM of vanadate, while phosphorylation of JNKs was not affected. Vanadate also enhances the phosphorylation of IkappaBalpha. These results suggest that the activation of AP-1 and NFkappaB by vanadate may be mediated through enhancement of phosphorylation of Erk/p38 kinases and IkappaBalpha, respectively.

Study on the genetic determinants responsible for expression and assembly of CS3 fimbriae.

Colonization factor antigens (CFAs) are important virulent factors and protective antigens of enterotoxigenic Escherichia coli (ETEC). Among the known CFAs of human origin, CS3 is a major fimbriae antigen. To study the genes associated with the expression and assembly of CS3 is a key step to delineate the regulating mechanism of expression and assembly of the fimbriae. In this paper we analyzed the restriction map of the CS3 genetic determinant and successfully cloned the CS3 subunit gene and the region encoding the auxiliary proteins. Expression results of the recombinant plasmids in minicells showed that the genetic determinant of CS3 fimbriae encodes six polypeptides with molecular weights 15, 17, 24, 27, 48, and 90 kDa. Western blotting and complementary expression analysis among the different mutants showed that the 15 kDa/17 kDa proteins compose the CS3 subunit and the 17 kDa protein is the precursor. The others may be necessary for the assembly of CS3 fimbriae. The relative locations of various genes of the CS3 genetic determinant were also determined.

[Effect of crushing of sciatic nerve on neuron of lumbar spinal cord].

In order to investigate the effect of nerve compression on neurons, the commonly used model of chronic nerve compression was produced in 48 SD rats. The rats were sacrificed in 1, 2, 3, 4, 5 and 6 months after compression, respectively. The number of neuron and ultrashruchure of alpha-motor neurons and ganglion cells of the corresponding spinal segment were examined. The results showed as following: After the sciatic nerve were crushed, the number of neuron and ultrastructure of alpha-motor neurons and ganglion cells might undergo ultrastructural changes, and even the death might occur. These changes might be aggravated as the time of crushing was prolonged and the compression force was increased. It was concluded that for nerve compression, decompression should be done as early as possible in order to avoid or minimize the ultructural changes of the neuron.

[Induction of apoptosis in human gastric carcinoma cell line MGC-803 by monoclonal antibody PD4].

OBJECTIVE: To examine if monoclonal antibody PD4 (McAb PD4) which identifies a surface antigen of 40 kD (P40) expressed in gastric carcinoma cells could induce tumor cell apoptosis. METHODS: The antibody was tested for its effect on cell cycle and cell injury in gastric carcinoma cell MGC-803 by flow cytometry, DNA electrophoresis and TdT assay. Fas antigen expression in MGC-803 was also examined. RESULTS: McAb PD4 could block tumor cell proliferation cycle and trigger cell death by inducing apoptosis, but expression of Fas antigen in MGC-803 cells was negative. CONCLUSION: P40 is a tumor associated antigen which is distinct from Fas antigen. Molecular cloning of P40 will clarify the pathway and mechanism of apoptosis induced by this monoclonal antibody.

[The clinical significance of multidrug resistance associated protein(MRP) gene expression in acute leukemia].

OBJECTIVE: To investigate the relationship between the expression of the multidrug resistance-associated protein(MRP) gene and clinical drug resistance in acute leukemia(AL). METHODS: Semi-quantitative reverse transcriptase polymerase chain reaction was used to examine the expression of MRP gene in 65 bone marrow aspirates of AL patients and 15 normal volunteer's peripheral blood mononuclear cells. The expression level of MRP were expressed as ratio of MRP/beta 2M(beta 2-microglobulin) and the ratio of MRP/beta 2M > or = 0.3 was defined as MRP positive. RESULTS: The level of MRP mRNA and the positive percentage of MRP in relapsed-refractory group were significantly higher than that in normal control group. In newly diagnosed group, the first complete remission rate of MRP negative patients(84%) was significantly higher than that of MRP positive(25%) patients. MRP expression level was different in leukemia sub-type. MRP and mdr-1 genes were examined simultaneously in 65 AL patients, the mean expression level and positive percentage of MRP and mdr-1 in clinical resistance group were significantly higher than those in non-resistance group. No significant correlation was found between the MRP and mdr-1 expression(rs = 0.1683). CONCLUSION: High expression of MRP leads to drug resistance and it is an unfavorable factor to prognosis.

[The effect of tamoxifen on experimental nasal hypersensitivity].

The effect of estrogen on nasal hypersensitivity was observed in female adult guinea pigs with high level of estrogen. The animal were divided into three groups: group A as normal control; in group B, tamoxifen was given before inducing high level estrogen and in group C, estrogen was given but without tamoxifen. The groups B and C were sensitized and then challenge with TDI as described previously. The result showed that tamoxifen may inhibit significantly the development of nasal hypersensitivity and reduce the pathologic changes of nasal mucosa. It is suggested that the mast cells may be the target of estrogen and tamoxifen might be useful in treatment of nasal hyperreaction concerning with the change of estrogen level.

[The influence of posture on nasal geometry].

To investigate the effect of posture on nasal cavity geometry, nasal cavity geometry were measured by acoustic rhinometry in 35 healthy adults (17 males and 18 females, 26-50 years old, mean age 32 years) after 5 min in changing postures of head and body. The minimum cross-sectional area and volume between the nostril and 8 cm posteriorly were measured on both sides. When changing from sitting to lateral recumbent with the wider side of the nasal cavity downwards both total volume and total minimum cross-sectional area decreased 18%, volume and minimum cross-sectional area of the wider sides showed that decreased 32% and 26%. When changing from lateral recumbent with the wider side of the nose downwards to lateral recumbent with the narrower side downwards, volume and minimum cross-sectional area of the wider sides and total volume showed that increased 41%, 24% and 12%. Our findings indicate that there is a complex response of the nasal cavity mucosa to posture changing. This factor should be taken into account in rhinological studies of environmental, clinical and pharmacological conditions.

[Expression and significance of vascular permeability factor in nasal polyps].

OBJECTIVE: To study the expression and significance of vascular permeability factor(VPF) in nasal polyps. METHODS: Expression of VPF and its receptor flk-1 in nasal polyps from 9 patients and inferior turbinates from 8 patients with chronic rhinitis were prospectively studied with immunohistochemistry. Each tissue section was observed under optical microscope. RESULTS: The VPF positivity in vascular endothelium and in gland cell was significantly higher in nasal polyps than in inferior turbinates (P < 0.01 and P < 0.05, respectively). The flk-1 labeling index of vascular endothelium was significantly higher in nasal polyps than that in inferior turbinates (P < 0.01). CONCLUSION: VPF may play a key role in the formation of heavy edema of nasal polyps.

[The role of acoustic rhinometry in the evaluation of the volume of nasopharynx before and after adenoidectomy].

OBJECTIVE: To determine the type of acoustic rhinometry curve in different kinds of nasal and nasopharyngeal diseases and the role of acoustic rhinometry in the evaluation of the changes of the volume of nasopharynx before and after adenoidectomy. METHODS: One hundred and fifty-one patients with nasal and nasopharyngeal diseases, including rhinitis, nasal polyps, obstructive sleep apnea syndrome, palatoschisis, atrophic rhinitis, adenoid vegetation, perforation of nasal septum, carcinoma of nasopharynx, and stricture of nasal limen, were measured with acoustic rhinometry. Meanwhile, 15 patients with adenoid vegetation were measured before and after adenoidectomy, the changes in volume recorded by acoustic rhinometry were compared with the adenoid volume obtained by the method of displacement. RESULTS: Acoustic rhinometry curve can be divided into normal and abnormal curves and the abnormal curves can be divided into four types according to the site of changes. Acoustic rhinometry curve altered with the changes in character, degree and site of nasal and nasopharyngeal diseases and could return to normal after surgery or medical treatment. The volume of nasopharynx of adenoid vegetation patients was significantly smaller than that of the normal control and it rose significantly and became closed to normal after adenoidectomy. The volume of adenoid and the calculated changes in volume of the nasopharynx was found to be interrelated and highly significant(r = 0.87, P < 0.001). CONCLUSION: The results indicate that acoustic rhinometry curve is a useful method in the assistant diagnosis and judgment of therapeutic effectiveness of many kinds of nasal and nasopharyngeal diseases and can provide reference for the evaluation of the volume of adenoids.

Selection of new Epitopes from monovalent displayed phage octapeptide library.

A library of 2 x 10(7) random octapeptides was constructed by use of phagemid-based monovalent phage display system. The randomly synthesized degenerated oligodeoxyribonucleotides (oligos) were fused to the truncated g III (p230-p403). Sequence analysis of 11 randomly chosen clones suggested that the degenerated inserts and its deduced amino acid (aa) sequences are randomly distributed. The library was used to select binding peptides to the monoclonal antibody (mAb) 9E10, which recognizes a continuous decapeptide epitope of denatured human c-myc protein. After four to five rounds of panning, most of the eluted clones could bind to 9E10. Sequence analysis of the selected positive clones indicated that the binding sequences could fall into two classes, one class (clone 1) shares a consensus motif, ISE x x L, with c-myc decapeptide; and the sequences of the other class are entirely different. The binding of both classes to 9E10 could be specifically inhibited by free c-myc decapeptide. The immunogenicity of the phage peptide was further investigated by construction of multivalent displayed phage peptides and immunization of animals with or without adjuvant. ELISA and competitive ELISA showed that anti-serum from both mice and rabbit immunized with either clone could bind to the original antigen, c-myc decapeptide. These results denote that in spite of the dissimilarity of the selected peptides with c-myc decapeptide, they are capable of inducing similar immune respones in vivo, thus actually mimicking the antigen epitope.

[Expression of intercellular adhesion molecules in nasal polyps tissue and its significance].

OBJECTIVE: To study the expression of intercellular adhesion molecule-1/lymphocyte function-associated antigen-1 in nasal polyps tissue and its significance. METHODS: Frozen sections of 9 normal nasal mucosa and 19 nasal polyp samples were studied with immunohistochemical technique and immunohistochemical and MGG dual staining method with ICAM-1 and LFA-1 monoclone antibodies. RESULTS: Expression of ICAM-1 and LFA-1 were stronger in nasal polyps tissue than in the normal nasal mucosa, and there is positive relationship between the expression of ICAM-1 and the expression of LFA-1 on eosinophils. CONCLUSION: High expression of ICAM-1 and LFA-1 in nasal polyps tissue showed that interaction of ICAM-1/LFA-1 may participate in the inflammatory reaction and prompt adhesion and migration of eosinophils. High expression of LFA-1+ on eosinophils may play an important role in the pathogens of nasal polyps.

[Effects of different amount of ginger juice on prepared products of Rhizoma Pinelliae].

Rhizoma Pinelliae were diged in Ginger juice of different propertion, the total alkaloid contents of these products were all higher than the raw Rhizoma Pinelliae, and related to the actions of anti-tumous cell. The contents of beta-sitosterol of these products were related to the amount of Ginger juice which is added in Pinelliae Rhizoma, but only the content of beta-sitosterol of the Pinelliae product which is prepared by same amount of Ginger juice was slight higher than the raw Rhizoma Pinelliae. The stimulations of five kinds of products on mice have no obvious relation to the amounts of Ginger juice which were added in Rhizoma Pinelliae, but the stimulation of the Pinellia product which was prepared by same amount of Ginger juice was the lowest of all products on mice. Dependent on above experiment results, we suggest that Rhizoma Pinelliae processed by Ginger juice should be prepared by same amount of Ginger juice with Rhizoma Pinelliae.

Intracellular Ca2+ thresholds that determine survival or death of energy-deprived cells.

Increase of intracellular ionized or free Ca2+ is thought to play a central role in cell death due to ATP depletion. However, concurrently operative mechanisms of injury that do not require intracellular Ca2+ increases have made it difficult to test this hypothesis or to determine the concentrations at which intracellular Ca2+ becomes lethal. The predominant Ca2+-independent mechanism of injury during ATP depletion involves the loss of cellular glycine. This type of damage can be fully inhibited by adding the amino acid exogenously. Using glycine to suppress Ca2+-independent plasma membrane damage, we have examined the effect of intracellular Ca2+ elevations on cell viability during ATP depletion. Madin-Darby canine kidney (MDCK) cells were depleted of ATP by incubation with a mitochondrial uncoupler in glucose-free medium. Free Ca2+ concentration in the medium was varied between 26 nmol/L and 1.25 mmol/L in the presence of a Ca2+ ionophore. Measurements with the Ca2+ probes fura-2, furaptra, and fura-2FF showed that intracellular Ca2+ was clamped at extracellular levels under these conditions. Cell survival during ATP depletion was indicated by viable cells recovered 24 hours later. The results show that ATP-depleted cells can sustain high levels of intracellular Ca2+ (100 micromol/L) for prolonged periods and remain viable if plasma membrane damage is prevented by glycine. Cell death was observed only when intracellular free Ca2+ was allowed to increase beyond 100 micromol/L, and this was associated with dramatic nuclear alterations: chromatin condensation, loss of nuclear lamins, and breakdown of DNA into large 50- to 150-kb fragments. Our studies demonstrate unexpectedly high resistance of cells to calcium cytotoxicity if glycine that is lost during ATP depletion is restored. In addition, they provide insights into novel mechanisms of nuclear disintegration and DNA damage that are triggered when the high thresholds of intracellular Ca2+ required for cell death are exceeded.

Adenosine Triphosphate↗

Development of porous defects in plasma membranes of adenosine triphosphate-depleted Madin-Darby canine kidney cells and its inhibition by glycine.

Studies during the past decade have led to the recognition of a fundamental, widely expressed mechanism of structural damage in energy-deprived cells, which is suppressed by physiologic levels of glycine and is independent of Ca2+ availability or alterations of cytosolic free Ca2+. To gain insight into this process, Madin-Darby canine kidney (MDCK) cells were depleted of adenosine triphosphate (ATP) by a mitochondrial uncoupler in glucose-free medium, and intracellular free Ca2+ was clamped at 100 nM to avoid calcium cytotoxicity. Although the ATP-depleted cells swelled and blebbed and their plasma membranes appeared to be under tension, they nevertheless became permeable to macromolecules. The plasma membranes of these cells retained structural continuity, as determined by morphologic observations, and confocal microscopy of a plasma membrane protein label (Biotin: Ultra Avidin-Texas Red) and a lipid label (NBD-sphingomyelin). Using fluoresceinated dextrans of graded molecular size, membrane permselectivity was examined noninvasively by confocal microscopy. Measured as inside/outside ratios of fluorescence intensity, the permeability indices showed progressively greater restriction to diffusion of increasingly larger dextran molecules across plasma membranes, with sharp break-points between 70,000 and 145,000 daltons (d). The results indicated that the membranes behaved as if they were perforated by water-filled channels or "pores," with size-exclusion limits of molecular dimensions. The membrane defects evolved from small pores permeable only to propidium iodide (668 d) and the smallest dextran (4,000 d), before enlarging with time to become permeable to larger dextrans. Inclusion of glycine during ATP depletion did not affect cell swelling or blebbing but completely prevented the development of permeability defects. Treatment of cells before ATP depletion with a membrane-impermeant homobifunctional "nearest neighbor" cross-linking agent, 3,3' dithiobis(sulfosuccinimidylpropionate), suppressed the development of permeability defects, even in the absence of glycine. These observations suggest that the cellular abnormality that is suppressed by glycine involves rearrangement of plasma membrane proteins to form water-filled pores large enough to leak macromolecules.
