[Reduction of effects of stimulating SmI and electroacupuncture produced by intraperitoneal injecting atropine in the adjuvant-induced acute arthritic rat].

Using the adjuvant-induced acute arthritic rat as a pain model, we have previously shown that the SmI area is implicated in the inhibitory effect of acupuncture on the afferent nociceptive information of parafascicular nuclei (Pf). The purpose of this study was to determine whether acetylcholine (ACh) participates in this modulatory process. Experiments were conducted with adjuvant-induced acute arthritic rats and extracellular recordings from pf were obtained through microelectrodes stereotaxically. The results showed that: (1) After intraperitoneal-injecting atropine (5mg/kg), the effect of stimulating SmI on nociceptive responses of Pf neurons was decreased, while it was not decreased if saline was administered. Compared in the same neurons, the difference of nociceptive responses at 0' to 15' after cessation of stimulating SmI was statistically significant. This finding indicated that ACh was involved in descending modulation of SmI. (2) The effect of acupuncture was decreased after intraperitoneal-injecting atropine (5mg/kg), while it was not decreased if saline was administered. The difference of nociceptive response was statistically significant at 0' to 10' and 20' to 40' after cessation of EA in comparison between the background of atropine and saline. It indicated that ACh was involved in acupuncture analgesia. These results suggest that ACh may be participated in acupuncture analgesia as a neurotransmitter of the corticofugal modulation of pain.

A new Ph1+, bcr cell line derived from a patient with ALL-L1 gained autonomy in culture concomitant to CD23 expression.

A permanent cell line, LEF1, has been established from the cells of an adult suffering from a Philadelphia positive acute lymphoblastic leukemia. The LEF1 cell line was obtained by maintaining peripheral blood cells from the patient in culture on a fibroblast feeder; subsequently, an autonomously growing cell population, independent of that feeder layer, developed. The karyotype of the cell line, 46, t(9;22)(q34;q11), was different from the karyotype at diagnosis which had 53 chromosomes and two Philadelphia chromosomes. Furthermore, compared with the initial leukemic blasts, the immortalized cell had three differences in surface phenotype (CD23+, CD11b-, CD10-). However, molecular studies indicated that the breakpoint in the 3' part of the first intron of the BCR gene was unchanged, confirming the leukemic origin of LEF1. The cell line was shown to be Epstein-Barr virus negative.

[The status of negative charge of the RBC and glomeruli and its relation to proteinuria in adriamycin nephrosis in rats].

The status of negative charge on RBC and glomeruli and its relation to proteinuria of adriamycin nephrosis in rats were studied by electron computerized picture analysis. The results showed that the negative charge (NC) on glomeruli decreased significantly at 48 hrs and 14 days after ADR injection although there was no proteinuria simultaneously. There was a positive relation between the changes of NC on glomeruli and those on RBC (r = 0.77, p less than 0.01), and there was negative relation between either of them and proteinuria respectively (r = -0.5140, p less than 0.05; r = -0.5231, p less than 0.01). The focal fusions of foot processes occurred prior to the onset of proteinuria.

[Mechanism of the antiasthmatic effect of total coumarins in the fruit of Cnidium monnieri (L.)Cuss].

Total coumarins in the fruit of Cnidium monnieri show a marked protective effect against bronchial asthma induced by inhalation of histamine in conscious guinea pig. In vitro these total coumarins can relax the contraction of the isolated trachea in guinea pigs induced by histamine as well as increase the perfusion rate of isolated lung preparation of guinea pig. These actions, however, may be blocked by propranolol. Besides, these coumarins cannot increase the heart rate and arterial blood pressure in anesthetized rats. All this shows that the antiasthmatic effect of total coumarins in the fruit of Cnidium monnieri is mediated by beta 2-receptor.

[Clinical study of 96 cases with chronic hepatitis B treated with jiedu yanggan gao by a double-blind method].

This paper reported 96 cases with chronic hepatitis B treated by a double-blind method. There were 51 cases of observation group(OG) and 45 cases of control group (CG). OG was treated with Jiedu Yanggan Gao consisting of Artemisia capillaris, Taraxacum mongolicum, Plantago seed, Cephalanoplos segetum, Hedyotis diffusa, Flos Chrysanthemi Indici, Smilax glabra, Astragalus membranaceus, Salviae miltiorrhizae, Fructus Polygonii Orientalis, Radix Paeoniae Alba, Polygonatum sibiricum, etc.). CG was prescribed with three charred medicinal herbs (charred Fructus Crataegi, charred Fructrus Hordei Germinatus, charred fermented mixture of several medical herbs and wheat bran). The average duration of treatment was five months. All 96 cases belong to the virus-duplication-type with positive HBsAg for over one year. Among them 65.5% of cases HBeAg, DNAP and HBV-DNA were positive. 20.8% of cases were positive in two out of the above tests. 13 data were compared statistically between two groups, and proved to be comparable (P greater than 0.05) before treatment. 27.3% and 66.7% of cases' ALT, AST returned to normal respectively in OG after treatment. However, in CG they were 9.1% and 22.2% (P less than 0.05). TTT returned to normal in 52% cases of OG and 44% in CG (P greater than 0.05). 20% cases HBeAg shifted to negative in OG, but 6.7% in CG. Cases with negative DNAP in OG occupied 34.2%, but 10.8% in CG. 31.6% cases' HBV-DNA changed to negative in OG, while 17.6% in CG. After comprehensive judgement, the total effective rate was 74.5% in OG and 24.4% in CG respectively (P less than 0.001). Eight cases were basically cured in OG and one case in CG. After one year's follow-up, one recurred in eight patients of OG, however the only one cured in CG still relapsed.

[Investigation on role of nucleus parabrachialis medialis in "renzhong" induced respiratory response in rabbits].

Experiments were performed on 27 rabbits, bivagotomized, gallamine triethiodide immobilized and artificially ventilated. The effects of electric stimulation of "Renzhong" (Du26) area, NPBM separately, and "Renzhong" area and nucleus parabrachialis medialis (NPBM) simultaneously, on phrenic nerve discharge were observed before and after electolytic and chemical lesion of NPBM. Evoked potential (EP) could be recorded in NPBM during the electric stimulation "Renzhong" area. Electric stimulation of the same site of NPBM where the EP was recorded could induce respiratory response and change the effect of electric stimulation of "Renzhong" area on respiration. But after electrolytic and chemical lesion of NPBM, stimulation of NPBM could no longer induce the respiratory response mentioned above and could not change the "Renzhong" effect on respiration either. These results suggest that NPBM might take part in respiratory effects of "Renzhong" area stimulation, but is not essential to it.

Transfer function analysis of the ventricular function: a new method for calculating right ventricular ejection fraction.

The relationship between the ventricular transfer function and ejection fraction has been investigated by the routine procedure of first-pass radionuclide angiocardiography. Ejection fraction has been shown to equal 1-e(-b), where b is the ratio of the R-R interval over the mean transit time difference between ventricular and atrial time-activity curves. To evaluate the effect of region of interest (ROI) on the right ventricular ejection fraction (RVEF), the results of the transfer function analysis (TFA) technique using precise ROI, TFA using rectangular ROI, and the routine method were compared. Regression analyses among RVEFs obtained from the above ROI methods yielded good correlations. Reliable RVEFs have been obtained even in the case of an improper bolus injection. Thus, the TFA technique is a new, simple, and reliable method for calculating RVEF without needing to outline the right ventricle precisely.

[Effects of concentration and ratio of N, P and K on seedlings of Panax quinquefolium L].

Results of the orthogonal experiment of N, P and K are as follows: (1) The highest growth rate occurs in treatment 4 (N100ppm, P25ppm, K250ppm) and other features are also good. So this treatment can be used in hydroponics of seedling of P. quinquefolium. (2) The influences of N, P and K concentration on the contents of ginsenosides in different parts of seedling are different. The content of ginsenosides in aerial part is 14.72-16.72%, higher than 3.24-4.00% in root.

[Erythrocyte deformability and extracorporeal thrombosis in patients with blood stasis syndrome].

The patients were divided into 3 groups: 127 cases of cor pulmonale, 47 cases of glomerulonephritis and 17 cases of coronary heart disease. Erythrocyte deformability indices (DI) and changes during extracorporeal thrombosis were investigated in all the 3 groups, in comparison with a group of 80 normal healthy individuals. The result showed that all the patients had notably lower DI (respectively 1.21 +/- 0.99, 0.73 +/- 0.93), and 0.84 +/- 0.81 for the groups in the given order), longer length of extracorporeal thrombi (L in mm, respectively 45.94 +/- 38.78, 62.88 +/- 43.65 and 59.74 +/- 40.99) and larger top inclination angle of blood plane in silicon-lined ring tube (respectively 15.66 degrees +/- 6.09 degrees, 13.21 degrees +/- 9.33 degrees and 12.65 degrees +/- 8.02 degrees) than individuals over 40 in the comparison group. In the group of cases of cor pulmonale, patients with abnormal pH and respiratory failure had apparent greater L than those with pH compensation and without respiratory failure; diminished DI were also observed in association with pH abnormality. In the group of nephritis, patients with renal failure and chronic nephritis showed evidently lower DI than those without renal failure but acute nephritis; increased L were also noted in association with renal failure. In addition, a similar comparison was done between individuals over 40 and individuals at or younger than the age of 40. It was found that people over 40 tended to have diminished DI, increased L and Wt of extracorporeal thrombi, and augmented initial inclination angle of blood plane though not as notably as what was observed in the 3 groups of victims of blood stasis.(ABSTRACT TRUNCATED AT 250 WORDS)

Analysis of junctional diversity in the preferential V delta 1-J delta 1 rearrangement of fresh T-acute lymphoblastic leukemia cells by in vitro gene amplification and direct sequencing.

To define the junctional diversity of T-cell antigen receptor delta gene rearrangements in fresh T-acute lymphoblastic cells and to correlate cell phenotype with the coding potential of rearrangements, we determined the junctional nucleotide sequences of 13 T-cell antigen receptor delta gene rearrangements involving the preferentially rearranged V (V delta 1) and J (J delta 1) segments using in vitro gene amplification and direct sequencing. We showed that, as in gamma delta+ cell lines, extensive junctional diversity exists in these clones and that this diversity is due both to random nucleotide deletions/additions and to the use of at least two D delta segments. We also showed that a high percentage of these rearrangements are potentially translatable (7:13) and that such functional rearrangements occur in both surface CD3+ and CD3- cells. Comparison of alpha beta versus gamma delta surface expression demonstrates that all CD3+ T acute lymphoblastic leukemias with a functional V delta 1-J delta 1 rearrangement express a surface gamma delta receptor and are recognized by the anti-delta monoclonal antibody delta TCS1, whereas a control CD3+ gamma delta+ leukemic case that had not undergone V delta 1 rearrangement was delta TCS1-. In addition, expression of this monoclonal antibody is not restricted by V gamma or C gamma usage or by the covalent or noncovalent link between gamma and delta chains.

Structural alterations of the BCR and ABL genes in Ph1 positive acute leukemias with rearrangements in the BCR gene first intron: further evidence implicating Alu sequences in the chromosome translocation.

In the Philadelphia positive bcr negative acute leukemias (Ph1+bcr- AL), the chromosomal breakpoints on chromosome 22 have been shown clustered within 10.8kb (bcr2) and 5kb (bcr3) fragments of the first intron of the BCR gene. We previously reported that the breakpoints were localized in Alu repeats on chromosomes 9 and 22 in a Ph1+bcr- acute lymphoblastic leukemia with a rearrangement involving bcr2. Molecular data of two other Ph1 translocations, one a Ph1+bcr- acute myeloblastic leukemia in the bcr2 region, and the other an acute lymphoblastic leukemia in the bcr3 region are presented. In the former, the breakpoints on chromosomes 9 and 22 are localized in Alu repeats, in regions with two inverted Alu sequences, as in our previously reported case. In the second leukemia, the breakpoints are not located in Alu sequences, but such repeats are found in their vicinity. The implications of these findings are discussed.

The rat nucleus paragigantocellularis as a relay station to mediate peripherally induced central effects of nicotine.

Previous electrophysiological studies have shown that intravenously administered nicotine in low doses indirectly excites noradrenergic locus coeruleus (LC) neurons. In the present study this effect of nicotine was attenuated (to about 25% of control response) by injection of lidocaine into the nucleus paragigantocellularis (PGi) whereas injection into the nucleus prepositus hypoglossi (PrH) had no effect in this regard. Whereas the basal firing rate of LC neurons was not affected by lidocaine injection into the PGi, more than 50% of the LC neurons displayed increased firing rate following lidocaine injection into the PrH. It is suggested that the peripherally induced effect of nicotine on LC neurons is indirectly mediated via activation of the PGi.

Rearrangements of the T cell receptor delta gene in T acute lymphoblastic leukemia cells are distinct from those occurring in B lineage acute lymphoblastic leukemia and preferentially involve one V delta gene segment.

Rearrangement of the TCR-delta gene was studied using J delta, C delta, and V delta probes in 61 cases of acute lymphoblastic leukemia (ALL) and several cases of chronic lymphoid neoplasms to define the specificity and the diversity of rearrangements occurring at the delta locus. TCR-delta rearrangements or deletions were found in all T (33 cases) and B lineage (28 cases) ALL but not in any case of B cell chronic proliferations (13 cases). The restriction patterns of rearrangement were clearly distinct between T and B ALL and use of one V delta probe showed that rearrangement of the V delta IDP2 gene segment which is also productively rearranged in the Peer cell line, occurred frequently in T-ALL but never in B lineage ALL. Studies of WT31 and delta TCS1 antibody reactivity showed that at least 4 of 13 CD3+ T-ALL cases expressed the delta protein. CD4 and/or CD8 Ag expression were observed in some of the gamma delta expressing T-ALL. These data show that particular TCR-delta gene rearrangements occur in neoplastic early B cells and that the combinatorial diversity of TCR-delta rearrangements in T cells is higher than initially expected. In addition this study shows that an important proportion of CD3 positive T-ALL cases express the gamma delta heterodimer.
