Respiratory-associated rhythmic firing of midbrain neurones in cats: relation to level of respiratory drive.

1. We recorded phrenic nerve activities and single unit firing of mesencephalic neurones in unanaesthetized supracollicularly decerebrated, paralysed and ventilated cats, in which vagi and carotid sinus nerves had been ablated. We made these measurements first at low levels of respiratory drive associated with normal PCO2 levels, then with increased respiratory drive and levels of phrenic activity produced by hypercapnia or by carotid sinus nerve stimulation. 2. We found that at least a quarter of the neurones in the central tegmental field of the mesencephalon, which were irregularly tonic or silent at low respiratory drives, developed a rhythmic increase of firing associated with each respiration. There appeared to be a threshold at about 50% of maximum respiratory activity, below which the respiratory-associated rhythm did not occur. Above this level, neuronal firing increased in graded fashion with increasing magnitude of respiratory activity. The latency from onset of phrenic activity to onset of increased neuronal firing was quite long (1.0 s) at drives just above the threshold but shortened to as little as 0.3 s as drive increased towards its maximum. 3. Cutting the spinal cord at C1-C2 had no effect on the ability of increased respiratory activity to generate a respiratory-associated rhythm in mesencephalic neurones. 4. Short-lasting anaesthesia with the agent Saffan caused mesencephalic neurones to lose the respiratory-associated rhythm with little change in phrenic activity and no change in respiratory cycle timing. 5. We also found a mesencephalic response to ventilator-induced chest expansion. The latency of the response from onset of expansion, indexed by fall of airway PCO2, to onset of neurone firing was shorter (0.2 s) than that found with the respiratory-associated rhythm. In seventeen neurones we found both the respiratory-associated rhythm and the independent ventilator-associated rhythm. 6. We interpret our findings to show that the respiratory-associated rhythmic firing of midbrain neurones is not primarily involved in generation or modulation of the motor function of the respiratory oscillator. We believe, instead, that these neurones are part of a sensory pathway conveying information about the magnitude of central neural respiratory drive, as well as spinally transmitted information from receptors in the chest wall, to thalamus and cortex. We suggest that the sensation ultimately generated may be that of 'air hunger' or dyspnoea.

Sequence requirements for transcriptional arrest in exon 1 of the human adenosine deaminase gene.

We have previously demonstrated that a transcriptional arrest site exists in exon 1 of the human adenosine deaminase (ADA) gene and that this site may play a role in ADA gene expression (Z. Chen, M. L. Harless, D. A. Wright, and R. E. Kellems, Mol. Cell. Biol. 10:4555-4564, 1990). Sequences involved in this process are not known precisely. To further define the template requirements for transcriptional arrest within exon 1 of the human ADA gene, various ADA templates were constructed and their abilities to confer transcriptional arrest were determined following injection into Xenopus oocytes. The exon 1 transcriptional arrest signal functioned downstream of several RNA polymerase II promoters and an RNA polymerase III promoter, implying that the transcriptional arrest site in exon 1 of the ADA gene is promoter independent. We identified a 43-bp DNA fragment which functions as a transcriptional arrest signal. Additional studies showed that the transcriptional arrest site functioned only in the naturally occurring orientation. Therefore, we have identified a 43-bp DNA fragment which functions as a transcriptional arrest signal in an orientation-dependent and promoter-independent manner. On the basis of our findings, we hypothesize that tissue-specific expression of the ADA gene is governed by factors that function as antiterminators to promote transcriptional readthrough of the exon 1 transcriptional arrest site.

Hepatitis B virus prevalence in industrialized cities in China.

The prevalence of HBV infection was investigated by radioimmunoassay for HBsAg, anti-HBs and anti-HBc in over 1000 workers in large scale factories located in four industrial cities (Jinxi, Shanghai, Wuxi and Xian) in 1987 to 1990. The age dependency of the prevalence was not evident. The overall prevalence rate of those positive for any of the three markers was 62.6%. The rate was significantly (p < 0.01) higher than the rates found in Beijing in a previous study but lower than the values observed in earlier studies.

Subjective symptom increase among dry-cleaning workers exposed to tetrachloroethylene vapor.

Subjective symptoms, hematology, serum biochemistry and other clinical signs were investigated in 56 dry-cleaning workers exposed to tetrachloroethylene at 20 ppm (as a geometric mean of 8-hr time-weighted average), and the results were compared with the findings in 69 non-exposed controls from the same factories. There were exposure-related increases in the prevalence of subjective symptoms during the work as well as in the past 3 month period, whereas there was no significant changes in hematology. Effects of the exposure on liver and kidney functions were also negative as judged by emission enzyme activities, BUN and creatinine in the serum.

A serological study of hepatitis C infection in plasmapheresis donors.

An epidemic of parenterally transmitted non-A, non-B hepatitis (PT-NANBH) occurred in plasmapheresis donors in Guan County, Hebei Province, China, in 1985. PT-NANBH was diagnosed by epidemiological studies and serological exclusion of HAV, HBV, CMV and EBV infections. Recently, 163 sera samples of 108 patients with PT-NANBH and 65 sera samples of 49 cases with elevated alanine aminotransferase (ALT) levels collected during the epidemic were tested by anti-HCV EIA (Chiron C100). The positive rates of anti-HCV in these two groups were 89.8% (97/108) and 93.9% (46/49), averaging 90.8%. The figures increased with the course of illness and persistance of ALT elevation, i.e., 17.6% and 55.6% within 1 month, 88.9% and 87.5% at 6 months and 100% and 100% after 2 years. Five patients with PT-NANBH and 1 with elevated ALT levels were followed up for 3 to 4 years. We demonstrated that anti-HCV remained positive after the disease had resolved and ALT levels had normalized.

[Clinical and consequence study on 51 cases of hepatitis C].

All the 51 patients had history of plasma donation before onset of the illness. All had no evidences of recent infection with following viruses: HAV, HBV, CMV and EBV and were confirmed recently as hepatitis C by the Chiron C-100 EIA test. The incubation period was 35 to 90 days, with an average of 57.9 +/- 19.8 days. The ratio of icteric to anicteric was 1.0:1.55. Only a few cases had fever in the acute phase (13.7%). 36 cases showed acute onset of illness; 15 cases, however, had ALT elevation before the onset. 33 cases became chronic hepatitis after 1 year (64.7%), 14 out of the 33 cases had persistently abnormal ALT and 19 cases showed repeated elevation of ALT. The chronicity rate in 2 years was 38.6%. These results indicate that hepatitis C has the features of high chronicity rate and prolonged clinical course.

[ELISA for detection of anti-urease antibodies of Campylobacter pylori and its application].

The urease extracted from 6 strains of Helicobacter pylori (HP) was used as antigen to detect sera anti-urease antibodies of HP in 136 patients with gastric diseases and 13 persons with normal gastric mucosa by enzyme-linked immunosorbent assay (ELISA). The antigen proved to be pure and specific for ELISA. For a titre of 450EU or over 450EU the test had a sensitivity of 91.96% and a specificity of 86.49%. The results of the ELISA showed excellent correlation with microbiological detection of HP and histological examination of the antrum. The level of sera antibody showed a relation to the bacterial amount of HP in gastric mucosa, but no connection with the severity of gastritis. While ELISA may be effective for the serodiagnosis of HP infection and is able to distinguish patients with normal antrum mucosa from those with gastritis. The test does not help to distinguish those with antrum gastritis from those with peptic ulcer. It also provides a reliable method for epidemiological investigation of HP infection.

Epidemiologic investigation on an outbreak of hepatitis C.

An outbreak of hepatitis in plasma donors occurred in a village of Hebei Province in the period of September to October 1985. The morbidity rate was 40.0% (26/65) in the plasma donors, which was significantly higher than that in whole blood donors (1/88) and in persons who were not blood donors (1/400). One to one paired survey was carried out, and the incidence was 46.4% (26/56) in the plasma donors, while there were no such outbreak in the control group. The distribution of cases showed positive correlation with the number of plasma donors from the production brigade. No secondary infection was found in their families. The peak of outbreak was about 2 months later than the peak of plasma donation. 26 cases of hepatitis in plasma donors all showed negative results for anti-HAV IgM, HBsAg, anti-HBC IgM, anti-CMV IgM and anti-EBV IgM. Sera of 25 cases were selected and sent to CDC, USA to confirm with Chiron C100 reagent. 24 cases were anti-HCV positive. This outbreak of hepatitis was demonstrated to be related to cross contamination during plasma donation.

Vaginal reconstruction using perineal-thigh flaps with subcutaneous pedicle.

A technique of vaginal reconstruction using bilateral, perineal-thigh flaps with subcutaneous pedicle is described. In this procedure, the flaps were raised bilaterally and introduced into an artificial space between the urinary bladder and rectum. The blood supply for the flaps flows from the perineal artery through anastomotic branches to the external pudendal artery. The authors used the technique on four patients, and all the flaps survived entirely. There was no complication. According to a more than two-year follow-up survey, the reconstructed vaginas are expansible and contract little. No stent is needed. There is good sensitivity in the wall of the artificial vagina because sensory nerves run through the flaps.

Establishment and preliminary application of spermidine radioimmunoassay with 125I-labelled monoclonal antibody and solid phase antigen.

Purified anti-spermidine monoclonal antibody was labelled with radioactive iodine by the Iodogen method and spermidine-bovine serum albumin (SPD-BSA) conjugate was used to coat polystyrene beads as solid phase antigen. The new solid phase 125I-labelled spermidine radioimmunoassay (RIA) depends on the competition between spermidine in the sample and the solid phase antigen for the limited amount of 125I-labelled monoclonal antibody. The sensitivity of this assay was 10 ng/ml higher than that of liquid RIA for spermidine with 14C-labelled spermidine. The coefficients of variation (CV) within and among batches were 4% and 13% respectively. The sample-batch capacity was increased from 20 (liquid RIA with 14C-labelled spermidine) to 150-200 by using this method. Because of its simplicity, the solid phase RIA kit is very convenient for population survey. This RIA could be used to determine spermidine in saliva for the diagnosis of precancerous lesions. In a preliminary study saliva spermidine levels in different populations were measured among 130 normal subjects, 202 esophageal epithelial hyperplasia cases treated with anti-tumor B for 5 years, 207 esophageal epithelial hyperplasia cases as control, and 55 esophageal cancer patients. The levels were 1,795 +/- 1,481, 3,470 +/- 6,981, 9,753 +/- 17,641 and 18,090 +/- 21,509 ng/ml, respectively, with the saliva spermidine levels in precancerous and cancer patients being significantly higher than that of normal subjects (P less than 0.001); the level in patients treated with anti-tumor B was significantly lower than that of controls (P less than 0.001). This decreased saliva spermidine content was coincident with the 47.3% reduction of canceration rate seen in precancerous patients after a 5-year treatment with anti-tumor B.

[Role of nucleus parabrachialis medialis in the "renzhong" induced pressor response in rabbits].

Experiments were performed on 18 rabbits, bivagotomized, gallamine triethiodide immobilized and artificially ventilated. The effects of electrical stimulation of "renzhong" (Du 26) area and NPBM, separately and simultaneously, on arterial blood pressure and heart rate were observed before and after electrolytic or chemical lesion of NPBM. Electrical stimulation of "renzhong" area or NPBM, separately could elicit an increase in arterial blood pressure; when both "renzhong" and NPBM were stimulated, the pressor response to NPBM stimulation could affect that of "renzhong" area's. But after electrolytic or chemical lesion of NPBM, stimulation of NPBM could no longer elicit pressor response. On the other hand, the pressor response induced by electrical stimulation of "renzhong" area decreased obviously, after lesion of NPBM or transection of brain stem between NPBM and Nucleus trigemini principalis. These results suggest that NPBM might take part in the pressor response of "renzhong" area stimulation.

[The changes of viscoelastic properties of whole blood in patients with chronic cor pulmonale].

The changes of blood viscoelasticity and apparent viscosities at low shear rate (0.512 S-1) and high shear rate (51.2 S-1) were studied in 61 patients with various severity of chronic cor pulmonale (CCP). The relation between blood viscoelasticity and apparent viscosities and PaO2 and PaCO2 was investigated and the data were collected before and after treatment. Grouped data: Group 1. A total of 61 patients with CCP were studied, 10 of them died during hospitalization. Sixty-one patients at the acute stage were divided into mild, severe and dead sub groups. Twenty-nine patients were improved after treatment. Group 2. Control group (74 normal people) matched group 1 in sex and age. The results demonstrated that eta 0.512, eta 51.2 and eta' were slightly increased in the mild group, but HCT, eta 0.512, eta 51.2, AI, eta' and eta" were elevated more significantly in the severe group. All variables mentioned above of died patients were elevated more remarked than that of the mild group. HCT, eta" in dead patients were slightly higher than those in severe group (P less than 0.05-0.01). It was clear that the more severe the patient, the more increased the hematocrit and RBC aggregation, the less the RBC deformability. HCT would be elevated when PaO2 was below 8 kPa (60 mmHg) and the HCT, eta 0.512, eta', eta" increased significantly (P less than 0.05-0.01) when PaCO2 was over 6.65 kPa (50 mmHg). There was a positive linear correlation between eta 0.512 and PaCO2.(ABSTRACT TRUNCATED AT 250 WORDS)

Specific binding and internalization of monoclonal antibody HI98-daunorubicin conjugate by human leukemic cells (HL60).

McAb-HI98 has been proved to bind specifically to HL60 cells. In this study, we further observed the specific binding of HI98-Daunorubicin (DNR) conjugate to HL60 cells, the transmembranal transport of HI98-DNR and the process of internalization by immunoelectron microscopy. After HL60 cells were incubated with HI98-DNR-Au, gold particles were first found on the surface membranes of HL60 cells, and then inside the cells. Internalization was found to occur through the formation of endocytic vesicles. Results indicate that the immunoconjugate selectively binds to target cells and enters the cells specifically and rapidly, thus enhancing intracellular drug concentration.

[Preliminary study on soilless cultivation techniques of Panax quinquefolium L].

Our experiment has shown 1. Vermiculite with sand (volume 1:1) is a good substrate of soil for American Ginseng. The yield and contents of ginsenoside of American ginseng in this soilless substrate are a little higher than those in ordinary soil. 2. This kind of substrate can be used continuously with fewer occurrences of insect pest and plant diseases and contributes significantly to the reduction of production cost.

The therapeutic and biological effects of total lymph node irradiation and autologous bone marrow infusion on malignant lymphoma patients.

Forty patients with malignant lymphoma were treated by 60Co total lymphoid irradiation (TLI): 21 cases received 6 Gy and 19 received 8 Gy. Ten also received autologous bone marrow infusion (ABMI). Acute radiation damage with digestive tract reaction and hemopoietic and immunological depression was observed. Bone marrow was depressed. WBC and platelets decreased rapidly. Lymphocytes showed quantitative and qualitative changes even at the early stage. All these symptoms subsided within 40 days. TLI accompanied by irradiation of the tumor site could result in effective control. The 1-, 3-, and 5-year survival rates of malignant lymphoma patients were 30/40 (75%), 14/24 (58%) and 4/12 (33%), respectively, while in those with Hodgkin's disease alone, the 1- and 3-year survival rates were 10/13 (76%) and 5/7 (71.4%), respectively. ABMI hastened hemopoietic reconstitution, which recovered relatively quickly after TLI.
