Clinical observation on 80 children with peptic ulcer treated primarily by traditional Chinese medicine.

Using the empirical recipe of Wang Pengfei (a very experienced TCM doctor) as the main treatment, the authors treated 80 children with peptic ulcer by means of promoting blood circulation, removing blood stasis as well as regulating the flow of qi, alleviating pain, tonifying the stomach and reinforcing the spleen. Measures for hemostasis, correction of anemia and other general treatment were also taken. After 8 weeks of treatment, a total effective rate of 92.5% was obtained.

Endogenous porphyrins in murine skin and transplanted PAM-212 squamous cell carcinoma tissues after injection of delta-aminolevulinic acid.

After intraperitoneal (IP) injection of delta-aminolevulinic acid (ALA), the endogenous porphyrins in murine skin and tumor tissues were determined by a method involving solvent and acid extractions. The results showed that the total amount of porphyrins in the tumor tissues after ALA injection was much higher than that in the skin from the same mice, although the amount of porphyrins in the skin from the ALA-injected mice was higher than that from the saline-injected (control) mice. The porphyrins in the tumor were mostly protoporphyrin and coproporphyrin, with only a small amount of uroporphyrin. The optimum period for porphyrin accumulation in the tumor as well as in the skin was 1 hour after the injection of ALA. As the period was extended to 3 and 6 hours, the amount of porphyrins in these tissues decreased considerably. These findings could be valuable for further application of ALA in the photodynamic therapy of skin cancer.

Beneficial effects of NTG on left ventricular volumes and systolic function in patients with myocardial infarction.

The effect of sublingual administration and intravenous drip of nitroglycerin (SLNTG and IVNTG) on left ventricular volumes (LVVs) were studied and systolic function was quantitatively assessed with two-dimensional echocardiography (2DE) in 28 patients with myocardial infarction (MI), including 13 cases without heart failure (MI-NHF) in one group and 15 with heart failure (MI-HF) in another. The results showed that after SLNTG and IVNTG in both groups the heart rate (HR) increased significantly (P < 0.05-0.001); the systolic blood pressure (SBP) decreased significantly (P < 0.05-0.001); the diastolic blood pressure (DBP) was not significantly changed (all P > 0.05); the left ventricular end-diastolic and end-systolic volumes (EDV and ESV) both significantly reduced [EDV: decreased by 5.8-11.6% (-13.2-19.0 ml), P < 0.05-0.001; ESV: decreased by 9.6-16.6% (-8.6-17.3 ml), P < 0.05-0.001)]; the left ventricular ejection fraction (LVEF) all significantly elevated (increased by 6.6%-9.4% (3.2%-3.4%), P < 0.05-0.001] except in MI-NHF group after IVNTG; the segmental EF of normal and hypokinetic segments also significantly increased (P < 0.05-0.001). The results suggest that NTG could beneficially reduce left ventricular EDV and ESV; and improve or strengthen its global and segmental systolic function in MI patients, particularly noticeable in patients with heart failure.

[Effects of lead on neurobehavior and neurochemistry in rats].

Effects of exposure to lead on neurobehavioral function, content of monoamine neurotransmitter in hippocampus, and brain level of lipid peroxide were studied in 45 SD rats. Results showed learning and memory abilities of the rats were affected in exposure to lead of 1333.3 mg/kg (with their blood lead level of 6.23 mumol/L), their motor activities changed and emotional status became unsteady with 266.7 mg/kg of lead (with blood lead of 4.20 mumol/L) and 1333.3 mg/kg of lead, respectively, and levels of dopamine neurotransmitter and its metabolites declined and those of lipid peroxide increased with increasing exposure to lead. It is postulated changes in levels of neurotransmitter, such as monoamine and lipid peroxide, could explain the mechanism for damage in neurobehavioral function caused by lead exposure.

Effect of retinoic acid isomers on proliferation, differentiation and PML relocalization in the APL cell line NB4.

Retinoic acids exert a wide physiological role in development and differentiation. Retinoic acids have also been used in the treatment of human cancers, particularly in acute promyelocytic leukemia (APL). A structure-function relationship of the RA isomers in terms of clinical effect has been observed since all-trans retinoic acid (ATRA) induces a high complete remission rate while 13-cis retinoic acid (13-cis RA) shows much poorer effect. In this study, we examined the effect of RA isomers, including ATRA, 13-cis RA and 9-cis RA, on the proliferation and differentiation of NB4 cells. A number of parameters such as cell growth curve, dynamics of cell cycle, expression of clusters of differentiation and reduction of nitro blue tetrazolium (NBT) as well as immunofluorescence staining of PML were used to evaluate the effects of three isomers at two concentrations (10(-8) M and 10(-7) M). It has been shown that during the first 48 h of RA treatment, the APL cell differentiation was coupled with the cell proliferation. Although similar effects of proliferation inhibition and differentiation induction were observed among the three isomers at 10(-7) M, significant differences appeared at a concentration of 10(-8) M, 9-cis RA showed a higher activity than that of ATRA, while ATRA showed better results than 13-cis RA. Our results provide further evidence that 9-cis RA could be a promising molecule in differentiation induction of malignant cells.

Differential expression of human tissue factor in normal mammary epithelial cells and in carcinomas.

BACKGROUND: Tissue factor (TF) is a glycoprotein which binds factor VIIa. The TF-VIIa complex serves as a potent initiator of the coagulation pathways. TF, an immediate early gene, may also play a role in cell growth. Expression of TF was correlated with some types of cancers. MATERIALS AND METHODS: Normal, immortalized, and tumor human mammary epithelial cells were used in the experiments. The differential display (DD) technique was used to identify genes differentially expressed in the cells. TF expression patterns were examined by Northern blot analysis, immunofluorescence staining of cultured cells, and immunohistochemical staining in human cryostat sections. RESULTS: In a 5-way display, an amplified polymerase chain reaction (PCR) product was found in normal and immortalized human mammary epithelial cells but not in the breast cancer cells. The PCR fragment was cloned and sequenced. The result showed that the fragment was identical to human tissue factor. Northern blot analysis showed that expression level of tissue factor mRNA remained high in growing, quiescent, and senescent normal mammary epithelial cells. Immunofluorescence staining also confirmed tissue factor expression pattern in the cell lines tested. Immunohistochemical staining showed that tissue factor was expressed in the normal luminal and myoepithelial cells of some ducts but not others. No staining was observed in invasive carcinoma cells. However, myoepithelial cell staining was seen in some residual ductal structures in invasive tumors. CONCLUSIONS: This study shows the use of DD to reveal the loss of TF expression pattern in human breast cancer cell lines. Immunohistochemical staining results showed breast carcinoma cells expressed little TF, if any, suggesting that TF is not required for breast tumor cell invasion. The results also indicated that TF expression was independent of the proliferation status of the expressing cells. The expression pattern of TF may be a meaningful marker in the development of breast cancer.

Effectiveness of three ribozymes for cleavage of an RNA transcript from human papillomavirus type 18.

We tested three hammerhead ribozymes for their ability to bind and cleave RNA transcripts derived from the E6 and E7 genes of human papillomavirus (HPV) type-18. Targets were located at nucleotides (nt) 123, 309, and 671 of the viral transcript. In vitro each ribozyme hybridized to its target site when the ribozyme:target ratio was 20:1 or greater and achieved maximal hybridization within 1 hour. HPV RNA from the HeLa cervical cancer cell line was cleaved effectively by each ribozyme. When HPV RNA and a ribozyme were expressed simultaneously in Escherichia coli, each ribozyme produced a significant reduction in the intracellular concentration of HPV RNA. In each assay the ribozyme directed to nt 309 was the most effective. A noncatalytic antisense molecule was used as a control and did not digest HPV RNA or reduce its concentration. The data imply that three different ribozymes each have potential for use in gene therapy of human tumors that express HPV-18 but that the ribozyme targeted to nt 309 is likely to be most effective.

[Characteristics and recent trends in endemicity of human parasitic diseases in China].

An analysis of the nationwide survey of the distribution of human parasites in the light of the literature previously published, two major trends in the endemicity of parasitic diseases in China was observed. For one trend, the infection rate of E. histolytica, Fasciolopsis buski and soil-transmitted helminthasis etc. declined markedly. The second trend was that the prevalence of food-transmitted parasitic diseases (Trichinella spiralis, Clonorchis sinensis, Paragonimiasis, cysticercosis) and hydatidosis was increasing. The relationship between the social economic factors and the two trends were discussed.

Preoperative segmental localization of focal hepatic lesion on MRI.

In order to evaluate the accuracy of preoperative segmental localization of hepatic mass lesions, 68 cases were prospectively studied and evaluated by the correlation of magnetic resonance imaging (MRI) and surgical findings. Right, middle and left hepatic veins were seen in 100%, 97% and 94% of the subjects respectively on MRI. The right and left portal veins were seen in 100% and 95.6% respectively. The accuracy in determining the segmental location of hepatic lesions was 89.7%. The accuracy could be increased to 91.3% if the lesion was located at single segment whereas it was only 86.4% when the lesion extended across several segments. It is possible to say that MRI is an important modality in the preoperative segmental localization of masses.

[Observations of blood circulation disturbance in random skin flap].

Thirty-nine rats were used to study the mechanism of random flap necrosis. A random flap (3 cm x 9 cm) created on the back of each rat was observed with naked eye, microscope, skin thermometer and laser Doppler flowmeter. The average surviving length of the flap was 4.69 +/- 0.47 cm. We found that the following observations indicated circulation disturbance in the "shock flap": After surgery, the color of the flap became dark; skin temperature kept falling; when the room temperature was about 20 degrees C, the flap temperature was below 28.5-29.0 degrees C; at one hour postoperation, LDF value decreased more than 50%; microcirculation of the flap stagnated without recovery. We conclude that necrosis of the flap can be fully predicted.

[Effects of low dose aspirin on platelet function and prostaglandins metabolism in systemic and coronary circulation in patients with ischemia heart disease].

Eleven patients with ischemia heart disease (IHD) were treated with low dose aspirin (ASA, 50mg/day) for more than two weeks (ASA group). 29 cases with IHD not taking ASA served as patient control (NASA group) and 13 cases without IHD not taking ASA as normal control. Blood samples for measurement of plasma (serum) TXB2 and 6-keto-PGF1 alpha were simultaneously taken from aortic root (AO) and coronary sinus (CS). The results showed: ASA group had lower plasma TXB2 level in AO blood than NASA group (P < 0.05), but there was no significant difference in plasma 6-keto-PGF1 alpha level between the two groups. Both of plasma and serum TXB2/6-keto-PGF1 alpha ratios in AO blood in ASA group were significantly lower than those in NASA group (P < 0.05 and P < 0.0005 respectively). Plasma TXB2 CS/AO ratio and 6-keto-PGF1 alpha CS/AO ratio in ASA group were significantly lower than those in NASA group (P < 0.05), but not different from those in control group. Both ASA and NASA groups had lower serum TXB2 CS/AO ratios than control group (P < 0.05). The results suggest that low dose aspirin inhibits selectively TXA2 synthesis in systemic circulation and inhibits synthesis and/or release of TXA2 and PGI2 equally (no selectivity) in coronary circulation, but could not completely inhibit intracoronary platelet activation.

[Preparation of 125I-F(ab')2-DNR-liposomes and radioimmunoassay of their specific conjugating ability].

The F(ab')2-DNR-liposomes were prepared using F(ab')2 obtained from digested monoclonal antibody against human white cells HI30 (CD45). Their affinity constant for peripheral blood mononuclear cells was determined by radioimmunoassay. Ka = 2.0 x 10(9). A good specific conjugation ability was demonstrated. In preparing the immunoliposomes, it was found that F(ab')2 coupled to the smaller liposomes with higher conjugation ratio.
