[The application of polymerase chain reaction-single strain conformation polymorphism in the pedigree analysis of familial hypercholerolemia patients].

OBJECTIVE: To discuss the value of polymerase chain reaction-single strand conformation polymorphism(PCR-SSCP) in the pedigree analysis of familial hypercholesterolemia(FH) patients. METHODS: For 4 patients with FH detected by PCR-SSCP and DNA sequence analysis (1 homozygote has point mutation in exon 7, 1 heterozygote in exon 14, and 2 heterozygotes in 3' part of exon 4 ), members of each pedigree, altogether 23 individuals, were analysed using PCR-SSCP. RESULTS: Every member of the 4 pedigrees was examined genetically. Besides the 4 probands, another 1 homozygote and 8 heterozygotes were found. CONCLUSION: The PCR-SSCP method can be used in the pedigree analysis of FH probands. The early diagnosis of siblings and relatives can help to provide genetic consultation and direction so as to pay attention to development of hypercholesterolemia.

[Latest progress in studies of self-setting calcium phosphate cement].

Self-setting calcium phosphate cement(CPC) is a non-ceramic form of hydrxyapatitic artificial bone material (HAP) which was first exclusively produced in America several years ago. CPC is free from the drawbacksto ceramic HAP, including sintering and difficulties in shaping. CPC has the characteristics of simple-producing and easy applyication. In 1991, the clinical application of CPC to repairing the calvarial bone defects was approved, and the results reportedly so far were good or excellent. This paper presents the latest progress in the studies of CPC, concerning the probing into the setting process, the producing of fast-setting and non-decayed types of CPC, the studies of organic compound CPC, the in vitro results of CPC as a drug delivery system, etc. As the research goes on deeply and broadly, CPC is hopefully becoming a standard material in repairing bone defects at the non- or low-bearing site in the future.

[Selection of high avermectins producing strain and identification of avermectin B1].

Three types of colony, powdery gray, white and bald, were isolated from Streptomyces avermitilis ATCC31272. Among them, only the powdery grey one produces avermectins. Sa-76 strain was selected from the powdery grey strain by mutation with high energy electric flow, and its avermectins titer attainde 100 micrograms/mL in shaking flask. Avermectin B1 was extracted and purified from the mycelia of Sa-76, and identified by UV, IR, 1H-NMR, 13C-NMR and mass spectra. After Sa-76 strain was treated twice with NTG, a strain named Sa-76-8 was selected with avermectins titer over 2000 micrograms/mL. The Sa-76-8 strain was treated with NTG once again, and a high avermectins producing strain named as Sa-76-9 with avermectins titer up to 3500-4000 micrograms/mL was selected.

[Estrogen receptor gene polymorphism and its association with clinicopathological manifestation of lupus nephritis].

OBJECTIVE: To investigate the distribution of estrogen receptor (ER) in patients with lupus nephritis (LN), and the association between ER gene polymorphism and the clinical and pathological features of the disease. METHODS: The Pvu II and Xba I restriction fragment length polymorphism (RFLP) of estrogen receptor gene were analyzed in 245 biopsy proven LN patients (58 males and 187 females) and 172 normal controls(101 males and 71 females) by PCR-RFLP. The clinical and pathological features of 49 male and 152 female LN patients with genotype PpXx or Ppxx, ppxx were then analyzed respectively. RESULTS: It was found that genotype PpXx and ppxx, Ppxx were three major genotypes of ER gene in both lupus patients group and control group. The distribution of ER gene polymorphism was quite different in lupus patients of different genders. The frequency of the PpXx genotype was significantly higher in male LN patients than in both the gender matched normal controls (P<0.05) and the female LN patients (P<0.05), while no difference was shown in the frequency of PpXx genotype between female LN patients and female controls. Interestingly, varied clinical and renal pathological features were also demonstrated in female patients with different genotypes of ER gene. CONCLUSION: The distribution of ER gene polymorphism in LN patients varies with gender. The PpXx genotype of ER gene may be associated with the susceptibility of LN in male. ER gene polymorphism is probably one of the genetic factors contributing to the development of clinical heterogeneity and sexually dimorphic manifestations of LN.

[Effects of ischemic preconditioning on apoptosis of hepatocytes in liver transplantation in rats].

OBJECTIVE: To evaluate the effects of ischemic preconditioning (IP) on hepatocyte apoptosis in ischemia/reperfusion injury of grafted liver and clarify its mechanism. METHODS: Orthotopic liver transplantation with cuff anastomosing method for the portal vein, suprahepatic vena cava and infrahepatic vena cava in the rats were divided into two groups: with and without IP treatment. The parameters of liver function were determined by auto-biochemical analyser. The concentrations of malondialdehyde (MDA) were measured with chromometry. The apoptotic hepatocytes in grafts liver were detected by flow cytometry combined with the in situ terminal deoxynucleotidyl transferase-mediated dUTP-FITC nick end-labeling (TUNEL) method. RESULTS: The levels of AST, ALT, LDH in the serum, and MDA and apoptotic cells in the liver were significantly increased compared with normal values after ischemia/reperfusion to the grafted liver. After IP treatment, the levels of AST, ALT, LDH, MDA and apoptotic cells were significantly decreased. CONCLUSION: IP can attenuate ischemia/reperfusion injury by alleviating lipid peroxide and decreasing apoptosis of hepatocytes.

Bone densitometry.

Assessment of BMD has become the essential part of evaluation of patients at risk of osteoporosis. It is likely that different BMD technologies will coexist in clinical practice in the future depending on varying clinical needs. DXA is currently the leading bone density technique because it has the capacity to measure axial and appendicular sites, superior monitoring capabilities, and more sophisticated reference databases and quality control procedures. More rapid and portable peripheral devices, such as peripheral DXA, peripheral instantaneous x-ray imager, and QUS, will likely be used for screening. This large variety of available bone densitometry techniques will provide a wide selection for clinicians to choose from. It may also add confusion, however, about which devices should be used and how to interpret data from different techniques. It is critical for clinicians to have a basic understanding of the technology being used, and the clinical advantages and disadvantages of each instrument, to choose properly which test to perform and interpret the obtained results. When combined with other major risks for fracture, such as age and prior fragility fractures, knowledge of the BMD is an invaluable tool in the assessment of patients with known or suspected osteoporosis.

[In vitro study on transduction of human O(6)-methylguanine-DNA-methyltransferase cDNA into human umbilical cord blood CD34(+) cells].

OBJECTIVE: To explore human umbilical cord blood hematopoietic progenitor cells transduced with human O(6)-methylguanine-DNA-methyltransferase (MGMT) gene increase resistance to 1,3-Bis(2-Chloroethyl)-1-Nitrosourea(BCNU). METHODS: The present authors obtained a full length cDNA fragment encoding the human MGMT from a patient with cholelithiasis liver tissue by RT-PCR method and confirmed by DNA sequencing. The fragment was cloned into pGEM-T vector and further subcloned into G1Na retrovirus vector. Then the G1Na-MGMT was transfected into the packaging cell lines GP+E86 and PA317 by LipofectAMINE method; using the medium containing BCNU for cloning selection and ping-ponging supernatant infection between ecotropic producer clone and amphotropic producer clone, the authors obtained high titer amphotropic PA317 producer clone with the highest titer up to 1.6x10(6) CFU/ml. Cord blood CD34(+) cell were transfected repeatedly with supernatant of retrovirus containing human MGMT cDNA under stimulation of hemopoietic growth factors. RESULTS: PCR, RT-PCR, Southern blot, Northern blot, Western blot and MTT analyses showed that MGMT gene had been integrated into the genomic DNA of cord blood CD34(+) cells and expressed efficiently in the transfected cells. The transgene recipient cells conferred 4 folds stronger resistance to BCNU than that of the non-transduced. CONCLUSION: The retrovirus vector-mediated transfer of MGMT drug resistance gene into human cord blood CD34(+) cells and expression could confer the resistance of transgene cells to BCNU toxicity.

In situ hybridization analysis of transforming growth factor-beta 1 RNA expression during mouse tooth development.

OBJECTIVE: The aim of this study is to investigate the temporal and spatial expression and localization of transforming growth factor-beta 1 mRNA during mouse tooth development. METHODS: The distribution pattern of TGF-beta 1 mRNA during tooth development was analyzed on 5 microns serial sections of paraformaldehyde-fixed and paraffin-embedded embryonic mouse heads or mandibles. RESULTS: The results show that TGF-beta 1 mRNA is expressed during mouse tooth development in a temporally and spatially regulated fashion. Local expression of TGF-beta 1 mRNA in the dental epithelium at budstaged (E13) and capstaged (E15) teeth were observed. During bell stage (E16-18), TGF beta 1 mRNA was very abundant in the ameloblast layer and dental papilla cells. The expression of TGF-beta 1 increased in the layer of odontoblast and ameloblast with the differentiation of these cells. CONCLUSION: These results suggest that TGF-beta 1 may have an important role in odontogenesis and that it acts as a paracrine and autoinducing factor.

Immunohistochemistry of bone sialoprotein and osteopontin during reparative dentinogenesis in vivo.

OBJECTIVE: To investigate the stage-specific and tissue expression of bone sialoprotein (BSP) and osteopontin (OPN) during reparative dentinogenesis in vivo. METHODS: Direct pulp-capping with Ca(OH)2 developed a model used for the investigation of reparative dentinogenesis in the exposed dental pulp. In this model, standardized class V cavities were prepared close to the gingival margin on the buccal surface of each tooth. Animals were sacrificed 3, 7, and 10 days post-operation. Immunohistochemical staining determined the tissue-specific expression of BSP and OPN during the process. RESULTS: Odontoblast-like cells and reparative dentin reacted positively with BSP and OPN. The expression of BSP reached a peak at day 7, then gradually decreased, while the expression of OPN showed no significant difference between day 7 and day 10. CONCLUSION: BSP and OPN may play different roles in reparative dentinogenesis. BSP may serve as a nucleator of hydroxyapatite crystal formation.

Immunolocalization of heat shock protein 70 during reparative dentinogenesis.

OBJECTIVE: To investigate the immunolocalization of heat shock protein 70 (hsp 70) during reparative formation and to discuss the role of heat shock response in dental pulp injury and repair. METHODS: A single cavity was prepared in the mesial surface of the first molars of both maxilla and mandible in Wistar rat. The animals were sacrificed at 3, 15, and 30 days post-operation. After the histological process, the paraffin sections were reacted with monoclonal antibodies against rat hsp 70 using the strept-avidin-biotin-peroxidase complex method. RESULTS: Immunolocalization demonstrated heavy staining for hsp 70 in normal pulp and at different stages of dental pulp repair. In normal pulp, immunoreactivity was visualized in the odontoblasts and the pulp fibroblast. In the group sacrificed at 3 days, heavy staining was located in the odontoblast process and cytoplasm. After 15 days, the newly formed odontoblast-like cells were strongly stained. At 30 days, the same staining intensity was observed in odontoblast-like cells and in pulp cells. No staining was seen in reparative dentin. CONCLUSION: These results demonstrated that heat shock protein 70 might play an important role as a molecular chaperone during reparative dentin formation.

[Effects of c-myc antisense oligodeoxynucleotide transfected by liposome on HL-60 cells].

OBJECTIVE: To investigate a delivery system for administering antisense oligodeoxynucleotide (AS-ODN) to HL-60 cells and study the antitumor effects of c-myc antisense oligodeoxynucleotide. METHODS: Human leukemia cell line HL-60 was transfected with a synthetic oligodeoxynucleotide(ODN) complementary to the 5' end region of c-myc gene. In addition, the changes of function and morphology were observed after transfection of cationic liposome. RESULTS: HL-60 cells incubated with c-myc AS-ODN/lipofectin complex were about 50% decreased in the 4th day as compared with control cells (P < 0.05). About 56% of HL-60 cells exposed to c-myc AS-ODN/lipofectin complex were NBT-positive and showed morphological changes with differentiated phenotype compared with 29% of untreated cells. The expression of the c-myc in the treated cells decreased markedly. The levels of c-myc protein after treatment decreased from (80.00 +/- 3.02)% to (41.67 +/- 4.56)% (P < 0.01). c-myc AS-ODN induced HL-60 cells apoptosis which morphological changes of apoptotic cells were investigated by fluorescence microsopy. A ladder-like pattern of DNA fragmentation was demonstrated on agorose gel electrophoresis and 17% apoptotic cells were found with flow cytometry. CONCLUSIONS: Antisense oligodeoxynucleotide can suppress the malignant phenotype of HL-60 cells and enhancement of the effect was observed by adding lipofectin.

Effect of histamine H3-receptor antagonist clobenpropit on spatial memory of radial maze performance in rats.

AIM: The effect of histamine H3-receptor antagonist, clobenpropit (VUF9153) on spatial memory deficits induced by scopolamine was investigated in rats. METHODS: Eight-Arm radial maze performance was used to measure spatial memory in rats, and the brain regions were subsequently dissected and histamine contents were determined by HPLC. RESULTS: Intracerebroventricular (icv) injection of clobenpropit (50 micrograms) ameliorated memory impairment induced by scopolamine in both parameters of radial maze performance. The amelioration induced by clobenpropit was antagonized by an H3-agonist, (R)-alpha-methylhistamine (5-10 micrograms). alpha-Fluoromethylhistidine (10, 20 micrograms), a histidine decarboxylase inhibitor, also effectively reversed clobenpropit-induced ameliorating effects. CONCLUSION: The brain histamine H3-receptor antagonists are highly related to the spatial memory, and this action may be due to cholinergic neurons.

Facilitating effects of histamine on spatial memory deficit induced by scopolamine in rats.

AIM: To investigate whether or not histamine was involved in scopolamine-induced spatial memory deficits evaluated in 8-arm radial maze performance of rats. METHODS: Eight-Arm radial maze performance was used to measure spatial memory in rats, and the brain regions were subsequently dissected and histamine contents were determined by HPLC. RESULTS: Intracerebroventricular (icv) injection of histamine (100 or 200 ng) or thioperamide (50 micrograms), and intraperitoneal (i.p.) injection of histidine (1000 mg/kg) ameliorated memory impairment induced by scopolamine regarding both parameters of radial maze performance. 2-Thiazolylethylamine, but not 4-methylhistamine showed the similar effect to histamine. Both histamine (200 ng, icv) and histidine (1000 mg/kg, i.p.) were equally effective in increasing the histamine content in the cortex, hippocampus, and hypothalamus. CONCLUSION: These results suggest that brain histamine plays an important role in learning and memory, and its action may be due to cholinergic neurons.
