Salt tolerance of an aqueous solution of a novel amphiphilic polysaccharide derivative.

An aqueous solution of an amphiphilic polysaccharide derivative, hydrophobically (stearyl alkyl group) and hydrophilically (sulfonic-acid salt group) modified hydroxyethylcellulose (HHM-HEC), showed increased viscosity, elasticity, and thixotropic properties in response to the addition of monovalent and divalent salts. Furthermore, the HHM-HEC solution had a transparent appearance at a NaCl concentration of 7 wt %. Since it showed superior salt tolerance to HEC, we focused attention on the two substituents of HHM-HEC and prepared HEC derivatives, namely, hydrophobically modified hydroxyethylcellulose (R-HEC), hydrophilically modified hydroxyethylcellulose (S-HEC), and nonmodified hydroxyethylcellulose (HEC). In addition, we used oscillatory, thixotropic, and fluorometric methods to compare the rheological properties of HHM-HEC with those of other derivatives in the presence of NaCl and ZnCl2, and attempted to elucidate the respective roles of the two substituents of HHM-HEC solution in the salt-tolerance mechanism. As the NaCl concentration in the HHM-HEC solution increased, the values of the elastic modulus G' and the viscous modulus G'' increased, and, moreover, the relative intensities of the first (I1 = 372 nm) and the third (I3 = 383 nm) vibronic bands of the pyrene monomer emission spectrum (the I1/I3 ratio) decreased. These results suggested that the added salt strengthened the three-dimensional network structure of the HHM-HEC polymer by the formation of cross-linkages through the association of hydrophobic substituents. This hydrophobic substituent was therefore essential in allowing HHM-HEC to exhibit a high viscosity in a salt solution. Although the R-HEC solution showed a higher viscosity than did the HHM-HEC solution in the absence of added salts, it became cloudy and lost its viscosity at high NaCl concentrations, apparently because of the shrinkage of its network structure. This signified that the hydrophilic substituent was essential for the sufficient solubility of HHM-HEC to show its rheological properties in a salt-rich solution. We propose to explain how the viscosity of HHM-HEC increases in the presence of salts as follows: Added salts weaken the electrostatic repulsion between the hydrophilic substituents, thereby enhancing the interactions of hydrophobic substituents and consequently increasing the rigidity of the HHM-HEC solution.

Molecular analysis of an inactive aflatoxin biosynthesis gene cluster in Aspergillus oryzae RIB strains.

To help assess the potential for aflatoxin production by Aspergillus oryzae, the structure of an aflatoxin biosynthesis gene homolog cluster in A. oryzae RIB 40 was analyzed. Although most genes in the corresponding cluster exhibited from 97 to 99% similarity to those of Aspergillus flavus, three genes shared 93% similarity or less. A 257-bp deletion in the aflT region, a frameshift mutation in norA, and a base pair substitution in verA were found in A. oryzae RIB 40. In the aflR promoter, two substitutions were found in one of the three putative AreA binding sites and in the FacB binding site. PCR primers were designed to amplify homologs of aflT, nor-1, aflR, norA, avnA, verB, and vbs and were used to detect these genes in 210 A. oryzae strains. Based on the PCR results, the A. oryzae RIB strains were classified into three groups, although most of them fell into two of the groups. Group 1, in which amplification of all seven genes was confirmed, contained 122 RIB strains (58.1% of examined strains), including RIB 40. Seventy-seven strains (36.7%) belonged to group 2, characterized by having only vbs, verB, and avnA in half of the cluster. Although slight expression of aflR was detected by reverse transcription-PCR in some group 1 strains, including RIB 40, other genes (avnA, vbs, verB, and omtA) related to aflatoxin production were not detected. aflR was not detected in group 2 strains by Southern analysis.

Characteristics of a protein error in determination of serum protein in the presence of inorganic salt.

There is a possibility that the color development of the dye-binding method based on a protein error of a pH indicator is affected by the coexisting inorganic salt. Thus, the author theoretically and experimentally investigated the effect of the inorganic salt on the protein error. In a theoretical analysis, the anion of an inorganic salt, like the dissociated dye and buffer anions, was assumed to react with the protein, forming a colorless anion-protein complex. The calculated results were compared with those obtained by experiments using three pH indicators and various kinds of inorganic salts. The calculated results obtained are as follows: (1) The color development decreases with increasing the concentration of the inorganic salt and the equilibrium constant of the reaction between the inorganic salt and protein; (2) The rate of the absorbance decrease is larger for a lower concentration of the inorganic salt than for a higher one; (3) The larger is the equilibrium constant, the larger is the absorbance decrease. The absorbance decrease was caused by the anion, and was increased by increasing the anion concentration. The magnitude of the effect of the anion was iodide > bromide > chloride, which was associated with their ionic radius. The difference in the effect of the anion was thought to indicate that the equilibrium constant, in other words, the bonding strength of the anion to protein is iodide > bromide > chloride.

Analysis of the difference in color development in the dye-binding method due to the kind of buffer solution.

In a dye-binding method using a pH indicator, color development has reportedly been affected by the kind of buffer solution used in the color reagent. This phenomenon was analyzed by using a calculation based on the assumption that the anion of the buffer solution also reacts with protein. Color development decreases with increases in the anion concentration of the buffer solution and in the equilibrium constant of the reaction between the anion and protein. The differences in color development due to the kind of buffer solution can be attributed to differences in the equilibrium constant of the reaction forming the anion-protein complex and to the concentration of the anion between the buffer solutions.

Semisynthesis of human ghrelin: condensation of a Boc-protected recombinant peptide with a synthetic O-acylated fragment.

The creation of peptide using a combination of recombinant expression and chemical synthesis can be a powerful tool for the production of a wide variety of polypeptides modified by phosphorylation, glycosylation, etc. We have developed a new method for the preparation of a recombinant peptide with a free N(alpha)-amino group and protected N(epsilon)-amino groups, and have used this method in the semisynthesis of human ghrelin. Ghrelin, a natural ligand for growth hormone secretagogue receptor, is a 28-residue peptide with an essential n-octanoyl modification on Ser3. A 7-residue N-terminal fragment of ghrelin containing the octanoyl modification was prepared by Fmoc chemistry. In the preparation of it, all reactions were performed on the 2-chlorotrityl resin. Additionally, TBDMS and tBu turned out to be the most effective protection groups for the Ser3 and the Ser2, Ser6, respectively. For preparation of a 21-residue C-terminal fragment, we established a two-step protease processing method for the partially protected segment. A recombinant precursor peptide was Boc protected and subsequently cleaved using two distinct proteases, OmpT and Kex2. The peptides were then coupled to each other and, after deprotection, resulted in fully active human ghrelin.

Measurement of the propelled liquid by isolated hamster ileum as a parameter to evaluate peristalsis.

We present a method to measure the volume of liquid propelled by peristaltic movements of isolated hamster ileum as a novel means to assess peristaltic activity. The oral and aboral ends of the dissected ileum were attached to cannulas fixed horizontally. The application of intraluminal pressure by raising the level of liquid in the bottle connected to the oral end evoked peristalsis and intermittent propulsion of the intraluminal liquid. The inhibition of intrinsic neurons by tetrodotoxin stopped propulsion; this indicated that the liquid propulsion was correlated with neuron-regulated peristalsis. The volume of liquid propelled by one complete peristaltic movement was significantly greater than that by incomplete peristalsis, whereas recordings of pressure changes were indistinguishable. Inhibitors of nitric oxide synthase decreased the volume of liquid propelled by peristaltic movements, suggesting a role of nitrergic neurons in peristalsis. Our data show that the method described above might be suitable for analyzing peristalsis.

A comparative histological study on the distribution of striated and smooth muscles and glands in the esophagus of wild birds and mammals.

Musculature and glands of the esophagus in various wild birds and mammals were examined histologically. Cervical and thoracic esophagi of all birds used (mallard, spot-billed duck, Ural owl and Hodgson's hawk-eagle) were comprised of smooth muscle fibers only. In contrast, esophagi of the nutria, Japanese raccoon dog, common raccoon and Japanese marten consisted largely of striated muscle fibers. In the masked palm civet, Japanese macaque and bottlenose dolphin, esophageal muscle layers consisted of both striated and smooth muscle fibers. Esophageal glands were observed except for the nutria and masked palm civet. These results show a wide variety of the structural composition in the esophagus of wild animals, particularly mammals, examined in this study.

Theoretical analysis concerning the characteristics of a dye-binding method for determining serum protein based on protein error of pH indicator: effect of buffer concentration of the color reagent on the color development.

In the dye-binding method based on protein error of a pH indicator, the color development has been reported to be markedly affected by the buffer concentration of the color reagent. In this study, the author analyzed this phenomenon by a theoretical calculation based on the chemical equilibrium of protein error. The calculation was performed on the assumption that both the dissociated dye anion and the anion contained in the buffer solution react with protein, forming a dye-protein complex and an anion-protein complex, respectively. The calculated results were compared with those obtained by the experiments using bromophenol blue, bromocresol green and bromocresol purple that are employed widely for determining the human serum albumin concentration clinically. The calculated results of this method are summarized as follows: (1) the color development decreases with the increase in the concentration of the anion contained in the buffer solution; (2) the calibration curve is more linear in the higher concentration of the anion than in the lower one. These calculated results agreed well with the experimental ones. From these results, it was concluded that the change in the color development by the buffer concentration of the color reagent is due to the change in the concentration of the buffer anion.

[A contribution of a pharmaceutical department to the safety management of an outpatient's cancer chemotherapy].

A concern about safety management is growing rapidly in the field of an outpatient's cancer chemotherapy. Therefore, the pharmaceutical department of Tokai University Hachioji Hospital has employed; (1) a system to check injection prescriptions accompanied by records of drug administration and (2) an injection drug preparation worksheet for a certain communication. The results that examined their usability were as follows: (1) a prescription check found a deviation of recipes from the protocol, then all of the deviations were revised, (2) use of the worksheet with an indicated cancellation line completely prevented preparation mistakes in the case of a prescription change. Consequently, the safety management of an outpatient's cancer chemotherapy has made progress due to an application of the system and worksheet by the pharmaceutical department.

Forecasting the number of inpatients with schizophrenia.

There has been much discussion in Japan regarding the reduction of psychiatric beds. For effective healthcare planning, reliable forecasting is important. The purpose of this study was to predict the number of future schizophrenic inpatients using quantitative methodology. Data was obtained from a survey of schizophrenic inpatients conducted annually at the end of March by the Niigata Prefecture from 1974 to 2003. The numbers of schizophrenic inpatients in different age groups over a long period of time were used in a precise time-series analysis to establish trends. Then these past trends were used to forecast inpatient numbers for future years. The pattern of ascents and declines of each inpatient group stratified by age appeared to be duplicated by the next older age group 10 years later. The numbers of inpatients with schizophrenia in 2013 and 2023 are projected to be 78.5% and 56.7% of the number of patients in 2003, respectively. By 2033, the number is forecast to decline to 41.0% of the number in 2003. This study forecasts that inpatients with schizophrenia will decrease substantially over the next several decades. Policy should be designed to reflect this trend.

The upper limit pH in the dye-binding method for the determination of serum protein via measurements of the absorbance increase produced by protein error.

In the dye-binding method for determining the albumin concentration, the absorbance increase due to the change of the color shade by protein error of a pH indicator can be measured by a spectrophotometer. This absorbance increase is observed only in a restricted pH region, but this pH region is not theoretically studied yet. Thus, the author investigated the upper limit pH (pHUL) at which the absorbance increase occurs by the theoretical calculation, and compared these results with those obtained experimentally using four pH indicators. The pHUL is not affected by the dye or protein concentrations, or by the formation constant of the dye-protein complex; but the value changes according to the acid-dissociation constant of the dye (KD) and the ratio of the molar absorptivities of the proton-dissociated dye anion (epsilonD) and the dye-protein complex (epsilonPD). The pHUL value can be calculated by the equation, found theoretically. The calculated pHUL values of BPB, BCG, BCP and BTB were 5.1, 4.8, 6.2 and 5.5, respectively. These values correlated with the experimental results of 4.5 for BPB, 4.7 for BCG, 5.9 for BCP and 5.2 for BTB, but were not associated with the pKD values of each dye. The pHUL of these dyes did not change significantly for various dye and protein concentrations, as was expected from the thoretical calculation.

Absolute eigenvalue diffusion tensor analysis for human brain maturation.

The absolute eigenvalues of the diffusion tensor of white matter in sixteen normal subjects in two groups representing the early developmental stage (ages 1-10 years, n=8) and young adult stage (ages 18-34 years, n=8) were assessed using a high-field (3.0 T) magnetic resonance (MR) system. All three eigenvalues, including the largest eigenvalue, decreased significantly with brain maturation. The rate of the decline in the two small eigenvalues was, however, much higher than that of the largest eigenvalue, resulting in an actual increase in fractional anisotropy, a commonly measured relative index. The data demonstrate that an increase in anisotropy associated with brain maturation represents a significant decline in the small eigenvalue components, rather than an increase in the largest eigenvalue. The observed pattern of eigenvalue changes is best explained by the simultaneous occurrence of two of several independent phenomena within the axonal microenvironment during the myelination process, namely, (1) decline in unrestricted water content in extra-axonal space, and (2) increase in apparent diffusivity within the axon.

Feasibility study of single region lambda chart analysis for pyramidal tract physiology.

Diffusion characteristics of the pyramidal tract were assessed in nine patients who had clinical evidence of pyramidal tract dysfunction, utilizing lambda chart analysis (LCA). The underlying pathologic process of tract dysfunction was varied and included Pelizaeus-Merzbacher disease (PMD), Alexander disease, adrenoleukodystrophy (adrenomyeloneuropathy (AMD) type and cerebral type), amyotrophic lateral sclerosis (ALS), and Wallerian degeneration (WD). While pyramidal tract diffusion characteristics in WD indicated a pathological process characterized by replacement of normal fibers by smaller cellular component such as degenerated small fibers and/or gliosis, pyramidal tract diffusion characteristics in patients with PMD, Alexander disease, and adreno leukodystrophy of the cerebral type indicated a pathological process characterized by replacement of normal fibers by larger cellular components such as spheroids or edematous space. Pyramidal tract diffusion characteristics of patients with ALS or adrenoleukodystrophy of AMD type were relatively intact suggesting a pathological process characterized by relatively preserved structural architecture. These findings are highly consistent with known pathophysiological indices and indicate the feasibility of the clinical utility of LCA for assessing pyramidal tract physiology.

Alteration of citrate metabolism in cluster roots of white lupin.

Organic acid excretion plays a key role in the superior P(i)-acquisition of barely soluble inorganic P sources from soils. Seedlings of white lupin (Lupinus albus L.) grown for 37 d in -P nutrient solution showed typical -P symptoms, such as low P content, increased root/shoot ratio and the development of cluster roots which released large amounts of citrate. Citrate concentration in the cluster roots was 21.5 micro mol (g FW)(-1), which corresponded to a 4.3- and 2.6-fold increase of +P and -P root apexes, respectively. Cluster roots possessed higher phosphoenolpyruvate carboxylase and phosphoenolpyruvate phosphatase activity than those in +P root apexes, which could result in increasing the supply of substrate for citrate synthase. On the other hand, the cytosolic pathway which converts citrate to 2-oxoglutarate consists of aconitase and NADP-specific isocitrate dehydrogenase activity that was lower in the cluster roots than in +P root apexes, and may contribute to citrate accumulation. Thus, metabolic balance with these alterations would play an important role in increasing citrate concentration in the cluster roots. The molecular characterization of NADP-specific isocitrate dehydrogenase indicated that the cytosolic isoenzyme functions as a hetero-dimer, and that the activity would be regulated by the transcript levels for both isoforms.

Effects of lidocaine into knee on QF strength and EMG in patients with ACL lesion.

PURPOSE: In our previous study, we found that injection of lidocaine into intact knees reduced the maximal voluntary contraction (MVC) and integrated electromyogram (I-EMG) of the quadriceps femoris (QF). This study was designed to investigate changes in the MVC and I-EMG of the QF in response to lidocaine, in patients with anterior cruciate ligament (ACL) lesion, to evaluate alpha-motoneuron activity innervating the QF. METHODS: The MVC of knee extension and I-EMG of the vastus medialis (VM), vastus lateralis (VL), and rectus femoris (RF) muscles were measured in eight patients with ruptured ACL, before and after lidocaine injection into the knee. RESULTS: There were no significant differences between preinjection and postinjection values of MVC (preinjection: 167 +/- 49 N.m; postinjection: 164 +/- 55 N.m) and I-EMG (preinjection: VL: 0.11 +/- 0.06, VM: 0.13 +/- 0.10, RF: 0.09 +/- 0.04) (postinjection: VL: 0.12 +/- 0.07, VM: 0.13 +/- 0.10, RF: 0.09 +/- 0.05). CONCLUSION: Our results indicated that hindrance of afferent feedback from the knee in patients with ACL rupture did not significantly change alpha-motoneuron activity. Lidocaine injection into the knee joint of the subjects in this study only attenuated afferent feedback from mechanoreceptors in the knee joint cavity, but not in the ACL, as afferent feedback from ACL was already lost due to ACL rupture. This indicated that attenuation of afferent feedback from mechanoreceptors in the knee joint cavity other than the ACL did not significantly decrease the activity of alpha-motoneurons innervating the QF during MVC exertion. Therefore, our findings provide evidence that afferent feedback from the ACL has a major influence on the MVC exertion of the QF.

High-energy, all-solid-state, ultraviolet laser power-amplifier module design and its output-energy scaling principle.

We demonstrate that a coaxially pumped, large-aperture ultraviolet power-amplifier module with solid-state tunable laser medium Ce(3+):LiCaAlF6 has 98-mJ, 290-nm, and 3-ns output pulses with a sufficient extraction efficiency of 25%. The detailed information of design parameters, including the gain-coefficient dependence on pump condition, is successfully accumulated for further energy scaling for a terawatt-class ultraviolet chirped pulse amplification laser system or a high-pulse-energy laser system.

Generation and characterization of human hybrid neurons produced between embryonic CNS neurons and neuroblastoma cells.

A human hybrid neuronal cell line A1 has been generated by somatic fusion between a human fetal cerebral neuron and a human neuroblastoma cell, and RT-PCR, immunochemical, and electrophysiological studies of the hybrid cells indicated that the cells express faithfully of morphological, immunochemical, physiological, and genetic features of human cerebral neurons. A1 hybrid neurons express neuron-specific markers such as neurofilament-L (NF-L), NF-M, NF-H, MAP-2, and beta tubulin III. A1 human hybrid neurons express messages for various cytokines and cytokine receptors which are similar to parental human CNS neurons and different from the other parental cell line, SK-SH-SY5Y neuroblastoma. A1 hybrid neurons also express messages for choline acetyltransferase (ChAT), tyrosine hydroxylase (TH), and glutamic acid decarboxylase (GAD), indicating that they could differentiate into various subsets of neuronal types. Whole-cell patch clamp experiments showed that A1 hybrid neurons expressed Na+ currents, which were completely blocked by tetrodotoxin. In addition, depolarizing and hyperpolarizing voltage clamp steps evoked respective outward and inward K+ currents in these cells. When A1 hybrid neurons were exposed to beta amyloid for 72 hr, there was three-fold increase in TUNEL positive cells over controls, indicating that beta amyloid is neurotoxic to A1 hybrid neurons. The present study indicates that the A1 human hybrid neuronal cell line should serve as a valuable in vitro model for studies of biology, physiology, and pathology of human neurons in health and disease.
