Intraspecific responses in grain quality of 10 wheat cultivars to enhanced UV-B radiation under field conditions.

Field studies were conducted to determine the potential for intraspecific responses in grain quality of 10 wheat (Triticum aestivum) cultivars to enhanced ultraviolet-B (UV-B, 280-315 nm) radiation. The plants were exposed to 5 kJm(-2) supplemental UV-B radiation, simulating a depletion of 20% stratospheric ozone. In wheat cultivars tested, intraspecific responses in 16 amino acid contents and total amino acid contents in grains were found. Out of tested wheat cultivars, Dali 905, Mianyang 20, Wenmai 3, Chuxiong 8807 and Huining 18 showed significant increase in proteins, and Yunmai 39 and Huining 18 showed significant increase while Dali 905 and Chuxiong 8807 showed significant decrease in total sugar, respectively. No wheat cultivar showed significant change in rough starch content. Out of 10 cultivars 9 had a positive quality response index (QRI), while only Yunmai 39 showed negative response (QRI -1.19). According to QRI, the tolerance of 10 wheat cultivars to UV-B radiation had the following sequence, Huining 18>Mianyang 20>Mianyang 26>Wenmai 3>Dali 905>Longchun 16>Fengmai 24>Liaochun 9>Chuxiong 8807>Yunmai 39. Intraspecific responses in grain quality of 10 wheat cultivars to enhanced UV-B radiation existed under field conditions.

Antiarrhythmic effects of ranolazine in a guinea pig in vitro model of long-QT syndrome.

Prolongation of the QT interval of the ECG is associated with increased risk of torsades de pointes ventricular tachycardia. Ranolazine, a novel antianginal agent, is reported to decrease the delayed rectifier potassium current, I(Kr), and to increase action potential duration (APD) and the QT interval. However, ranolazine is also reported to reduce late sodium current (late I(Na)), a depolarizing current that contributes to prolongation of the plateau of the ventricular action potential. We hypothesized that ranolazine would decrease APD and the occurrence of arrhythmias when late I(Na) is increased. Therefore, we measured the effects of ranolazine alone and in the presence of anemone toxin (ATX)-II, whose action mimics the sodium channelopathy associated with long-QT3 syndrome, on epicardial monophasic action potentials and ECGs recorded from guinea pig isolated hearts. Ranolazine (0.1-50 microM) prolonged monophasic APD at 90% repolarization (MAPD(90)) by up to 22% but did not cause either early afterdepolarizations (EADs) or ventricular tachycardia (VT). ATX-II (1-20 nM) markedly increased APD and caused EADs and VT. Ranolazine (5-30 microM) significantly attenuated increases in MAPD(90) and reduced episodes of EADs and VT produced by ATX-II. Ranolazine also attenuated the synergistic effect of MAPD(90) increase caused by combinations of ATX-II and blockers of I(K) [E-4031; 1-[2-(6-methyl-2-pyridyl)ethyl]-4-methylsulfonylaminobenzoyl)piperidine]. Thus, although ranolazine alone prolonged APD, it reduced APD and ventricular arrhythmias caused by agents that increased late I(Na) and decreased I(K).

Differential expression of genes during aflatoxin B(1)-induced hepatocarcinogenesis in tree shrews.

AIM: Through exploring the regulation of gene expression during hepatocarcinogenesis induced by aflatoxin B(1) (AFB(1)), to find out the responsible genes for hepatocellular carcinoma (HCC) and to further understand the underlying molecular mechanism. METHODS: Tree shrews (Tupaia belangeri chinensis) were treated with or without AFB(1) for about 90 weeks. Liver biopsies were performed regularly during the animal experiment. Eight shares of total RNA were respectively isolated from 2 HCC tissues, 2 HCC-surrounding non-cancerous liver tissues, 2 biopsied tissues at the early stage (30th week) of the experiment from the same animals as above, 1 mixed sample of three liver tissues biopsied at the beginning (0th week) of the experiment, and another 1 mixed sample of two liver tissues from the untreated control animals biopsied at the 90th week of the experiment. The samples were then tested with the method of Atlas(TM) cDNA microarray assay. The levels of gene expression in these tissues taken at different time points during hepatocarcinogenesis were compared. RESULTS: The profiles of differently expressed genes were quite different in different ways of comparison. At the same period of hepatocarcinogenesis, the genes in the same function group usually had the same tendency for up- or down-regulation. Among the checked 588 genes that were known to be related to human cancer, 89 genes (15.1%) were recognized as "important genes" because they showed frequent changes in different ways of comparison. The differentially expressed genes during hepatocarcinogenesis could be classified into four categories: genes up-regulated in HCC tissue, genes with similar expressing levels in both HCC and HCC-surrounding liver tissues which were higher than that in the tissues prior to the development of HCC, genes down-regulated in HCC tissue, and genes up-regulated prior to the development of HCC but down-regulated after the development of HCC. CONCLUSION: A considerable number of genes could change their expressing levels both in HCC and in HCC-surrounding non-cancerous liver tissues. A few modular genes were up-regulated only in HCC but not in surrounding liver tissues, while some apoptosis-related genes were down-regulated in HCC and up-regulated in surrounding liver tissues. To compare gene-expressing levels among the liver tissues taken at different time points during hepatocarcinogenesis may be helpful to locate the responsible gene (s) and understand the mechanism for AFB(1) induced liver cancer.

Effects of retinoic acid on the beta-catenin/TCF pathway in cultured porcine tracheobronchial epithelial cells.

The effects of retinoic acid on the beta-catenin/TCF pathway in cultured porcine tracheobronchial epithelial cells (TBEC) were investigated. After TBEC were treated with retinoic acid at various concentrations, mRNA and protein changes of beta-catenin in cytoplasm, nucleus and whole cell of the TBEC were observed by immunocytochemical stain, RT-PCR and Western blotting. And the changes of the target gene cyclinD1 of beta-catenin/TCF pathway were also observed. It was found that there was no significant difference in beta-cat mRNA level after retinoic acid treatment. However, the expression of beta-catenin in the whole cell and cytoplasm was elevated with the increase of retinoic acid concentration (P<0. 01). The nuclear protein beta-catenin and target gene cyclinD1 of beta-catenin/TCF pathway was decreased (P<0.05). It was indicated that retinoic acid could increase beta-catenin level of the whole cell protein and decrease nuclear beta-catenin, downregulating beta-cat/TCF signaling activity and reducing target gene cyclinD1 protein level. As a result, retinoic acid can downregulate beta-catenin/TCF pathway in porcine tracheobronchial epithelial cell, suggesting that retinoic acid can inhibit the proliferation and accelerate differentiation of tracheobronchial epithelial cells.

Effect of lithium on cell cycle progression of pig airway epithelial cells.

To investigate the effect of lithium on cell cycle progression of airway epithelial cells, primary pig tracheobronchial epithelial cells were incubated with lithium chloride (LiCl) at different concentrations (0, 5 mmol/L, and 10 mmol/L) and time (12 h, 16 h and 24 h). After the treatment, cells were counted, cell cycle profile was measured by BrdU labeling and flow cytometry, and expression of cyclin D1 and cyclin B1 were detected by Western blotting. The results showed that after 24h of 10mmol/L but not 5mmol/L LiCl treatment, proliferation of cells was slowed down as manifested by delayed confluence and cell number accumulation (P<0.05). Lithium did not change the percentage of cells in S phase (P>0.05), but 24 h incubation with 10 mmol/L LiCl induced a G2/M cell cycle arrest. Furthermore, 10mmol/L LiCl elevated cyclin D1 expression after 12h treatment, while expression of cyclin B1 increased more significantly after 24h incubation. These data demonstrate that lithium inhibits proliferation of pig airway epithelial cells by inhibiting cell cycle progression, and suggest that lithium-sensitive molecule(s) such as glycogen synthase kinase 3 may have a role in the regulation of growth of airway epithelial cells.

Cloning, expression, and purification of soluble human interleukin-4 receptor in Streptomyces.

Interleukin-4 (IL-4) is a pleiotropic cytokine which plays a pivotal role in shaping immune responses and mediating important proinflammatory functions in asthma. This cytokine exerts its biological effects through binding to its receptor (IL-4R) complex, with the alpha chain as the high-affinity binding subunit. Soluble IL-4R (sIL-4R) lacks the transmembrane and cytoplasmic domains, so it cannot induce cellular activation. By acting as a decoy to circulating IL-4 and neutralizing its activity, the high specificity and affinity of sIL-4R make it ideal as an IL-4 antagonist. In this study, a sIL-4R cDNA encoding the extracellular domain of IL-4R alpha chain was cloned into a Streptomyces-Escherichia coli shuttle plasmid pSGLgpp and expressed secretly in Streptomyces lividans TK 24. On SDS-PAGE gel, the expressed sIL-4R protein showed a Mw of 24 kDa, agreeable with the predicted size. The N-terminal sequence of the protein was also determined, confirming its identity and indicating that no degradation occurred at the N-terminus. With DEAE-Sepharose Fast Flow and Superdex HR 75 columns, the protein was purified and used on HPLC analysis, the purity reaching about 90%. The results of the ligand-binding blot and ELISA showed that such protein has biological activity of binding with ligand IL-4.

Atrial natriuretic peptide-C receptor-induced attenuation of adenylyl cyclase signaling activates phosphatidylinositol turnover in A10 vascular smooth muscle cells.

Atrial natriuretic peptide (ANP)-C receptor activation has been shown to inhibit adenylyl cyclase (AC) activity as well as to stimulate phospholipase C (PLC) signaling pathways. The present studies were undertaken to investigate whether ANP-C receptor-mediated decreased cAMP levels contribute to the activation of PLC signaling. C-ANP(4-23) [des(Gln(18),Ser(19), Glu(20),Leu(21),Gly(22))ANP(4-23)-NH(2)], a ring-deleted peptide of ANP that interacts specifically with ANP-C receptor, stimulated inositol 1,4,5-tris-phosphate (IP(3)) production (PLC activity) in A10 vascular smooth muscle cells in a concentration- and time-dependent manner. The maximal stimulation observed was about 75% at 2 h of treatment, with an apparent EC(50) of about 20 to 30 nM. Pertussis toxin treatment of the cells completely abolished the C-ANP(4-23)-mediated stimulation of IP(3) production. Forskolin (FSK), a stimulator of adenylyl cyclase, dibutyryl cAMP (db cAMP), and isoproterenol (ISO), a beta-adrenergic agonist that stimulates adenylyl cyclase activity and cAMP levels, inhibited IP(3) production by about 35, 30, and 50%, respectively, whereas dideoxyadenosine (DDA), an inhibitor of adenylyl cyclase activity, and oxotremorine stimulated IP(3) production by about 90 and 80%, respectively, in these cells, suggesting a functional interaction between these two signaling pathways. Treatment of the cells with antisense oligonucleotide of ANP-C receptor that attenuated ANP-C receptor-mediated inhibition of adenylyl cyclase resulted in a complete attenuation of C-ANP(4-23)-induced stimulation of IP(3) formation, whereas FSK, db cAMP, and ISO-mediated decrease and oxotremorine and endothelin-1 (ET-1)-induced increase in IP(3) production was not affected by this treatment. Furthermore, C-ANP(4-23)-induced increase in IP(3) formation was significantly potentiated by DDA and inhibited by FSK and db cAMP, whereas ET-1-induced increase in IP(3) production was not affected by FSK. In addition, N-[2-(4-bromocinnamylamino)ethyl]-5-isoquinoline (H-89), an inhibitor of protein kinase A, completely abolished C-ANP(4-23) and not ET-1-induced stimulation of IP(3) production. These results indicate that ANP-C receptor activation by C-ANP(4-23) and resulting decrease in cAMP levels may be responsible for the activation of phosphatidylinositol (PI) turnover signaling, suggesting a cross-talk between ANP-C receptor-mediated adenylyl cyclase and PLC signaling pathways.

[Methylation status of multidrug resistance (mdr1) gene and its correlation with expression of mdr1 gene in patients with hematologic malignancies].

To investigate the correlation between methylation and expression of multidrug resistance (mdr1) gene, restriction endonuclease HpaII combined with competitive PCR technique was used to quantitatively detect the methylation status of two CCGG sites located at -110 and -50 bp (region I and II) up to the transcription start site in mdr1 promoter in 54 AL and 9 MM patients. Semi-quantitative RT-PCR was used to detect the expression level of mdr1 gene. The results showed that inverse correlation between methylation rate of either region or total methylation rate and expression of mdr1 gene was observed. The correlation in the region I (r = -0.64) was closer than that in the region II (r = -0.4). High expression rate of mdr1 ascended significantly in low methylation group (n = 36) (P < 0.001). In comparison with chemotherapy sensitive group (n = 8), the methylation rate in refractory AL patients (n = 16) was lower (P = 0.05) in the region I, P < 0.05 in the region II and total regions. Comparing with the untreated patients (n = 36), the methylation rate in the region I and total methylation rate were lower in the patients with chemotherapy (n = 14) (P < 0.05). The methylation rate in the region II was also decreased after chemotherapy, however, no statistical significance was shown (P > 0.05). Increased mdr1 expression level accompanying with decreased methylation rate after chemotherapy was found, although no significant difference was shown (P = 0.06). It is concluded that the expression level of mdr1 gene was associated with the methylation status of CCGG in -110 and -50 bp upstream to the transcription start site, especially the -110 site. In both the patients treated with chemotherapy and the refractory patients, the methylation level of mdr1 gene decreased relatively. The rising expression of mdr1 gene after chemotherapy was associated with the decrease of methylation level.

[The study of proliferation of cells at the invasive tumor front of squamous cell carcinoma of tongue].

OBJECTIVE: To evaluate the value of the invasive front grading (IFG), the proliferative activity of cells at the invasive tumor front (ITF) of squamous cell carcinoma (SCC) of tongue. METHODS: All the tumors were graded using IFG grading. The expression of PCNA, Ki67 and staining and measurement of AgNOR number were evaluated at the ITF and the other parts of the same tumor. RESULTS: The expression of PCNA (P < 0.001), Ki67 (P < 0.001), AgNOR number (P < 0.001) at the ITF were higher than those of other parts. The IFG score was significantly correlated with the expression of PCNA (P < 0.05), AgNOR number (P < 0.01) at the ITF. The complete IFG score (P < 0.01) was significant in both univariate and multivariate cox analysis; the AgNOR number at the ITF (P < 0.05) was only significant in univariate analysis (P < 0.05). CONCLUSIONS: The ITF of SCCs of tongue differentiate poorer than the other parts and is active in proliferation. The complete IFG score and the AgNOR number at the ITF could be a valuable indication to predict the prognosis of SCCs of tongue.

[Relationship of matrilysin with invasion, metastasis, and prognosis in laryngeal cancer].

BACKGROUND & OBJECTIVE: Matrilysin (matrix metalloproteinases-7, MMP-7) might play an important role in invasion and metastasis of tumor and is thought to be related to prognosis of many kinds of carcinomas. This study was designed to investigate the significance of MMP-7 expression in the invasion and metastasis of laryngeal cancer and the relationship of MMP-7 expression with the prognosis of laryngeal cancer. METHODS: The expression of MMP-7 in 70 samples with paraffin-embedded laryngeal cancer and corresponding para-tumor normal mucosas were using immunohistochemical technique. The mRNA expression of MMP-7 were examined by reverse transcription-polymerase chain reaction (RT-PCR) in 24 frozen specimens of laryngeal cancer and corresponding para-tumor normal mucosas. RESULTS: (1) The immunohistochemical staining of MMP-7 was observed in 77.1% (54/70) of cancer tissues, which was significantly higher than that (5.7%, 4/70) of para-tumor normal mucosas (P< 0.01). The mRNA expression of MMP-7 was detected in 75.0% (18/24) of cancer tissues, which was also significantly higher than that (4.2%, 1/24) of para-tumor normal mucosas (P< 0.01). (2) The expression of MMP-7 in protein level was hot only higher in T3-T4 group than that in T1-T2 group (P< 0.01) but also higher in nodal metastases group than that in group without nodal metastases (P< 0.01). (3) The mRNA expression of MMP-7 was significantly higher in T3-T4 group than that in T1-T2 group (2.908 +/- 0.891 vs.1.662 +/- 0.508, P< 0.01) and was significantly higher in nodal metastases group than that in group without nodal metastases (2.958 +/- 0.827 vs. 1.887 +/- 0.769,P< 0.01). (4) Kaplan-Meier survival curve showed that the prognosis of group with MMP-7 protein high expression was poorer than that in group of MMP-7 protein weak or negative expression (log-rank=4.7559, 8.9513; P=0.0292, 0.0028). No significant difference was found between group with MMP-7 protein weak expression and group with that negative expression (log-rank=0.0314, P= 0.8593). CONCLUSION: MMP-7 possesses close relationship with the invasion, metastasis, and prognosis of laryngeal cancer, and it may be served as a marker in estimating the invasive and metastatic potency and prognosis of laryngeal cancer.

[Constructing shuttle plasmid fusion expressing Ag85B-ESAT-6 on the surface of Mycobacterium].

OBJECTIVE: To construct the E. coli.-BCG (Bacille Calmette-Guerin) shuttle vector expressing Mycobacterium tuberculosis secreted protein Ag85B-ESAT-6 on the surface of Mycobacterium vaccae. METHODS: The gene fragment containing 19 000 antigen (19-ss) were amplified by polymerase chain reaction (PCR) from the Mycobacterium tuberculosis H(37)Ra. We cloned the 19ss gene into the E. coli.-BCG shuttle vector pOLYG and named the pCW, which can shuttle and express exogenous antigen gene on cell wall of Mycobacterium. Then Mycobacterium tuberculosis secret protein Ag85B and ESAT-6 gene were cloned into the vector and determined by indirect immunofluorescence. RESULTS: The sequence of 19-ss gene was identified with Genbank reported by sequencing. The constructed E. coli.-BCG shuttle vector using 19ss gene had the function of shuttle between E. coli. and Mycobacteria. By indirect immunofluorescence technique the secreted protein Ag85B-ESAT-6 can be fused and expressed on surface of Mycobacterium vaccae. CONCLUSION: The E. coli.-BCG shuttle vector is constructed successfully which could express exogenous antigen gene as a chimeric exported membrane.

[Influence of epidermal growth factor and gonadotrophin on the in vitro maturation of human oocytes].

OBJECTIVE: To study the effect of epidermal growth factor(EGF) and different concentrations of gonadotrophin (Gn) on the in vitro maturation of human oocytes. METHODS: EGF was added to the vitro culture medium in order to observe the effect of Gn combined with or without EGF on the result of in vitro maturation. The concentrations of hCG and FSH were changed respectively to observe the difference between the results. RESULTS: Adding EGF to the culture medium improved the maturation rate of oocyte significantly (P < 0.05). There was no difference between the results with different concentrations of hCG and FSH in the culture medium. CONCLUSION: EGF can improve the results of the in vitro maturation of human oocytes by increasing the maturation rate significantly. Increasing the concentration of Gn does not influence the results of in vitro maturation.

[Re-expression of p16 gene in myeloma cell line U266 by arsenic trioxide].

BACKGROUND & OBJECTIVE: DNA methylation status regulates gene expression and is associated with oncogenesis. Demethylation of DNA has been proposed as a possible new strategy for cancer prophylaxis and treatment. S-adenosylmethionine is required as methyl donor for both arsenic metabolism and DNA methylation. The present study was designed to explore the possibility and mechanism of re-expression of the silenced p16 gene in human myeloma cell line U266 cells by arsenic trioxide (As2O3). METHODS: The U266 cell line in which the p16 gene is silenced due to hypermethylation was treated with different concentration of As2O3. The PCR technique combined with HpaII and its isoschizomer MspI was used to assess p16 gene methylation status. The mRNA expression levels of p16 and DNA methyltransferases (DNMTs) genes were determined with RT-PCR technique. Western blot was performed for P16 protein expression analysis. RESULTS: (1)The characteristic hypermethylation with losing expression of p16 gene in U266 cells was confirmed. After agarose gel electrophoresis, the genomic DNA digested with MspI showed a "smear" pattern, however the HpaII digested DNA showed a strong single band. There was no PCR amplified product of p16 gene when MspI digested DNA was used as the templates, whereas a 340 bp p16 gene product was found in HpaII digested DNA sample as the undigested DNA was amplified. Also, neither p16 mRNA nor P16 protein was detectable when assessed with RT-PCR and Western blot, respectively. (2)When the genomic DNA from cells treated with (0.5-2.0) micromol/L As2O3 was digested with HpaII and then analyzed by electrophoresis, a "smear" pattern was observed. The 340 bp product could not be amplified if such digested DNA was used as PCR templates. Detectable expression of both p16 mRNA and P16 protein was found in the cells treated with 1.0 micromol/L and 2.0 micromol/L As2O3. (3)Expression levels of DNMT 3A and 3B mRNA were increased in the treated cells and were dependent on As2O3 concentration, however no significant difference was found between each two groups (P >0.05). CONCLUSION: As2O3 could induce p16 gene re-expression in human myeloma cell line U266 through DNA demethylation.

[Expression and significance of EGF mRNA and EGFR mRNA in hepatocellular carcinoma].

BACKGROUND & OBJECTIVE: Many evidences demonstrated that the epidermal growth factor receptor (EGFR) subfamily played an important role in they initiation and progression of various cancers. But it is not clear whether there is a relationship between EGFR and hepatocellular carcinoma (HCC). The aims of the present study were to explore the expression and significance of the epidermal growth factor (EGF) mRNA and EGFR mRNA in human HCC tissues. METHODS: Reverse transcription polymerase chain reaction (RT-PCR) was employed to determine the expression of EGF mRNA and EGFR mRNA in 60 HCC tissues and their adjacent liver tissues. RESULTS: The positive rate of EGF mRNA was significantly lower in the HCC tissue (60%, 36/60) than in the adjacent liver tissue (80%, 48/60) (P< 0.05). The positive rate of EGFR mRNA in the HCC tissue (60%, 36/60) was markedly higher than in the adjacent liver tissue (41.67%, 25/60) (P< 0.05). The expression of EGFR mRNA in the HCC tissue was significantly correlated with the clinical stage, the portal vein tumor thrombus, the presence of extrahepatic metastasis, and the recurrence of tumor, the number of tumor, but not correlated with the diameter of tumor, the level of serum alpha-fetoprotein (AFP), the differentiation of tumor and the liver cirrhosis in the adjacent tissue. The detectable rate of EGF mRNA was correlated with the diameter of tumor but not correlated with the clinical stage, the portal vein tumor thrombus, the presence of extrahepatic metastasis, the recurrence of tumor, the number of tumor, the level of serum AFP, the differentiation of tumor and the liver cirrhosis in the adjacent tissue. CONCLUSIONS: EGF may not be involved in the initiation and progression of HCC, whereas, EGFR relates to the initiation and progression and the recurrence of HCC. EGFR can be considered as a marker for predicting the metastasis and recurrence of HCC.

[Computer tomography characteristics of chronic invasive fungal rhinosinusitis in early stage].

OBJECTIVE: To discuss the computer tomography (CT) characteristics of chronic invasive fungal rhinosinusitis in early stage and their diagnostic value. METHOD: The CT characteristics of 18 cases of chronic invasive fungal rhinosinusitis in early stage and 35 noninvasive fungal rhinosinusitis diagnosed with pathologic examination were retrospectively analyzed and compared. RESULT: On sinus CT scan of chronic invasive fungal rhinosinusitis in early stage, 78% (14/18) were single-sinus lesioned. The maxillary sinus was the most commonly lesioned sinus, followed by the sphenoid sinus and ethmoid sinus. 22% (4/18) were poly-sinus lesioned. All infected sinus showed complete or subtotal heterogeneous opacity. In 62% (12/18) cases the lesion expanded into the adjacent sinus or nasal cavity. 78%(14/18)had calcification spots or points (CT number between 80 Hu and 160 Hu) in the soft tissue window. Bony destruction occurred in 6 cases (33%), in which lamina papyracea and middle turbinates were mostly destructed simultaneously each in 2 cases. Chronic invasive fungal rhinosinusitis had more poly-sinus lesioned cases and bony destructed cases than noninvasive fungal rhinosinusitis and its bone lesion degree was higher too. CONCLUSION: Chronic invasive fungal rhinosinusitis in early stage has more poly-sinus lesioned cases and higher bony destruction ratio and degree. But its total CT characteristics are similar with noninvasive fungal rhinosinusitis. So sinus CT scan has no specific meaning in diagnosing of chronic invasive rhinosinusitis in early stage.
