Super-enhancer trapping by the nuclear pore via intrinsically disordered regions of proteins in squamous cell carcinoma cells.

Master transcription factors such as TP63 establish super-enhancers (SEs) to drive core transcriptional networks in cancer cells, yet the spatiotemporal regulation of SEs within the nucleus remains unknown. The nuclear pore complex (NPC) may tether SEs to the nuclear pore where RNA export rates are maximal. Here, we report that NUP153, a component of the NPC, anchors SEs to the NPC and enhances TP63 expression by maximizing mRNA export. This anchoring is mediated through protein-protein interaction between the intrinsically disordered regions (IDRs) of NUP153 and the coactivator BRD4. Silencing of NUP153 excludes SEs from the nuclear periphery, decreases TP63 expression, impairs cellular growth, and induces epidermal differentiation of squamous cell carcinoma. Overall, this work reveals the critical roles of NUP153 IDRs in the regulation of SE localization, thus providing insights into a new layer of gene regulation at the epigenomic and spatial level.

[Emergence of AmpC enzyme and antibiotics resistance in gram-negative bacilli].

OBJECTIVE: To investigate the relationship between emergence of AmpC enzyme and drug resistance in the gram-negative bacilli in Xiangya Hospital and to provide information for the treatment of antibiotics. METHODS: The bacteria were identified; the susceptibility was determined by Microscan microbiological identification system; and the AmpC enzyme was detected by disc diffusion method. RESULTS: Of the 204 gram-negative strains, the positive rate of AmpC enzyme was 19.61%; AmpC-positive strains resisted to ceftriaxone, piperacillin, ceftazidime, cefotaxime, cefoperazone, aztreonam, piperacillin/tazobactam, cefuroxime, ceftazidime/clavulanic, and cefotaxime/clavulanic. But AmpC-negative strains were susceptible to those antibiotics. There were significant differences, between the two groups (All P < 0.05). Both groups were susceptible to cefepime, imipenem, meropenem, cefoperazone/sulbactam, levofloxacin, and amikacin. CONCLUSION: The detection of AmpC enzyme is helpful to the screen of drug resistant strains. Most bacilli with AmpC enzyme show the resistance to third-generation cephosporins, aztreonam, and the beta-lactamase inhibitors, but susceptible to imipenem, cefepime, meropenem, cefoperazone/sulbactam, levofloxacin, and amikacin, which provides the information for the antibiotic therapy.

[Determination of three nitroimidazole residues in royal jelly by high performance liquid chromatography-tandem mass spectrometry].

A method for analysis of trace metronidazole (MTZ), dimetridazole (DMZ) and ronidazole (RNZ) residues in royal jelly was developed by high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). After samples were dissolved in sodium hydroxide solution to disassociate target analytes from matrix, liquid-liquid extraction methods by ethyl acetate solvent were used. Matrix effects were minimized and good quantitation results were obtained by using highly-selective reaction monitoring (H-SRM) technology when deuterated dimetridazole (dimetridazole-D3) was selected as internal standard. Limits of detection (LODs) were 1.0 microg/kg for DMZ, 0.5 microg/kg for MTZ and RNZ (S/N > 5). Limits of quantitation (LOQs) were 2.0 microg/kg for DMZ, 1.0 microg/kg for MTZ and RNZ (S/N > 10). The linear ranges were 2.0 - 200 microg/L for all target analytes. Recoveries and relative standard deviations (RSDs) were in the ranges of 96.6% - 110.6% and 2.1% -7.4%, respectively. This method is suitable for statutory residue testing in the National Residue Surveillance Plan in China and meets the requirement for export.

[Determination of metabolites of nitrofuran antibiotics in royal jelly by high performance liquid chromatography-tandem mass spectrometry].

Nitrofurans are a group of widely used veterinary antibiotics that have been banned in many countries. This has generated a great deal of interests and demands for assay of nitrofurans in animal food products. To our knowledge, this is the first time that a high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) method has been successfully developed for simultaneously analyzing the metabolites of four nitrofuran antibiotics (furazolidone, furaltadone, nitrofurazone and nitrofurantoin) in royal jelly. Trichloroacetic acid solution was used both to precipitate proteins and to provide acidic reaction condition. Four isotope internal standards were utilized to improve the quantitative precision. The limits of detection (LODs) were 0.03 microg/kg for the metabolite of furaltadone and 0.05 microg/kg for the other three metabolites. The limits of quantitation were 0.20 microg/kg for the metabolite of furaltadone and 0.25 microg/kg for the other three metabolites. The linear range was 0.4-20 ng/mL for all the target analytes. The recoveries calibrated by internal standard were in the range of 97.7%-104.8 with the relative standard deviations (RSDs) of 2.7-9.7. It showed that this method could meet the requirements of national monitoring plan in China and the Minimum Required Performance Limits (MRPL) set by the European Union.

[Determination of lincosamide residues in honey using high performance liquid chromatography-electrospray tandem mass spectrometry].

An analytical method for the determination of lincosamide antibiotic residues in honey was developed. In the procedure a solid-phase extraction for the isolation of lincomycin and clindamycin from diluted honey samples was employed. The residues of lincomycin and clindamycin were subsequently analyzed by reversed-phase high performance liquid chromatography (RP-HPLC) coupled with electrospray tandem mass spectrometry. Mass spectral acquisition was applied with selective reaction monitoring of two diagnostic transition reactions. The qualitative analysis was based on the retention time, the precursor ion and two product ions, and the quantitation was carried out with intension of the characteristic ion m/z 126. The linear ranges were from 1.0 microg/L to 200 microg/L for lincomycin and clindamycin and the correlation coefficients (r2) were all greater than 0.996. The average recoveries for lincomycin and clindamycin ranged from 80% to 110% in replicate sets of honey samples spiked with the drug concentrations of 1.0, 5.0 and 20.0 microg/kg, and the relative standard deviations (RSDs) were less than 8% for intra-day and 15% for inter-day determinations. The method detection limit and quantitation limit were 0.1 micro/kg and 0.5 microg/kg, respectively. The method is simple and suitable for routine analysis.

Effects of different ingredients of zedoary on gene expression of HSC-T6 cells.

AIM: To investigate the effects of four different ingredients of zedoary (Curcuma aromatica oil, Curcumol, beta-elemence, and Curcumin) on the gene expressions of hepatic stellate cells (HSCs), and to explore the molecular mechanism of zedoary against hepatic fibrosis at gene network level. METHODS: We detected the mRNA sequences of 50 liver fibrosis-related genes in GenBank and designed oligonucleotide probes. We synthesized oligonucleotides with PE8909 DNA synthesizing instrument, and carried out oligonucleotide microarray with OGR-04 dropping instrument and aldehyded glass chip. Cultured HSC-T6 cells were treated with different concentrations of Colchicine, Curcuma aromatica oil, Curcumol, beta-elemence, and Curcumin. According to the experiment of cell toxicity, we took the appropriate concentrations of medicines that resulted in over 50% of cell survival as experiment concentrations. We collected the cells at 1, 6, 12, and 24 h, and extracted total RNA with TRIzol reagent, then labeled cDNAs with Cy3-dUTP and Cy5-dUTP. These labeled cDNAs were hybridized to an oligonucleotide microarray which was washed several times and scanned by scanner GenePix 4000B. Different gene expressions of HSC-T6 cells were analyzed by ImaGene 4.2 software. RESULTS: After HSC-T6 cells were cultured in a medium containing 6.25 microg/mL Colchicine for 12 h, expression of TIMP-1 decreased 2.2-folds. After HSC-T6 cells were cultured in a medium containing 78.125 microg/mL of Curcuma aromatica oil for 24 h, the expression of TIMP-2 and IL-6 decreased 2.3- and 2.2-folds, respectively. Moreover, after HSC-T6 cells were cultured in a medium containing 1.5625 microg/mL of Curcumol for 12 h, the expression of TGFbeta1 and P450a decreased 2.3- and 2.1-folds, respectively. CONCLUSION: Our results may show the possible molecular mechanism of Curcuma aromatica oil and Curcumol against hepatic fibrosis.

The HMX homeodomain protein MLS-2 regulates cleavage orientation, cell proliferation and cell fate specification in the C. elegans postembryonic mesoderm.

The proper formation of a complex multicellular organism requires the precise coordination of many cellular events, including cell proliferation, cell fate specification and differentiation. The C. elegans postembryonic mesodermal lineage, the M lineage, allows us to study mechanisms coordinating these events at single cell resolution. We have identified an HMX homeodomain protein MLS-2 in a screen for factors required for M lineage patterning. The MLS-2 protein is present in nuclei of undifferentiated cells in the early M lineage and in a subset of head neurons. In the M lineage, MLS-2 activity appears to be tightly regulated at the fourth round of cell division, coincident with the transition from proliferation to differentiation. A predicted null allele of mls-2, cc615, causes reduced cell proliferation in the M lineage, whereas a semi-dominant, gain-of-function allele, tm252, results in increased cell proliferation. Loss or overexpression of mls-2 also affects cleavage orientation and cell fate specification in the M lineage. We show that the increased cell proliferation in mls-2(tm252) mutants requires CYE-1, a G1 cell cycle regulator. Furthermore, the C. elegans Myod homolog HLH-1 acts downstream of mls-2 to specify M-derived coelomocyte cell fates. Thus MLS-2 functions in a cell type-specific manner to regulate both cell proliferation and cell fate specification.

[Relationship between species distribution and habitat effective temperature in subalpine meadow of Mt. Xiaowutai].

By the methods of sugar-inversion, plant community field survey and laboratory analysis, this paper measured the effective temperature of habitats, relative coverage of species, and aboveground biomass in subalpine meadow of Mt.Xiaowutai, and analyzed the relationship between species distribution and habitat effective temperature in landscape scale. The results showed that among 97 vascular plant species registered, the distribution patterns of about 36 % of total species were influenced by the effective temperature of the habitats, and the relative coverage of about 20% of them correlated significantly with the effective temperature. Eight species showed a negative correlation between their relative coverage and effective temperature, and eleven species had a positive correlation between their relative coverage or aboveground biomass and effective temperature. The eleven species with a positive correlation had certain drought resistance feature or a tendency distributing towards warmer habitats. Concerning the effectiveness of species feature applied in the analysis, relative coverage could be used to determine the relationship between species distribution and effective temperature efficiently, if the species frequency ranged from 90 % to 20 %. But, for the dominant species with their frequency over 90 %, aboveground biomass seemed to be more suitable for the analysis.

Platelet adhesive resistance of polyurethane surface grafted with zwitterions of sulfobetaine.

A possible approach to improve the blood compatibility of poly(etherurethane)s (PU) involves the covalent attachment of key molecular on its surface. Recently, polymer tailed with zwitterions was found having good blood compatibility. The purpose of present study was to design and synthesis a novel nonthrombogenic biomaterial by modifying the surface of poly(etherurethane) with zwitterions of sulfobetaine via HDI spacer. The films of polyurethane were grafted with sulfobetaine by a three-step procedure. In the first step, the film surfaces were treated with hexamethylene diisocyanate (HDI) in toluene at 50 degrees C in the presence of di-n-butyl tin dilaurate(DBTDL) as a catalyst. The extent of the reaction was measured by ATR-IR spectra; a maximum number of free NCO group was obtained after a reaction time of 2.5 h. In the second step, the primary amine group of N,N-diethylethylenediamine (DEA) or N,N-dimethylethylenediamine (DMA) was allowed to react in toluene with isocyanate groups bound on surface. In the third step, two kinds of sulfobetaines were formed in the surface through the ring-opening reaction between tertiary amine of DMA or DEA and 1,3- propanesultone (PS). The reaction process was monitored with ATR-IR spectra and XPS spectra. The wettability of films was investigated by water contact angle measurement. A platelet adhesion experiment was conducted as a preliminary test to confirm the improved blood compatibility of PU. The number of platelets adhering to PU decreased greatly compared to original after 1 h and 3 h of contact with human plate-rich plasma.

Blood compatibility of polyurethane surface grafted copolymerization with sulfobetaine monomer.

Surface modification is an effective way to improve the hemocompatibility and remain bulk properties of biomaterials. Recently, polymer tailed with zwitterions was found having good blood compatibility. In this study, the grafting copolymerization of sulfobetaine onto polyurethane surface was obtained through two steps. In the first step, polyurethane film coupled with vinyl groups was obtained through the reaction between the carboxyl group of acrylic acid (AA) and the NH-urethane group of polyurethane by dicyclohexylcarbodiimide (DCC). In the second step, sulfobetaine was grafted copolymerization on the surface using AIBN as an initiator. The reaction process was monitored with ATR-IR spectra and X-ray photoelectron spectroscopy (XPS) spectra. The wettability of films was investigated by water contact angle measurement. The blood compatibility of the grafted films was evaluated by platelet adhesion in platelet rich plasma (PRP) and protein absorption in bovine fibrinogen (BFG). Low platelet adhesion was observed on the grafted films incubated in PRP for 1 and 3 h, respectively. The protein absorption was reduced on the grafted films after incubated in bovine fibrinogen for 2 h. All of these results revealed that the improved blood compatibility was obtained by grafting copolymerization with zwitterionic monomer of sulfobetaine onto polyurethane film. In addition, introducing vinyl groups onto surface through DCC and AA is a novel method to functionalize polyurethane for further modification.

A real-time PCR assay for the detection and quantitation of Campylobacter jejuni using SYBR Green I and the LightCycler.

Campylobacter jejuni is recognized as a leading human food-borne pathogen. Traditional biochemical identification for C. jejuni is not reliable due to special growth requirements and the possibility that this bacterium can enter a viable but nonculturable (VNC) state. Nucleic acid-based tests have emerged as a useful alternative to traditional testing. In this article, we present fluorescent quantitative PCR assay for quantitative detection of C. jejuni, the assay was carried out using a LightCycler instrument and product formation was monitored continuously with the fluorescent double-stranded DNA binding dye SYBR Green I. When this assay was applied, the assay positive for all of the isolates of C. jejuni tested (11 isolates, including type strain ATCC33560) and negative for all other Campylobacter spp. (three isolates) and several other bacteria (five species tested). The total assay could be completed in 60 min with a detection limit of approximately 1 CFU, and a correlation coefficient was 1.000. Result indicated that fluorescent quantitative detection methods provided a special, sensitive, rapid, reproducible and accurate method for quantitative detection of C. jejuni.

Application of real-time PCR for quantitative detection of Campylobacter jejuni in poultry, milk and environmental water.

Campylobacter jejuni is a leading human food-borne pathogen. The rapid and sensitive detection of C. jejuni is necessary for the maintenance of a safe food/water supply. In this article, we present a real-time polymerase chain reaction (PCR) assay for quantitative detection of C. jejuni in naturally contaminated poultry, milk and environmental samples without an enrichment step. The whole assay can be completed in 60 min with a detection limit of approximately 1 CFU. The standard curve correlation coefficient for the threshold cycle versus the copy number of initial C. jejuni cells was 0.988. To test the PCR system, a set of 300 frozen chicken meat samples, 300 milk samples and 300 water samples were screened for the presence of C. jejuni. 30.6% (92/300) of chicken meat samples, 27.3% (82/300) of milk samples, and 13.6% (41/300) of water samples tested positive for C. jejuni. This result indicated that the real-time PCR assay provides a specific, sensitive and rapid method for quantitative detection of C. jejuni. Moreover, it is concluded that retail chicken meat, raw milk and environmental water are commonly contaminated with C. jejuni and could serve as a potential risk for consumers in eastern China, especially if proper hygienic and cooking conditions are not maintained.

Medical diagnosis with C4.5 Rule preceded by artificial neural network ensemble.

Comprehensibility is very important for any machine learning technique to be used in computer-aided medical diagnosis. Since an artificial neural network ensemble is composed of multiple artificial neural networks, its comprehensibility is worse than that of a single artificial neural network. In this paper, C4.5 Rule-PANE which combines artificial neural network ensemble with rule induction by regarding the former as a preprocess of the latter, is proposed. At first, an artificial neural network ensemble is trained. Then, a new training data set is generated by feeding the feature vectors of the original training instances to the trained ensemble and replacing the expected class labels of the original training instances with the class labels output from the ensemble. Additional training data may also be appended by randomly generating feature vectors and combining them with their corresponding class labels output from the ensemble. Finally, a specific rule induction approach, i.e., C4.5 Rule, is used to learn rules from the new training data set. Case studies on diabetes, hepatitis, and breast cancer show that C4.5 Rule-PANE could generate rules with strong generalization ability, which profits from artificial neural network ensemble, and strong comprehensibility, which profits from rule induction.

Urban garbage disposal and management in China.

This paper, probing into the present situation of urban domestic garbage by analyzing its growing trend, compositional change and regional difference, reveals the problems existing in its disposal and management in China. Meanwhile, a questionnaire was conducted in five big cities around China for surveying urban residents' attitudes towards garbage disposal and management policies and measures. Results showed the output of urban domestic garbage in Chinese cities is ever increasing, and the recoverable materials and energy in garbage composition are also increasing. The population growth, economic development, and increase of residents' expenditure level are the main factors influencing the growing output and changing composition of the garbage. Information acquired from the questionnaire showed that majority of the urban residents are in favor of the garbage reduction policies and managerial measures and are willing to collaborate with municipal government in battling against garbage. Based on the analysis and questionnaire, some policymaking-oriented suggestions such as operating the garbage disposal from a social welfare service to a sector of profit-gaining enterprises, transferring the garbage management from passive end control to active source control, promoting the classified garbage collection in cities around China, and charging garbage fees for its cleanup and disposal, have also been put forward in the paper.
