Diversity and evolution of conotoxins based on gene expression profiling of Conus litteratus.

Cone snails are attracting increasing scientific attention due to their unprecedented diversity of invaluable channel-targeted peptides. As arguably the largest and most successful evolutionary genus of invertebrates, Conus also may become the model system to study the evolution of multigene families and biodiversity. Here, a set of 897 expressed sequence tags (ESTs) derived from a Conus litteratus venom duct was analyzed to illuminate the diversity and evolution mechanism of conotoxins. Nearly half of these ESTs represent the coding sequences of conotoxins, which were grouped into 42 novel conotoxin cDNA sequences (seven superfamilies), with T-superfamily conotoxins being the dominant component. The gene expression profile of conotoxin revealed that transcripts are expressed with order-of-magnitude differences, sequence divergence within a superfamily increases from the N to the C terminus of the open reading frame, and even multiple scaffold-different mature peptides exist in a conotoxin gene superfamily. Most excitingly, we identified a novel conotoxin superfamily and three novel cysteine scaffolds. These results give an initial insight into the C. litteratus transcriptome that will contribute to a better understanding of conotoxin evolution and the study of the cone snail genome in the near future.

SeqVis: visualization of compositional heterogeneity in large alignments of nucleotides.

UNLABELLED: Most phylogenetic methods assume that the sequences evolved under homogeneous, stationary and reversible conditions. Compositional heterogeneity in data intended for studies of phylogeny suggests that the data did not evolve under these conditions. SeqVis, a Java application for analysis of nucleotide content, reads sequence alignments in several formats and plots the nucleotide content in a tetrahedron. Once plotted, outliers can be identified, thus allowing for decisions on the applicability of the data for phylogenetic analysis. AVAILABILITY: http://www.bio.usyd.edu.au/jermiin/programs.htm.

Opposite functions of histamine H1 and H2 receptors and H3 receptor in substantia nigra pars reticulata.

The substantia nigra pars reticulata (SNr) is a key basal ganglia output nucleus. Inhibitory outputs from SNr are encoded in spike frequency and pattern of the inhibitory SNr projection neurons. SNr output intensity and pattern are often abnormal in movement disorders of basal ganglia origin. In Parkinson's disease, histamine innervation and histamine H3 receptor expression in SNr may be increased. However, the functional consequences of these alterations are not known. In this study, whole cell patch-clamp recordings were used to elucidate the function of different histamine receptors in SNr. Histamine increased SNr inhibitory projection neuron firing frequency and thus inhibitory output. This effect was mediated by activation of histamine H1 and H2 receptors that induced inward currents and depolarization. In contrast, histamine H3 receptor activation hyperpolarized and inhibited SNr inhibitory projection neurons, thus decreasing the intensity of basal ganglia output. By the hyperpolarization, H3 receptor activation also increased the irregularity of the interspike intervals or changed the pattern of SNr inhibitory neuron firing. H3 receptor-mediated effects were normally dominated by those mediated by H1 and H2 receptors. Furthermore, endogenously released histamine provided a tonic, H1 and H2 receptor-mediated excitation that helped keep SNr inhibitory projection neurons sufficiently depolarized and spiking regularly. These results suggest that H1 and H2 receptors and H3 receptor exert opposite effects on SNr inhibitory projection neurons. Functional balance of these different histamine receptors may contribute to the proper intensity and pattern of basal ganglia output and, as a consequence, exert important effects on motor control.

Effect of total phenolics from Laggera alata on acute and chronic inflammation models.

The anti-inflammatory effect of total phenolics from Laggera alata (TPLA) was evaluated with various in vivo models of both acute and chronic inflammations. In the acute inflammation tests, TPLA inhibited significantly xylene-induced mouse ear oedema, carrageenan-induced rat paw oedema and acetic acid-induced mouse vascular permeability. In the carrageenan-induced rat pleurisy model, TPLA significantly suppressed inflammatory exudate and leukocyte migration, reduced the serum levels of lysozyme (LZM) and malondialdehyde (MDA), increased the serum levels of superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), and also decreased the contents of total protein, nitric oxide (NO) and prostaglandin E(2) (PGE(2)) in the pleural exudates. In the chronic inflammation experiment, TPLA inhibited significantly cotton pellet-induced rat granuloma. These results indicated that TPLA possesses potent anti-inflammatory activity on acute and chronic inflammation models. Its anti-inflammatory mechanisms are probably associated with the inhibition of prostaglandin formation, the influence on the antioxidant systems, and the suppression of LZM release. Furthermore, the total phenolic content of Laggera alata and its main component type was quantified, and its principle components were isolated and authenticated. Acute toxicity studies revealed that TPLA up to an oral dose of 8.5 g/kg body weight was almost nontoxic in mice.

Biomolecule-assisted synthesis and electrochemical hydrogen storage of Bi2S3 flowerlike patterns with well-aligned nanorods.

Bi2S3 flowerlike patterns with well-aligned nanorods were synthesized using a facile solution-phase biomolecule-assisted approach in the presence of L-cysteine (an ordinary and cheap amino acid), which turned out to serve as both the S source and the directing molecule in the formation of bismuth sulfide nanostructures. Emphatically, no nauseous scent (H2S) appeared in our experiments, which could not be avoided in other previous reports. The morphology, structure, and phase composition of the as-prepared Bi2S3 products were characterized using various techniques (scanning electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, transmission electron microscopy, selected area electron diffraction, and high-resolution transmission electron microscopy). The formation mechanism for the bismuth sulfide flowerlike assemblies with well-arranged nanorods was also discussed. In addition, other Bi2S3 homogeneous nanostructures (e.g., networklike nanoflakes, nanorod-based bundles, and nanoflakes) were obtained through varying the experimental parameters. Interestingly, we have found that these synthesized bismuth sulfide nanostructures using the biomoleucle-assisted approach could electrochemically charge and discharge with the capacity of 142 (mA h)/g (corresponding to 0.51 wt % hydrogen in single-walled carbon nanotubes) under normal atmosphere at room temperature. A novel two-plateau phenomenon was observed in the synthesized Bi2S3 nanostructures, suggesting that there were two independent steps in the charging process. It has been demonstrated that the bismuth sulfide's morphology and the constant charge-discharge current density had a noticeable influence on their capacity of electrochemical hydrogen storage. These differences in hydrogen storage capacity are likely due to the size and density of space/pores as well as the morphology of different Bi2S3 nanostructures. The novel Bi2S3 nanomaterials may find potential applications in hydrogen storage, high-energy batteries, luminescence, optoelectronic and catalytic fields, as well as in the studies of structure-property relationships. This facile, environmentally benign, and solution-phase biomolecule-assisted method can be potentially extended to the preparation of other metal chalcogenides including FeS, CuS, NiS, PbS, MnS, and CoS nanostructures.

Electrochemical properties of self-assembled monolayers of polyaniline: effects of the thiol substituent and reduced dimensionality.

Self-assembled monolayers and bulk films of thioanilines, polymerized on gold and platinum surfaces, have been characterized and compared to bulk polyaniline (PANI) films. In a previous study [Kuwabata; et al. Langmuir 1999, 15, 6807-6812], only one redox couple was observed in the cyclic voltammetric profile of a polymerized monolayer of thioaniline on gold, in contrast to the known profiles of bulk PANI, which exhibit two couples. We observe two couples in both a polymerized thioaniline monolayer and a bulk polythioaniline (S-PANI) film, but the 200 mV window between the couples (the width of the region of high conductivity) in the S-PANI films is much smaller than the 600 mV window in bulk PANI. We ascribe this difference to the presence of the thiol substituent. The windows of high conductivity of the polymerized thioaniline monolayer and the bulk S-PANI film are the same within the limits of our experiment, implying that the difference in the dimensionality of the films (a 2D monolayer vs 3D bulk films) has a limited effect on the films' voltammetric profiles.

Antidepressant-like effects of PDE4 inhibitors mediated by the high-affinity rolipram binding state (HARBS) of the phosphodiesterase-4 enzyme (PDE4) in rats.

RATIONALE: Phosphodiesterase-4 (PDE4) has two conformation states based on rolipram binding, the high-affinity rolipram binding state (HARBS) and the low-affinity rolipram binding state (LARBS); their functions remain to be fully explained. OBJECTIVE: Experiments were carried out to determine the roles of the HARBS and LARBS in the mediation of antidepressant-like effects on behavior. MATERIALS AND METHODS: Two animal models sensitive to antidepressant drugs, the forced-swim test (FST), and the differential-reinforcement-of-low-rate (DRL) 72-s operant schedule, were used to examine the antidepressant-like effects of rolipram, CDP840, and piclamilast, PDE4 inhibitors that interact differentially with the HARBS and LARBS, and MEM1018 and MEM1091, two novel PDE4 inhibitors. Drug discrimination vs rolipram and rolipram competition binding assays also were carried out. RESULTS: In the FST, rolipram and piclamilast, both at 0.1 mg/kg, produced an antidepressant-like effect, i.e., reduced immobility and increased swimming, whereas, 1 mg/kg of CDP840 or 0.5 mg/kg of MEM1018 or MEM1091 was required to produce a similar effect. Consistent with this, only rolipram and piclamilast produced antidepressant-like effects in rats under the DRL schedule of reinforcement, as evidenced by decreased response rates and increased reinforcement rates. In addition, in rats trained to discriminate rolipram from its vehicle, only rolipram and piclamilast substituted. Finally, [(3)H]rolipram and [(3)H]piclamilast binding analysis revealed that CDP840 and the two novel PDE4 inhibitors MEM1018 and MEM1091 exhibited a lower affinity for the HARBS than did rolipram. CONCLUSION: These results suggest that the HARBS of PDE4 is the primary conformation important for antidepressant-like effects on behavior.

Hepatoprotective effect of total flavonoids from Laggera alata against carbon tetrachloride-induced injury in primary cultured neonatal rat hepatocytes and in rats with hepatic damage.

The hepatoprotective activities of total flavonoids of Laggera alata (TFLA) were evaluated by carbon tetrachloride (CCl(4))-induced injury in primary cultured neonatal rat hepatocytes and in rats with hepatic damage. In vitro, TFLA at a concentration range of 1-100 microg/ml improved cell viability and inhibited cellular leakage of two enzymes, hepatocyte aspartate aminotransferase (AST) and alanine aminotransferase (ALT), caused by CCl(4). In vivo, oral treatment with TFLA at doses of 50, 100, and 200 mg/kg significantly reduced the levels of AST, ALT, total protein, and albumin in serum and the hydroxyproline and sialic acid levels in liver. Histopathological examinations revealed that liver damage were improved when treated with TFLA. Meanwhile, 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide radicals scavenging activities of TFLA were also determinated. To understand the exact components of TFLA responsible for the hepatoprotective effect, nine flavonoid compounds were isolated and identified from TFLA. In conclusion, the present investigation was the first to verify the hepatoprotective effect of L. alata in vitro and in vivo. The hepatoprotective action of TFLA is likely related to its potent antioxidative and anti-inflammatory activity. Neutralizing reactive oxygen species by nonenzymatic mechanisms and enhancing the activity of original natural hepatic-antioxidant enzymes may be the main mechanisms of TFLA against CCl(4)-induced injury.

Potential acidifying capacity of deposition experiences from regions with high NH4+ and dry deposition in China.

Acid rain may cause soil acidification possibly leading to indirect forest damage. Assessment of acidification potential of atmospheric deposition is problematic where dry and occult deposition is significant. Furthermore, uncertainty is enhanced where a substantial part of the potential acidity is represented by deposition of ammonium (NH(4)(+)) since the degree of assimilation and nitrification is not readily available. Estimates of dry deposition based on deposition velocity are highly uncertain and the models need to be verified or calibrated by field measurements of total deposition. Total deposition may be monitored under the forest canopy. The main problem with this approach is the unknown influence of internal bio-cycling. Moreover, bio-cycling may neutralize much of the acidity by leaching of mainly K(+). When the water percolates down into the rooting zone this K(+) is assimilated again and acidity is regenerated. Most monitoring stations only measure deposition. Lacking measurements of output flux of both NH(4)(+) and NO(3)(-) from the soil one cannot assess current net N transformation rates. Assumptions regarding the fate of ammonium in the soil have strong influence on the estimated acid load. Assuming that all the NH(4)(+) is nitrified may lead to an overestimation of the acidifying potential. In parts of the world where dry deposition and ammonium are important special consideration of these factors must be made when assessing the acidification potential of total atmospheric loading. In China dry and occult deposition is considerable and often greater than wet deposition. Furthermore, the main part of the deposited N is in its reduced state (NH(4)(+)). The IMPACTS project has monitored the water chemistry as it moves through watersheds at 5 sites in China. This paper dwells at two important findings in this study. 1) Potassium leached from the canopy by acid rain is assimilated again upon entering the mineral soil. 2) Nitrification apparently mainly takes place in forest floor (H- and O-) horizon as NH(4)(+) that escapes this horizon is efficiently assimilated in the A-horizon. This suggests that the potential acidification capacity of the deposition may be found in the throughfall and forest floor solution by treating K(+) and NH(4)(+), respectively, as acid cations in a base neutralization capacity (BNC) calculation.

Synthesis and infrared and fluorescent properties of rare earth complexes with a new aryl amide podand.

Solid complexes of rare earth nitrates and picrates with a new aryl amide podand, 1,4-bis{[(2'-benzylamino-formyl)phenoxyl]methyl}-naphthaline (L) were synthesized. The complexes were characterized by elemental analysis, IR and molar conductivity measurements. At the same time, the fluorescent properties of the Eu(III) and Tb(III) nitrates and picrates complexes in solid state were also investigated. Under the excitation, these complexes exhibited characteristic emissions of europium and terbium ions. The counter anion factor influencing the fluorescent intensity was also discussed.

Air pollution and lung cancer risks in China--a meta-analysis.

Lung cancer is a serious health problem in China, as in the rest of the world. Many studies have already proved that air pollution as well as other environmental factors can increase the risk of lung cancer. Based on epidemiological studies carried out in China, this paper proposes odds ratios (OR) to evaluate the risk of lung cancer from indoor air pollution for the Chinese population by applying the method of meta-analysis. For domestic coal use for heating and cooking, the pooled OR values are 1.83 (95% CI: 0.62-5.41) and 2.66 (1.39-5.07) for women and both sexes, respectively. For indoor exposure to coal dust, the OR values are 2.52 (95% CI: 1.94-3.28) and 2.42 (1.62-3.63) for women and both sexes, respectively. Cooking oil vapor is another factor increasing lung cancer risk. The OR values are 2.12 (95%CI: 1.81-2.47), 1.78 (1.50-2.12) and 6.20 (2.88-13.32) for nonsmoking women, women, and both sexes, respectively. Regarding environmental tobacco smoke, the pooled OR values are 1.70 (95% CI: 1.32-2.18) and 1.64 (1.29-2.07) for nonsmoking women and both sexes, respectively. Funnel plots with statistical test have been applied to examine the publication bias, and the results implied that the analysis of coal consumption and cooking oil pollution might be affected by publication bias. The meta-analysis results confirm the association between lung cancer and indoor air pollution for the Chinese population.

Evaluation of antiinflammatory activity of the total flavonoids of Laggera pterodonta on acute and chronic inflammation models.

The antiinflammatory effect of the total flavonoids of Laggera pterodonta (TFLP) was evaluated with various in vivo models of both acute and chronic inflammation. In the acute inflammation tests, TFLP significantly inhibited xylene-induced mouse ear oedema, carrageenan-induced rat paw oedema and acetic acid-induced mouse vascular permeability. In the carrageenan-induced rat pleurisy model, TFLP efficiently suppressed inflammatory exudate and leukocyte migration, reduced the serum levels of lysozyme (LZM) and malondialdehyde (MDA), increased the activity of serum superoxide dismutase (SOD), and also decreased the contents of total protein, nitric oxide (NO) and prostaglandin E2 (PGE2) in the pleural exudates. No marked effect of TFLP on the activity of serum glutathione peroxidase (GSH-PX) was observed. In the chronic inflammation experiment, TFLP inhibited cotton pellet-induced rat granuloma. The antiinflammatory mechanisms of TFLP are probably associated with the inhibition of prostaglandin formation, influence on the antioxidant systems and the suppression of LZM release. The acute toxicity study revealed that TFLP was nontoxic in mice up to an oral dose of 7.5 g/kg body weight.

Effect of a Chinese herbal formula, Shi-Bi-Lin, on an experimental model of allergic rhinitis.

BACKGROUND: Jia Wei Cang Er Zi San, a traditional Chinese herbal formula, has been used to treat allergic rhinitis (AR) for several centuries. However, its effect on experimental animal models and its therapeutic mechanism remain unclear. OBJECTIVE: To study the effect of Shu-Bi-Lin, a modified Jia Wei Cang Er Zi San, on an animal model of AR. METHODS: Shu-Bi-Lin was administered to the guinea pig model of AR. Meanwhile, an antihistamine-treated group for the treatment control, an ovalbumin-sensitized and untreated group for the positive control, and a sham-sensitized, sham-challenged group for the sham control were studied in parallel. Symptomatic and some pathophysiologic variables were evaluated. RESULTS: Sneezing and nasal scratching after challenges were significantly ameliorated in the Shu-Bi-Lin-treated group compared with the ovalbumin-sensitized and untreated group, but rhinorrhea volume was not reduced. Shu-Bi-Lin significantly suppressed the production of IgG1 in the passive cutaneous anaphylaxis test. The thromboxane B2 level in nasal lavage fluid was significantly deceased in the Shu-Bi-Lin-treated group; however, the reduction in histamine and peptide leukotriene levels did not reach statistical significance. In addition, eosinophil infiltration and endothelial nitric oxide synthase immunoreactivity in the nasal tissues were reduced in the Shu-Bi-Lin-treated group. CONCLUSIONS: Shu-Bi-Lin could alleviate the nasal symptoms of AR, and its mechanism might be related to its inhibitory effect on type I anaphylaxis reactions and eosinophil infiltration in the nasal tissues, as well as the inhibition of some mediators related to AR.

[Effects of exogenous phytohormones on hairy root growth and biosynthesis of anthraquinones in the hairy root culture of Polygonum multiflorum].

The effects of exogenous phytohormones on hairy root growth and biosynthesis of anthraquinones in the hairy root cultures of Polygonum multiflorum Thunb. were studied. The results showed that the 2,4-D, NAA and 6-BA all have obvious effects on the growth of hairy root cultures and the biosynthesis of anthraquinones. The growth of hairy root and biosynthesis of anthraquinones were strongly restrained by 2,4-D. However, NAA and 6-BA of appropriate concentration were favourable to hairy root growth and anthraquinones production.

Synthesis and cytotoxicity of racemic isodeoxypodophyllotoxin analogues with isoprene-derived side chains.

A series of isodeoxypodophyllotoxin (5) analogues, 26-38, with various isoprene-derived side chains at the E-ring were designed and synthesized. For comparison, compound 39, with a benzyloxy group on the E-ring, and six D-ring opened analogues, 40-45, were also prepared. All the synthetic compounds were evaluated for their cytotoxic activities in vitro against seven cultured human tumor cell lines. Compounds 27, 43, and 44 were more cytotoxic than etoposide on BEL-7404, A549, and HL-60 cell lines, respectively. However, none of the synthetic isodeoxypodophyllotoxins were more cytotoxic than podophyllotoxin (1).
