VEGF C2578A polymorphism does not contribute to amyotrophic lateral sclerosis susceptibility in sporadic Chinese patients.

A linkage and association of the VEGF (vascular endothelial growth factor) C2578A polymorphism and amyotrophic lateral sclerosis (ALS) has been found in some studies. We analysed the C2578A polymorphism in sporadic ALS patients from a Chinese population. The polymorphism was analysed in 115 patients and 200 healthy individuals by amplifying across position 2705 to 2494 of the promoter region of the VEGF gene. It was found that the frequency of the allele A was 24% in ALS patients and 28% in healthy individuals (p = 0.264). Comparing the background of this polymorphism in healthy individuals between Chinese and Caucasians, significant decreases were found in the frequencies of the A/A genotype and allele A (p(s)<0.001). We concluded that VEGF C2578A polymorphism did not confer a susceptibility to sporadic Chinese ALS patients, which was in disagreement with that reported previously in Caucasian populations and might be ascribed to the different genetic background between Chinese and Caucasians.

Ca1.2 and CaV1.3 neuronal L-type calcium channels: differential targeting and signaling to pCREB.

Neurons express multiple types of voltage-gated calcium (Ca2+) channels. Two subtypes of neuronal L-type Ca2+ channels are encoded by CaV1.2 and CaV1.3 pore-forming subunits. To compare targeting of CaV1.2 and CaV1.3 L-type Ca2+ channels, we transfected rat hippocampal neuronal cultures with surface-epitope-tagged sHA-CaV1.2 or sHA-CaV1.3a constructs and found that: (i) both sHA-CaV1.2 and sHA-CaV1.3a form clusters on the neuronal plasma membrane surface; (ii) when compared with sHA-CaV1.2 surface clusters, the sHA-CaV1.3a surface clusters were 10% larger and 25% brighter, but 35% less abundant; (iii) 81% of sHA-CaV1.2 surface clusters, but only 48% of sHA-CaV1.3a surface clusters, co-localized with synapsin clusters; (iv) co-expression with GFP-Shank-1B had no significant effect on sHA-CaV1.2 surface clusters, but promoted formation and synaptic localization of sHA-CaV1.3a surface clusters. In experiments with dihydropyridine-resistant CaV1.2 and CaV1.3a mutants we demonstrated that CaV1.3a L-type Ca2+ channels preferentially mediate nuclear pCREB signaling in hippocampal neurons at low, but not at high, levels of stimulation. In experiments with primary neuronal cultures from CaV1.3 knockout mice we discovered that CaV1.3 channels play a more important role in pCREB signaling in striatal medium spiny neurons than in hippocampal neurons. Our results provide novel insights into the function of CaV1.2 and CaV1.3 L-type Ca2+ channels in the brain.

Serum level of homocysteine is correlated to carotid artery atherosclerosis in Chinese with ischemic stroke.

OBJECTIVES: To investigate the relationship between serum level of homocysteine (Hcy) and carotid artery atheroscleosis (CAA). METHODS: Both sides of the common carotid artery and internal carotid artery in 126 Chinese patients with ischemic stroke were measured by B-mode ultrasound. The patients were divided into groups: normal, A, B, C and D according to the severity of CAAs. With fasting serum, Hcy as well as folate, vitamin B(12) and lipids were detected. The mean +/- SD age was 64 +/- 13 years (range 39-87 years). RESULTS: In a logistic regression model, the Hcy concentrations were associated with an elevated risk of CAAs independent of all traditional risk factors, and when CAAs became severer, the serum Hcy was higher. The levels of the normal group and A, B, C, D groups were 13.22 +/- 6.15 micromol/L, 16.29 +/- 9.81 micromol/l, 19.49 +/- 11.16 micromol/l, 27.21 +/- 17.47 micromol/l, and 24.14 +/- 8.64 micromol/l, respectively. Rank test showed a significant difference between normal and other groups (p<0.05). The levels of folate and vitamin B(12) were negatively correlated with the Hcy concentrations. Spearman correlation coefficient were -0.23 and -0.42 (p<0.05). CONCLUSIONS: Hyperhomocysteinaemia is an independent risk factor of CAAs and the degree of CAAs is highly correlated with the level of Hcy in serum. The causes of hyperhomocysteinemia may be the result from the decrease in folate and vitamin B(12). Clinical trials are now required to evaluate the effect of treatment with these vitamins on the primary and secondary prevention of cerebral vascular diseases.

Study of transcriptional regulation using a reporter gene assay.

Study of gene expression can be facilitated by using a reporter gene assay. Instead of directly measuring the level of target gene mRNA, one can clone the promoter region of the gene of interest in front of a reporter gene and measure the reporter gene expression as a reflection of the expression of the gene of interest. We describe a simple lacZ-fusion system to measure the activity of the reporter gene product beta-galactosidase. Different strategies of making the fusion construct and their applications are also discussed. This method is particularly useful to dissect the promoter region of the gene of interest and is also used in other experimental protocols such as the yeast two-hybrid analysis.

[Study on expression of brain aging-relative genes HSP86 and HSP84 and effects of acupuncture in the SAMP10 mouse].

OBJECTIVE: To probe the mechanism of acupuncture for anti-aging. METHODS: In the senescence accelerated mouse the SAMP10 and the SAMR1, by using RT-PCR and DIG-labeled Northern blot technique, the expression differences of HSP84 and HSP86 genes in whole brain, cortex and hippocampus in the 4 groups,8-month SAMR1 control group, 8-month SAMP10 control group, 8-month SAMP10 acupuncture group and 8-month SAMP10 non-point acupuncture group were investigated. RESULTS: In the SAMP10 control group, the expression of HSP84 and HSP86 were down-regulated in the whole brain, the cortex and the hippocampus, and they were up-regulated after acupuncture, tending to the normal group. CONCLUSION: Brain aging of the SAMP10 mouse is related with abnormal expression of HSP84 and HSP86 genes, and acupuncture can strengthen the protection of cells, inhibit apoptosis and anti-oxidative stress through regulating expression of HSP84 and HSP86, hence anti-aging.

[Effects of Bushen Tiaochong Recipe contained serum on proliferation and secretion of ovarian granulocyte cultured with excessive androgen].

OBJECTIVE: To observe the effect of Bushen Tiaochong Recipe (BTR) on proliferation and secretion of ovarian granulocyte (OGC) cultured with excessive androgen by serum pharmacologic technique. METHODS: Follicular stimulating hormone (FSH) contained and BTR contained (high, moderate or low dose) serum prepared on 6-week-old Sprague Dawley rats by serum pharmacologic method were respectively added into cultured OGC cell-line after treatment with testosterone propionat (TP). Then the cells were incubated for 48 hrs. The quantity of 3 H-thymidine incorporated DNA was determined with a scintillation counter; the cell cycle distribution and proliferation index (PI) were estimated with flow cytometer (FCM); and the contents of estrodial and progestogen in culture fluid and intracellular cAMP were detected by radioimmuoassay. RESULTS: Excessive androgen obviously inhibited the proliferation and secretion of OGC, and reduced the intracellular cAMP. BTR contained serum could improve above-mentioned inhibition of androgen to promote the cell proliferation and secretion of E2 and P in dose-dependant manner. CONCLUSION: BTR could alleviate the inhibitory action of excessive androgen on OGC, the ovarian function may be adjusted by BTR through promoting follicular development, increasing the levels of estrogen and progestogen, improving the ratio of estrogen/androgen.

[Effects of acupuncture on expressions of transcription factors NF-E2, YB-1, LRG47 in the SAMP10 mouse].

OBJECTIVE: To explore the mechanism of acupuncture in delaying aging. METHODS: Using SAMP10 mice and normal control SAMR1 as model and applying RT-PCR and DIG probed Northern blot techniques to observe expression of NF-E2, YB-1, LRG47 genes in whole brain, cortex and hippocampus in the 8-month SAMR1 control group, 8-month SAMP10 control group, 8-month SAMP10 acupuncture group and 8-month SAMP10 non-point acupuncture group. RESULTS: In the SAMP10 control group, the expression of NF-E2, YB-1 and LRG47 were down-regulated in the whole brain, cortex and hippocampus, and after acupuncture they were up-regulated and tended to normal. CONCLUSION: Aging of the SAMP10 mouse brain is related with expression of NF-E2, YB-1 and LRG47 genes, and acupuncture can regulate the expression of NF-E2, YB-1 and LRG47 genes, improving the functions of erythrocyte series, increasing proliferation of cells and immune function of cells in anti-bacteria, hence anti-aging.

[Neutral position magnetic resonance imaging for diagnosis of Hirayama disease].

OBJECTIVE: To investigate the sensitivity and specificity of neutral position cervical MRI in the diagnosis of Hirayama disease. METHODS: Neutral position cervical MRI of 18 patients and 31 young normal control subjects were evaluated for localized lower cervical cord atrophy, asymmetric cord flattening, abnormal cervical curvature, loss of attachment (LOA) and noncompressed intramedullary high signal intensity on T2-weighted MRI. The difference in frequency of these findings between the control and patient groups was examined by means of the x' test. The sensitivity and specificity of these MRI findings in the diagnosis of Hirayama disease were calculated. Multiple logistic regression was performed to explore the relationship with Hirayama disease. RESULTS: (1) There was significant difference in the frequency of localized lower cervical cord atrophy, asymmetric cord flattening and LOA between the patient and control groups (P < 0.05). (2) After adjusting localized lower cervical cord atrophy, asymmetric cord flattening, abnormal cervical curvature and LOA, the analysis showed that LOA was the only significantly important predictor of the disease, with odds ratio of 49.38 (95% CI: 3.97 - 614.73). Sensitivity and specificity of LOA were 88.0% and 90.6% respectively. (3) Sensitivity and specificity of localized lower cervical cord atrophy were 84.6% and 80.6%. Sensitivity and specificity of asymmetric cord flattening were 87.5% and 73.2%, but our case-control study did not fine any significant association between localized lower cervical cord atrophy and Hirayama disease and also between asymmetric cord flattening and Hirayama disease. CONCLUSIONS: LOA between posterior dural sac and subjacent lamina is the most valuable finding in the diagnosis of Hirayama disease at neutral position MR imaging.

Determination of curvature and twist by digital shearography and wavelet transforms.

A new technique based on digital shearography for determining the transient curvature and twist of a continuously deforming object from a series of speckle patterns is presented. The intensity variation of each pixel is analyzed along the time axis by using a complex Morlet wavelet transform. The absolute sign of the phase variation is determined by introduction of a temporal carrier when the speckle patterns are captured by a high-speed camera. A high-quality spatial distribution of the deflection derivative is extracted at any instant without the need for temporal or spatial phase unwrapping. The continuous Haar wavelet transform is subsequently processed as a differentiation operator to reconstruct the instantaneous curvature and twist of a continuously deforming object.

Temperature dependence and thermodynamic properties of Ca2+ sparks in rat cardiomyocytes.

To elucidate the temperature dependence and underlying thermodynamic determinants of the elementary Ca2+ release from the sarcoplasmic reticulum, we characterized Ca2+ sparks originating from ryanodine receptors (RyRs) in rat cardiomyocytes over a wide range of temperature. From 35 degrees C to 10 degrees C, the normalized fluo-3 fluorescence of Ca2+ sparks decreased monotonically, but the Delta[Ca2+]i were relatively unchanged due to increased resting [Ca2+]i. The time-to-peak of Ca2+ sparks, which represents the RyR Ca2+ release duration, was prolonged by 37% from 35 degrees C to 10 degrees C. An Arrhenius plot of the data identified a jump of apparent activation energy from 5.2 to 14.6 kJ/mol at 24.8 degrees C, which presumably reflects a transition of sarcoplasmic reticulum lipids. Thermodynamic analysis of the decay kinetics showed that active transport plays little role in early recovery but a significant role in late recovery of local Ca2+ concentration. These results provided a basis for quantitative interpretation of intracellular Ca2+ signaling under various thermal conditions. The relative temperature insensitivity above the transitional 25 degrees C led to the notion that Ca2+ sparks measured at a "warm room" temperature are basically acceptable in elucidating mammalian heart function.

Profiling of objects with height steps by wavelet analysis of shadow moiré fringes.

A temporal wavelet analysis algorithm is proposed for shadow-moiré-based three-dimensional surface profiling on objects having discontinuous height steps. A grating is positioned close to an object, and its shadow is observed through the grating. The moiré fringe patterns vary when the grating is in-plane rotating. A series of fringe patterns are captured by a CCD camera at different rotating angles. Phase values are evaluated point by point with the continuous wavelet transform. From the phase values of each point on the object, the distance between the object and the grating can be retrieved. The surface profile is obtained without temporal or spatial phase unwrapping. This technique is applicable to objects with discontinuous height steps, which are impossible to measure with conventional shadow moiré topography. Two specimens are tested to demonstrate the validity of the method: One is an object with a height step of 1.6 mm, and another is a small coin with unevenness of less than 0.2 mm. The experimental results are compared with test results by use of the mechanical stylus method.

Wavelet analysis of speckle patterns with a temporal carrier.

A novel temporal phase-analysis technique that is based on wavelet analysis and a temporal carrier is presented. To measure displacement on a vibrating object by using electronic speckle pattern interferometry, one captures a series of speckle patterns, using a high-speed CCD camera. To avoid ambiguity in phase estimation, a temporal carrier is generated by a piezoelectric transducer stage in the reference beam of the interferometer. The intensity variation of each pixel on recorded images is then analyzed along the time axis by a robust mathematical tool, i.e., a complex Morlet wavelet transform. After the temporal carrier is removed, the absolute displacement of a vibrating object is obtained without the need for temporal or spatial phase unwrapping. The results obtained by a wavelet transform are compared with those from a temporal Fourier transform.

Association of CaV1.3 L-type calcium channels with Shank.

Neurons express multiple types of voltage-gated calcium (Ca2+) channels. Two subtypes of neuronal L-type Ca2+ channels are encoded by CaV1.2 and CaV1.3 pore-forming subunits. Both CaV1.2 and CaV1.3 subunits contain class I PDZ (postsynaptic density-95/Discs large/zona occludens-1) domain-binding consensus at their C termini. In yeast two-hybrid screen of rat brain cDNA library with the C-terminal bait of CaV1.3a (long C-terminal splice variant) L-type Ca2+ channel subunit, we isolated multiple clones of postsynaptic adaptor protein Shank. We demonstrated a specific association of CaV1.3a C termini, but not of CaV1.2 C termini, with Shank PDZ domain in vitro. We further demonstrated that the proline-rich region present in C termini of CaV1.3a subunit binds to Shank Src homology 3 domain. We established that CaV1.3a and Shank localized to postsynaptic locations in cultured rat hippocampal neurons. By expressing epitope-tagged recombinant CaV1.3 subunits in rat hippocampal neuronal cultures, we demonstrated that the presence of Shank-binding motifs in CaV1.3a sequence is both necessary and sufficient for synaptic clustering of CaV1.3 L-type Ca2+ channels. In experiments with dominant-negative peptides and dihydropyridine-resistant CaV1.3a mutants, we demonstrated an importance of Shank-binding motif in CaV1.3a sequence for phosphorylated cAMP response element-binding protein (pCREB) signaling in cultured hippocampal neurons. Our results directly link CaV1.3 neuronal L-type Ca2+ channels to macromolecular signaling complex formed by Shank and other modular adaptor proteins at postsynaptic density and provide novel information about the role played by CaV1.3 L-type Ca2+ channels in pCREB signaling.

Two intra-amygdaloid pathways to the central amygdala exhibit different mechanisms of long-term potentiation.

Synaptic plasticity in the amygdala is thought to underlie aversive or rewarding learning and emotional memories. In this study, different mechanisms were found to underlie synaptic plasticity in lateral (LA) and basolateral (BLA) amygdala pathways to the primary output nucleus of the amygdala, the central amygdala (CeA). Specifically, 1) long-term potentiation (LTP) at the BLA-CeA synapses was independent of inhibition and mediated through N-methyl-d-aspartate receptors (NMDARs) and L-type voltage-gated calcium channels (VGCCs), and 2) LTP in the LA-CeA pathway was gated by inhibition and mediated through VGCCs but not NMDARs.

Synthesis of monodentate chiral spiro phosphonites and the electronic effect of ligand in asymmetric hydrogenation.

New monodentate chiral phosphonites were synthesized from enantiomerically pure 1,1'-spirobiindane-7,7'-diol. The phosphonites 2 were efficient ligands for the Rh-catalyzed asymmetric hydrogenation of alpha- and beta-dehydroamino acid derivatives, providing the amino acids in high enantioselectivities. The study of electronic effect showed that the electron-withdrawing substitutent on the P-phenyl ring of the phosphonite ligand dramatically decreased both the reactivity and enantioselectivity of the ligand.
