Identification of the Jak/Stat proteins as novel downstream targets of EphA4 signaling in muscle: implications in the regulation of acetylcholinesterase expression.

Eph receptors and their cognate ligands ephrins are important players in axon guidance and neural patterning during development of the nervous system. Much of our knowledge about the signal transduction pathways triggered by Eph receptors has been related to the modulation of actin cytoskeleton, which is fundamental in mediating the cellular responses in growth cone navigation, cell adhesion, and cell migration. In contrast, little was known about whether long term activation of Eph receptor would regulate gene expression. Here we report a novel signaling pathway of EphA4, which involves activation of the tyrosine kinase Jak2 and the transcriptional activator Stat3. Transfection of COS7 cells with EphA4, but not the kinase-dead mutant, induced tyrosine phosphorylation of Jak2, Stat1, and Stat3. Treatment of cultured C2C12 myotubes with ephrin-A1 also induced tyrosine phosphorylation of Stat3, which was abolished by the Jak2 inhibitor AG490. Moreover, Jak2 was co-immunoprecipitated with EphA4 in muscle, and both proteins were concentrated at the neuromuscular junction (NMJ) of adult muscle. By using microarray analysis, we have identified acetylcholinesterase, the critical enzyme that hydrolyzed the neurotransmitter acetylcholine at the NMJ, as a downstream target gene of the Jak/Stat pathway in muscle. More importantly, ephrin-A1 increased the expression of acetylcholinesterase protein in C2C12 myotubes, which was abolished by AG490. In contrast, ephrin-A1 reduced the expression of fibronectin mRNA in C2C12 myotubes independently of Jak2. Finally, the expression level of acetylcholinesterase in limb muscle of EphA4 null mice was significantly reduced compared with the wild-type control. Taken together, these results have identified Jak/Stat proteins as the novel downstream targets of EphA4 signaling. In addition, the present study provides the first demonstration of a potential function of Eph receptors and Jak/Stat proteins at the NMJ.

Collagen synthesis is not altered in women with stress urinary incontinence.

AIMS: The objective of this study was to demonstrate that weakened pelvic floor support of the lower genitourinary tract in women with stress urinary incontinence (SUI) is due, in part, to decreased collagen synthesis and secretion and/or an altered ratio of collagen III/I synthesis by the fibroblasts of the endopelvic fascia and skin compared to that of women without evidence of pelvic floor weakening. METHODS: Endopelvic fascia and skin biopsies were obtained from women with SUI (n = 14) and women without evidence of SUI or genital prolapse (n = 12). Fibroblast cultures established from the biopsies were incubated with 3H-proline in medium containing ascorbic acid for 3 hr. Conditioned medium was collected and cells were harvested. The radiolabeled collagens were precipitated and digested with collagenase. The collagen synthesized (as a percent of total protein) was determined. Collagen alpha1(III) was separated from collagen alpha1(I) and alpha2(I) by interrupted SDS-PAGE and the amount of (3)H-proline in each band was determined. RESULTS: Collagen synthesis, expressed as percent of total protein synthesis, was not significantly different between fibroblasts obtained from women with or without SUI. The mean of collagen III/I synthesized in fibroblasts was not significantly different between fibroblasts obtained from women with or without SUI. CONCLUSIONS: These data suggest that the lower collagen content in the endopelvic fascia and skin of women with SUI is not due to reduced collagen synthesis or selective reduction in synthesis of either collagen I or collagen III, compared to women without pelvic floor weakening.

Castleman's disease of the urachus.

Castleman's disease (angiofollicular lymphoid hyperplasia) is a rare heterogeneous group of lymphoproliferative disorders of uncertain cause. Most cases occur as mediastinal masses, although extrathoracic involvement has been reported. Castleman's disease involving the genitourinary tract, one of the most infrequent organ systems affected, has so far only been described in pararenal and retroperitoneal locations. We describe a patient with an unusual case of unicentric Castleman's disease that was located in a urachal remnant and bore a malignant appearance. The patient initially presented with abdominal pain and underwent laparoscopic radical excision of the urachal mass. Pathologic examination revealed Castleman's disease, hyaline-vascular type, with atypical lymphoid proliferation and follicular dendritic cell dysplasia. No evidence of concomitant lymphoma or sarcoma was found. Two years after surgical removal of the tumor, no recurrence was evident. To our knowledge, this is the first case of Castleman's disease presenting as a urachal tumor.

A family-based genetic association study of variants in estrogen-metabolism genes COMT and CYP1B1 and breast cancer risk.

In this paper, we report findings from a family-based association study examining the association between polymorphisms in two key estrogen-metabolism genes CYP1B1 (codon 432 G --> C and codon 453 A --> G variants) and COMT (codon 158 G --> A variant) and female breast cancer. We conducted the study among 280 nuclear families containing one or more daughters with breast cancer with a total of 1124 family members (702 with available constitutional DNA and questionnaire data and 421 without). These nuclear families were selected from breast cancer families participating in the Metropolitan New York Registry (MNYR) - one of the six centers of NCI's Breast Cooperative Family Registry. We used likelihood-based statistical methods to examine the allelic associations. We found none of the variant alleles of the CYP1B1 and COMT genes to be associated with breast cancer in these families. This was consistent with results from matched case-control analyses using all available sib-ships in these families. However, we found that parental carrier status of the CYP1B1 codon 453 variant G allele and the COMT codon 158 variant A allele was associated with breast cancer risk in daughters (independent of the daughters' own genotype). In conclusion, findings from this family-based study indicate that a woman's own CYP1B1 or COMT genotypes are not associated with her breast cancer risk. Although the study found that parental carrier status of certain CYP1B1 or COMT genotypes might be associated with daughter's breast cancer risk, the biological basis as well as independent confirmation of this finding need to be investigated in future larger family-based studies before making meaningful inferences.

Validity of a food-frequency questionnaire for a large prospective cohort study in Bangladesh.

We have developed a thirty-nine-item semi-quantitative food-frequency questionnaire (FFQ) to assess the dietary consumption of 11,746 men and women in a prospective cohort study that evaluates the health effects of As from drinking water in Bangladesh. In order to validate the FFQ, two 7 d food diaries (FD) were completed for 189 randomly selected cohort participants in two different seasons of the year. Nutrient values were converted based on both the United States Department of Agriculture's National Nutrient Database and a food composition table for the Indian subcontinent. Pearson product-moment and Spearman non-parametric rank correlation coefficients comparing food and nutrient consumptions estimated from FFQ and 7 d FD were calculated based on log-transformed consumption values with or without adjustment for total energy and correction for within-individual variation. Correlations of macronutrients and common micronutrients including total fat, monounsaturated fat, polyunsaturated fat, saturated fat, protein, carbohydrate, dietary fibre, Na, K, vitamin B6, vitamin B12, riboflavin, Mn, thiamin and Fe were moderately good, ranging from 0.30 to 0.76. However, correlations of other micronutrients were weak (<0.30). Large seasonal variations in intakes of retinol equivalents and vitamin C were observed. This analysis documents the degree of validity of the FFQ in measuring specific nutrient intakes in the study population. To our knowledge, the present study is the first to document the validity of a FFQ with the use of 7 d FD in a Bangladeshi population.

Genome-scale gene function prediction using multiple sources of high-throughput data in yeast Saccharomyces cerevisiae.

Characterizing gene function is one of the major challenging tasks in the post-genomic era. To address this challenge, we have developed GeneFAS (Gene Function Annotation System), a new integrated probabilistic method for cellular function prediction by combining information from protein-protein interactions, protein complexes, microarray gene expression profiles, and annotations of known proteins through an integrative statistical model. Our approach is based on a novel assessment for the relationship between (1) the interaction/correlation of two proteins' high-throughput data and (2) their functional relationship in terms of their Gene Ontology (GO) hierarchy. We have developed a Web server for the predictions. We have applied our method to yeast Saccharomyces cerevisiae and predicted functions for 1548 out of 2472 unannotated proteins.

Osmotic regulation of cell function and possible clinical applications.

Inflammation and immunosuppression can cause acute respiratory distress syndrome, multiple organ failure, and sepsis, all of which are lethal posttraumatic complications in trauma patients. Prevention of the inflammation and immunosuppression has been a main focus of trauma researcher for many years. Recently, hypertonic resuscitation has attracted attention as a possible therapeutic approach to counteract such deleterious immune responses in trauma patients. We have begun to understand how hypertonic fluids affect immune cell signaling, and a number of experimental and clinical studies have started to reveal valuable information on the clinical efficacy and the limitations of hypertonic resuscitation fluids. Knowledge of how osmotic cues regulate immune cell function will enable us to fully exploit the clinical potential of hypertonic resuscitation to reduce inflammatory and anergic complications in trauma patients.

Heat shock treatment protects against angiotensin II-induced hypertension and inflammation in aorta.

Angiotensin II (Ang II) is a potent vasoconstrictor and induces inflammation and end-organ injury through its activation of the proinflammatory transcription factor, nuclear factor-kappaB (NF-kappaB). Heat shock (HS) treatment with subsequent expression of heat shock proteins (Hsps) is an effective strategy for tissue protection against oxidative injuries. Recently, HS and Hsps have been shown to interact with NF-kappaB in tissue injury. In this study, we investigated whether HS could protect against Ang II-induced hypertension and inflammation by inhibiting NF-kappaB. Sprague-Dawley rats were divided into control and HS groups. Control and 24-hour post-heat shocked rats were treated with Ang II. At days 1, 3, 5, 7, 11, and 14 after Ang II administration, systolic blood pressures were measured by tail-cuff plethysmography, and aorta tissues were collected. Aorta NF-kappaB deoxyribonucleic acid-binding activity was measured by electrophoretic mobility shift assay, and NF-kappaB p65 subunit, Hsp70, Hsp27, and interleukin-6 (IL-6) expressions were measured by Western analysis. HS treatment significantly decreased Ang II-induced hypertension. The activation of NF-kappaB in aorta by Ang II was suppressed by HS treatment. The elevated expression of IL-6 induced by Ang II treatment was also decreased by HS treatment. Although Ang II treatment induced an increase in Hsp70 and Hsp27, HS treatment induced a greater elevation of Hsp70 and Hsp27 expression. HS treatment protects against Ang II-induced hypertension and inflammation. This protection may relate to the interaction of Hsps and the NF-kappaB pathway.

NMR studies of protein-nucleic acid interactions.

Protein-DNA and protein-RNA complexes play key functional roles in every living organism. Therefore, the elucidation of their structure and dynamics is an important goal of structural and molecular biology. Nuclear magnetic resonance (NMR) studies of protein and nucleic acid complexes have common features with studies of protein-protein complexes: the interaction surfaces between the molecules must be carefully delineated, the relative orientation of the two species needs to be accurately and precisely determined, and close intermolecular contacts defined by nuclear Overhauser effects (NOEs) must be obtained. However, differences in NMR properties (e.g., chemical shifts) and biosynthetic pathways for sample productions generate important differences. Chemical shift differences between the protein and nucleic acid resonances can aid the NMR structure determination process; however, the relatively limited dispersion of the RNA ribose resonances makes the process of assigning intermolecular NOEs more difficult. The analysis of the resulting structures requires computational tools unique to nucleic acid interactions. This chapter summarizes the most important elements of the structure determination by NMR of protein-nucleic acid complexes and their analysis. The main emphasis is on recent developments (e.g., residual dipolar couplings and new Web-based analysis tools) that have facilitated NMR studies of these complexes and expanded the type of biological problems to which NMR techniques of structural elucidation can now be applied.

Cancer burden from arsenic in drinking water in Bangladesh.

We assessed the potential burden of internal cancers due to arsenic exposure in Bangladesh. We estimated excess lifetime risks of death from liver, bladder, and lung cancers using an exposure distribution, death probabilities, and cancer mortality rates from Bangladesh and dose-specific relative risk estimates from Taiwan. Results indicated at least a doubling of lifetime mortality risk from liver, bladder, and lung cancers (229.6 vs 103.5 per 100 000 population) in Bangladesh owing to arsenic in drinking water.

[Comparative study of kidney-tonifying recipes and spleen-invigorating recipe on regulating pattern of the expression of T lymphocyte apoptosis signaling-related genes in corticosterone-treated rats].

OBJECTIVE: To investigate the regulating pattern of kidney-tonifying recipes and spleen-invigorating recipe on the expression of T lymphocyte apoptosis signaling-related genes in corticosterone-treated rats. METHODS: We analyzed the activation-induced T lymphocyte apoptpsis rate through TUNEL-labelled flow-cytometry assay, quantified the mRNA expression level of T cell apoptosis-regulating genes by using the fluoresecence real-time PCR, analyzed the activity of caspase-8 and caspase-3 by colorimetric assay. RESULTS: (1) T lymphocyte apoptosis rate of corticosterone-treated rats was higher than that of the normal control rats; T lymphocyte apoptosis rates of Youguiyin treated rats and Bushen Yishou Capsule treated rats were lower than that of the corticosterone-treated group; T lymphocyte apoptosis rate of Sijunzi Decoction treated rats displayed no significance compared with that of the corticosterone-treated group. (2) Pro-apoptotic genes such as Fas, FasL, TNFR1, and Bax displayed increased expression level in corticosterone-treated group compared with normal control group. Two kidney-tonifying recipes down-regruated the expression level of FasL and TNFR1, at the same time, both recipes up-regulated the expression level of Bcl-2. The spleen-invigorating recipe had up-regulating effect on the expression level of Bcl-2. And all these results had statistical significance. (3) The activities of caspase-8 and caspase-3 were increased in corticosterone-treated rats compared with normal control; both kidney-tonifying recipes had significant effect on decreasing the activities of these two apoptosis executioners. CONCLUSION: The excessive apoptosis of T cell in corticosterone-treated rats could be effectively modulated with two kidney-tonifying recipes. Down-regulating the expression of FasL and TNFR1 and up-regulating the expression of Bcl-2 as well as decreasing the activities of caspase-8 and caspase-3 may be the unique regulating pattern of tonifying-kidney method with respect to T cell apoptosis related genes in corticosterone-treated rats.

[Transitional CK19 positive cells-a new possible marker of hepatic precancerous lesion].

OBJECTIVE: To investigate the relationship between Transitional CK19 positive cells and hepatic precancerous lesion in chronic hepatitis B patients. METHODS: We observed the expression of CK19 in liver tissue of chronic hepatitis B patients by LSAB immunohistochemical staining, and examined serum AFP and ultrasonography one time per 3 months for one year. RESULTS: We observed a population of CK19 positive cells-with size and structure between those of human oval cells and mature hepatocytes-that usually occurred along with oval-cell proliferation. It was suggested that these transitional cells may partly account for the elevation of serum AFP. One patient occurred hepatic carcinoma, another patient had low-echogenic nodules in liver parenchyma within the 1 year follow-up period. CONCLUSION: Transitional CK19 positive cells could be regarded as a new possible pathological marker of hepatic precancerous lesion.

[Molecular mechanism of epimedium flavonoids in immune homeostasis remodeling in aged rats revealed by lymphocyte gene expression profile].

OBJECTIVE: To elucidate the gene regulatory pattern of Epimedium flavonoids (EF) in immune homeostasis remodeling in the aged rats. METHODS: (1) To quantitatively analyse the apoptosis percentage of lymphocyte in spleen of aged, young and EF treated rats using flow cytometry. (2) To analyse the lymphocyte gene expression profiles of different groups using gene chips (Rat Genome U34A). RESULTS: (1) Detection of lymphocyte apoptosis percentage showed that there was significant difference in comparing between aged group and young group, between EF treated group and aged group (P < 0.01). (2) As compared with that in the young group, in the aged group, 116 genes were up-regulated and 215 down-regulated. As compared with that in the old group, in the EF treated group, 447 genes were up-regulated and 456 down-regulated, which involved the aspects as cell apoptosis and cell proliferation regulation, etc. CONCLUSION: (1) The expression pattern characterized by up-regulation of apoptosis promoting genes expression and down-regulation of apoptosis inhibiting genes expression, is the important gene background of immuno-homeostasis imbalance in the aged. (2) The role of EF is to reverse the abnormal changes of gene expressions with opposite functions, i.e. the apoptosis promoting and inhibiting, proliferation enhancing and antagonizing genes, to reconstruct a beneficial equilibrium of gene expression and thus to further remodel the immuno-homeostasis in the aged.

[Correlative study between expression of BRI gene and metastatic potential in human non-small cell lung cancer].

OBJECTIVE: To investigate the potential relationship between BRI gene expression and metastatic potential in human non-small cell lung cancer (NSCLC). METHODS: Using semi-quantitative reverse-transcription polymerase chain reaction (RT-PCR) and Northern blot hybridization techniques, differential expression of the BRI gene in human lung adenocarcinoma cell lines AGZY-83-a and Anip-973 was investigated. Having a much higher metastatic potential, Anip-973 was isolated from AGZY-83-a parental cell line. In addition, the other 6 non-small cell lung cancer cell lines (SPC-A-1, A549, 95D, TKB-18, GLC-82, PAa) and 30 samples of lung cancer tissues with matched corresponding adjacent normal tissues were also analyzed. RESULTS: There were significant differences in BRI gene expression between the two cell lines. BRI was preferentially expressed in Anip-973 cells compared to its parental cell line AGZY-83-a, and was also up-regulated in the other 6 lung cancer cell lines, correlating possibly with their metastatic potentials. BRI gene over-expression was observed in 30 lung cancer tissues compared with its corresponding adjacent normal tissues. A relative over-expression of BRI mRNA (tumor/normal >or= 2) was observed in 6 of 8 cancer samples with lymph node metastasis and 10 of 22(45.5%) samples without lymph node metastasis. Furthermore, two mRNA transcripts of BRI gene were observed: a 2.0 kb transcript which was mainly observed in normal lung tissues and a 1.6 kb transcript which was present as a dominant species in cancer tissues. CONCLUSION: BRI mRNA expression is significantly up-regulated in NSCLC cell lines and clinical tumor samples. An alternatively spliced 1.6 kb mRNA is a major transcript of the gene in NSCLCs, suggesting that differential RNA processing and expression of BRI gene may play a role in the tumorigenesis and/or be related to the metastatic potential of human lung cancer.

Expression pattern of UidA gene under the control of rice glutelin GluA-2 gene upstream sequence in transgenic rice endosperm.

In order to study the expression pattern of rice glutelin endosperm specific promoter in Chinese cultivar Zhonghua 8 (Oryza sativa L. subsp japonica), UidA gene was fused with rice glutelin GluA-2 gene 5' upstream sequence 2.3 kb and 750 bp upstream respectively and transferred into rice by Agrobacterium mediated transformation. Southern blot indicated that UidA gene was integrated into the genome of transgenic plants as single copy. Northern blot demonstrated that the expression of UidA gene and endogenous GluA-2 gene reached their highest level at 13-15 days and 11-13 days after pollination respectively, and then declined. Histochemical staining of immature transgenic rice seeds showed UidA gene was specifically expressed in endosperm and the highest level GUS expression was observed in aleurone layer. Quantitative analysis of GUS activity showed seeds GUS activity of that 2.3 kb transgenic plant was about two to three folds of those of 750 bp transgenic plant. Sequence analysis suggested that the G-box located in the -2,170 bp (from transcription start site) may be a quantitative cis-element.

[Study on the molecular epidemiology of SHV type beta-lactamase-encoding genes of multiple-drug-resistant acinetobacter baumannii].

OBJECTIVE: To analyse the plasmid-mediated SHV type beta-lactamases-encoding genes sequence and to identify its subtype of multiple-drug-resistant acinetobacter baumannii isolated from Huzhou district, Zhejiang province, China. METHODS: Sixty strains of acinetobacter baumannii were isolated from hospitalized patients between Jul, 2000 and Dec, 2002. Susceptibility of antimicrobial agents and confirmatory tests for Extended-spectrum beta-lactamases (ESBLs) were tested by microdilute method. SHV type beta-lactamases-encoding genes were tested by polymerase chain reaction (PCR). SHV sequences of acinetobacter baumannii HZ02 and HZ10 strains were detected by ABI automated sequencer and were analysed to compare with SHV genes that had been published in GenBank. RESULTS: Eighteen (30.0%) strains of acinetobacter baumannii isolated between Jun, 2001 and Jan, 2002 were carrying SHV beta-lactamases resistant gene of plasmids. Detected SHV sequences of acinetobacter baumannii HZ02 strain and HZ10 strain had 825 and 833 nucleotides respectively and had the same gene sequence as the gene encoding SHV-12 subtype of ESBLs discovered in Switzerland. CONCLUSIONS: Thirty percentage of the clinically isolated acinetobacter baumannii were carrying SHV type (extended-spectrum) beta-lactamases resistant gene of plasmids and causing an outbreak in hospital and was discovered to have carried the strains of SHV-12 subtype producing ESBLs gene in acinetobacter baumannii which was the first reported case in the world.
