Design and synthesis of Mg2+-selective fluoroionophores based on a coumarin derivative and application for Mg2+ measurement in a living cell.

Novel Mg2+ fluorescent molecular probes (KMG-20-AM and KMG-27-AM; where AM is an acetoxymethyl group) based on a coumarin possessing a charged beta-diketone structure were designed and synthesized. These fluorescent probes produced a red shift from 425 to 445 nm in the absorption spectra after formation of a complex with Mg2+. The fluorescence spectra of these probes also showed a red shift from 485 to 495 nm and an increasing fluorescence intensity after formation of a complex with Mg2+. The optimum experimental conditions were excitation wavelength of 445 nm and a monitored wavelength of 500 nm, where these probes functioned as an indicator showing an image of increasing fluorescence in the presence of Mg2+. These probes showed a "seesaw-type" fluorescent spectral change with the isosbestic point at 480 nm due to the light excitation at 445 nm, which indicates that ratiometry can be used for the measurement. The molecular probes formed a 1:1 complex with Mg2+ and the dissociation constant (Kd) was 10.0 mM for KMG-20. The association constants of the probes with Mg2- were approximately 3 times higher than that with Ca2+, which showed that the selectivity of Mg2+ versus Ca2+ for these probes was over 200 times higher than that for commercially available Mg2+ fluorescent molecular probes such as mag-fura-2, Magnesium Green. As an application of these probes, intracellular fluorescent imaging of Mg2+ was demonstrated using a fluorescent microscope. After the addition of KMG-20-AM and KMG-27-AM into PC12 cells, a strong fluorescence was observed in the cytoplasm and a weak fluorescence in the nuclei region. After treatment with a high-K+ medium, the fluorescence intensity increased due to increasing intracellular Mg2+. The real image of Mg2+ release from the magnesium store was successfully observed with these Mg2+ fluorescent probes.

Effects of SK-896, a new human motilin analogue ([Leu13]motilin-Hse), on postoperative ileus in dogs after laparotomy.

The effects of SK-896, a new human motilin analogue ([Leu13]motilin-Hse), on digestive tract motility in postoperative ileus were evaluated in a dog model of ileus after laparotomy. SK-896 was intravenously administered at 0.17, 0.33 and 0.67 microg/kg starting soon after operation and then at 6-h intervals, for a total of 9 times. SK-896 progressively, dose-dependently and significantly increased the duodenal motility from 1 h after operation. The recovery time of the gastrointestinal-interdigestive migrating complex (GI-IMC) activity, which is an indicator of normal gastrointestinal tract activity after laparotomy, was 56.5 +/- 5.0 h in the control group. SK-896 significantly shortened this recovery time. On the other hand, the plasma SK-896 concentrations declined diexponentially after administration, and can be described by a linear pharmacokinetic model within the dose range used. In addition, the pharmacokinetics of SK-896 did not change significantly at any postoperative time. There was no correlation between the plasma SK-896 concentrations and the intensity of duodenal motility, because the activity in the duodenum decreased transiently 13 h after laparotomy and increased with time thereafter. The changes in the activity are considered to reflect the progressive changes in the state of ileus. In conclusion, SK-896 increased the duodenal motility significantly, shortening the recovery time of GI-IMC-like activity in dogs with post-laparotomy ileus. Therefore, it is expected from these results that SK-896 would be useful and effective for the treatment of gastroparalysis after abdominal surgery.

Antiproteinuric effect of KD3-671, an angiotensin II type 1 receptor antagonist, in rats with accelerated passive Heymann nephritis.

The antiproteinuric effect of KD3-671 (2-propyl-8-oxo-1-[(2'-(H-tetrazole-5-yl)biphenyl-4-yl)methyl]-4,5,6,7-tetrahydrocycloheptimidazole), an angiotensin II type 1 receptor antagonist, was compared with that of enalapril, an angiotensin 11-converting enzyme inhibitor, using an experimental model of membranous nephropathy. KD3-671 (3, 10 and 30 mg/kg per day) and enalapril (30 mg/kg per day) were given p.o. for 40 days, respectively. KD3-671 (30 mg/kg per day) inhibited the elevation of proteinuria and plasma total cholesterol. On the other hand, enalapril showed only a tendency to diminish these parameters. KD3-671 had an antiproteinuric effect in rats with accelerated passive Heymann nephritis. These findings provide considerable encouragement for the clinical development of KD3-671.

Induction of cyclooxygenase-2 expression in glomeruli by aggregated protein.

Cyclooxygenase has two isozymes, a constitutive type (cyclooxygenase-1) and an inducible type (cyclooxygenase-2). The aim of the present study was to determine whether cyclooxygenase-2 is associated with the increased production in prostaglandin E2 in glomeruli by aggregated protein. Mice were injected with aggregated bovine serum albumin. Glomeruli were isolated using sieves and a magnet. Production of prostaglandin E2 was increased in glomeruli after injection of aggregated bovine serum albumin. RT-PCR analysis indicated enhanced expression of cyclooxygenase-2 mRNA in aggregated bovine serum albumin-loaded glomeruli. Western blotting analysis indicated an increase in cyclooxygenase-2 protein in glomeruli by aggregated bovine serum albumin. Glomeruli were incubated with indomethacin, NS-398 or niflumic acid in the presence of arachidonic acid. Indomethacin resulted in remarkable reduction of prostaglandin E2 levels in aggregated bovine serum albumin-loaded glomeruli. Niflumic acid also inhibited prostaglandin E2 production, and its inhibitory rate was more than that of NS-398. In conclusion, aggregated protein induces cyclooxygenase-2 in glomeruli, suggesting that cyclooxygenase-2 is involved in the process of disposal of aggregated protein in glomeuli.

Immunolocalization of multispecific organic anion transporters, OAT1, OAT2, and OAT3, in rat kidney.

Recently, a family of multispecific organic anion transporters has been identified, and several isoforms have been reported. However, the physiologic and pharmacologic roles of each isoform, except OAT1, in the transepithelial transport of organic anions in the kidney remain to be elucidated. To address this issue, it is essential to determine the intrarenal distribution and membrane localization of each OAT isoform along the nephron. In this study, the intrarenal distributions of rOAT1, rOAT2, and rOAT3 were investigated by an immunofluorescence method that used frozen rat serial kidney sections. Confocal microscopic analysis showed that immunoreactivity for rOAT1 was detected exclusively in the proximal tubules (S1, S2, S3) in the cortex with basolateral membrane staining. rOAT2 was detected in the apical surface of the tubules in the medullary thick ascending limb of Henle's loop (MTAL) and cortical and medullary collecting ducts (CD). rOAT3 was localized in the basolateral digitation of the cell membrane in all the segments (S1, S2, and S3) of the proximal tubules, MTAL, cortical TAL, connecting tubules, and cortical and medullary CD. These results on the distribution of each OAT isoform will facilitate the understanding of the role of OATs in the renal processing of organic anions.
