Integrated optical filter arrays fabricated by using the combinatorial etching technique.

A combinatorial etching technique is developed to fabricate integrated narrow bandpass filters on a single substrate. It is highly efficient for fabrication of integrated filter arrays in optical regions. A monolithic filter array has been fabricated by using the technique with a two-step deposition process. The filter contains 32 elements in the near-infrared region. The relative full width at half-maximum (FWHM) sigmalambda/lambda of the filter elements is less than 0.2%. Such a narrow bandpass filter array can be utilized in many optical applications.

LFA-1 and VLA-4 involved in human high proliferative potential-endothelial progenitor cells homing to ischemic tissue.

Cumulative evidences have revealed that endothelial progenitor cell (EPC) transplantation can promote the neovascularization in ischemic tissue, but the mechanism of EPCs homing to the site of ischemia is poorly understood. In this study, to investigate the mechanism of human umbilical cord blood-derived high proliferative potential-endothelial progenitor cells (HPP-EPCs) homing to ischemic tissue we evaluated the expression of lymphocyte function-associated antigen-1 (LFA-1, or CD11a/CD18) and very late antigen-4 (VLA-4, or CD49d/CD29) in EPCs and the changes of expression level of their ligands, intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1), in ischemic tissue and performed the adhesion and migration assays to analyze the interaction between the receptors and ligands. Furthermore, we studied the roles of LFA-1 and VLA-4 in EPC homing in an ischemic model of mice. The results show that LFA-1 andVLA-4 were expressed in HPPEPCs and ICAM-1 and VCAM-1 were expressed in vessel endothelium in ischemic tissues. The pre-incubation of HPP-EPCs with neutralizing antibodies against CD11a or CD49d reduced adhesion and migration of HPP-EPCs in vitro and reduced recovery of hind-limb blood flow, capillary density and incorporation of HPP-EPC into ischemic tissues in vivo. Furthermore, the pre-incubation of HPP-EPCs with the combination of CD11a and CD49d antibodies led to synergistically negative effects on adhesion and transmigration of HPP-EPCs in vitro, and on the homing of HPP-EPCs to ischemic tissue and on neovascularization capacity in vivo. These results indicate that LFA-1 andVLA-4 are involved in HPP-EPC homing to ischemic tissues.

Zinc(II)-selective ratiometric fluorescent sensors based on inhibition of excited-state intramolecular proton transfer.

To develop a zinc(II)-selective emission ratiometric probe suitable for biological applications, we explored the cation-induced inhibition of excited-state intramolecular proton transfer (ESIPT) with a series of 2-(2'-benzenesulfonamidophenyl)benzimidazole derivatives. In the absence of Zn(II) at neutral pH, the fluorophores undergo ESIPT to yield a highly Stokes' shifted emission from the proton-transfer tautomer. Coordination of Zn(II) inhibits the ESIPT process and yields a significant hypsochromic shift of the fluorescence emission maximum. Whereas the paramagnetic metal cations Cu(II), Fe(II), Ni(II), Co(II), and Mn(II) result in fluorescence quenching, the emission response is not altered by millimolar concentrations of Ca(II) or Mg(II), rendering the sensors selective for Zn(II) among all biologically important metal cations. Due to the modular architecture of the fluorophore, the Zn(II) binding affinity can be readily tuned by implementing simple structural modifications. The synthesized probes are suitable to gauge free Zn(II) concentrations in the micromolar to picomolar range under physiological conditions.

[Insulin mitigates the effect of free fatty acid to cause endothelial dysfunction in rat aortic rings].

OBJECTIVE: To investigate whether free fatty acid (FFA) directly and independently causes endothelial dysfunction and whether enhancing insulin action mitigates FFA-induced endothelial dysfunction. METHODS: Twenty-nine S-D rats were divided into 4 groups and infused by jugular catheterization with different solutions: 20% intralipid + heparin (0.72 IU/min) for 4 hours (FFA group, n = 9), short-acting insulin by hyperinsulinemic euglycemic clamp for 4 hours (Ins group, n = 5), intralipid + heparin and insulin by hyperinsulinemic euglycemic clamp for 4 hours (Ins + FFA group, n = 10), and normal saline (18 microliter/min) for 4 hours(C group, n = 5). After the infusion, the rats were killed and their aortic rings were resected and put into organ bath and connected to a force transducer and amplifier. PGF2a was added into the organ bath to constrict the rings until the constriction tension reached a balance. Then acethylcholine (Ach) and sodium nitroprusside (SNP) of increasing concentrations were added to relax the rings to record the endothelial dependent vasodilatation (EDV) and endothelium independent vasodilatation (EIV). RESULTS: The EDV value was 85.0% +/- 3.2% in C group and 54.8% +/- 2.5% in FFA group (C vs FFA, P = 0.002); 80.6% +/- 1.8% in Ins group (C vs Ins, P = ns); 72.8% +/- 2.1% in Ins + FFA group (FFA + Ins vs C P = 0.02, FFA + Ins vs FFA, P = 0.005). The EIV response in the three experimental groups was not different from that in the control group. CONCLUSION: FFA damages the endothelisal dependant vasodilatation of aorta, thus causing endothelial dysfunction by direct interaction with the endothelium. FFA may be proatherogenic. Hyperinsulinemia has salutary effects on endothelial function in the context of elevated FFA.

Effect of acupuncture on free radicals in rats with early experimental spinal cord injury.

Effect of acupuncture on free radicals after spinal cord injury was observed in rats with experimental spinal cord injury (SCI). Results indicated that within 24 hours after SCI malondialdehyde (MDA) increased progressively, 2 hours after SCI it reached the peak; and the superoxide dismutase (SOD) activity decreased significantly at the same hours, the decrease being the most marked 2-6 hours after SCI. The MDA content in the acupuncture group was significantly lower (P < 0.05) and the SOD activity higher (P < 0.01) than that of the control group respectively. It is suggested that acupuncture inhibits production of MDA and increases the SOD activity.
