[Studies on the changes in rice straw composition in relay treatment of chemical-microbial process by FTIR spectroscopy].

Direct burning of crop straw in the field has given or is giving rise to a serious pollution of atmosphere. The difficult decomposing of the crop straw by soil microorganisms is one of the reasons the crop straw is not popularly used in agriculture. Fourier transform infrared spectroscopy (FTIR) was used to study the changes in straw composition during the relay treatment of chemical-microbial process. The results showed that the method of FTIR spectra could indicate the changes in straw composition during the treatment processes. After the relay treatment of chemical-microbial process, the contents of cellulose, semi-cellulose, and silicon, and C/N ratio were decreased significantly, while the water soluble substances were increased, which was in accordance with the results of chemical analysis. The method to treat crop straw proposed in this paper could provide a practicable way in agricultural utilization of crop straw.

Enantioselective responses to a phosphorothioate analogue of lysophosphatidic acid with LPA3 receptor-selective agonist activity.

The metabolically stabilized LPA analogue, 1-oleoyl-2-O-methyl-rac-glycerophosphothioate (OMPT), is a potent agonist for the LPA(3) G-protein-coupled receptor. A new enantiospecific synthesis of both (2R)-OMPT and (2S)-OMPT is described. Calcium release assays in both LPA(3)-transfected insect Sf9 and rat hepatoma Rh7777 cells showed that (2S)-OMPT was 5- to 20-fold more active than (2R)-OMPT. Similar results were found for calcium release, MAPK and Akt activation, and IL-6 release in human OVCAR3 ovarian cancer cells.

Synthesis of migration-resistant hydroxyethoxy analogues of lysophosphatidic acid.

[reaction: see text] The susceptibility of lysophosphatidic acid (LPA) to intramolecular acyl migration impedes the determination of specific receptor activation by the sn-1 and sn-2 LPA regioisomers. An efficient enantioselective synthesis of hydroxyethoxy (HE)-substituted analogues of sn-1-acyl and 2-acyl LPA derivatives that possess palmitoyl and oleoyl chains is described. While the palmitoyl derivatives fail to activate calcium release in cells transfected with LPA(2) or LPA(3) G-protein-coupled receptors, the LPA(3) receptor is activated by both 1-HE and 2-HE oleoyl LPA analogues with a potency 10-fold lower than that of the parent oleoyl LPA.

High-quality-factor Bragg onion resonators with omnidirectional reflector cladding.

We propose to approximate a spherically symmetric Bragg resonator in an onionlike geometry. We develop a transfer-matrix theory for calculation of the quality factors and analyze the effect of the onion stem on cavity Q factors. We find that it is possible to achieve significant inhibition of spontaneous emission in an onion resonator with omnidirectional cladding layers.

Engineering transverse Bragg resonance waveguides for large modal volume lasers.

We recently analyzed a new class of laser amplifier based on transverse Bragg reflection. We show that the unique properties of Bragg confinement make it possible through modal loss discrimination to achieve single-transverse-mode operation with transverse modal size that is an order of magnitude larger than in lasers that depend on total internal reflection for transverse confinement.

Loss analysis of air-core photonic crystal fibers.

By using a multipole moment approach, we analyze the loss of an air-core photonic crystal fiber and demonstrate that it is possible reduce the transmission loss that is due to photon radiation leakage through the photonic crystal cladding to a level below 0.01 dB/km, with eight rings of air holes. An analogy is drawn between air-core photonic crystal fiber modes and Bragg fiber modes. The influence of material absorption in the silica glass is discussed.

Systematic design of single-mode coupled-resonator optical waveguides in photonic crystals.

By establishing a direct relation between the dispersion and the field profile of a coupled-resonator optical waveguide (CROW) and those of its constituent cavities, we present a systematic method for the design of a single-mode CROW and for control of its dispersion. The procedure includes the design of a single-mode cavity and control of its frequency by engineering its structure. Then, by chaining these cavities in the proper direction and at an appropriate distance, we achieve the desired dispersion for the CROW.

Interferon gamma repression of collagen (COL1A2) transcription is mediated by the RFX5 complex.

Interferon gamma (IFN-gamma) plays an important physiological role during inflammation by down-regulating collagen gene expression and activating major histocompatibility II (MHC-II) complex. The activation of MHC-II by IFN-gamma requires activation of a trimeric DNA binding transcriptional complex, RFX5 complex, containing RFXB (also called RFXANK or Tvl-1), RFXAP, as well as RFX5 protein. Previously, we demonstrated that RFX5 binds to the collagen transcription start site and represses collagen gene expression (Sengupta, P. K., Fargo, J., Smith, B. D. (2002) J. Biol. Chem. 277, 24926-24937). In this report, we have examined the role of RFXB and RFXAP proteins within the RFX5 complex to regulate collagen gene expression. The data show that all three RFX5 complex proteins are required for maximum repression. Expression of proteins with mutations known to be important for RFX5 complex formation does not repress collagen promoter activity. Two mutated forms of RFX5 act as dominant negative proteins activating collagen expression and reversing IFN-gamma down-regulation of collagen expression in human lung fibroblasts. IFN-gamma increases expression and nuclear translocation of RFX5. RFXB has a naturally occurring splice variant isoform (RFX SV). Interferon increases expression of the long form of RFXB and decreases expression of RFX SV with the same kinetics as collagen gene expression. Overexpression of the splice variant form reverses the IFN-gamma induced collagen repression in human lung fibroblasts. Finally, all three RFX5 complex proteins increase at the collagen transcription start site with IFN-gamma treatment using chromatin immunoprecipitation analysis. Thus, these studies suggest an important role for RFX5 complex in collagen repression.

Lipid phosphate phosphatases regulate lysophosphatidic acid production and signaling in platelets: studies using chemical inhibitors of lipid phosphate phosphatase activity.

Blood platelets play an essential role in ischemic heart disease and stroke contributing to acute thrombotic events by release of potent inflammatory agents within the vasculature. Lysophosphatidic acid (LPA) is a bioactive lipid mediator produced by platelets and found in the blood and atherosclerotic plaques. LPA receptors on platelets, leukocytes, endothelial cells, and smooth muscle cells regulate growth, differentiation, survival, motility, and contractile activity. Definition of the opposing pathways of synthesis and degradation that control extracellular LPA levels is critical to understanding how LPA bioactivity is regulated. We show that intact platelets and platelet membranes actively dephosphorylate LPA and identify the major enzyme responsible as lipid phosphate phosphatase 1 (LPP1). Localization of LPP1 to the platelet surface is increased by exposure to LPA. A novel receptor-inactive sn-3-substituted difluoromethylenephosphonate analog of phosphatidic acid that is a potent competitive inhibitor of LPP1 activity potentiates platelet aggregation and shape change responses to LPA and amplifies LPA production by agonist-stimulated platelets. Our results identify LPP1 as a pivotal regulator of LPA signaling in the cardiovascular system. These findings are consistent with genetic and cell biological evidence implicating LPPs as negative regulators of lysophospholipid signaling and suggest that the mechanisms involve both attenuation of lysophospholipid actions at cell surface receptors and opposition of lysophospholipid production.

Synthesis of monofluorinated analogues of lysophosphatidic acid.

Lysophosphatidic acid (LPA, 1- or 2-acyl-sn-glycerol 3-phosphate) displays an intriguing cell biology that is mediated via interactions both with G-protein coupled seven transmembrane receptors and with the nuclear hormone receptor PPARgamma. Synthesis and biological activities of fluorinated analogues of LPA are still relatively unknown. In an effort to identify receptor-selective LPA analogues and to document in detail the structure-activity relationships of fluorinated LPA isosteres, we describe a series of monofluorinated LPA analogues in which either the sn-1 or the sn-2 hydroxy group was replaced by fluorine, or the bridging oxygen in the monophosphate was replaced by an alpha-monofluoromethylene (-CHF-) moiety. The sn-1 or sn-2 monofluorinated LPA analogues were enantiospecifically prepared from chiral protected glycerol synthons, and the alpha-monofluoromethylene-substituted LPA analogues were prepared from a racemic epoxide with use of a hydrolytic kinetic resolution. The sn-2 and sn-1 fluoro LPA analogues were unable to undergo acyl migration, effectively "freezing" them in the sn-1-O-acyl or sn-2-O-acyl forms, respectively. The alpha-monofluoromethylene LPA analogues were unique new nonhydrolyzable ligands with surprising enantiospecific and receptor-specific biological readouts, with one compound showing a 1000-fold higher activity than native LPA for one receptor subtype.

Synthesis of alpha-fluorinated phosphonates from alpha-fluorovinylphosphonates: a new route to analogues of lysophosphatidic acid.

A versatile, efficient method for the preparation of alpha-monofluoromethylene (-CHF-) phosphonates from alpha-fluorovinylphosphonate provides access to a class of lysophosphatidic acid (LPA) receptor-subtype agonists. In addition, sn-2 O-methylation of alpha-monofluoromethylene phosphonates using trimethylsilyldiazomethane generated sn-1-acyl, 2-O-methyl alpha-monofluoromethylene derivatives. Finally, a novel method for the selective etherification of 1,2-diols was developed and a new class of sn-1 O-methyl, 2-acyl alpha-monofluoromethylene LPA analogues was prepared. In vivo breast imaging with diffuse optical tomography based on higher-order diffusion equations.

We report on in vivo absorption and scattering imaging of a human breast cyst and implant, using a reconstruction algorithm based on our third-order diffusion equations. To validate these in vivo images, a series of phantom experiments were conducted, in which we used low-absorbing and low-scattering heterogeneities to mimic a breast cyst or implant. These heterogeneities or targets were composed of pure water or a mixture of water and very dilute Intralipid (0.05% and 0.1%). The phantom experiment confirmed the quantitative imaging capability of our improved algorithm for reconstructing heterogeneities where the conventional diffusion approximation is inadequate. Pilot clinical results from female volunteers indicate that enhanced diffuse optical tomography can quantitatively image findings such as breast cysts or implants in which the absorption and scattering coefficients are usually low.

Minocycline blocks 6-hydroxydopamine-induced neurotoxicity and free radical production in rat cerebellar granule neurons.

Neurotoxicity induced by 6-hydroxydopamine (6-OHDA) is believed to be due, in part, to the production of reactive oxygen species (ROS). Anti-oxidants by inhibiting free radical generation, protect neurons against 6-OHDA-induced neurotoxicity. In this study, we investigated whether or not minocycline, a neuroprotective compound, could directly protect neurons against 6-OHDA-induced neurotoxicity and inhibit 6-OHDA-induced free radical production in cultured rat cerebellar granule neurons (CGN). We now report that exposure of CGN to 6-OHDA (100 microM) resulted in a significant increase in free radical production with death of 86% of CGN. Pretreatment with minocycline (10 microM) for 2 h prevented 6-OHDA-induced free radical generation and neurotoxicity. Furthermore, minocycline also attenuated H(2)O(2)-induced neurotoxicity. Our results suggest that minocycline blocks 6-OHDA-induced neuronal death possibly by inhibiting 6-OHDA-induced free radical generation in CGN. Both the antioxidative and neuroprotective effects of minocycline may be beneficial in the therapy of Parkinson's disease and other neurodegenerative diseases.

Transverse Bragg resonance laser amplifier.

We propose and analyze a new type of optical amplifier that is formed by addition of gain in the periodic cladding of a transverse Bragg resonance waveguide [Opt. Lett. 27, 936 (2002)]. Using the coupled-wave formalism, we calculate the mode profiles, the exponential gain constant, and, for comparison, the gain enhancement compared with those of conventional semiconductor optical amplifiers. In contrast with coupled-mode theory, in one-dimensional structures (e.g., the distributed-feedback laser) the exponential gain constant in the longitudinal direction is involved in both longitudinal and transverse confinement, and its solution has to be achieved self-consistently, together with the quantized guiding channel width.

Catalytic gasification of coal using eutectic salts: recovery, regeneration, and recycle of spent eutectic catalysts.

Catalyst recovery studies were conducted for gasified chars produced from steam gasification of Illinois #6 coal catalyzed with two different catalyst systems. A ternary (43.5 mol% Li2CO3-31.5 mol% Na2COr-25 mol% K2CO3) and a binary (29 mol% Na2CO3-71 mol% K2CO3) eutectic catalyst system were used for gasifying coal. Various extraction schemes, such as water extraction, H2SO4 extraction, and acetic acid extraction, were evaluated with respect to their extraction efficiencies. Effects of major process variables, such as solvent-to-char ratio, mixing time, temperature, and concentration, on the extraction efficiency were evaluated. A process schematic for the entire catalyst recovery, regeneration, and recycle scheme was developed and the preliminary process economics were determined based on these extraction schemes. H2SO4 extraction was found to be the most desirable. It also turned out to be more attractive than a once-through throwaway system.

Mesh-based enhancement schemes in diffuse optical tomography.

Two mesh-based methods including dual meshing and adaptive meshing are developed to improve the finite element-based reconstruction of both absorption and scattering images of heterogeneous turbid media. The idea of dual meshing scheme is to use a fine mesh for the solution of photon propagation and a coarse mesh for the inversion of optical property distributions. The adaptive meshing method is accomplished by the automatic mesh refinement in the region of heterogeneity during reconstruction. These schemes are validated using tissue-like phantom measurements. Our results demonstrate the capabilities of the dual meshing and adaptive meshing in both qualitative and quantitative improvement of optical image reconstruction.

Phase-contrast imaging of tissue using near-infrared diffusing light.

We describe a new model-based image reconstruction method that can extract the spatial variation of tissue refractive index with near-infrared diffusing light. The model of light propagation is a diffusion approximation to the radiative transfer equation for highly scattering media with spatially varying refractive index. The spatial map of refractive index is recovered using a finite element-based Newton-type reconstruction algorithm. The imaging method is validated using several tissue-like phantom experiments. Initial in vivo imaging of breast tissue is also presented, demonstrating the ability of this new imaging method for tumor detection.
