A partially zipped SNARE complex stabilized by the membrane.

The SNARE complex acts centrally for intracellular membrane fusion, an essential process for vesicular transport in cells. Association between vesicle-associated (v-) SNARE and target membrane (t-) SNARE results in the coiled coil core that bridges two membranes. Here, the structure of the SNARE complex assembled by recombinant t-SNARE Sso1p/Sec9 and v-SNARE Snc2p, which are involved in post-Golgi trafficking in yeast, was investigated using EPR. In detergent solutions, SNAREs formed a fully assembled core. However, when t-SNAREs were reconstituted into the proteoliposome and mixed with the soluble SNARE motif of Snc2p, a partially zipped core in which the N-terminal region is structured, whereas the C-terminal region is frayed, was detected. The partially zipped and fully assembled complexes coexisted with little free energy difference between them. Thus, the core complex formation of yeast SNAREs might not serve as the energy source for the fusion, which is different from what has been known for neuronal SNAREs. On the other hand, the results from the proteoliposome fusion assay, employing cysteine- and nitroxide-scanning mutants of Sso1p, suggested that the formation of the complete core is required for membrane fusion. This implies that core SNARE assembly plays an essential role in setting up the proper geometry of the lipid-protein complex for the successful fusion.

Supramolecular aggregates constructed from gold nanoparticles and l-try-CD polypseudorotaxanes as captors for fullerenes.

A series of cyclodextrin-based polypseudorotaxanes (PPRs) were constructed by threading native beta-cyclodextrin or l-tryptophan-modified beta-cyclodextrin onto the amino-terminated PPG chains of different lengths. Subsequently, these PPRs were further assembled to form netlike supramolecular aggregates through the linkage of gold nanoparticles, and the resulting water-soluble Au-PPR aggregates were comprehensively characterized by FT-IR, UV, NMR, fluorescence spectroscopy, powder X-ray diffraction patterning, TG-DTA, and transmission electron microscopy. The results showed that the size and sedimentation rate of the Au-PPR aggregates were mainly dependent on the lengths of the PPG chains. Significantly, the Au-PPR aggregate 8, involving many l-tryptophan residues, showed not only a satisfactory water solubility but also a good capturing ability for fullerenes in aqueous solution. The 8-fullerene conjugate thus formed exhibited a good DNA cleavage ability under light irradiation.

Anisotropy in the wetting of rough surfaces.

Surface roughness amplifies the water-repellency of hydrophobic materials. If the roughness geometry is, on average, isotropic then the shape of a sessile drop is almost spherical and the apparent contact angle of the drop on the rough surface is nearly uniform along the contact line. If the roughness geometry is not isotropic, e.g., parallel grooves, then the apparent contact angle is no longer uniform along the contact line. The apparent contact angles observed perpendicular and parallel to the direction of the grooves are different. A better understanding of this problem is critical in designing rough superhydrophobic surfaces. The primary objective of this work is to determine the mechanism of anisotropic wetting and to propose a methodology to quantify the apparent contact angles and the drop shape. We report a theoretical and an experimental study of wetting of surfaces with parallel groove geometry.

Assembly behavior of inclusion complexes of beta-cyclodextrin with 4-hydroxyazobenzene and 4-aminoazobenzene.

To further reveal the factors governing the supramolecular assembly of beta-cyclodextrin (beta-CD) inclusion complexes, two aggregates (1 and 2) were prepared from the inclusion complexes of beta-CD with 4-hydroxyazobenzene and 4-aminoazobenzene, respectively, and their binding behavior were investigated by means of X-ray analysis, UV-vis, NMR, and circular dichroism spectra in both solution and the solid state. The obtained results indicated that the beta-CD/4-hydroxyazobenzene complex 1 could form head-to-head dimers (triclinic system, space group P1) in the solid state, which were further self-assembled to a linear supramolecular architecture by the intra- and interdimer hydrogen bond interactions as well as the intradimer pi-pi interactions. However, when the included guest 4-hydroxyazobenzene was switched to a 4-aminoazobenzene, the resultant beta-CD/4-aminoazobenzene complex 2 (monoclinic system, space group P2(1)) could be self-assembled to a wave-type supramolecular aggregate under similar conditions. Furthermore, the combination of crystallographic and spectral investigations jointly revealed the inclusion complexation geometry of beta-CD with 4-hydroxyazobenzene and 4-aminoazobenzene in both solution and the solid state, which demonstrated that the disparity of substituents in the azobenzenes played an important role in the inclusion complexation and molecular assembly, affecting not only the structural features of aggregates but also the binding abilities of azobenzenes with beta-CD.

Gene delivery to embryonic stem cells.

Since the establishment of embryonic stem (ES) cells and the identification of tissue-specific stem cells, researchers have made great strides in the analysis of the natural biology of such stem cells for the development of therapeutic applications. Specifically, ES cells are capable of differentiating into all of the cell types that constitute the whole body. Thus, ES cell research promises new type of treatments and possible cures for a variety of debilitating diseases and injuries. The potential medical benefits obtained from stem cell technology are compelling and stem cell research sees a bright future. Control of the growth and differentiation of stem cells is a critical tool in the fields of regenerative medicine, tissue engineering, drug discovery, and toxicity testing. Toward such a goal, we present here an overview of gene delivery in ES cells, covering the following topics: significance of gene delivery in ES cells, stable versus transient gene delivery, cytotoxicity, suspension versus adherent cells, expertise, time, cost, viral vectors for gene transduction (lentiviruses, adenoviruses, and adeno-associated viruses, chemical methods for gene delivery, and mechanical or physical gene delivery methods (electroporation, nucleofection, microinjection, and nuclear transfer).

Possible mechanism of referred pain in the perineum and pelvis associated with the prostate in rats.

PURPOSE: Since persistent pain in the perineum and pelvic floor associated with chronic prostatitis /chronic pelvic pain syndrome has been hypothesized to be referred pain, it might also be explained by neural mechanisms. MATERIALS AND METHODS: Dual retrograde fluorescent labeling and immunohistochemistry were identified as methods with which to investigate the neurogenic aspect of this status. The dual distribution of dorsal root ganglia (DRG) cells was determined after double retrograde fluorescent staining of the prostate and pelvic floor, and the prostate and perineum somatic nerves. Calcitonin gene-related peptide (CGRP) and substance P (SP) in dual labeled cells were determined by immunohistochemistry, giving possible insight into the cause of pelvic pain. RESULTS: Fluorescent double labeled cells were found in the lumbar and sacral DRG, while double labeled cells were distributed predominantly in L6 to S1 and L1 to L2 segment DRG in groups 1 and 2, respectively. On immunohistochemistry some of them were confirmed to contain CGRP and SP. Thus, there are crossover pathways between the prostate and pelvic floor. CONCLUSIONS: The findings that we present confirm that the peripheral process of DRG cells dichotomizes to the prostate, sphincter and somatic parties simultaneously. Some of these cells contain CGRP and SP, which indicate that referred pain in the perineum and pelvic floor may be caused by an axon reflex in the peripheral process of DRG neurons.

Effect of combined cyclic stretch and fluid shear stress on endothelial cell morphological responses.

Endothelial cells in vivo are normally subjected to multiple mechanical stimuli such as stretch and fluid shear stress (FSS) but because each stimulus induces magnitude-dependent morphologic responses, the relative importance of each stimulus in producing the normal in vivo state is not clear Using cultured human aortic endothelial cells, this study first determined equipotent levels of cyclic stretch, steady FSS, and oscillatory FSS with respect to the time course of cell orientation. We then tested whether these levels of stimuli were equipotent in combination with each other by imposing simultaneous cyclic stretch and steady FSS or cyclic stretch and oscillatory FSS so as to reinforce or counteract the cells' orientation responses. Equipotent levels of the three stimuli were 2% cyclic stretch at 2%/s, 80 dynes/cm2 steady FSS and 20 +/- 10 dynes/cm2 oscillatory FSS at 20 dyne/cm2-s. When applied in reinforcing fashion, cyclic stretch and oscillatory, but not steady, FSS were additive. Both pairs of stimuli canceled when applied in counteracting fashion. These results indicate that this level of cyclic stretch and oscillatory FSS sum algebraically so that they are indeed equipotent. In addition, oscillatory FSS is a stronger stimulus than steady FSS for inducing cell orientation. Moreover, arterial endothelial cells in vivo are likely receiving a stronger stretch than FSS stimulus.

[Complication of laparoscopic radical hysterectomy and lymphadenectomy: an analysis of 23 cases].

OBJECTIVE: To explore complications of laparoscopic radical hysterectomy and lymphadenectomy in patients with cervical and endometrial carcinoma, and its prevention. METHODS: From July 2000 to December 2004 at our institution, a total of 278 laparoscopic radical hysterectomy and lymphadenectomy procedures were performed in 242 patients with cervical carcinoma and 36 patients with endometrial carcinoma. We reviewed the clinical data of patients who underwent laparoscopic radical hysterectomy and lymphadenectomy to check their complications and other factors related to this open surgical procedure. RESULTS: All but 4 surgical procedures were completed laparoscopically. Paraaortic lymphadenectomy was performed in 108 patients and pelvic lymphadenectomy was performed in all of 278 patients. Major and minor intraoperative complications occurred in 4.7% (13/278) of patients. The overall conversion rate was 1.4% (4/278), consisting of 3 emergency and 1 elective conversions. Vascular injuries occurred in 7 patients, five of them were repaired or treated laparoscopically and one left external iliac vein injury required laparotomy, another patient underwent laparotomy to control bleeding. Operative cystotomies occurred in 4 patients and all were repaired laparoscopically. One patient underwent laparotomy because of hypercapnia. Another patient underwent laparotomy because of ascending colon injury. Postoperative surgery complications occurred in 3.6% (10/278) of patients. Three patients had a ureterovaginal fistula and three patients had a vesicovaginal fistula after the operation that required reoperation. There was one patient with ureterostenosis while three patients with urinary retention. CONCLUSIONS: Laparoscopic radical hysterectomy in combination with lymphadenectomy is becoming a routine procedure in the armamentarium of many gynecologists. Complications that are unique to laparoscopy exist but they decrease with more practice and experience.

Closely linked polymorphic marker: successful application in preimplantation genetic diagnosis for beta-thalassemia.

OBJECTIVE: To evaluate the applicability of the polymorphic marker closely linked with beta-globin gene for the preimplantation genetic diagnosis (PGD) in couples at risk of having child with beta-thalassemia. METHODS: Single cell multiplex nested PCR which coamplifies the beta-globin gene and the closely linked polymorphic marker, HumTHO1 gene, was applied in six clinical PGD cycles for four couples with beta-thalassemia. RESULTS: In six clinical PGD cycles, a total of 44 embryos were biopsied and 44 blastomeres were obtained. Forty-one blastomeres were amplified and thirty-five embryos were given definite diagnoses. Fourteen embryos were transferred back to the uterus of the patients and one pregnancy went on well and ended with one live healthy birth, which confirmed the results of PGD. The average amplification efficiency of single blastomere was 89.7% and the average allele drop-out(ADO) rate was 14.4%. The coamplification of HumTHO1 could help to detect the existence of ADO and contamination. CONCLUSION: This is the first report on unaffected pregnancy resulting from PGD using multiplex nested PCR in China. The simultaneous amplification of polymorphic marker closely linked to beta-globin gene(HumTHO1) could help to resist the risk of misdiagnosis in PGD caused by ADO and contamination.

[Causes and prevention of anastomotic leakage after esophagectomy and reconstruction through different routes for esophageal cancer].

OBJECTIVE: To compare the anastomotic leakage rates after esophagectomy and reconstruction through different routes for esophageal cancer and analyze the causes for higher anastomotic leakage rate after esophagectomy, systemic lymph node dissection and reconstruction through retrosternal route and its prevention. METHODS: Data of 1105 cases of esophagectomy were reviewed retrospectively. Patients in group A (n=229) underwent esophagectomy through left thoracotomy and intrathoracic anastomosis, patients in group B (n=716), esophagectomy through right anterio-lateral thoracotomy and cervical reconstruction through posterior mediastinal route, patients in group C (n=160) esophagectomy, systemic lymph node dissection and cervical anastomosis through the retrosternal route. RESULTS: The leakage rate was significantly higher (19.4%) in group C than that in group B (11.9%, P< 0.05) and much significantly higher than that in group A (2.2%, P< 0.01). In group C, there was no significant difference in leakage rate between the patients with hand-sewn or mechanical anastomosis (22.2% vs.11.6%, P=0.133), between the patients who had whole stomach or tube-typed gastric reconstruction (25% vs.15.6%, P=0.146). The leakage rate was significantly decreased from 23.3% to 9.1% after prolonged nasal-gastric drainage for seven days (P< 0.05). CONCLUSION: The high anastomotic leakage rate after retrosternal reconstruction is mainly due to compression of the stomach in the anterior mediastinum. Prolonged nasogastric drainage is an effective way to decrease the leakage rate after systemic lymphadenectomy.

[Prevention efficiency of Exserohilum monoceras with chemical herbicides against Echinochloa crus-galli in paddy field].

This paper studied the impact of several herbicides on the conidium germination of Exserohilum monoceras strain X27 in petri dish, and the synergistic effects of the pathogen and chemical herbicide quinclorac or propanil against Echinochloa crus-galli in greenhouse. The prevention efficiency of the tank-mixture of pathogen and quinclorac was also evaluated in paddy field. The results showed that test herbicides except quinclorac and bensulfulfuronmethyl could inhibit conidium germination and hypha growth to different degree. A significant synergism was observed between the pathogen and quinclorac. Adding quinclorac could obviously increase the prevention efficiency against Echinochloa crus-galli. Under field condition, the single use of pathogen could only control about 60% of the weed, while the efficiency of pathogen-quinclorac mixture could reach 90%.

[Inhibitory effects of human papilloma virus 18 E6 gene in HeLa cell transfected with shRNA].

OBJECTIVE: To investigate the inhibition of HPV18E6 gene in HeLa cell transfected with plasmid expressing human papilloma virus 18 E6 (HPV18E6) short hairpin RNA (shRNA). METHODS: We synthesized two HPV18E6 shRNA frames and sub-cloned them into pSUPER which can express shRNA in mammalian cells to construct pE6-1shRNA and pE6-2shRNA which were mutant in E6 shRNA frame. The pE6-1shRNA, pE6-2shRNA and pcDNA3.1 were co-transfected into HeLa cells by cationic liposome respectively and the positive transfectants were selected by G418. The HPV18E6 mRNA and protein expression level was detected by semi-quantitative RT-PCR and streptavidin-peroxidase conjugated method (SP) to assay the inhibitory effects of pE6shRNA. RESULTS: We successfully constructed several new HeLa cell lines transfected with pE6-1shRNA and pE6-2shRNA. In the HeLa cells without transfection and the HeLa cells transfected with pE6-1shRNA plasmid, the HPV18E6 mRNA levels were 1.14 +/- 0.45, 0.76 +/- 0.28 respectively, and the difference of HPV18E6 mRNA levels was significant (P < 0.05). The inhibition efficiency of HPV18E6 gene mRNA was 33.3% and the HPV18E6 protein levels were declined after transfection with pE6-1shRNA. In the HeLa cells transfected with pE6-2shRNA and pSUPER plasmids, HPV18E6 mRNA and protein expression levels were not different from those in wild HeLa cells. CONCLUSIONS: The pE6-1shRNA plasmid can inhibit HPV18E6 expression in HeLa cells, which is persistent, specific and heritable.
