[Observation on clinical therapeutic effect of transcutaneous point electric stimulation on periarthritis of shoulder at different stages].

OBJECTIVE: To observe the therapeutic effect of transcutaneous point electric stimulation on periarthritis of shoulder at different stages and compare with electroacupuncture. METHODS: Three hundred and sixty cases of periarthritis of shoulder at different stages were divided into a treatment group (n=186) treated with transcutaneous point electric stimulation, and a control group (n=174) treated with electroacupuncture. Same acupoints were selected in the two groups. RESULTS: The total effective rate of transcutaneous point electric stimulation was 96.6% at the adhesion prophase and 96.9% at the adhesive stage, but electroacupuncture stimulation was 93.5% and 97.9%, respectively, with no significant differences between the two groups; transcutaneous point electric stimulation not only could relieve pain, but also significantly improve dysfunction of shoulder joints at the adhesive stage. CONCLUSION: Transcutaneous point electric stimulation is an effective and convenient therapy for periarthritis of shoulder.

[Immunotherapeutic efficacy of both helper T lymphocytes and cytotoxic T lymphocytes epitopes augmented dendritic cells tumor vaccine on gastric cancer].

OBJECTIVE: To investigate the immunotherapy efficacy of both helper T lymphocytes (Th) and cytotoxic T lymphocytes (CTL) epitopes augmented dendritic cells (DCs) tumor vaccine on gastric cancer. METHODS: Naïve spleen T cells were stimulated by mixed peptides (a mixture of Th epitope MAGE-3 (22-36)) primed DCs per week in vitro. After 4 cycles of restimulation, peptide specific T cells were harvested and subgroups of which were determined with flow cytometry. Cytokines secreting profiles by CD4+ T cells and cytotoxicities of CD8+ T cells on tumor cells were assessed. The protective immunity by referred DCs tumor vaccines was also monitored. RESULTS: Both Th and CTL epitopes primed DCs could elicit both CD4+ T cells and CD8+ T cells in vitro,of which CD4+ T cells released high amount of Th1 type cytokines (IFN-gamma, IL-2) on recognizing specific antigen, as well as CD8+ T cells exhibited efficient tumor-killing capacity. The effects induced by DCs pulsed with single epitope (Th or CTL epitope) in vivo were less effective than those induced by DCs pulsed with mixture epitopes. CONCLUSIONS: Both Th and CTL epitopes augmented DCs tumor vaccine can induce CD4+ Th1 and CD8+ CTL mediated immune responses to eradicate gastric cancer cells.

[Isolation of BADH gene promoter from Suaeda liaotungensis and its sequence analysis].

In this study, the 5' -flanking proximal region of stress-induced gene encoding betaine aldehyde dehydrogenase was isolated by Adaptor-PCR and TAIL-PCR from halophyte Suaeda liaotungensis. 1993 bp sequence was obtained by sequencing. The transcription start site, which localized at 62 bases upstream of the start ATG, was predicted using TSSP-TCM program. The functional elements were analysed by PLACE programm. The SlBADH gene promoter contains the basic elements: TATA-box, CAAT-box, and stress-induced elements: salt responsed element, cold, dehydration, ABA and frozen responsed elements, WUN responsed elements and HSE. Obtaining the promoter of betaine aldehyde dehydrogenase gene from Suaeda liaotungensis provides a foundation for analyzing the stress-induced promoter elements, studying the relationship between structure and founction of the promoter, and investigating the molecular mechanism of BADH gene regulation.

[Mesenchymal stem cells in hematopoietic regulation--review].

As the progenitor of most cell components in the hematopoietic microenvironment, mesenchymal stem cells (MSC) exhibit self-renewal and multilineage differentiation capacity. Through direct interaction with hematopoietic cells, secreting extracellular matrix and factors, MSC maintain the integrity of hematopoietic microenvironment and regulate hematopoiesis accurately. This review summarized the function of MSC in hematopoietic regulation, such as secretion of cytokines supporting hematopoiesis, MSC expression and adhesion molecules interacting with hematopoietic cells, and supportive effects of transplantation combining MSC with HSC on hematopoietic reconstruction, and its clinical perspectives.

[Impact of mesorectal involvement on the outcome of patients with rectal cancer].

OBJECTIVE: To assess the relationship between the incidence and prognostic significance of mesorectal involvement. METHODS: 328 cases of rectal cancer resected with total or subtotal mesorectal excision in our hospital from Jan. 1997 to Dec. 1998 were followed up and analyzed in this study. The neoplastic foci were identified at the pathologic examination of the mesorectum. RESULTS: Neoplastic mesorectal metastasis was found in 234 cases (71.3%); node involvement in 59.8% and microscopic foci involvement in 36% of all cases (isolated in 11.6%, microfoci alone without any kind of other mesorectal involvement). Microscopic deposits were found in 10.3% of TNM Stage I tumors, in 18.4% of Stage II and in 45.1% of Stage III cancers. Five-year disease-free survival rate (49.6% vs. 91.4%) were observed in patients with mesorectal involvement, compared with those without deposits. CONCLUSION: The incidence of neoplastic foci in the mesorectum seem to affect prognosis, even in early staged tumors. The presence of mesorectal foci should be considered an index in modifying the conventional staging of the rectal tumor.

[Impact of acupuncture to IGF-I expression in spared dorsal root ganglion of cats].

OBJECTIVE: To explore the relationship between Insulin-like growth factor-I (IGF-I) and acupuncture promoting the spinal cord plasticity, the changes of IGF- I expressing in spared dorsal root ganglia (DRG,L6) after operation and acupuncture were investigated. METHODS: 25 adult cats were divided into 5 groups: normal control group; 7th day and 14th day group after unilateral partial rhizotomy (unilateral L1-L5,L7-S2 DRG Were transected, but L6 DRG was spared); 7th day and 14th day group of acupuncture stimulating the spared DRG (electro-needle stimulation was performed by following unilateral partial root rhizotomy). Animals survived for 7 or 14 days after operation respectively. Unilateral L6 dorsal root ganglia of each group were made into 20 microm frozen sections. By immunohistochemistry ABC method, the sections were stained with specific IGF-I (1:200) antibody. The distribution and the number of IGF-I positive neurons in spared DRG (L6) that located the operated/acupuncture side of each animal were observed and counted. RESULTS: For 7th day group after acupuncture stiumlation, the number of IGF-I positive neurons of spared DRG of acupuncture side showed significantly more than that of 7th day operation group (P<0.05), but still less than that of normal group (P < 0.05); In 14th day group, IGF- I expression in neuron of L6 DRG also increased apparently more than that of 14th day operation group, with coming back to normal level. After acupuncture stimulating the spared DRG for 14 days, the numbers of IGF- I positive neurons in spared DRG increased significantly more than that of 7th day group after acupuncture (P<0.05). CONCLUSION: Acupuncture can significantly increase the number of IGF- I positive neurons. Our results indicate that the expression changes of IGF-I in spared DRG associate with acupuncture promoting the spinal cord plasticity.

[Expression of heparanase and angiopoietin-2 mRNA in endometriosis].

OBJECTIVE: To investigate the relationship between heparanase (Hpa) and angiopoietin-2 (Ang-2) gene expression and the development of endometriosis (EM). METHODS: Expression of Hpa gene and Ang-2 mRNA in the eutopic and ectopic endometrium collected from 86 patients with endometriosis (EM group) and the normal endometrium from 30 women without endometriosis (control group) was determined by RT-PCR. RESULTS: (1) Hpa gene expression was detected in 53 ectopic endometrium and 47 eutopic endometrium specimens in the EM group and 8 normal endometrium specimens in the control group. There was no significant difference in Hpa gene expression between ectopic and eutopic endometrium in the EM group (P > 0.05). However, the Hpa gene expression in normal endometrium in the control group was significantly lower than that in ectopic and eutopic endometrium in the EM group (P < 0.05). (2) Ang-2 gene expression was detected in 61 ectopic endometrium and 56 eutopic endometrium specimens in the EM group and 10 normal endometrium specimens in the control group. There was no significant difference in Ang-2 gene expression between ectopic and eutopic endometrium in the EM group (P > 0.05), however, the Ang-2 positive rate in normal endometrium in the control group was significantly lower than that in ectopic and eutopic endometrium in the EM group (P < 0.05). (3) The expression of Hpa gene in stage III-IV endometriosis was significantly higher than that in stage I-II endometriosis (P < 0.05). (4) The difference of Ang-2 mRNA expression between stage III-IV and stage I-II endometriosis was not significant (P > 0.05). (5) The expression of Hpa gene was shown to be significantly correlated to the expression of Ang-2 gene in ectopic endometrium specimens from patients with endometriosis (P < 0.05). CONCLUSION: Hpa and Ang-2 genes are highly expressed in both ectopic and eutopic endometrium of patients with endometriosis. Over-expression of these two genes may play a role in the development and progression of endometriosis.

[Clinicopathologic features of gallbladder adenocarcinoma with marked stromal fibrosis--a report of 19 cases].

BACKGROUND & OBJECTIVE: Macropathologic types of gallbladder cancer are mostly polyp type, intumescent type, and cauliflower form lump. Its histological types include well or poorly differentiated adenocarcinoma, mucinous adenocarcinoma, and undifferentiated cancer. This research was to explore the clinicopathologic features of gallbladder adenocarcinoma with marked stromal fibrosis. METHODS: Pathology of 19 cases of gallbladder adenocarcinoma with marked stromal fibrosis was observed using a light microscopy and SP immunohistochemistry. Clinicopathologic features of 19 patients were analyzed. RESULTS: Most of the patients had long-term history of cholecystitis gallbladder calculus. B ultrasound showed that the gallbladder wall was irregularly thickened or presented nodosity. Observed with naked eyes, gallbladder adenocarcinoma with marked stromal fibrosis did not form cancer nodule and extrude into the gallbladder lumen, the gallbladder wall showed regional thickening, a few cases showed diffuse irregular thickening. Observed under a light microscope, the adenocarcinoma cells were mostly arranged as single layers, seldom arranged as multiple layers, and formed adenoid structures with different sizes, various shapes, and irregular arrangement; the nuclei were heterogenic with haryomitosis presented in a few cases; inflammatory cells were infiltrated in hyperplastic fibrous connective tissue of some cases. According to immune phenotyping, CK (AE1/AE3), CK (AE1), CK7 (OV-TL12/30), CK8 (C51), CK18 (Dc-10), CK19 (RCK108), and EMA (Mc-5) showed strong expression, CEA (COL-1), CK20 (Ks20. 4), and MUC-5AC (CLH2) showed moderate expression, and MUC-2 (B306. 1) showed weak expression; CK17 (E3) showed focal expression. CONCLUSIONS: The clinical manifestation, macropathologic type, histological characteristics of gallbladder adenocarcinoma with stromal fibrosis are different from other types of adenocarcinoma. Its genesis may be related to chronic cholecystitis: long-term inflammation causes regional hyperplasia and heterogeneity of the gland body, lead to focal or regional thickening of the gallbladder wall, and result in gallbladder adenocarcinoma with stromal fibrosis finally.

[A study on the expression of HSP70 in endothelial cells pretreated with ethanol and fluid shear stress].

OBJECTIVE: To detect whether ethanol can affect the expression of HSP70 in endothelial cells under fluid shear stress. METHODS: Ethanol at different concentrations was added to the culture medium of endothelial cells, EA. Hy926, which was treated statically or under 1Pa fluid shear stress. After the incubation of 1 h, 2 h, 4 h and 6 h, the expression of HSP70 was detected by immunohistochemical method(SP). RESULTS: In the control group, the expression of HSP70 was negative under static state, while it was positive under 1Pa fluid shear stress lasting 4 h even without ethanol. No statistic difference was found between the 50 mg/dL ethanol group and the control group with the same treatment time of fluid shear stress. HSP70 expression was found under static state with 150 mg/dL ethanol after 4 h or 300 mg/dL ethanol after 2 h respectively. The expression increased greatly under 1Pa fluid shear stress in the same ethanol concentrations. CONCLUSION: Medium to high ethanol concentration in coordination with fluid shear stress can strongly stimulates the expression of HSP70 by a kinetic mechanism of time-dependent.

[Progesterone-modulated proteins in human endometrial cancer cell line Ishikawa].

OBJECTIVE: To identify proteins associated with growth inhibitory effects of progesterone in Ishikawa endometrial adenocarcinoma cell line (Ishikawa). METHODS: The total cellular proteins of the control and megestrol acetate (MA)-treated Ishikawa cells were separated by two-dimensional gel electrophoresis. After colloidal Coomassie G-250 staining and scanning, the gel images were analyzed by PDQuest software. Matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-TOF MS) and Mascot software were used to identify the differentially expressed protein. RESULTS AND CONCLUSIONS: Well-resolved and reproducible 2-dimensional patterns of both control and MA-treated Ishikawa cells were obtained. Forty-one proteins displayed differential expression after MA treatment, among which 23 were up-regulated and 18 down-regulated. Peptide mass fingerprints (PMF) by MALDI-TOF-MS analysis and search in NCBInr resulted in the identification of 8 proteins. The up-regulated proteins identified were catechol-O-methyltransferase (COMT), glutathione S-transferase, keratin 8, splicing factor 3a (subunit 3), chaperonin containing TCP1 (subunit 6A isoform a, CCT6A), RAB6A protein and AHA1. The down-regulated protein was peptidyl prolyl isomerase A (isoform 1). These findings can be helpful in further investigation of the molecular mechanism of progesterone.

[Protective effect of protein kinase C and mitogen-activated protein kinases and its mechanism in liver ischemic preconditioning].

OBJECTIVE: To investigate the protective effects of protein kinase C (PKC) and mitogen-activated protein kinases (MAPKs) their and mechanisms in liver ischemic preconditioning. METHODS: In rat models of liver ischemia-reperfusion (IR) and ischemic preconditioning (IP), the liver function was evaluated by examining serum alanine aminotransferase and aspartate aminotransferase levels, and the morphological changes of the liver cells were observed under microscope. PKC activator phorbol 12-myristate 13-acetate(PMA) and inhibitor chelerythrine(CHE), as well as MEK inhibitor PD98059, were utilized to analyze the phosphorylation of PKC and P44/42 MAPKs. RESULTS: Compared with the control rats, the liver function was best protected in rats of IP group, but not in those of IP group with PD98059 or CHE treatment. The rats in IR group showed improved liver function after PMA treatment. Similarly, the phosphorylation of PKC and P44/42 MAPKs was correlated with the liver function, and highly enhanced PKC and P44/42 MAPKs activity was observed in IP and IR+PMA groups, but decreased activity in IR and IP+CHE groups. CONCLUSION: Phosphorylation of PKC and MAPKs plays a pivotal role in the preservation of the hepatocytes during IP.

[Preliminary study on expression of oviductin in mouse cervix].

In the present study,in order to investigate the pattern of the expression and location of oviductin using RT-PCR and Western blot,a 410bp cDNA fragment was amplified from mouse cervix tissue. Sequencing analysis and homology comparison with BLAST alignment showed that this fragment amplified from mouse cervix was homologous to the region from +310 to +714 of the mouse OGP gene in Genebank by the degree up to 100%. Western blot showed 60 KD and 30 KD components from mouse cervix same as those from mouse oviduct mucosa epithelium. Semi-quantitative RT-PCR analysis showed that expression of OGP was significantly higher at the estrous stage than that of non-estrous stage and almost no expression in the immature mouse. Those results suggest that the mRNA level of mouse cervix oviductin was related to estrous cycle, implied with estrogen-dependence. To further confirm the results,in situ hybridization histochemistry analysis showed that OGP mRNA was expressed at columnar epithelium of mouse cervix. To clarify the possible significance of oviductin expressed at the cervix, the function of oviductin on the mobility of human sperm was studied using "loss-of-function". The VAP, VCL, ALH, VSL showed no significant differences of the mobility of the human sperm in the conditioned medium blocked by the anti-CTP rOGP (rabbit Oviductin) antiserum compared with that in the conditioned medium of the normal rabbit serum. These results suggest that rOGP may not affect the mobility of human sperm in vitro.

[Relationship between the polymorphism of alpha-adducin gene and the two phenotypes of constitutions in patients with essential hypertension classified by TCM].

OBJECTIVE: To investigate the relationship between the polymorphism of alpha-adducin (ADD1) gene and the two phenotypes of constitution in patients with essential hypertension, the Yang-hyperactive (YH) type and phlegm-dampness (PD) type, classified by traditional Chinese medicine (TCM) approach. METHODS: Two hundred and seven patients differentiated by TCM approach as YH type (113 cases) or PD type (94 cases) were observed, with the systolic blood pressure (SBP), diastolic blood pressure (DBP), body mass index (BMI), fasting blood glucose (FBG), serum creatinine (Cr), uric acid (UA), total cholesterol (TC) and triglycerides (TG) as the criteria of observation. Gly460Trp polymorphism of the ADD1 gene was detected by MALDI-TOF mass spectrometry. Results The levels of BMI, DBP, FBG and UA, etc. in the PD group were significantly higher than those in the YH group respectively. The rate of GG, GT and TT type of ADD1 gene was 29.2%, 41.6% and 29.2% in the YH group, 28.7%, 48.9% and 22.3% in the PD group, showing no significant difference in ADD1 genotype distribution between the two groups, while there was also no difference in the hypertension phenotype distribution among different genotypes (both P > 0.05). For the patients with TT genotype, there were significant differences between the YH group and the PD group in BMI (24.11 +/- 3.04 kg/m2 vs 26.20 +/- 2.30 kg/m2), DBP (96.79 +/- 4.05 mmHg vs 99.56 +/- 3.90 mmHg), FBG (5.01 +/- 0.53 mmol/L vs 5.51 +/- 1.07 mmol/L) and UA level (302.22 +/- 71.95 micromol/L vs 358.25 +/- 88.75 micromol/L, all P < 0.05). CONCLUSION: There was no relation between ADD1 gene polymorphism and the TCM genotype of constitution in patients with essential hypertension. However, it is likely that for hypertension patients with TT genotype, those of PD type are more susceptible to cardiovascular disease and have worse prognosis than those of YH type.

Surface-exchange of NOx and NH3 above a winter wheat field in the Yangtze Delta, China.

A four-dynamic-chamber system was constructed to measure NOx and NH3 surface-exchange between a typical wheat field and the atmosphere in the Yangtze Delta, China. The average fluxes ofNO, NO2 and NH3 were 79, -5.6 and -5.1 ngN/(m2 x s), and 91, -1.8 and 23 ngN/(m2 x s), respectively for the wheat field and the bare soil. The NO flux was positively correlated with soil temperature and the fluxes of NO2 and NH3 were negatively correlated with their ambient concentrations during the investigated period. The compensation point of NO2 between the wheat field and the atmosphere was 11.9 microg/m3. The emissions of NO-N and NH3-N from the urea applied to the wheat field were 2.3% and 0.2%, respectively, which indicated that the main pathway of N loss from the investigated winter wheat field was NO. Application of a mixture of urea and lignin increased the emissions of NO, but also greatly increased the yield of the winter wheat.
