[Y-SNP analysis of Bouyei, Gelao, Mulao, Maonan, and Zhuang populations in Guizhou].

To understand the patrilineal genetic structure of Baiyue ethnic group in Guizhou province, we studied the frequencies of Y-chromosome haplotypes which consisted of 10 single nucleotide polymorphisms (SNPs) by using the PCR-RFLP method. Five haplotypes were found in Baiyue ethnic group in Guizhou, among which H8 was the most common, whereas that of Miao in Guizhou tended to be H8, H11 and H12 haplotypes. Compared with Miao in Guizhou, the Guizhou Baiyue (excluding Sui and Dong) was significantly different and could be regarded as an independent ethnic group. Differences were also found in the same ethnic group among different areas.

[Polymorphism analysis of seven Y-STR loci in Shui population in Guizhou, China].

The objective of this study was to investigate the polymorphism of seven Y-specific STR loci in Shui ethnic population of Guizhou, China, and to obtain the polymorphism information in this minority. One trinucleotide STR locus and six tetranucleotide STR loci were simultaneously amplified with fluorescently labeled primers, and genotypes were determined with ABI PRISM 377 DNA Sequencer. Allele frequencies, genetic diversity and haplotype diversity were calculated. Among 94 unrelated males, 6, 4, 6, 2, 3, 5, 4 alleles were observed in loci DYS19, DYS389 I, DYS389 II, DYS390, DYS391, DYS392 and DYS393, respectively. Altogether, 27 haplotypes were identified for the seven Y-STR loci. The genetic diversity values for each locus ranged from 0.124 (DYS389 I) to 0.630 (DYS19). The haplotype diversity value was 0.868. High haplotype diversities were found in Shui population of Guizhou. The study suggests that these seven Y-STR loci are valuable Y-specific markers for establishing a Y-STR database, understanding ethical origin and migrations and for personal identification.

Up-regulation interleukin-6 and interleukin-8 by activated protein C in lipopolysaccharide-treated human umbilical vein endothelial cells.

OBJECTIVE: To investigate the effect of activated protein C (APC) on inflammatory responses in human umbilical vein endothelial cells (HUVEC) stimulated with lipopolysaccharide (LPS). METHODS: The second passage of collagenase digested HUVEC was divided into the following groups: serum free medium control group (SFM control), phosphate buffer solution control group (PBS control), LPS group with final concentration of 1 microg/ml (LPS group), APC group with final concentration of 7 microg/ml, Pre-APC group (APC pretreatment for 30 min prior to LPS challenge), and Post-APC group (APC administration 30 min after LPS challenge). Supernatant was harvested at 0, 4, 8, 12 and 24 h after LPS challenge. Interleukin-6 (IL-6) and Interleukin-8 (IL-8) levels were analyzed with ELISA. Cells were harvested at 24 h after LPS challenge, and total RNA was extracted. Messenger RNA levels for IL-6 and IL-8 were semi-quantitatively determined by RT-PCR. RESULTS: Compared with control group, IL-6 and IL-8 levels steadily increased 4 to 24 h after LPS stimulation. APC treatment could increase LPS-induced IL-6 and IL-8 production. The mRNA levels of IL-6 and IL-8 exhibited a similar change. CONCLUSION: APC can further increase the level of IL-6 and IL-8 induced by LPS. The effect of these elevated cytokines is still under investigation.

Type I interferon production by tertiary lymphoid tissue developing in response to 2,6,10,14-tetramethyl-pentadecane (pristane).

Lymphoid neogenesis is associated with antibody-mediated autoimmune diseases such as Sjogren's syndrome and rheumatoid arthritis. Although systemic lupus erythematosus is the prototypical B-cell-mediated autoimmune disease, the role of lymphoid neogenesis in its pathogenesis is unknown. Intraperitoneal injection of 2,6,10,14-tetramethyl-pentadecane (TMPD, pristane) or mineral oil causes lipogranuloma formation in mice, but only TMPD-treated mice develop lupus. We report that lipogranulomas are a form of lymphoid neogenesis. Immunoperoxidase staining of lipogranulomas revealed B cells, CD4(+) T cells, and dendritic cells and in some cases organization into T- and B-cell zones. Lipogranulomas also expressed the lymphoid chemokines CCL21, CCL19, CXCL13, CXCL12, and CCL22. Expression of the type I interferon (IFN-I)-inducible genes Mx1, IRF7, IP-10, and ISG-15 was greatly increased in TMPD- versus mineral oil-induced lipogranulomas. Dendritic cells from TMPD lipogranulomas underwent activation/maturation with high CD86 and interleukin-12 expression. Magnetic bead depletion of dendritic cells markedly diminished IFN-inducible gene (Mx1) expression. We conclude that TMPD-induced lupus is associated with the formation of ectopic lymphoid tissue containing activated dendritic cells producing IFN-I and interleukin-12. In view of the increased IFN-I production in systemic lupus erythematosus, these studies suggest that IFN-I from ectopic lymphoid tissue could play a role in the pathogenesis of experimental lupus in mice.

Effects of intravenous administration of human bone marrow stromal cells after intracerebral hemorrhage in rats.

OBJECT: The goal of this study was to investigate whether human bone marrow stromal cells (hBMSCs) administered by intravenous injection have a beneficial effect on outcome after intracerebral hemorrhage (ICH) in rats. METHODS: An ICH was induced in 54 adult male Wistar rats by a stereotactically guided injection of autologous blood into the right striatum. Intravenous infusion of the hBMSCs (3, 5, or 8 million cells) was performed 1 day after ICH, and for each dose group there was a control group that received injections of vehicle. Neurological function, which was evaluated using the Neurological Severity Score (NSS) and the corner turn test, was tested before and at 1, 7, and 14 days after ICH. After 14 days of survival, the area of encephalomalacia was calculated and histochemical labeling was performed. For all three groups, there were no statistical differences in either the NSS or corner turn tests after 1 day. After 7 and 14 days, however, the three groups that received the hBMSCs showed significant improvement in functional scores compared with the control group. In addition, after 14 days there was significantly more striatal tissue loss in the placebo groups compared with each of the three treatment groups. The region of injury in the treated animals demonstrated a significantly increased presence of hBMSCs, immature neurons, neuronal migration, synaptogenesis, and newly formed DNA. CONCLUSIONS: Intravenous administration of hBMSCs significantly improves neurological function in rats subjected to ICH. This improvement in the treated animals is associated with reduced tissue loss and increased local presence of the hBMSCs, mitotic activity, immature neurons, synaptogenesis, and neuronal migration.

[Bone marrow stromal cells co-modified with BMP-2 and bFGF gene].

AIM: To establish modified bone marrow stromal cells(BMSCs) which can express BMP-2 and bFGF stably. METHODS: BMP-2 and bFGF gene were amplified by RT-PCR, and then cloned into the expression vector pcDNA3.0. After being confirmed by DNA sequencing, pcDNA3.0-BMP-2 and pcDNA3.0-bFGF were co-transfected into rat BMSCs with Lipofectamine 2000 reagent. The expression of BMP-2 and bFGF gene in rat BMSCs was detected by RT-PCR, Western blot, immunohistochemical staining and ELISA. RESULTS: BMP-2 and bFGF gene were cloned, and their sequences were identical with those in GenBank. The expression plasmids, pcDNA3.0-BMP-2 and pcDNA3.0-bFGF, were constructed and co-transfected into rMSCs successfully. RT-PCR showed the mass transcription of BMP-2 and bFGF mRNA in transfected BMSCs. Western blot, immunohistochemical staining and ELISA confirmed the expression of BMP-2 and bFGF genes in transfected cells and in the supernatant. CONCLUSION: We have constructed the optimal rat BMSCs which can be used in bone tissue engineering.

[Analysis of polymorphisms in Y-DNA haplotypes and mtDNA haplogroups in Yao ethnic group from Guizho].

The frequencies of Y-chromosome haplotypes consisting of 12 single nucleotide polymorphisms (SNP) and mitochondrial DNA (mtDNA) haplotypes consisting of 9 SNPs were investigated using PCR-RFLP in 97 Yaos from Guizhou to study the patrilineal and matrilineal genetic structure and the origin of Yao Ethnic Group from Guizhou. Results showed that all 97 samples were classified into 4 Y-DNA haplotypes (H7, H8, H9 and H11). The major haplotype H7 of Hmong-Mien Population was highly prevalent (92.4%) in Yaos from Guizhou. Eight mtDNA haplotypes were identified through mtDNA analysis and all the haplotypes could be classified into 5 haplogroups (B4, B5, D4, D5 and N*) that were defined previously. The frequency of a 9-bp deletion in the human mtDNA Co II/tRNALys intergenic region was 58.2% in the 97 samples. These data suggest that the patrilineal genetic structure of Yao ethnic group from Guizhou was simple and Yaos from Guizhou had the typical genetic character of the Hmong-Mien Population and some admixture with other populations. The matrilineal genetic structure of Yaos from Guizhou was relatively complex and the 9-bp deletion was a characteristic genetic marker of the matrilineal genetic structure of Yaos from Guizhou.

[Growth and osteogenesis characteristics of cultured canine mesenchymal stem cells under osteogenic induction].

To investigate the growth and osteogenesis characteristics of cultured canine mesenchymal stem cells (cMSCs) under osteogenic induction. We found the cMSCs were isolated from adult canine using density gradient separation method. The cMSCs attachment formed soon after seeding and grew into colonies with the appearance of fibroblastic cells. The osteogenic induction compound of Dexamethasone (Dex), beta-sodium glycerphosphate (beta-GP), ascorbic acid (AA) was added to passaged cMSCs and the proliferation and osteogenic differentiation of them was studied. The morphology of cells was observed by light micrograph and transmission electron microscope. The proliferation and growth characteristics of cMSCs were observed during primary and passage cultures through MTT. The differentiation were assayed by alkaline phophatase (ALP) and osteocalcin (OCN). We found the cMSCs have an active proliferative ability in primary and passage culture, and cMSCs under osteogenic induction have the typical characteristic of a secretory cell; the osteogenic induction compound may induce cMSCs to differentiate to osteoblasts. There are higher expression of ALP and OCN in passage 3 cMSCs under osteogenic induction than that of the osteoblasts osteogenic induction condition. Our research suggest the cMSCs in our culture system are mainly undifferentiated osteoprogenitors and can differentiate to osteoblast under osteogenic induction.

[The study of osteogenous differentiation of MSCs transfected with different gene].

This experiment is sought to study the contribution of different gene in osteogenous differentiation of bone marrow stromal cells (MSCs) and optimize the seed cells of bone tissue engineering. Firstly, we obtained the full length gene of BMP-2, VEGF165 and bFGF by RT-PCR, and cloned into the expression vector pcDNA3. 0. After to be transfected, the MSCs cell lines which could express each of the target protein were selected out by G418. RT-PCR and immunohistochemistry were used to confirm the exogenous gene expression in MSCs. MTT showed that almost all of the MSCs which have been transfected with exogenous gene had more strong proliferative potential than the untransfected group did. There was a notable increase of ALP activity in transfected cell compared with the control group. The concentration of OCN in cell culture medium had a increase in some degree except of VEGF group. The outstanding osteogenous differentiation could be observed in BMP-2 and bFGF transfected MSCs. The results showed that the MSCs modified by BMP-2 and bFGF would be more effective in bone tissue engineering field.

[Study of immunization protocol of ovarian carcinoma associated 6B11 anti-idiotypical minibody].

OBJECTIVE: To probe better immunization dose and interval of anti-idiotypical minibody (6B11 minibody) in animals and to guide the clinical trials of ovarian epithelial cancer treatment. METHODS: Minibody was induced with isopropylthiogalactoside (IPTG) in E.coli and analyzed with direct enzyme linked immunosorbent assay (ELISA). Normal BALB/c mice were randomly divided into groups A, B, C, D, E, F, immunized with the murine monoclonal anti-idiotypic antibody 6B11 minibody 150 microg, 100 microg, 50 microg, and mice IgG 100 microg, human IgG 100 microg, phosphate buffered saline (PBS) 100 microl at d(1), d(14), d(28) respectively. Anti-anti-idiotypic antibody (Ab3) level was tested and antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) effect of mouse spleen cells to human ovarian carcinoma SKOV3 cells were measured using (51)Cr-release assay. UV absorption values were tested at 490 nm. RESULTS: 6B11 minibody was successfully expressed on E.coli and could react with a murine monoclonal antibody COC166-9. Ab3 could not be detected until one week after d(14)'s boost, its level gradually elevated and peaked one week after d(28)'s boost, and maintained at high level for about six weeks then declined dramatically seven weeks after d(28)'s boost. When boosted again at seven weeks after d(28)'s boost, the Ab3 level elevated quickly one week after, and maintained at this level at least for two weeks. The Ab3 level of group B was higher than groups A and C, mean A (absorption) value during six weeks after d(28)'s boost was 1.05 in group A, 1.06 in group B, 0.94 in group C, and there was no significant difference (P > 0.05). At four different effector cell/target cell (E/T) ratios, the ADCC was highest in group A and lowest in group C; when the E/T ratio was 1:125, (51)Cr releasing rate was 23% in group A, 17% in group B, and 12% in group C, the difference between groups A, B, C had statistical significance (P < 0.05). When serum was diluted at 1:50, the CDC effects were higher in group A and B than in group C ((51)Cr releasing rate was 47%, 39%, 26% respectively; P < 0.05), but there was no significant difference between groups A and B (P > 0.05). CONCLUSION: 6B11 anti-idiotypic minibody of ovarian carcinoma can be used as tumor vaccine, the suitable immunization protocol in animals may be: prime at d(1), d(14) and d(28) at a dose of 100 microg; prime interval is six weeks.

A multicentral randomized control study on clinical acupuncture treatment of Bell's palsy.

OBJECTIVE: To confirm the clinical therapeutic effects of acupuncture and moxibustion on Bell's palsy. METHODS: 480 cases from 4 hospitals were enrolled for this study, among whom 439 cases completed the whole course of the study. The patients were randomly divided into the following 3 groups, a control group(treated with prednisone, vitamin B1, vitamin B12 and dibazol), an acu-moxibustion group (treated with filiform needle plus moxibustion), and a basic treatment plus acu-moxibustion group (treated with oral medicine like those in the control group plus acupuncture, and with moxibustion like in the acu-moxibustion group). The whole treatment course lasted 4 weeks. The therapeutic effects were evaluated according to the symptoms and signs, House-Brackmann grading scale and facial disability indexes(FDI). RESULTS: All the 4 centers (hospitals) completed this study well, with no statistically significant difference found among the 4 centers in therapeutic effects. The patients with different conditions were well distributed in the 3 groups, thus the basic general data were comparable (P > 0.05). The therapeutic effects of the two treatment groups were better than the control group (respectively P < 0.05 and P < 0.01), and it was the best in the acu-moxibustion group (P < 0.01). CONCLUSION: Acupuncture and moxibustion may exert definite therapeutic effects on Bell's palsy, better than that of the basic treatment group or the basic treatment plus acu-moxibustion group.

[The effect of mechanical strain on proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells from rats].

In order to investigate the effects of cyclic biaxial mechanical strain on proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells (MSCs)from rats. The MSCs were isolated from bone marrow in the 9-month-old female Sprague-Dawley rats by a density gradient method. After the third passage,1 x 10(5) cells were seeded on the flexible silicone membrane in the cyclic biaxial mechanical strain unit. The amplitude of mechanical strain applied to the cells were 4000 microstrain, at a frequency of one hertz (1 Hz), for periods of 3 repetitions of 2 hours' mechanical strain and a break of 2 hours. The osteoblastic marker such as osteopontin (OPN),alkaline phosphatease (ALP), osteocalcin (OCN), type I collagen (Col I) were detected by RT-PCR. The proliferation of all cells were studied with Flow Cytometry (FCM). ALP and OPN levels significantly increased in the mechanical strained groups and reached the top at 3th day after mechanical strain. COL I level was increase only at 3th day after mechanical strain, and the change of OCN was not obvious in every group. The proliferation index of the medchanical strained groups was increased, compared with the control. These results showed that mechanical strain can improve MSCs proliferation and make MSCs differentiate to osteoblast.

[The effects of post coronary stenting triple antiplatelet therapies on platelet functions].

OBJECTIVE: To explore the effects of triple antiplatelet therapy on platelet aggregation and activation in patients who underwent coronary stenting. METHODS: 120 in-hospital coronary heart disease patients with coronary stenting were randomized to two groups receiving either triple antiplatelet drugs of aspirin and clopidogrel combined with cilostazol or dual antiplatelet drugs of aspirin and clopidogrel. On the first day after stenting cilostazol was added to the triple group patients who were previously administered aspirin and clopidogrel. Expressions of PAC-1 and CD62p which indicate platelet activation were assessed with flow cytometry and the maximal platelet aggregation rate (MPAR) induced by 5 and 20 micromol/L ADP was measured at the day before receiving cilostazol and the fifth day after stenting, respectively. RESULTS: The baseline clinical characteristics did not differ significantly between the two groups. There were no significant differences in the baseline level of MPAR CD62p and PAC-1 at the first day after stenting between the two groups. The margins between the two measurements were [(6.44 +/- 14.44)% vs (5.41 +/- 13.77)%, P > 0.05] for DeltaMPAR induced by 5 micromol/L ADP, [(8.50 +/- 15.50)% vs (7.84 +/- 14.21)%, P > 0.05] for DeltaMPAR induced by 20 micromol/L ADP, [(5.12 +/- 11.25)% vs (1.08 +/- 4.97)%, P < 0.05] for DeltaCD(62)p and [(12.12 +/- 12.30)% vs (2.22 +/- 15.15)%, P < 0.01] for DeltaPAC-1 in the triple and dual group patients, respectively. Among the above measurements, DeltaCD62p and DeltaPAC-1 in the triple group patients were significantly higher than those in the dual group patients although DeltaMPAR did not significantly differ between the two groups at the fifth day after stenting. Subgroup analysis for patients with acute coronary syndrome (ACS) showed that DeltaMPAR induced by 5 micromol/L ADP [(8.68 +/- 10.35)% vs (2.92 +/- 13.06)%, P = 0.018], DeltaMPAR induced by 20 micromol/L [(11.05 +/- 11.14)% vs (5.16 +/- 13.27)%, P = 0.019], DeltaCD62p [(5.57 +/- 12.08)% vs (1.35 +/- 4.42)%, P = 0.028] and DeltaPAC-1 [(11.62 +/- 12.73)% vs (1.29 +/- 15.73)%, P = 0.001] in triple group were significantly higher than that in dual group. At 3-month clinical follow-up, the rate of major adverse cardiac and cerebral events was 0 in the triple group and 3.3% (2/60) in the dual group, and the rate of hemorrhage was 5% (3/60) in the triple group and 3.3% (2/60) in the dual group, the differences were not statistically significant. CONCLUSIONS: Compared with dual antiplatelet regimen with aspirin plus clopidogrel, triple antiplatelet therapy with aspirin and clopidogrel combined with cilostazol is more efficient in inhibiting platelet activation and aggregation after coronary stent implantation. Large scale clinical trials are needed to confirm efficacy and safety of the triple antiplatelet regimen.

Hypometabolism exceeds atrophy in presymptomatic early-onset familial Alzheimer's disease.

UNLABELLED: The aim of the present study is to compare brain atrophy with hypometabolism as preclinical markers of Alzheimer's disease (AD) by studying presymptomatic individuals from families with known early-onset autosomal dominant AD (FAD) carrying mutations in the Presenilin 1 gene. METHODS: Seven asymptomatic at-risk FAD individuals (age, 35-49 y; 4 women; education >/= 12 y) and 7 matched healthy control subjects received complete clinical, neuropsychologic, MRI, and (18)F-FDG PET examinations. Regions of interest (ROIs: whole brain [WB], hippocampus [Hip], entorhinal cortex [EC], posterior cingulate cortex [PCC], inferior parietal lobule [IPL], and superior temporal gyrus (STG]) were drawn on the MRI scans of all subjects and used to measure volumes on MRI and glucose metabolism (MRglc) from the MRI-coregistered, atrophy-corrected PET scans. RESULTS: Compared with controls and after correcting for head size, MRI volume reductions in FAD subjects were restricted to the IPL (18%, P < 0.02). After atrophy correction and adjusting for pons MRglc, PET MRglc reductions were found in all FAD subjects compared with controls in the WB (13%), bilaterally in the IPL (17%) and in the STG (12%), and in the left EC (21%), PCC (20%), and Hip (12%) (P values < 0.05). PET MRglc measurements were consistently less variable than MRI measures, yielding significantly larger effect sizes in separating FAD from controls. CONCLUSION: Presymptomatic FAD individuals show widespread MRglc reductions consistent with the typical AD PET pattern in the relative absence of structural brain atrophy. These data further suggest that PET MRglc measures may serve as biomarkers for the preclinical diagnosis of AD.

The maize Knotted1 gene is an effective positive selectable marker gene for Agrobacterium-mediated tobacco transformation.

We have assessed the use of a homeobox gene knotted1 (kn1) from maize as a selectable marker gene for plant transformation. The kn1 gene under the control of cauliflower mosaic virus 35S promoter (35S::kn1) was introduced into Nicotiana tabacum cv. Xanthi via Agrobacterium-mediated transformation. Under nonselective conditions (without antibiotic selection) on a hormone-free medium (MS), a large number of transgenic calli and shoots were obtained from explants that were infected with Agrobacterium tumefaciens LBA4404 harboring the 35S::kn1 gene. On the other hand, no calli or shoots were produced from explants that were infected with an Agrobacterium strain harboring pBI121 (nptII selection) or from uninfected controls cultured under identical conditions. Relative to kanamycin selection conferred by nptII, the use of kn1 resulted in a 3-fold increase in transformation efficiency. The transgenic status of shoots obtained was confirmed by both histochemical detection of GUS activity and molecular analysis. The results presented here suggest that kn1 gene could be used as an effective alternative selection marker with a potential to enhance plant transformation efficiency in many plant species. With kn1 gene as a selection marker gene, no antibiotic-resistance or herbicide-resistance genes are needed so that potential risks associated with the use of these traditional selection marker genes can be eliminated.
