The Ca2+/calcineurin-regulated cup gene family in Dictyostelium discoideum and its possible involvement in development.

Changes in free intracellular Ca2+ are thought to regulate several major processes during Dictyostelium development, including cell aggregation and cell type-specific gene expression, but the mechanisms involved are unclear. To learn more about Ca2+ signaling and Ca2+ homeostasis in this organism, we used suppression subtractive hybridization to identify genes up-regulated by high extracellular Ca2+. Unexpectedly, many of the genes identified belong to a novel gene family (termed cup) with seven members. In vegetative cells, the cup genes were up-regulated by high Ca2+ but not by other ions or by heat, oxidative, or osmotic stress. cup induction by Ca2+ was blocked completely by inhibitors of calcineurin and protein synthesis. In developing cells, cup expression was high during aggregation and late development but low during the slug stage. This pattern correlates closely with reported levels of free intracellular Ca2+ during development. The cup gene products are highly homologous, acidic proteins possessing putative ricin domains. BLAST searches failed to reveal homologs in other organisms, but Western analyses suggested that Cup-like proteins might exist in certain other cellular slime mold species. Localization experiments indicated that Cup proteins are primarily cytoplasmic but become cell membrane-associated during Ca2+ stress and cell aggregation. When cup expression was down-regulated by antisense RNA, the cells failed to aggregate. However, this developmental block was overcome by partially up-regulating cup expression. Together, these results suggest that the Cup proteins in Dictyostelium might play an important role in stabilizing and/or regulating the cell membrane during Ca2+ stress and/or certain stages of development.

Rice dwarf phytoreovirus segment S6-encoded nonstructural protein has a cell-to-cell movement function.

Rice dwarf virus (RDV) is a member of the genus Phytoreovirus, which is composed of viruses with segmented double-stranded RNA genomes. Proteins that support the intercellular movement of these viruses in the host have not been identified. Microprojectile bombardment was used to determine which open reading frames (ORFs) support intercellular movement of a heterologous virus. A plasmid containing an infectious clone of Potato virus X (PVX) defective in cell-to-cell movement and expressing either beta-glucuronidase or green fluorescent protein (GFP) was used for cobombardment with plasmids containing ORFs from RDV gene segments S1 through S12 onto leaves of Nicotiana benthamiana. Cell-to-cell movement of the movement-defective PVX was restored by cobombardment with a plasmid containing S6. In the absence of S6, no other gene segment supported movement. Identical results were obtained with Nicotiana tabacum, a host that allows fewer viruses to infect and spread within its tissue. S6 supported the cell-to-cell movement of the movement-defective PVX in sink and source leaves of N. benthamiana. A mutant S6 lacking the translation start codon did not complement the cell-to-cell movement of the movement-defective PVX. An S6 protein product (Pns6)-enhanced GFP fusion was observed near or within cell walls of epidermal cells from N. tabacum. By immunocytochemistry, unfused Pns6 was localized to plasmodesmata in rice leaves infected with RDV. S6 thus encodes a protein with characteristics identical to those of other viral proteins required for the cell-to-cell movement of their genome and therefore is likely required for the cell-to-cell movement of RDV.

NS-3 protein of the Junonia coenia densovirus is essential for viral DNA replication in an Ld 652 cell line and Spodoptera littoralis larvae.

The genome of Junonia coenia densovirus (JcDNV) shares with members of the genus Densovirus the property of possessing structural (VP) and nonstructural (NS) genes in opposite orientations. The three NS genes located in the 5' half on one strand encode three NS proteins assumed to be involved in viral DNA replication: NS-1 and NS-2, which are common to all DNVs, and a 28-kDa polypeptide, NS-3, with a unique sequence. Whereas the essential role played by JcDNV NS-1 in viral DNA replication has been clearly established (C. Ding, M. Urabe, M. Bergoin, and R. M. Kotin, J. Virol. 76:338-345, 2002), nothing is known of the biological function(s) of NS-3. To investigate this function, we designed constructs derived from pBRJ, a plasmid encompassing an infectious sequence of JcDNV DNA (M. Jourdan, F. X. Jousset, M. Gervais, S. Skory, M. Bergoin, and B. Dumas, Virology 179:403-409, 1990), with partial or complete deletion of NS-3 sequence or with the ATG initiation codon mutated by site-directed mutagenesis. Transfection of these constructs to sensitive Ld 652 cells or Spodoptera littoralis larvae prevented the accomplishment of a productive cycle. We clearly established that the blocking of the replicative cycle in the absence of NS-3 expression occurred at the level of viral DNA replication. Replication of viral DNA could be restored by cotransfecting Ld 652 cells with a plasmid expressing JcDNV-NS-3 protein in trans. Time course analysis showed that NS-3 is produced early (6 h posttransfection) in the replicative cycle, and its production parallels that of replicative-form viral DNA. Finally, we present evidence that NS-1 and NS-2 proteins are synthesized at apparently the same levels whether or not NS-3 is expressed.

Atorvastatin reduction of intravascular thrombosis, increase in cerebral microvascular patency and integrity, and enhancement of spatial learning in rats subjected to traumatic brain injury.

OBJECT: Atorvastatin, a beta-hydroxy-beta-methylglutaryl coenzyme A reductase inhibitor, has pleiotropic effects, such as promoting angiogenesis, increasing fibrinolysis, and reducing inflammatory responses, and has shown promise in enhancing recovery in animals with traumatic brain injury (TBI) and stroke. The authors tested the effect of atorvastatin on vascular changes after TBI. METHODS: Male Wistar rats subjected to controlled cortical impact injury were perfused at different time points with fluorescein isothiocyanate (FITC)--conjugated dextran 1 minute before being killed. Spatial memory function had been measured using a Morris Water Maze test at various points before and after TBI. The temporal profile of intravascular thrombosis and vascular changes was measured on brain tissue sections by using a microcomputer imaging device and a laser confocal microscopy. The study revealed the following results. 1) Vessels in the lesion boundary zone and hippocampal CA3 region showed a variety of damage, morphological alterations, reduced perfusion, and intraluminal microthrombin formation. 2) Atorvastatin enhanced FITC-dextran perfusion of vessels and reduced intravascular coagulation. 3) Atorvastatin promoted the restoration of spatial memory function. CONCLUSIONS: These results indicated that atorvastatin warrants investigation as a potential therapeutic drug for TBI.

[Establishment and comparison of two intraperitoneally transplanted human ovarian carcinoma models with immune reconstitution in severe combined immunodeficient mice].

BACKGROUND & OBJECTIVE: Ovarian carcinoma is leading cause of death in gynecologic malignancies. The survival rate cannot be improved after routine surgery, chemotherapy, and radiotherapy. Therefore biotherapy becomes the fourth treatment pattern for ovarian carcinoma. Adequate experimental models for the development of biologic therapeutic strategies are needed. Our purpose was to establish and compare two intraperitoneally transplanted human ovarian carcinoma models with human immune reconstitution in severe combined immunodeficient (SCID) mice. METHODS: Six and ten C.B17/SCID mice were intraperitoneally injected with human ovarian adenocarcinoma SKOV3 and SKOV3.ip1 cells, respectively. Their biological, histological, and immunological features were compared. Ascites of the mice in SKOV3.ip1 group was injected into other six mice. All the 22 SCID mice were intraperitoneally injected with human peripheral blood lymphocytes (PBL) to establish immune reconstituted model. RESULTS: The taken rates of the SKOV3 and SKOV3.ip1 groups were both 100%. The latent periods of tumor growth were 20-41 days and 22-30 days, respectively (P >0.05). While the mean survival time were 50-78 days and 32-43 days, respectively (P< 0.0001). 83.3% (5/6) of the mice injected with ascites of the mice in SKOV3.ip1 group successfully formed new tumor mass and ascites. Autopsy showed the tumors of the two models were widespread in pelvic cavity. The SKOV3.ip1 group also had 0.35-5.60 ml bloody ascites that was similar to the clinical behavior of most patients, while only 0.2 ml in one mice of SKOV3 group. Histological results showed the tumors of the two groups remained the characteristics of serous papillary adenocarcinoma of human ovary, and immunohistochemistry staining showed the ovarian associated antigen OC166-9 were both positive. Human IgG were detected in 72.7% (16/22) of the mice, and human CD4(+) and CD8(+) T cells were positive in 54.5% (12/22) of the mice. CONCLUSION: The two intraperitoneally transplanted human ovarian carcinoma models had been established in human PBL reconstituted SCID mice. The SKOV3.ip1 model may be an ideal animal model for biotherapy research of ovarian carcinoma as it simulates the intraperitoneally disseminating behavior of human ovarian carcinoma in the patients with immune function, and it took relatively shorter time to be established.

Kinetics of perrhenate uptake and comparative biodistribution of perrhenate, pertechnetate, and iodide by NaI symporter-expressing tissues in vivo.

UNLABELLED: Pertechnetate (as (99m)TcO(4)(-)), (123)I(-), and (131)I(-) have a long and successful history of use in the diagnosis and therapy of thyroid cancer, with uptake into thyroid tissue mediated by the sodium-iodide symporter (NIS). NIS has also emerged as a potential target for radiotherapy of nonthyroid malignancies that express the endogenous or transfected symporter. Perrhenates (as (188)ReO(4)(-) and (186)ReO(4)(-)) are promising therapeutic substrates of NIS, although less is known about their behavior in vivo. In this study, we endeavored to characterize the biologic behavior of perrhenate, especially in relation to iodide and pertechnetate, to better explore its possible therapeutic role. METHODS: We describe the simultaneous biodistribution and uptake in vivo of iodide, pertechnetate, and perrhenate in groups of healthy CD1 mice, either with or without coadministration of perchlorate (ClO(4)(-)), a potent NIS inhibitor. Animals administered single radiopharmaceuticals were imaged as a means of illustrating these findings. Kinetic properties of perrhenate were compared with those of iodide in a stably transfected NIS-bearing Madin-Darby canine kidney (MDCK) cell line. RESULTS: Biodistributions of iodide, pertechnetate, and perrhenate in live mice were remarkably similar. Activity in salivary gland and stomach was severalfold greater than in blood, remained elevated over the initial 2 h, and subsequently washed out. A similar pattern characterized pertechnetate and perrhenate uptake by the thyroid, in which the 2-h concentration was slightly more elevated than at the 20-min time point. However, uptake subsequently decreased by 19 h. In contrast, iodide continued to increase through the 19-h time point, presumably as a result of organification. The addition of perchlorate sharply decreased uptake of all 3 radiopharmaceuticals by the stomach, salivary glands, and thyroid and resulted in their rapid clearance, paralleling blood-pool clearance. In tissues that do not express NIS (liver, muscle, spleen), uptake of all 3 radiopharmaceuticals was low and rapidly decreased over time, paralleling blood-pool clearance. Similar findings were seen in kidney, where only minimal amounts of NIS are expressed in tubular cells. In stably transfected MDCK cells, steady-state accumulation of iodide was approximately 4-fold higher than that of perrhenate at 30 min. No active transport was demonstrated in nontransfected MDCK cell lines or after perchlorate administration. Uptake values measured at different concentrations of substrate demonstrated saturation kinetics. Apparent maximal velocity values for perrhenate and iodide were 25.6 +/- 1.4 and 106 +/- 3.2 pmol/ micro g, respectively, and corresponding affinity constant values were 4.06 +/- 0.87 and 24.6 +/- 1.81 micro mol/L. CONCLUSION: Perrhenate is avidly taken up by NIS in a manner similar to iodide and pertechnetate in vivo, with the exception of organification of iodide by the thyroid. By more fully appreciating the behavior of perrhenate, especially in relation to iodide and pertechnetate, we can better realize its potential role in the diagnosis and therapy of NIS-bearing tissues.

[In vivo study of antitumor immune responses induced by anti-idiotypic minibody vaccine of ovarian cancer].

BACKGROUND & OBJECTIVE: 6B11 anti-idiotypic minibody can be constructed by fusion of anti-idiotypic single chain antibody (6B11scFv) to human IgG(1) hinge and CH3, which can simulate the function of ovarian carcinoma antigen. It has immune activity of both 6B11 and human immunoglobulin IgG(1) Fc. This study was designed to evaluate whether anti-tumor immune response can be induced in BALB/c mice immunized with 6B11anti-idiotypic minibody and explore its probability as ovarian cancer vaccine. METHODS: BALB/c mice were immunized repeatedly by 6B11 anti-idiotypic minibody. Competition inhibition test, indirect ELISA test, and immune flow cytometry were used for analyzing the serum characterization of anti-anti-idiotypic antibody (Ab3) and the changes of T lymphocyte phenotype of spleen. RESULTS: The specific antitumor immune response could be induced in BALB/c mice after immunized with anti-idiotypic minibody without carrier proteins and adjuvant. The Ab3 maintained at a high level till 30 day after the last immunization. It stimulated proliferation of CD4+ T cells and CD8+ T cells from the spleen of BALB/c mice on 4th day, 14th day, 24th day, and 30th day after the last immunity respectively. CONCLUSIONS: 6B11 anti-idiotypic minibody can induce both humoral and cellular immunity against ovarian carcinoma in vivo, which provided experimental evidence for clinical use of fusion protein 6B11 minibody as anti-idiotype vaccines against ovarian carcinoma.

[Inhibitory effect of endoplasmic reticulum-retained anti-type IV collagenase intrabody on invasion and proliferation of cancer cell].

BACKGROUND & OBJECTIVE: Invasion and metastasis are significant characteristics of cancer cells, Type IV collagenase (matrix metalloproteinase-2, and-9) plays an important role in cancer invasion and metastasis. This study was designed to block the secretion of type IV collagenase via intracellular antibody (intrabody) methods, and inhibit cancer invasion and metastasis. METHODS: We constructed expression plasmid pcDNA3.1-ER.scFv coding for an endoplasmic reticulum (ER)- retained, single chain antibody (ER.scFv) against the type IV collagenase. The intrabody gene was transfected into human giant cell pulmonary carcinoma PG cells. Western blot was performed to detect the expression of pcDNA3.1-ER.scFv. Co-immunoprecipitation was used to analyse the interaction between ER. scFv and target protein in PG cells. The secretion of type IVcollagenase was detected by gelatin zymography. Cell behavior was examined by invasion and proliferation assay in vitro. RESULTS: ER. scFv expressed in PG cells, and can recognize and combine its cognate target protein matrix metalloproteinase-9. Intrabody gene transfection significantly blocked the function and activity of type IV collegenase. The ER.scFv-transfected PG cells were less invasive than parental or vector control cells, the suppression rate of ER.scFv-transfected PG cells was 76.3%(P< 0.05). ER. scFv showed anti-proliferative effect on PG cells cultured on Matrigel. CONCLUSIONS: ER-retained intrabody technology may restrain the activity of type IV collagenase in protein processing and secretory pathway, and inhibits cancer cell invasion and proliferation. Anti-type IV collagenase intrabody may be further used in cancer gene therapy.

Efficacy of acupuncture and moxibustion in treating Bell's palsy: a multicenter randomized controlled trial in China.

BACKGROUND: Bell's palsy involves acute facial paralysis due to inflammation of the facial nerve. Acupuncture and moxibustion (acu-moxi) is beneficial in treating facial palsy. In order to verify the efficacy of acu-moxi on Bell's palsy, a randomized single-blind, multicenter clinical trial was performed. METHODS: A total of 480 patients from four clinical centers were involved in this trial, of whom 439 completed the trial and 41 did not. All patients were randomly assigned to either the control group or to one of two treatment groups. The control group was treated with prednisone, vitamin B1, vitamin B12, and dibazole; the treatment groups were treated either with acu-moxi alone or in combination with prednisone, Vitamin B1, vitamin B12, and dibazole. Symptoms and signs, the House-Brackmann scale, and facial disability index (FDI) scores were assessed and determined both pre- and post-treatment to evaluate the effectiveness of the treatment methods. RESULTS: The characteristics of the control and two treatment groups were comparable without statistically significant differences before treatment. There were significant differences between the control and treatment groups after treatment (chi(2) = 15.265, P = 0.018). According to evaluations based on the House-Brackmann scale and FDI scores, the effectiveness of treatment in the two treatment groups was better than in the control group and was most effective in patients receiving acu-moxi treatment alone (Z = -2.827, P = 0.005). CONCLUSION: The efficacy of acu-moxi treatment for Bell's palsy is verified scientifically.

Two hundred and fifty-eight cases of hemoptysis treated by injection of vitamin K3 into Chize point.

Injection of vitamin K3 into Chize (LU 5) was used from 1980 to 1992 in 258 inpatients suffering from diseases of the respiratory system accompanied with hemoptysis. As a result, the effective rate was 83% for mild hemoptysis and 72.1% for moderate hemoptysis in the treatment group, as compared to 48.5% and 45.2% respectively in control group A, and to 83.8% and 75% in control group B. There was a significant difference between the treatment group and control group A (P < 0.01). However, the effective rate was only 31.8% for severe hemoptysis in the treatment group, as compared to 81.3% in control group B with a significant difference. It is therefor concluded that injection of vitamin K3 into Chize is a good therapy for mild or moderate hemoptysis but not for severe cases.

The experimental study on treatment of glucocorticoid-induced ischemic necrosis of femoral head by gu fu sheng capsule.

OBJECTIVE: To observe the effects of Gu Fu Sheng Capsule ([symbol: see text] GFSC) on nitric oxide (NO) level and fibrinolytic activity in rabbits with glucocorticoid-induced ischemic necrosis of femoral head (GINFH) so as to explain the therapeutic mechanism of GFSC. METHODS: 48 rabbits were randomly divided into 4 groups: GFSC group A, Ma Shi Gu Pian ([symbol: see text] MSGP) group B, model group C, and control group D. The models of GINFH were produced by injection of glucocorticoid hormone for 8 weeks. From the 9th week, GFSC decoction was given to the rabbits of the group A (45 g x kg(-1) x d(-1)) and MSGP extract was given to the group B (0.17 g kg(-1) x d(-1)) and the same volume of normal saline to group C and D by intragastric infusion. The content of plasma nitric oxide (NO), and activities of tissue-type plasminogen activator (t-PA), and plasminogen activator inhibitor (PAI) were detected after the hormone was injected for 4 and 8 weeks and after the drugs were given for 4 weeks. RESULTS: Compared with the group D, the contents of NO and t-PA activity in the group C decreased significantly (P<0.01), but the activity of PAI increased significantly (P<0.01). Compared with the group C, the contents of NO and t-PA activity in the group A and B increased significantly (P<0.05), but the activity of PAI decreased significantly (P<0.05), with no significant difference between the two treatment groups (P>0.05). CONCLUSION: GFSC could increase the level of NO, protect endotheolial cells and improve fibrinolytic activity, which might be the mechanism of GFSC in curing GINFH.

[Spatial and temporal variation of soil temperature extremum under plastic mulch in Xinjiang].

The upper and lower limit values of soil temperature affect crop growth and development greatly. Observations on the soil maximal and minimal temperatures under different mulching and cropping conditions in Xinjiang showed that during crop growth period in 1998 and 1999, soil temperature extremums were both at 0 cm, and varied with different observation time. The soil minimal temperature under plastic mulch was higher than that without mulch, indicating that plastic mulch could remarkably increase soil temperature. The diurnal variation of soil minimal temperature could be expressed by quadratic function, while the maximal temperature at 14:00 and 20:00 could be expressed by ellipse function and linear function of soil depth, respectively. Soil temperature extremums correlated linearly with air temperature under different conditions. The correlation between soil minimal temperature and air temperature was higher for bare soil than for mulched soil, and higher for maize field than for cotton field, while the correlation between soil maximal temperature and air temperature was lower than that between soil minimal temperature and air temperature.

[Laser fluorescence excited spectrum of NO via alpha2sigma<--chi2pi transition].

Two-photon fluorescence excited spectrum of NO induced by Nd: YAG laser pumped optical parameter generator/amplifier as excitation source was obtained in the range of 420-472 nm. With this technique, the structure of the energy levels of NO molecule in alpha2sigma electronic state was investigated. The peaks of the spectrum were attributed to alpha2sigma(v' = 0,1)<--chi2pi(v" = 0) transition. The near square dependence of fluorescence signal on the laser intensity indicates a two-photon process. The ground-vibrational-state oscillation frequency and the force constant of alpha2sigma state were calculated. The fluorescence lifetime of alpha2sigma(v' = 0) state under the pressure of 266 Pa was also obtained by measuring fluorescence decay curve of alpha2sigma(v' = 0) states. It is about 53.76 ns. Fitting the curve of the fluorescence radiant lifetime versus pressure, the spontaneous radiant lifetimes and the rate coefficient of nonradiative transition relaxation of alpha2sigma(v' = 0,1) states were deduced.

Evidence that transgenes encoding components of the Wnt signaling pathway preferentially induce mammary cancers from progenitor cells.

Breast cancer is a genetically and clinically heterogeneous disease, and the contributions of different target cells and different oncogenic mutations to this heterogeneity are not well understood. Here we report that mammary tumors induced by components of the Wnt signaling pathway contain heterogeneous cell types and express early developmental markers, in contrast to tumors induced by other signaling elements. Expression of the Wnt-1 protooncogene in mammary glands of transgenic mice expands a population of epithelial cells expressing progenitor cell markers, keratin 6 and Sca-1; subsequent tumors express these markers and contain luminal epithelial and myoepithelial tumor cells that share a secondary mutation, loss of Pten, implying that they arose from a common progenitor. Mammary tumors arising in transgenic mice expressing beta-catenin and c-Myc, downstream components of the canonical Wnt signaling pathway, also contain a significant proportion of myoepithelial cells and cells expressing keratin 6. Progenitor cell markers and myoepithelial cells, however, are lacking in mammary tumors from transgenic mice expressing Neu, H-Ras, or polyoma middle T antigen. These results suggest that mammary stem cells and/or progenitors to mammary luminal epithelial and myoepithelial cells may be the targets for oncogenesis by Wnt-1 signaling elements. Thus, the developmental heterogeneity of different breast cancers is in part a consequence of differential effects of oncogenes on distinct cell types in the breast.

Phosphoinositide 3-kinase promotes adult subventricular neuroblast migration after stroke.

Rodent adult subventricular zone (SVZ)-derived progenitor cells abandon the rostral migratory stream (RMS)/olfactory complex postmiddle cerebral artery occlusion (MCAo) and migrate into compromised tissue, possibly playing a role in brain recovery. Using SVZ tissue explants from the adult rat, we investigated the role of the phosphoinositide 3-kinase (PI3K) signal transduction pathway in the migration of SVZ cells. Stroke significantly (P <.01) increased migratory speed (198 +/- 39 microm/day) of neuroblasts out of the SVZ explants compared with the speed (99 +/- 20 microm/day) in the normal SVZ (nSVZ) explants within the first 3 days of incubation. Three-dimensional laser scanning confocal microscopy revealed formation of neuroblast encompassing chain-like astrocyte structures extruding from both normal and stroke explants. Western blots showed that stroke SVZ (sSVZ) explants increased Akt phosphorylation. Treatment of sSVZ explants with the selective PI3K inhibitor LY294002 significantly (P <.01) attenuated neuroblast migration and Akt phosphorylation, whereas treatment with LY294002 did not affect the number of bromodeoxyuridine (BrdU)- and caspase-3-immunoreactive cells, indicating that stroke-enhanced neuroblast migration is independent of cell proliferation and survival. PI3K catalyzes phosphatidylinositol-3,4,5-triphosphate (PIP(3)) which facilitates Akt phosphorylation. Thus, our data demonstrate that the PI3K/Akt signal transduction pathway mediates neuroblast migration after stroke.

Chirality transfer from guest chiral metal complexes to inorganic framework: the role of hydrogen bonding.

The structure elucidation of a new zinc phosphate [Co(II)(en)(3)][Zn(4)(H(2)PO(4))(3)(HPO(4))(2)(PO(4))(2 H(2)O)(2)] (1) reveals that the racemic cobalt complex templates the zinc phosphate framework in such a way that the local C(2) point symmetry of the structural motif of the inorganic framework conforms with that of the cobalt complex pairing with it, in essence transferring its chirality to the inorganic host. An analysis of hydrogen bonding between the guest molecules and the inorganic host framework reveals that hydrogen bonding is responsible for the stereospecific structural arrangement. Upon examining previously reported chiral metal-complex-templated structures of metal phosphates, it is revealed that such hydrogen bonding is the common origin for inducing chirality transfer in metal-phosphate frameworks templated with chiral metal complexes.
