Myostatin induces cyclin D1 degradation to cause cell cycle arrest through a phosphatidylinositol 3-kinase/AKT/GSK-3 beta pathway and is antagonized by insulin-like growth factor 1.

Myostatin is a transforming growth factor beta superfamily member and is known as an inhibitor of skeletal muscle cell proliferation and differentiation. Exposure to myostatin induces G1 phase cell cycle arrest. In this study, we demonstrated that myostatin down-regulates Cdk4 activity via promotion of cyclin D1 degradation. Overexpression of cyclin D1 significantly blocked myostatin-induced proliferation inhibition. We further showed that phosphorylation at threonine 286 by GSK-3beta was required for myostatin-stimulated cyclin D1 nuclear export and degradation. This process is dependent upon the activin receptor IIB and the phosphatidylinositol 3-kinase/Akt pathway but not Smad3. Insulin-like growth factor 1 (IGF-1) treatment or Akt activation attenuated the myostatin-stimulated cyclin D1 degradation as well as the associated cell proliferation repression. In contrast, attenuation of IGF-1 signaling caused C2C12 cells to undergo apoptosis in response to myostatin treatment. The observation that IGF-1 treatment increases myostatin expression through a phosphatidylinositol 3-kinase pathway suggests a possible feedback regulation between IGF-1 and myostatin. These findings uncover a novel role for myostatin in the regulation of cell growth and cell death in concert with IGF-1.

Analysis and design of terahertz photonic crystal fibers by an effective-index method.

An effective-index method (EIM) is used to analyze and design photonic crystal fibers (PCFs) for the terahertz radiation. By building an analogy between a conventional optical fiber and a PCF, the EIM solves the effective index of the fiber cladding and the effective modal index of a PCF analytically. The EIM is first validated by comparison with available data in the reference, showing that the role of material dispersion is negligible at higher frequencies. Terahertz PCFs with flattened dispersion are designed based on this method and the scaling property of the Maxwell equations.

Comorbidity between and risk factors for severe hearing and memory impairment in older Americans.

OBJECTIVE: Comorbidities complicate our understanding of impairments and their risk factors. The objective of the current study was to assess the comorbidity between hearing impairment (HI) and memory impairment (MI) in older Americans and to assess to what extent they share common risk factors. METHOD: Data were examined from 5252 participants aged 65 years or older from the Third National Health and Nutrition Examination Survey (1988-1994). RESULTS: The prevalence of HI and MI was 21.4 (SE 0.8)% and 8.3 (0.6)%, respectively, but the prevalence of concurrent impairment was only 2.1 (0.2)%. African Americans had higher odds of MI than whites (OR=3.75, 95% CI: 2.81, 5.00), but not of HI (OR=0.62, 95% CI: 0.42, 0.91). Leisure physical activity was associated with lower odds of MI, good lung function was associated with lower odds of HI, and moderate alcohol use was associated with lower odds of both HI and MI. Compared with individuals free of HI, the OR of MI was 1.07 (95% CI: 0.78, 1.47) for individuals with HI. CONCLUSION: Although prevalence was relatively high, comorbidity was surprisingly low, and the two impairments were not significantly associated. In addition to age, other factors appear to play critical but different roles in the development of these impairments.

Effect of polypeptides in bee venom on growth inhibition and apoptosis induction of the human hepatoma cell line SMMC-7721 in-vitro and Balb/c nude mice in-vivo.

Polypeptides in bee venom (PBV) produced a significant growth inhibition against SMMC-7721 human hepatoma cell line. Analysis of the mechanisms of cell death indicated that PBV induced an apoptotic cell death. SMMC-7721 cells exposed to PBV (10.0 microg mL(-1)) produced an insignificant morphological change. Analysis of the cytotoxicity with the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium) assay confirmed that the cytotoxic effects of PBV were dose- and timedependent. The result of Ki67 immunohistochemistry demonstrated that the proliferation of SMMC-7721 cells treated with PBV (10.0 mug mL(-1)) was inhibited. The apoptotic cell death was then confirmed by annexin V, propidium iodide staining and DNA fragmentation analysis. In in-vivo experiments, treatment with PBV (1.5 or 3 mg kg(-1)) resulted in a significant retardation of SMMC-7721 cell growth in Balb/c nude mice. These findings suggested that PBV could be used as a chemotherapeutic agent against tumours.

Evaluation of modifying collagen matrix with RGD peptide through periodate oxidation.

The aim of the study is to evaluate the effect of modifying collagen matrices with Arg-Gly-Asp (RGD) peptide through periodate oxidation. The collagen matrices were modified with RGD peptide, by periodate activation. The modified collagen matrices and unmodified matrices were characterized by scanning electron microscopy (SEM), differential scanning calorimetry (DSC), and electron spectroscopy for chemical analysis (ESCA). Mesenchymal stem cells (MSCs) were used to evaluate the cell compatibility of collagen matrices. In terms of cell growth, the MSCs attached much better on the modified matrix than on the unmodified one. But there was no significant difference between two groups regarding the MSC proliferation. Compared to the unmodified matrices, the mechanical strength of the modified matrix decreased sharply, and its 3D structure was destroyed. Introducing specific RGD receptor-mediated adhesion sites on matrices obviously enhanced the MSC adhesion on collagen matrices, but the coupled method of periodate oxidation would likely result in the declination of the mechanical strength of the matrix, as well as the destruction of the matrix structure. This would affect the cell growth on the matrix, and decrease the histocompatibility of the matrices.

How does a suicide attempter eat differently from others? Comparison of macronutrient intakes.

OBJECTIVE: Issues of the subtle effects of diet and nutrition on suicidal behaviors are rarely examined. This study describes the differences in dietary intakes of macronutrients between individuals who had ever made a suicide attempt and those who never had. METHODS: We examined the data of 7631 non-institutionalized adults ages 17 to 39 y who completed a mental disorder diagnostic interview and a 24-h dietary recall as a part of the Third National Health and Nutrition Examination Survey, 1988 to 1994. Multivariable adjusted means of daily caloric intake and energy percentage from macronutrients were calculated for individuals who had a history of lifetime suicide attempt(s) (n = 408) and compared with those who did not. RESULTS: After adjustment for covariates, including socioeconomic indicators, health risk factors, and history of medical and psychiatric illnesses, we observed that the means of daily caloric intake, cholesterol, and energy percentages from carbohydrate, total fat, saturated fat, monounsaturated fat, and protein did not differ significantly between attempters and non-attempters. However, energy percentage from polyunsaturated fat was lower in attempters than in non-attempters. Mean percentages +/- standard error for smoking attempters, non-smoking attempters, smoking non-attempters, and non-smoking non-attempters were 5.5 +/- 0.4 (P = 0.01), 6.7 +/- 0.2 (P = 0.02), 7.5 +/- 1.4 (P = 0.78), and 7.2 +/- 0.1 (reference), respectively, in men, and 6.3 +/- 0.4 (P = 0.01), 7.2 +/- 0.2 (P = 0.07), 7.3 +/- 1.0 (P = 0.89), and 7.5 +/- 0.1 (reference), respectively, in women. Dietary fiber intake was also lower in attempters than in non-attempters. These differences were independent of daily caloric intake. CONCLUSIONS: Individuals with a history of suicide attempt have low intakes of polyunsaturated fat and fiber. Potential deleterious effects of low intake of polyunsaturated fat and fiber among suicide attempters merit further investigation.

Study on modification of collagen with chondroitin sulfate on the microcosmic level.

To observe the changes during the process of modificating collagen with chondroitin sulfate (CS), a series of experiments was designed. Collagen scaffold was constructed by lyophilization. CS was attached into the scaffold through crosslinking. Solid-liquid phase separation method was used to shape 3D porous structure. Both kinds of scaffolds were investigated by Transmission Electron Microscopy (TEM), Scanning Electron Microscopy (SEM), and Electron Spectroscopy for Chemical Analysis (ESCA), Amino Acid Analyses (AAA), and so on for structures and characteristics. The results showed that CS could improve the properties of collagen both in biocompatibility and mechanical property. It increased the polar groups on the surface of scaffold and protected amino acid residues of collagen fiber. At the same time, the structure of scaffold became more regular because of the increase of polar groups. It was indicated that those were the real reasons for the improvement of modification.

[Individualized energy metabolic monitoring and its clinical application].

The nutritional status affects the factors of patient outcome such as the therapeutic intervention, the length of hospitalization and prognosis. Personalization of nutritional support treatments can minimize the complications that may arise from overfeeding and underfeeding. Appropriate nutritional support requires an individual nutrition assessment based on each patient's caloric and regularly monitored respiratory quotient to accommodate continuing changes in the patient's condition.

The presequence translocase-associated protein import motor of mitochondria. Pam16 functions in an antagonistic manner to Pam18.

Transport of preproteins into the mitochondrial matrix requires the presequence translocase of the inner membrane (TIM23 complex) and the presequence translocase-associated motor (PAM). The motor consists of five essential subunits, the mitochondrial heat shock protein 70 (mtHsp70) and four cochaperones, the nucleotide exchange-factor Mge1, the translocase-associated fulcrum Tim44, the J-protein Pam18, and Pam16. Pam16 forms a complex with Pam18 and displays similarity to J-proteins but lacks the canonical tripeptide motif His-Pro-Asp (HPD). We report that Pam16 does not function as a typical J-domain protein but, rather, antagonizes the function of Pam18. Pam16 specifically inhibits the Pam18-mediated stimulation of the ATPase activity of mtHsp70. The inclusion of the HPD motif in Pam16 does not confer the ability to stimulate mtHsp70 activity. Pam16-HPD fully substitutes for wild-type Pam16 in vitro and in vivo but is not able to replace Pam18. Pam16 represents a new type of cochaperone that controls the stimulatory effect of the J-protein Pam18 and regulates the interaction of mtHsp70 with precursor proteins during import into mitochondria.

Pam16 has an essential role in the mitochondrial protein import motor.

Mitochondrial preproteins destined for the matrix are translocated by two channel-forming transport machineries, the translocase of the outer membrane and the presequence translocase of the inner membrane. The presequence translocase-associated protein import motor (PAM) contains four essential subunits: the matrix heat shock protein 70 (mtHsp70) and its three cochaperones Mge1, Tim44 and Pam18. Here we report that the PAM contains a fifth essential subunit, Pam16 (encoded by Saccharomyces cerevisiae YJL104W), which is selectively required for preprotein translocation into the matrix, but not for protein insertion into the inner membrane. Pam16 interacts with Pam18 and is needed for the association of Pam18 with the presequence translocase and for formation of a mtHsp70-Tim44 complex. Thus, Pam16 is a newly identified type of motor subunit and is required to promote a functional PAM reaction cycle, thereby driving preprotein import into the matrix.

[Progress in researches on sperm antigen fertilin beta].

Fertilin beta plays an important role in fertilization by its disintegrin domain as a sperm-specific antigen. This paper reviews its structure, localization and roles in fertilization, and suggests that fertilin beta, as an important target antigen, has a very promising value in the development of human immunocontraceptive vaccine.

[Present research on the male germ cell differentiation in vitro].

Several culture systems such as tissue and organ culture system, seminiferous tubules culture system, co-culture systems of Sertoli cells and germ cells for male germ cell differentiation studies in vitro have been established. The importance of the blood-testis barrier and the polarity of testicular cells have been emphasized recently. And then the bicameral chambers culture system that can mimic the compartmental structure of testes and the calcium algmate culture system that can provide three-dimensional support have been primarily set up. These culture systems are powerful tools for facilitating the understanding to the spermatogenesis. The entire meiotic part of spermatogenesis in some animals has been achieved in vitro by different laboratories, but little is known about the regulation mechanism of the meiotic step of spermatogenesis in detail. This review focuses on the present studies on the differentiation of male germ cells, especially the meiotic step of the differentiation.

[Immunomagnetic beads sorting and functional identification of human spermatogonial stem cells].

OBJECTIVE: The very nature of spermatogonial stem cells (SSC) is still poorly understood. The objective of this study is to explore the specific markers of human SSC and search for the suitable method for their isolation and functional identification. METHODS: Adults testicular cell suspensions were sorted by immunomagnetic beads method using alpha6,beta1 integrin and Thy-1 markers. The light-scattering properties and DNA Ploidy of the resultant subpopulations were analyzed by flow cytometry. The efficacies of the sorting on human SSC were evaluated by germ cell transplantation. RESULTS: (1) The alpha6+ Thy-1+ c-kit- and beta1+ Thy-1+ c-kit- cells were relatively uniform subpopulations in size and morphology, which represented about 2%-3% and 0.5%-1% of the unsorted testis cells, respectively. The analysis of light-scattering properties showed that both of the subpopulations had low side light-scattering properties. The DNA Ploidy analysis showed significant changes of these two cell subpopulations in DNA Ploidy. The percentage of diploid cells in alpha6+ Thy-1+ c-kit- cell subpopulation significantly increased to 51.2% and synthesis phase and tetraploid cells disappeared. (2) The functional evaluation showed that the SSC in the alpha6+ Thy-1+ c-kit- cells were enriched 40 times and the SSC in the beta1+ Thy-1+ c-kit- cells 20 times that of the unsorted cells. CONCLUSION: The alpha6,beta1 integrin and Thy-1 may be used for the SSC isolation as positive markers. The immunomagnetic beads sorting using alpha6,beta1 integrin and Thy-1 markers can result in significant enrichment of human SSC. It will open up a wide prospect for the researches on the biology of human SSC and the treatment of male sterility.

A J-protein is an essential subunit of the presequence translocase-associated protein import motor of mitochondria.

Transport of preproteins into the mitochondrial matrix is mediated by the presequence translocase-associated motor (PAM). Three essential subunits of the motor are known: mitochondrial Hsp70 (mtHsp70); the peripheral membrane protein Tim44; and the nucleotide exchange factor Mge1. We have identified the fourth essential subunit of the PAM, an essential inner membrane protein of 18 kD with a J-domain that stimulates the ATPase activity of mtHsp70. The novel J-protein (encoded by PAM18/YLR008c/TIM14) is required for the interaction of mtHsp70 with Tim44 and protein translocation into the matrix. We conclude that the reaction cycle of the PAM of mitochondria involves an essential J-protein.
