[Roles of ERK1/2 MAPK in vitamin E succinate-induced apoptosis in human gastric cancer SGC-7901 cells].

In order to investigate the roles of ERK1/2 mitogen-activated protein kinase in vitamin E succinate (VES)-induced apoptosis in human gastric cancer SGC-7901 cells, apoptosis was observed by DAPI staining and the phosphorylation of ERK1/2 by VES at different doses and different time points was measured by western blot. The results showed that VES obviously induced cells to undergo apoptosis and apoptotic rate after 24 h and 48 h of treatment with VES at 20 micrograms/ml. VES was 14.2% and 89.4%, respectively. The expression of p-ERK was evidently reduced by VES at 5, 10 and 20 micrograms/ml for 24 h. ERK1/2 was immediately activated by VES at 20 micrograms/ml, but the expression was decreased for 2 h, then increased again and reached the top level for 12 h. The data implicated that ERK1/2 pathway might be involved in VES-induced apoptosis, but in the proliferation eventually.

[Effects of soil water stress and shading on growth characteristics of ginger].

Water stress decreased the chlorophyll contents, stomatic density, and stomatic size of ginger leaves, and reduced the root activity and magnitude of bleeding sap of ginger roots. Under the same water contents, shading increased the root activity and the chlorophyll contents and stomatic size of upper cuticulae of ginger leaves, but decreased the stomatic density and size of their lower cuticulae. Water stress influenced the growth and yield of ginger significantly, but the effects of light intensity on ginger growth and yield were different under different water contents. Under normal water content, ginger grew better and yield was higher in natural sunlight, while under water stress, ginger grew better and yield was higher in shading.

[The determination of konjac glucomannan in konjac refined powder and monosaccharide compositions by HPLC].

OBJECTIVE: To establish a quantitative method for the content determination and monosaccharide composition analysis of Konjac glucomannan (KGM) in Konjac refined powder by pre-column derivatization high performance liquid chromatographic method (HPLC). METHOD: The two derivatives combined reducing monosaccharides with 1-phenyl-3-methyl-5-pyrazolone (PMP) were separated by reverse-phase HPLC using a developed fragment gradient elution process, and monitored by ultraviolet detector at 250 nm. The broad reagent peak of PMP was separated very well from all the PMP-sugars, and good separation was achieved for derivatives of mannose and glucose. The quantitative methods of two reducing monosaccharides were studied by the method combined internal and external standard; while the KGM content in Konjac refined powder was determined. RESULT: Linearity of glucose was good (r = 0.9990) in range of 1.002-8.016 nmol; while mannose (r = 0.9994) in range of 1.001-8.008 nmol. The average recovery of this method was 98.1%, RSD of repeatability was 1.72%. KGM content in Konjac refined powder was 79.5%, ratio of glucose to mannose in KGM was 1:1.51. CONCLUSION: This method is a sample, convenient and rapid method that can determine KGM content and analyze monosaccharide compositions in KGM, which will be helpful to quality assessment of Konjac refined powder.

[The new safe strategy on the marker genes in transgenic plant].

The presence in transgenic plants of antibiotic and herbicide resistant selective marker genes might be an unpredictable hazard to the ecosystem as well as to human health. There are two applicable strategies can be used to resolve this problem. One is to remove the resistant marker genes before the transgenic plants are released to field. The other is to develop and use safe marker genes to produce transgenic plants. The present paper reviewed three technique systems employed to remove resistant marker genes in transgenic plants and several safe marker genes used in plant transformation.

[Changes in mechano-receptive properties of Abeta-fiber induced by antidromical electrical stimulation of the cutaneous nerve from adjacent spinal segment].

The purpose of the present study was to observe whether primary afferent Abeta-fiber is involved in the information transmission between peripheral terminals of adjacent dermatomes. The dorsal cutaneous nerve branches of spinal nerves from T(8) to T(12) segments were cut proximally. One peripheral stump end of the cut nerves was dissected into a few filaments for the examination of mechanoreceptive properties of single Abeta-fibers and their discharges were observed while the other end was stimulated antidromically. Fifty Abeta-units were recorded in forty-two intact rats. After an electrical stimulation (0.45 mA, 0.1 ms, 20 Hz, for 10 s) was delivered to the stimulated nerve, the size of the receptive field of 60.6% (n=33) Abeta-fibers extended. The mean area of receptive fields of all examined units enlarged from 8.94+/-6.51 mm(2) to 20.34+/-16.17 mm(2) (P<0.01) and the shapes of the receptive fields of 81.8% (n=20) units changed from a dot, round or ellipse with its long axis in parallel with the longitudinal axis of the body to an oblique ellipse with the longitudinal axis of the body. The mechanoreceptive threshold of 68.0% (n=50) units decreased with a reduction in mean threshold from 2.37+/-1.24 to 2.29+/-1.24 mN (P<0.05). The duration of these changes in mechano-receptive properties increased from 52.23+/-9.27 to 56.93+/-15.76 min. Meanwhile, increasing discharge was found in 50.0% (n=50) units but lasted only for 1.52+/-0.46 min. The changes in mechanoreceptive properties appeared simultaneously with discharge changes but had longer duration than that of discharge change (P<0.01). Discharges changes usually appeared in those units with the changes in mechanoreceptive properties following an antidromical electrical stimulation of adjacent spinal segment. These results suggest that low-threshold mechanoreceptive Abeta-fibers are affected by antidromical electrical stimulation of the cutaneous nerve from an adjacent spinal segment, indicating that information transmission occurs between the two endings of peripheral afferent nerves from adjacent spinal segments without any involvement of the central nervous system, and that Abeta-fibers are involved in the process of information transmission between peripheral terminals from adjacent spinal segments.

Genome-wide scan for familial nasopharyngeal carcinoma reveals evidence of linkage to chromosome 4.

Nasopharyngeal carcinoma (NPC) occurs with high frequency in Asian populations, especially among people of Cantonese ancestry. In areas with high incidence, NPC clusters in families, which suggests that both geography and genetics may influence disease risk. Although the HLA-Bw46 locus is associated with increased risk of NPC, no predisposing genes have been identified so far. Here we report the results of a genome-wide search carried out in families at high risk of NPC from Guangdong Province, China. Parametric analyses provide evidence of linkage to the D4S405 marker on chromosome 4 with a logarithm of odds for linkage (lod) score of 3.06 and a heterogeneity-adjusted lod (hlod) score of 3.21. Fine mapping with additional markers flanking D4S405 resulted in a lod score of 3.54 and hlod score of 3.67 for the region 4p15.1-q12. Multipoint nonparametric linkage analysis gives lod scores of 3.54 at D4S405 (P = 5.4 x 10(-5)) and 4.2 at D4S3002 (P = 1.1 x 10(-5)), which is positioned 4.5 cM away from D4S405. When Epstein Barr virus antibody titer was included as a covariate, the lod scores reached 4.70 (P = 2.0 x 10(-5)) and 5.36 (P = 4.36 x 10(-6)) for D4S405 and D4S3002, respectively. Our findings provide evidence of a major susceptibility locus for NPC on chromosome 4 in a subset of families.

[Study of treatment of refractory rheumatoid arthritis with autologous peripheral blood stem cell transplantation].

OBJECTIVE: To explore the efficacy, safety and immune reconstitution of autologous peripheral blood stem cell transplantation (APBSCT) using T cell depleted grafts in the treatment of refractory rheumatoid arthritis (RA). METHODS: One patient with RA was treated with APBSCT. The method included mobilization with 2 g/m(2) cyclophosphamide (CY) and subcutaneous injection of granulocyte-colony stimulating factor (G-CSF). Immunomagnetic selection of CD34(+) cells from the leukapheresis products was performed to deplete potentially autoreative lymphocytes. The conditioning regimen consisted of intravenous administration of 2 g/m2 CY and 90 mg/kg ATG, with subsequent reinfusion of the graft. G-CSF was used to help hematopoietic and immunologic reconstitution. Phenotype of the peripheral blood lymphocytes was analyzed to observe the immunologic reconstitution after transplantation. RESULTS: The patient completed the mobilization, conditioning regimen and transplantation successfully. The hematologic recovery was rapid and the patient achieved clinical remission. The erythrocyte sedimentation rate (ESR), C-reactive protein (CRP) decreased to normal level and the rheumatoid factor (RF) turned negative after a follow-up of 12 months. An ongoing course of immunologic reconstitution was observed. CONCLUSION: APBSCT is effective and safey for refractory RA, and can induce improvement of disease activity. The course of immunologic reconstitution after transplantation remains to be observed in long-term followup.

[Effects of electrical stimulation of the dorsal cutaneous branches of spinal nerves on the discharge activity of remote mechanoreceptive units in rats].

The method of isolating filaments of the dorsal cutaneous branches was used to observe the effects of antidromic electrical stimulation of the spinal nerves on the discharge of remote A delta and C mechanoreceptive units in rats. Seventy-nine mechanoreceptive units were recorded from the T12 nerve filaments after stimulation of the dorsal cutaneous branches of T9 spinal nerve. It was found that the discharge frequency of 59.3% (16/27) A delta-units and 71.2% (37/52) C-units significantly increased during 90-120 s after the stimulation. Sixty-four mechanoreceptive units were recorded from the T12 nerve filaments after stimulation of the dorsal cutaneous branches of T8 spinal nerves. The discharge frequency of 47.8% (11/23) A delta-units and 36.6% (15/41) C-units significantly increased during 120-150 s after the stimulation. In addition, the threshold of the majority of these mechanoreceptors (78.3%, 18/23) decreased after the stimulation. The results suggest that antidromic electrical stimulation of the dorsal cutaneous branches of spinal nerves leads to sensitization of A delta and C mechanoreceptive units of the remote peripheral nerve endings, which results in an increase in afferent discharge of these units.

Identification, characterization, and localization of a novel kidney polycystin-1-polycystin-2 complex.

The functions of the two proteins defective in autosomal dominant polycystic kidney disease, polycystin-1 and polycystin-2, have not been fully clarified, but it has been hypothesized that they may heterodimerize to form a "polycystin complex" involved in cell adhesion. In this paper, we demonstrate for the first time the existence of a native polycystin complex in mouse kidney tubular cells transgenic for PKD1, non-transgenic kidney cells, and normal adult human kidney. Polycystin-1 is heavily N-glycosylated, and several glycosylated forms of polycystin-1 differing in their sensitivity to endoglycosidase H (Endo H) were found; in contrast, native polycystin-2 was fully Endo H-sensitive. Using highly specific antibodies to both proteins, we show that polycystin-2 associates selectively with two species of full-length polycystin-1, one Endo H-sensitive and the other Endo H-resistant; importantly, the latter could be further enriched in plasma membrane fractions and co-immunoprecipitated with polycystin-2. Finally, a subpopulation of this complex co-localized to the lateral cell borders of PKD1 transgenic kidney cells. These results demonstrate that polycystin-1 and polycystin-2 interact in vivo to form a stable heterodimeric complex and suggest that disruption of this complex is likely to be of primary relevance to the pathogenesis of cyst formation in autosomal dominant polycystic kidney disease.

Polycystin-2 expression is increased following experimental ischaemic renal injury.

BACKGROUND: Mutations in PKD2 account for 15% of patients with autosomal dominant polycystic kidney disease. Expression of the PKD2 protein, polycystin-2, is developmentally regulated, suggesting a major role for this protein during nephrogenesis. However, the regulation of polycystin-2 expression in the adult kidney has not been previously explored. METHODS: We have utilized an established model of renal ischaemic injury to study polycystin-2 expression in adult rat kidney for up to 120 h following ischaemia. RESULTS: Our results indicate that polycystin-2 expression is increased in the post-ischaemic kidney by up to 5-fold, with a peak in expression at 48 h reperfusion. This time course mirrored the increase in cell proliferation observed. In the non-ischaemic kidney, polycystin-2 expression was highest in distal nephron segments but faint proximal tubular staining was also observed. No expression was seen in glomeruli. In the ischaemic kidney, polycystin-2 expression was greatly increased but the increase in expression was not restricted to segments with the highest number of proliferating cells. Moreover, polycystin-2 was detectable mainly intracellularly following ischaemia. Consistent with this, polycystin-2 was completely sensitive to endoglycosidase H during renal recovery, suggesting that it remains largely retained within the endoplasmic reticulum under these conditions. CONCLUSIONS: Our results provide the first evidence that polycystin-2 is increased following renal ischaemia, but show that this increase is not restricted to actively proliferating cells. The increase in polycystin-2 may relate instead to the process of cellular repair or differentiation following injury.

RRR-alpha-tocopheryl succinate inhibits human gastric cancer SGC-7901 cell growth by inducing apoptosis and DNA synthesis arrest.

AIM: To investigate the effects of growth inhibition of human gastric cancer SGC-7901 cell with RRR-alpha-tocopheryl succinate (VES), a derivative of natural Vitamin E, via inducing apoptosis and DNA synthesis arrest. METHODS: Human gastric cancer SGC-7901 cells were regularly incubated in the presence of VES at 5, 10 and 20mg x L(-1) (VES was dissolved in absolute ethanol and diluted in RPMI 1640 complete condition media correspondingly to a final concentration of VES and 1 mL x L(-1) ethanol), succinic acid and ethanol equivalents as vehicle (VEH) control and condition media only as untreated (UT) control. Trypan blue dye exclusion analysis and MTT assay were applied to detect the cell proliferation. Cells were pulsed with 37kBq of tritiated thymidine and (3H) TdR uptake was measured to observe DNA synthesis. Apoptotic morphology was observed by electron microscopy and DAPI staining. Flow cytometry and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay were performed to detect VES-triggered apoptosis. RESULTS: VES inhibited SGC-7901 cell growth in a dose-dependent manner. The growth curve showed suppression by 24.7%, 49.2% and 68.7% following 24h of VES treatment at 5, 10 and 20 mg x L(-1), respectively, similar to the findings from MTT assay. DNA synthesis was evidently reduced by 35%, 45% and 98% after 24h VES treatment at 20mg x L(-1) and 48 h at 10 and 20mg x L(-1), respectively. VES induced SGC-7901 cells to undergo apoptosis with typically apoptotic characteristics, including morphological changes of chromatin condensation, chromatin crescent formation/margination, nucleus fragmentation and apoptotic body formation, typical apoptotic sub-G1 peak by flow cytometry and increase of apoptotic cells by TUNEL assay in which 90% of cells underwent apoptosis after 48 h of VES treatment at 20 mg x L(-1). CONCLUSION: VES can inhibit human gastric cancer SGC-7901 cell growth by inducing apoptosis and DNA synthesis arrest. Inhibition of SGC-7901 cell growth by VES is dose- and time-dependent. Therefore VES can function as a potent chemotherapeutic agent against human gastric carcinogenesis.

Effects of cryopreservation and phenylacetate on biological characters of adherent LAK cells from patients with hepatocellular carcinoma.

AIM: To improve the preparation of adherent lymphokine-activated killer (A-LAK) cells and to study the effects of cryopreservation and phenylacetate (PA) on biological characters of A-LAK cells. METHODS: A-LAK cells were obtained from peripheral blood mononuclear cells (PBMCs) of the patients with hepatocellular carcinoma (HCC) by using L-phenylalanine methyl ester (PME) to deplete immunosuppressive monocytes. Proliferative activity of SMMC7721 cell line after treatment with phenylacetate (PA) was observed. A-LAK cells were treated with the supernatant of SMMC7721 cells that had been pretreated with PA. The changes of proliferation, cytotoxicity and phenotype of A-LAK cells were investigated after cryopreservation. RESULTS: The expansion of A-LAK cells (96.79 +/- 69.10 folds on Day 14) was significantly higher than that of non-adherent LAK (NA-LAK) cells (22.77 +/- 13.20) as well as conventional LAK cells (4.64 +/- 0.91). PA significantly suppressed the growth of SMMC7721 cells, and the inhibitor ratio was 46%. The supernatant of cultured tumor cells intensively suppressed the proliferation and cytotoxicity of A-LAK cells, but the suppressive effect of the supernatant was previously decreased after treatment with PA. Impairments in proliferation and cytotoxicity of A-LAK cells immediately after thawing of cryopreservation and recovery after reincubation with IL-2 were observed. The cytotoxicity of thawed A-LAK cells on Day 5 was significantly higher than that of fresh A-LAK before freezing (54.8 +/- 10.2% vs 40.5 +/- 6.4%). No significant change in the percentage of lymphocyte subsets was identified in frozen A-LAK cells as compared with that in the fresh control cells. CONCLUSION: A-LAK cells can be simply prepared by using PME, and showed a synergistic anti-tumor effect with the combination of PA. Cryopreservation can increase the immunoactivities of A-LAK cells from the patients with hepatocellular carcinoma.

The effects of vitamin E succinate on the expression of c-jun gene and protein in human gastric cancer SGC-7901 cells.

AIM: To investigate the effects of vitamin E succinate (VES) on the expression of c-jun gene and protein in human gastric cancer SGC-7901 cells. METHODS: After SGC-7901 cells were treated with VES at different doses (5,10,20 mg x L(-1)) at different time, reverse transcription-PCR technique was used to detect the level of c-jun mRNA; Western Blot was applied to measure the expression of c-jun protein. RESULTS: After the cells were treated with VES at 20 mg x L(-1) for 3 h, the expression rapidly reached its maximum that was 3.5 times of UT control (P<0.01). The level of c-jun mRNA was also increased following treatment of VES for 6 h. However, the expression after treatment of VES at 5 mg x L(-1) for 24 h was 1.6 times compared with UT control (P<0.01). Western blot analysis showed that the level of c-jun protein was obviously elevated in VES-treated SGC-7901 cells at 20 mg x L(-1) for 3 h. The expression of c-jun protein was gradually increased after treatment of VES at 20 mg x L(-1) for 3, 6, 12 and 24 h, respectively, with an evident time-effect relationship. CONCLUSION: The levels of c-jun mRNA and protein in VES-treated SGC-7901 cells were increased in a dose- and time-dependent manner; the expression of c-jun was prolonged by VES, indicating that c-jun is involved in VES-induced apoptosis in SGC-7901 cells.

Roles of Fas signaling pathway in vitamin E succinate-induced apoptosis in human gastric cancer SGC-7901 cells.

AIM: To investigate the roles of Fas signaling pathway in vitamin E succinate-induced apoptosis in human gastric cancer SGC-7901 cells. METHODS: Human gastric cancer SGC-7901 cells were treated with VES at 5, 10, 20 mg x L(-1), succinic acid and vitamin E as vehicle control and condition media only as untreated (UT) control. Apoptotic morphology was observed by DAPI staining. Western blot analysis was applied to measure the expression of Fas, FADD and caspase-8 proteins. After the cells were transiently transfected with Fas and FADD antisense oligonucleotides, respectively, caspase-8 activity was determined by flurometric method. RESULTS: The morphologically apoptotic changes were observed after VES treatment by DAPI staining. 23.7 % and 89.6 % apoptosis occurred after 24 h and 48 h of 20 mg x L(-1) VES treatment, respectively. The protein levels of Fas, FADD and caspase-8 were evidently increased in a dose-dependent manner after 24 h of VES treatment. The blockage of Fas by transfection with Fas antisense oligonucleotides obviously inhibited the expression of FADD protein. After SGC-7901 cells were transfected with Fas and FADD antisense oligonucleotides, caspase-8 activity was obviously decreased (P<0.01), whereas Fas blocked more than FADD. CONCLUSION: VES-induced apoptosis in human gastric cancer SGC-7901 cells involves Fas signaling pathway including the interaction of Fas, FADD and caspase-8.
