[Associated study on interleukin 10 gene promoter polymorphisms related to hepatitis B virus infection in Chinese Han population].

OBJECTIVE: To investigate the association that the polymorphisms of interleukin 10 gene (IL10) promoter region are related to the susceptibility and clinical phenotypes of hepatitis B virus (HBV) in Chinese Han population. METHODS: With polymerase chain reaction with restriction fragment length polymorphism (PCR-RFLP) method, the single nucleotide polymorphisms (SNP) of the promoter region of IL10 gene at position -1082G/A, -819T/C, -592A/C were detected in 231 patients with chronic hepatitis B, 165 individuals spontaneously recovered from HBV infection and 135 normal controls. RESULTS: No significant difference was found in frequencies of genotypes and alleles of IL10 gene promoter region at position -1082G/A, -819T/C, -592A/C among normal controls, individuals spontaneously recovering from HBV infection and patients with chronic hepatitis B (P>0.05), also between patients with HBV infection with HBV-DNA<1x10(3)copies/mL and those with HBV-DNA> or =1 x 10(3)copies/mL (P>0.05). However, frequencies of TT genotype at position -819T/C and AA genotype at position -592A/C in chronic hepatitis B were significantly higher than that in asymptomatic HBV carriers (P<0.05). CONCLUSION: It is possible that genetic polymorphisms of IL10 promoter region are not associated with both susceptibility of HBV infection and HBV-DNA replication after infected HBV in Chinese Han population. However, the polymorphisms of the promoter region IL10 at position -819T/C and -592A/C are related to inflammatory reaction to liver of the patients with HBV infection.

[Treatment of acute myocardial infarction with autologous bone marrow stem cells mobilization combined with recombinant growth factor in rat].

OBJECTIVE: To observe the regeneration cardiomyocytes and neovascularization after mobilizing autologous bone marrow stem cells by granulocyte colony stimulating factor (G-CSF) alone or G-CSF combined with recombinant human growth hormone (rhGH) in Wistar rats with acute myocardial infarction (AMI). METHODS: AMI rat model was reproduced by liquid nitrogen cryoinjury. The rats were randomly divided into six groups: mobilization group (N group, n=8), sham operation group (SO group, n=6), myocardial infarction group (MI group, n=8), G-CSF group (G group, n=8), rhGH group (GH group, n=8) and G-CSF combined with rhGH group (GG group, n=8). White blood cell (WBC) count and mononuclear cells proportion (MNC%) in peripheral blood were determined with ABX blood cell analyzer to estimate bone marrow stem cells mobilization. Four weeks after intervention, the rats were sacrificed, their respective body and heart weight were obtained, and the hearts were harvested for hematoxylin and eosin (HE) and immunohistochemical examination. RESULTS: (1)Comparing with baseline values, after 6 days administration of G-CSF, the WBC and MNC% increased in N and G groups (both P<0.01); WBC increased (P<0.01) but no difference of MNC% in MI group (P>0.05); WBC and MNC% were significantly higher in G group than those in MI group (all P<0.05). (2)Body and heart weights in GH and GG groups were higher than those in SO, MI and G groups respectively (all P<0.05). The ratio of heart and body weight was higher in GC group than that in MI,G and SO groups (P<0.05). (3)There were no significant differences in infarct size among MI, G, GH, and GG groups (P>0.05). (4)The capillary densities were higher in G, GH and GG groups than those in MI and SO groups; the density in GG group was higher than that in G and GH groups (all P<0.01). (5)BrdU positively stained neonatal cells were observed in G, GH and GG groups. Of them some developed into the endothelial cells. BrdU and cTnI double positive stained cells were observed in G and GG groups, which implied these cells might have differentiated into cardiac myocyte like cells. CONCLUSION: (1)G-CSF can mobilize bone marrow stem cells into peripheral blood in normal and cardiac infarct rats. The mobilized stem cells may enter into the infarct zone and induce the regeneration of cardiac myocyte like cells and vascular endothelial cells. (2)rhGH may promote the regeneration of capillary in the zone of infarction, but does not induce regeneration of cardiac myocyte like cells. (3)The combination of G-CSF with rhGH might promote more capillary regeneration than either of them used alone.

[BRG1 expression in prostate carcinoma by application of tissue microarray].

OBJECTIVE: To investigate the expression of gene BRG1 in prostatic intraepithelial neoplasia and adenocarcinoma, and the relationship between gene BRG1 expression and the clinicopathological features of prostate carcinoma. METHODS: Gene BRG1 expression was evaluated in 37 cases of human prostate carcinoma, 13 human prostatic intraepithelial neoplasia (PIN) and 14 human benign prostatic hyperplasia (BPH) by using immunohistochemistry (EnVision method) and tissue microarray. RESULTS: The positive rates of BRG1 protein were 81.08% (30/37), 38.46% (5/13) and 14.28% (2/14) in prostate carcinoma, PIN and BPH, respectively, significantly higher in the first group than in the latter two (P < 0.05). There was no statistically significant difference in BRG1 gene expression either between PIN and BPH (P > 0.05) or between the groups of the moderate differentiation (the Gleason histologic grading: 5-7) and the lower one (the Gleason histologic grading: 8-10) (P > 0.05). CONCLUSION: BRG1 may play an important role in the development of prostate carcinoma. Tissue microarray technology, with the advantages of high throughput, conciseness, rapidity, high efficiency, low cost, and nice reproducibility, has significant practical value and broad application prospects in pathology.

[The effects of flutamide on the full gene expression of early testis development of embryonic period male SD].

OBJECTIVE: To study the effects of Flutamide (Flu) on the full gene expression of early testis development of embryonic period in male SD rats. METHODS: Flu was injected subcutaneously (sc) with NS at doses of 6.25 mg/kg x d from 12th to 17th gestation days (GD), and in control group with tales doses NS. Fifty pregnant SD rats were sacrificed on GD 20. The mRNA were extracted respectively from fetus testis with treated and untreated Flu, then transcribed reversely to cDNA and labeled with cy5 and cy3 fluorescence. Subsequently, cDNA probes were hybridized to the Mouse40S cDNA microarray and fluorescence signals were analyzed. Meanwhile, we took out of the right testes of male offspring to make the detection of immunohistochemistry (IHC). Furthermore, we determined the level of testosterone by enzyme-linked immunosorbentassay (ELISA). RESULTS: Compared with the control group, a total of 31 identified genes expressed differently, including 14 down-regulated and 17 upregulated genes. However, the function of 5 down-regulated and 10 upregulated difference expression genes are unknown in Genbank. This finding has provided important reference basis for the molecular mechanism of reproductive development of Flu in the future. The activity of two important enzymes was cut down by IHC. The level of testosterone was decreased by ELISA. CONCLUSION: The results indicated that Flu administrated to F0 pregnant rats can damage apparently to the reproductive development of embryonic period male SD rats in many ways, and can cause the abnormality of sexual differentiation and sexual development in rats.

[Correlation of XPC Ala499Val and Lys939Gln polymorphisms to risks of esophageal squamous cell carcinoma and gastric cardiac adenocarcinoma].

BACKGROUND & OBJECTIVE: Xeroderma pigmentosum group C(XPC) gene is involved in nucleotide excision repair (NER). Single nucleotide polymorphisms (SNP) in XPC gene may affect DNA repairing capacity and genetic susceptibility to cancer. This study was to investigate the correlation of XPC exon 8 Ala499Val and exon 15 Lys939Gln SNPs to the susceptibility of esophageal squamous cell carcinoma (ESCC) and gastric cardiac adenocarcinoma (GCA) in a population at a high incidence region of Hebei Province. METHODS: XPC exon 8 Ala499Val and exon 15 Lys939Gln SNPs were genotyped by polymerase chain reaction-restrictive fragment length polymorphism (PCR-RFLP) analysis in 327 ESCC patients, 253 GCA patients, and 612 healthy controls. RESULTS: The number of the subjects with family history of upper gastrointestinal cancer (UGIC) was significantly higher in ESCC and GCA groups than in control group. Family history of UGIC may increase the risk of developing ESCC and GCA [age and gender adjusted odds ratio (OR) =1.76 and 1.77, 95% confidence interval (CI) = 1.34-2.32 and 1.31-2.39]. The overall allelotype and genotype distributions of XPC exon 8 Ala499Val in ESCC patients were not significantly different from those in healthy controls (P>0.05). T allelotype frequency of XPC exon 8 in GCA patients was 26.5%, which was significantly lower than that in healthy controls (Chi2=6.12, P=0.01). The C/T genotype frequencies of XPC exon 8 in GCA patients and healthy controls were 35.6% and 46.1% respectively. Compared with individuals with C/C genotype, individuals with C/T genotype had significantly lower risk in developing GCA (OR=0.62, 95% CI=0.45-0.84). When stratified for smoking status and family history of UGIC, compared with individuals with C/C genotype, individuals with C/T genotype in smoker group and in the group without family history of UGIC had lower risk in developing GCA (OR=0.57, 95% CI=0.36-0.91 and 0.37-0.88). The overall allelotype and genotype distributions of XPC exon 15 Lys939Gln in ESCC and GCA patients were not significantly different from those in healthy controls (P>0.05). When stratified for smoking status and family history of UGIC, compared with individuals with A/A genotype, individuals in non-smoker group with C/C genotype had higher risk in developing ESCC (OR=2.05, 95% CI=1.15-3.66). The haplotype distribution of ESCC patients was not significantly different from that of healthy controls (P>0.05), while the haplotype distribution of GCA patients was significantly different from that of healthy controls (P=0.02). Compared with A/T haplotype, A/C and C/C haplotypes significantly increased the risk of developing GCA (OR=1.35 and 1.46, 95% CI=1.01-1.81 and 1.06-2.00). CONCLUSIONS: In the high incidence region of Hebei Province, C/T genotype of XPC exon 8 may decrease the risk of developing GCA. Lys939Gln SNP in exon 15 may have no influence on the risk of ESCC and GCA, but when stratified for smoking status, C/C genotype of XPC exon 15 may increase the risk of developing ESCC in non-smoking population. While A/C and C/C haplotypes may increase the risk of developing GCA.

[Effects of different interventional modalities on vascular endothelial dysfunction of obese rats].

OBJECTIVE: To observe the effect of diet-control only, diet-control with swimming training or with polysaccharide sulfate (PSS), a kind of blood lipid-lowering drug on the serum lipid level and vascular endothelial function in obese rats fed by fat-rich-diet. METHODS: Forty Wistar rats were randomly divided into the following 5 groups: group F (n = 8), group N (n = 8), group A (n = 8), group B (n = 8) and group C (n = 8), where the rats were given fat-rich-diet, basic-diet, 12 weeks of diet-control after 8 weeks of fat-rich-diet, 12 weeks of diet-control with swimming training after 8 weeks of fat-rich-diet and 12 weeks of diet-control with PSS after 8 weeks of fat-rich-diet, respectively. All rats were sacrificed after 12 weeks of intervention. Then the levels of Lee index, serum total cholesterol (TC), triglyceride (TG), plasma endothelin (ET), nitric oxide (NO) and von Willebrand Factor (vWF) were measured. The protein expression of intercellular adhesion molecule-1 (ICAM-1) in artery endothelium was evaluated by immunohistochemistry (IHC) and the gene expression of ICAM-1 was examined by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). RESULTS: After the interventions for 12 weeks, the levels of serum TC, TG and ET decreased in group A (P < 0.05). The levels of Lee index, TC, TG, ET, vWF, ICAM-1 protein and ICAM-1 mRNA decreased in group B and C (P < 0.05). Three interventions increased serum NO production (P < 0.05) in group B. CONCLUSIONS: Diet-control could a meliorate the hyperlipidemia and vascular function. Diet-control with swimming training and diet-control with PSS could result in weight loss of rats and meliorate the hyperlipidemia, vascular endothelial function, coagulatory activities and adhesive dysfunction. The effects of diet-control with swimming on vascular endothelial function were prominent.

[A prospective control study on treatment of stress urinary incontinence with alpha-adrenoceptor agonists and pelvic floor muscle exercises].

OBJECTIVE: To evaluate the therapeutic results of alpha-adrenoceptor agonists and pelvic floor muscle exercises (PFME) for stress urinary incontinence. METHODS: A prospective, single-center, random control study was carried out in 46 patients with stress urinary incontinence (SUI). The patients were divided into two groups, 22 in alpha-adrenoceptor agonists group vs 24 in PFME group. Clinical outcomes and the scores of quality of life were evaluated and compared between the two groups. RESULTS: Two patients in alpha-adrenoceptor agonists group and four patients in PFME group were failed to complete the study. According to patient's subjective scores, excellent effective rate was 15% (3/20) and effective rate was 85% (17/20) in alpha-adrenoceptor agonists group versus effective rate of 55% (11/20) and no-effective rate of 45% (9/20) in PFME group. According to objective 1 h pad test criteria, the efficacy was significantly better in alpha-adrenoceptor agonists group (P < 0.01). The scores of quality of life were improved in all patients of two groups after treatment, and were significantly better in alpha-adrenoceptor agonists group (P < 0.05). There was no difference in sexual life score in PFME group and alpha-adrenoceptor agonists group. CONCLUSIONS: Both alpha-adrenoceptor agonists and PFME are effective non-surgical treatments for stress urinary incontinence. However, alpha-adrenoceptor agonists have better effect than PFME.

[Research in the relation between telomerase reverse transcriptase expression in spermatogenic cells and serum levels of estradiol of fluorotic rats].

OBJECTIVE: To study the relation between telomerase reverse transcriptase (TERT) activity expression in spermatogenic cells and serum levels of estradiol of fluorotic rats. METHODS: We randomly divided thirty SD male rats into control group, low-dose group and high-dose group, then inject sodium fluoride (0, 10, 20 mg/kg bw) into celiac of rats. We respectively observed changes of estrogen and TERT using methods of radioimmunoassay, in situ hybridization. In addition, we observed the quality of spermatozoa. RESULTS: The level of estrogen, the expression of telomerase and the number and the livability of the spermatozoon in low-dose and high-dose fluorotic rats were lower than those of control rats (P < 0.05). Therefore, the above indexes decreased with the increase of dosage. In addition, sperm aberration of each fluorotic group was higher than control group (P < 0.05). And it increased with the increase of dosage. The content of E2 in serum of different fluoride treatment groups was positively correlated with the expression of telomerase in seminiferous tubule significantly, respectively (low-dose fluoride treatment groups, r = 0.941, P < 0.01, high-dose fluoride treatment groups, r = 0.929, P < 0.01). CONCLUSION: NaF possibly damaged the male reproductive system by the approach of E2/ER-TERT-spermatozoon, relation between TERT expression in spermatogenic cells and serum levels of estradiol is positive correlation.

[Correlation of p73 polymorphisms to genetic susceptibilities to esophageal carcinoma and gastric cardiac carcinoma].

BACKGROUND & OBJECTIVE: p73 is a tumor suppressor gene. The two polymorphisms in the non-coding region of the exon 2 of p73 gene, named G4C14-A4T14, can form a stem-loop structure, and may influence p73 expression. This study was to investigate the correlation of p73 G4C14-A4T14 polymorphisms to susceptibilities to esophageal squamous cell carcinoma (ESCC) and gastric cardiac adenocarcinoma (GCA) in a high incidence region of Hebei Province. METHODS: We conducted a population-based case-control study in 348 ESCC patients, 259 GCA patients, and 630 healthy controls. p73 genotypes were determined by polymerase chain reaction-restrictive fragment length polymorphism (PCR-RFLP). RESULTS: Family history of upper gastrointestinal cancer (UGIC) significantly increased the risk of developing ESCC and GCA [the age, sex, and smoking status adjusted odds ratio (OR)=1.68, 95% confidence interval (CI)=1.28-2.20; OR=1.68, 95% CI=1.24-2.26, respectively]. The overall distribution of p73 genotype and allelotype in cancer patients and controls were not significantly different. Stratification analysis by smoking status and family history of UGIC found that the GC/AT genotype significantly increased the risk of developing GCA among the subjects without family history of UGIC (OR=1.71, 95% CI=1.14-2.57), while p73 G4C14-A4T14 polymorphisms did not increase the risk of developing ESCC and GCA in other subgroups. CONCLUSION: In p73 G4C14-A4T14 polymorphisms, the GC/AT genotype increases the risk of developing GCA among the subjects without family history of UGIC.

[Clinical application of HLA sequencing in unrelated umbilical cord blood transplantation].

From June 1998 to July 2004, Guangzhou umbilical cord blood bank provided unrelated umbilical cord blood for 54 patients to more than 21 transplantation centers. HLA sequencing-based typing (SBT) was used to re-analyze the results of HLA antigens and alleles so as to investigate the relationship between HLA alleles and GVHD. The information about 48 out of 54 patients was obtained after 6 months of follow up. SBT was used to identify HLA-A, B, DRB1 alleles in 48 patients received the unrelated umbilical cord blood units, and the obtained results were compared with the results of HLA-SSP Low Resolution Typing. The results showed that the difference of GVHD incidence between less than 2 mismatched HLA sites and less than 3 sites was statistically significant (P < 0.05). In the results from single factor analysis and high-resolution typing of HLA-A, B and DRB1 alleles, the mismatch between HLA-B and HLA-DRB1 alleles was found to be a significant factor for the occurence of GVHD. It is concluded that SBT plays an important role in umbilical cord blood transplantation, and the incidence of GVHD is higher in the transplantation with HLA-DRB1 alleles mismatching.

[Calibration transfer used for the interpretation of RS-FTIR spectrum].

A technique of analysis for RS-FTIR (Remote Sensing Fourier Transform Infrared) spectrum was built based on the principle of calibration transfer. A system of four-component gas mixtures, whose FTIR spectra were overlapped seriously, was determined as the object in the present research. Its FTIR data from EPA and field measurement were used to set up calibration and prediction models, respectively. With OSC (orthogonal signal correction), the model built with data of EPA predicted values of field measurement. The RMSEP values of acetone, methanol, benzene and chloroform were 0.0085, 0.0180, 0.0640 and 0.0028, respectively, while without OSC correction the corresponding values were 0.0856, 0.0479, 1.0653 and 0.0142, respectively. With optimization, the Kennard-Stone method was used for subset selection, and the iteration in the process of OSC was carried out 3 times, yielding the best performance. The result indicates that this technique can overcome the limitation of background and calibration model, which restricts the application of RS-FTIR in air monitoring.

[Isolation, in vitro culture and identification of cardiac stem cells from neonatal SD rats].

OBJECTIVE: To isolate and culture cardiac stem cells (CSCs) in vitro and evaluate their potential of differentiation into functional cardiac myocytes. METHODS: Myocardial tissues obtained from neonatal SD rats were cut into pieces of 0.5-1.0 mm(3), and digested twice for 5 min at 37 degrees C; with 0.2% trypsin and 0.1% collagenase II. The remaining tissues were cultured in complete explant culture medium (CEM) at 37 degrees C; in the presence of 5% CO(2). About a week later, a layer of fibroblast-like cells was generated from the adherent explants. These cells were passaged and seeded at about 1x10(6) cells/ml in poly-D-lysine-coated multi-well plates in cardiosphere-growing medium. When beating of the cultured cells was observed (at week 2), flow cytometry and immunohistochemistry were performed for identification of the primary and passaged cells. RESULTS: The primary cells were successfully cultured from the digested myocardial tissue, and flow cytometry demonstrated the phenotype of c-kit(+)CD31(+)CD34(-)CD45(-)CTnT(-). After cell passage for about two weeks, single beating cells and cell clusters with synchronized contraction were seen microscopically, and their phenotype was converted to c-kit(+)CD31(-)CD34(-)CD45(-) CTnT(+). Immunohistochemistry staining identified CTnT expression in the passaged cells but not in the primary cells. CONCLUSIONS: A cell population with the phenotype c-kit(+)CD31(+)CD34(-)CD45(-)CTnT(-) has been obtained from neonatal SD rat heart, which possesses the potential to differentiate in vitro into beating cardiac myocytes and express CTnT protein.

[Protecting effect of brevifolin and 8,9-single-epoxy brevifolin of Phyllanthus simplex on rat liver injury].

OBJECTIVE: To investigate the effect of protecting liver of brevifolin and 8,9-single-epoxy brevifolin of Phyllanthus simplex. METHOD: Rats were administered with CCl4 (ip) or alcohol (ig) to establish acute or chronic liver injured model, respectively. ALT, AST and TBIL in serum were measured using colorimetric analysis to evaluate liver function. MDA content or SOD activity in serum and liver tissue was measured by thiobarbituric acid chromatometry and xanthine oxidase methods, respectively. The hemorheological parameters were observed. RESULT: Brevifolin and 8,9-single-epoxy brevifolin reduced the increase of ALT induced by CCl4, but they did not influence the increase of AST. And it could inhibit the pathologic increase of serum TBIL induced by alcohol. They could ameliorate the MDA increase or SOD decrease in serum and liver tissue in rats with liver injury, and decrease abnormal changed hemorheological parameters. CONCLUSION: Brevifolin and 8,9-single-epoxy brevifolin show protective effective against acute and chronic liver injuries, and the mechanism is relevant to antagonizing the lipid peroxidation of free radical and improving the blood circulation.

Colon-specific delivery tablets of sodium 4-aminosalicylic acid.

AIM: To prepare a new oral colon-specific delivery formulation and to investigate the release profile in vitro and the colon-specific delivery property in vivo in dogs. METHODS: Sodium 4-aminosalicylic acid was selected as the model drug. The combination of Eudragit RL30D and RS30D were used as sustained-release film, and Eudragit FS30D used as enteric film, which was expected to release drug depending on pH and time. The release profile of tablets was studied in three phosphate buffers with the pH 6.5, 7.0 or 7.4 for 12 h after a simulated gastric presoak for 2 h in 0.1 mol x L(-1) HCl. The tablets were radiolabelled with 99mTc to make their release times and positions in the gastrointestinal tract be followed using a gamma camera. RESULTS: For the in vitro study, there was no drug released in 0.1 mol x L(-1) HCl for 2 h, and release occurred slowly when pH was above 6.5. Drug was released faster while pH was higher. For the in vivo study, the coated tablets remained intact in the upper gastrointestinal tract, and drug release began after the colonic arrival. The uncoated tablets, however, disintegrated in the stomach of the dogs rapidly. CONCLUSION: The coating could protect the drug until the tablets reached the ascending colon, where drug was released slowly for over 10 h.
