Localization of a 24-kilodalton glycoprotein in adult Schistosoma mansoni using immunofluorescence and immunoelectron microscopy.

Studies on schistosome protective immune responses have focused mainly on antigens of the parasite's syncytial surface. One of the characterized schistosome antigens, a 24-kDa glycoprotein, has been considered important in mechanisms of immune evasion by the parasites. In the present study, using affinity-purified antibodies to the 24-kDa protein for immunofluorescence and immunoelectron microscopy, we demonstrated an association of the 24-kDa antigen with the discoid bodies (the major syncytial inclusion bodies; DBs) and the surface membrane complex (most likely the apical plasma membrane) of adult Schistosoma mansoni. This is consistent with previous observations that the 24-kDa antigen appeared to be localized to the syncytial membrane and DB fractions. The present results also support the suggestion that the DBs are the precursor organelles of the apical plasma membrane.

[Effects of baicalin and liquid extract of licorice on sorbitol level in red blood cells of diabetic rats].

10 streptozotocin-induced diabetic rats were treated with baicalin 150 mg/kg.d and liquid extract of licorice (LEL) 7.5 ml/kg.d (5 by each medicine) for a week. The results indicated that orally baicalin and LEL could dramatically reduce the sorbitol levels in red blood cells (123.7 +/- 28.6 to 72.5 +/- 21.8 and 116.9 +/- 25.2 to 80.5 +/- 23.8 nmol/g.Hb, respectively) without affecting blood glucose levels significantly. It is suggested that both medicines act by inhibiting aldose reductase.

[Resolution of the enantiomers of some beta-amino alcohols using chiral derivatization and reversed-phase liquid chromatography].

A convenient and rapid procedure for enantiomeric resolution of beta-amino alcohol drugs, such as oxprenolol, propafenone, timolol and phenylephrine, is described. The method is based on derivatization with the chiral reagent 2,3,4,6-tetra-O-acetyl-beta-D-glucopyranosyl isothiocyanate (GITC), and resolution of the resulting diastereomeric thioureas by reversed-phase HPLC using methanol-aqueous monobasic ammonium phosphate mixture as the mobile phase. The salt forms of these drugs can be directly derivatized with GITC reagent instead of being converted into their free bases and without protection of the phenolic hydroxyl group of phenylephrine.

[A pharmacognostical study on the fruits of Cnidium monnieri (L.) Cuss].

The fruits of three species are used as the Chinese drug "Shechuangzi", namely Cnidium monnieri, C. monnieri var. formosana and C. japonicum, among which only C. monnieri is the real sort. The authors have distinguished them by macroscopic and microscopic structures and TLC. The total coumarin, trace element and amino acid contents are reported.

[Effect of PGR-1 (plant growth regulator-1) on variations in mineral content and nutritive value of Areca catechu L].

The investigation was conducted to find out the effect of PGR-1 (spurting) on variations in the mineral contents and nutritive values of the leaf, flower and root of Areca catechu in different phenophases of th year. The result shows that PGR-1 helps to expand th leaf area of different leaf segments by 20.8 to 37.47 cm2 and increase the dry matter by 4.66 to 6.56g, so as to promote the nutritive value of inflorescence and thus raise the flowering and fruitication rate.

Mechanism of testicular atrophy induced by di-n-butyl phthalate in rats. Part 1.

Repeated oral doses of di-n-butyl phthalate (DBP) to male rats caused a decrease in testicular fructose and glucose and a sloughing of the germ cells on the first day of treatment. On day 2, more severe sloughing was seen and was accompanied by decreases in testicular iron and zinc levels and increases in the level of inositol and cholesterols. The sloughing was followed by atrophy, accompanied by dissociation of the germ cells from the Sertoli cells and reduction of triglycerides, cholesterols and phospholipids containing choline and ethanolamine residues in the testis.

Damage thresholds of skin irradiated by ultraviolet lasers.

The effects of pulsed radiation on animal models were studied. Erythemic responses to laser radiation were observed macroscopically and were examined histologically by both light and electron microscopy. Based on statistical analysis of the biological data presented in this study, skin damage thresholds were calculated for short pulses with lasers at wavelengths of 265 nm and 308 nm.

Differentiation of hog cholera and bovine virus diarrhoea viruses in pigs using monoclonal antibodies.

Monoclonal antibodies against hog cholera and bovine viral diarrhoea viruses were assayed on organ tissue sections of experimentally infected animals. The animals had been infected simultaneously with both viruses. The antibodies were tested using an indirect immunofluorescence test and an indirect enzyme immunoassay with a biotin/streptavidin/peroxidase detection system. A polyclonal hyperimmune serum was used as a control in direct immunofluorescence tests. Both techniques based on monoclonal antibodies were more sensitive and more specific than the conventional test, the enzyme immunoassay being more sensitive than the immunofluorescence test. Small amounts of BVD viral antigen were demonstrable with monoclonal antibodies in most organ tissues.

A study of the pattern of deaths by diseases in major Chinese cities.

"In research in recent years, the co-authors of this article have discovered that changes in PDD [patterns of disease and death] in urban and rural areas [in China] follow to a large degree an identical pattern except for a difference in the timing of these changes (approximately 5 to 10 years). This study is based on statistics obtained in 14 cities with a population of more than one million, complete registration systems and higher data reliability. It is intended to ascertain how the pattern of deaths by diseases changes, and to predict the tendency of such changes through dynamic research on such patterns in those cities." The authors analyze changes in specific causes of death and their impact on life expectancy in China.

[Experimental study of qi deficiency syndrome and Codonopsis pillosulae and Astragalus injection on the immune response in mice].

According to TCM theory "hunger impairs one's Qi", the authors had made mouse model of Qi deficiency syndrome (QDS) through controlling amount of food fed. Compared with mice of normal group, the body weight, thymus index and spleen index obviously decreased in mice of QDS group. The similar effects on humoral immunity and cell-mediated immunity were also obtained. It was found that the Codonopsis pilosulae and Astragalus Injection (CPAI) possessed the promoting action of thymus index, spleen index, humoral immunity and cell mediated immunity on QDS mice and normal mice except spleen index. However, QDS mice were more sensitive to CPAI than the normal mice. 4 x 10(9) SRBC/mouse was administered to normal donor mice by i.p., the super-optimal immunization could obviously induce generation of Ts cells. The CPAI could also reduce generation of Ts cells. This paper had proved that the inherent relation between Qi and immune response of organism, and that the CPAI could promote immune response by means of reducing Ts cells.

Effects of serotonin (5HT) and complement C3 on the synthesis of the surface membrane precursors of adult Schistosoma mansoni.

The syncytial surface epithelium of Schistosoma mansoni plays an important role in immune evasion. This syncytium is covered by an unusual double-membrane complex consisting of an apical plasma membrane (APM) and an overlying envelope (En) that have been shown to have different rates of synthesis and turnover. It has been suggested that discoid bodies (DBs) and multilamellar bodies (MLBs), the major syncytial inclusion bodies of schistosomes, may be the precursors of the APM and En, respectively. In this ultrastructural study, we examined the effects of serotonin (5HT) and complement C3, which have been shown to stimulate synthesis and turnover of the APM and En, respectively, on the synthesis of DBs and MLBs in vitro. With short-time incubations (20 or 40 min), 5HT stimulated the synthesis of the DBs by 2-fold, whereas C3 accelerated synthesis of the MLBs by 2-fold. Furthermore, when microtubules within the cytoplasmic connections between the syncytium and the underlying cell bodies (the site of membrane synthesis) were disrupted with colchicine, the DBs and MLBs synthesized in response to 5HT or C3 accumulated in the cell bodies. This suggests that the transport of the organelles to the syncytium is dependent upon the microtubules but not the signaling mechanism in response to 5HT or C3. These observations also support the suggestion that the DBs and MLBs are synthesized in subsyncytial cell bodies and serve as precursors of the APM and En, respectively. The rapid synthetic response to 5HT and C3 is also consistent with rapid synthesis and turnover of the APM/En, as suggested by previous studies.
