Flow-cytometric separation and enrichment of hepatic progenitor cells in the developing mouse liver.

Stem cells responsible for tissue maintenance and repair are found in a number of organs. However, hepatic stem cells assumed to play a key role in liver development and regeneration remain to be well characterized. To address this issue, we set up a culture system in which primitive hepatic progenitor cells formed colonies. By combining this culture system with fluorescence-activated cell sorting (FACS), cells forming colonies containing distinct hepatocytes and cholangiocytes were identified in the fetal mouse liver. These cells express both CD49f and CD29 (alpha6 and beta1 integrin subunits), but do not mark for hematopoietic antigens such as CD45, TER119, and c-Kit. When transplanted into the spleen, these cells migrated to the recipient liver and differentiated into liver parenchymal cells. Our data demonstrate that hepatic progenitor cells are enriched by FACS and suggest approaches to supplanting organ allografting and improving artificial-organ hepatic support.

Altered expression and functional profile of lysophosphatidic acid receptors in mitogen-activated human blood T lymphocytes.

Lysophosphatidic acid (LPA) from platelets and mononuclear phagocytes regulates T cell functions through endothelial differentiation gene-encoded G protein-coupled receptors (Edg Rs). Human blood unactivated CD4+ T cells express predominant ly Edg-4 LPA R over marginal levels of Edg-2 LPA R, as assessed by semiquantitative PCR and Western blots. After mitogen activation, the CD4+ T cells express Ed g-2 Rs at approximately one half the level of Edg-4 Rs. Secretion of IL-2 by unactivated Edg-4 R-predominant CD4+ T cells incubated with anti-CD3 plus anti-CD28 antibodies was suppressed significantly and by up to 60% by 10-10 M to 10-6 M LPA, whereas secretion of IL-2 by mitogen-activated Edg-2 R and Edg-4 R codominant CD4+ T cells was enhanced by up to twofold by the same concentrations of LPA. The possibility that the two Edg Rs transduce different LPA signals to CD4+ T cells was supported by findings that IL-2 secretion was inhibited by mouse anti-Edg-4 R monoclonal antibody, but enhanced by mouse anti-Edg-2 R monoclonal antibody. The separate effects of each LPA R were studied in Jurkat T cell transfectants expressing principally human Edg-2 Rs (Jurkat-T-2) or Edg-4 Rs (Jurkat-T-4) and stimulated with anti-CD3 plus phorbol myristate acetate. LPA and anti-Edg-4 R antibody suppressed IL-2 secretion by stimulated Jurkat-T-4 cells, whereas LPA and anti-Edg-2 R antibody enhanced IL-2 secretion by stimulated Jurkat-T-2 cells. Activation-induced alterations in the relative levels of Edg-2 and -4 Rs on CD4+ T cells thus reverse the effects of LPA on T cell receptor-stimulated generation of IL-2.

Bone mineral density variations along the lumbosacral spine.

Vertebral bone quality is essential in vertebral strength and the stability of spinal screw fixation. A major factor of bone quality is the density of bone mineral and its distribution throughout the bone. There have been studies regarding bone mineral density variations in the cervical and thoracolumbar spine, but bone mineral density variations in the lumbosacral spine have not been documented. The purpose of the current study was to quantify bone mineral density variations at different lumbosacral levels and within vertebra, especially along the pathways of lumbar pedicle screws. Bone mineral density variations within the vertebrae along the lumbosacral spine were measured in 13 specimens from young male cadavers using peripheral quantitative computed tomography. Measurements included bone mineral density variations at different lumbosacral levels and in transverse layers and vertical columns within each vertebral body. These original data showed that the bone mineral density increased gradually from lumbar to S1 vertebrae, possibly reflecting a caudally increasing load on different lumbosacral levels. The highest bone mineral density in the lumbosacral spine is found at the pedicles and regions closest to pedicle bases, supporting the use of pedicle screw fixation.

Associations between drinking water and urinary arsenic levels and skin lesions in Bangladesh.

The present study examined the associations between drinking water and urinary arsenic levels and skin lesions among 167 residents of three contiguous villages in Bangladesh. Thirty-six (21.6%) had skin lesions (melanosis, hyperkeratosis, or both), of which 13 (36.1%) occurred in subjects who were currently drinking water containing concentrations of arsenic < 50 micrograms/L. The risk for skin lesions in relation to the exposure estimates based on urinary arsenic was elevated more than 3-fold, with the odds ratios for the highest versus the lowest quartiles being 3.6 (95% confidence interval, 1.2 to 12.1) for urinary total arsenic and 3.2 (95% confidence interval, 1.1 to 10.0) for urinary creatinine-adjusted total arsenic. The risks for skin lesions in relation to the exposure estimates based on arsenic in drinking water were less strongly elevated, with the odds ratios for the highest versus the lowest quartiles of exposure being 1.7 (95% confidence interval, 0.6 to 5.1) for drinking-water arsenic and 2.3 for cumulative arsenic index. The study suggests that arsenic exposure is associated with skin lesions in the Bangladesh population and that urinary arsenic may be a stronger predictor of skin lesions than arsenic in drinking water in this population.

Three leflunomide metabolite analogs.

The title compounds, 1-cyano-2-hydroxy-N-[4-(methylsulfonyl)phenyl]but-2-enamide, C(12)H(12)N(2)O(4)S, PHI492, 1-cyano-2-hydroxy-N-[3-(methylsulfonyl)phenyl]but-2-enamide, C(12)H(12)N(2)O(4)S, PHI493, and N-[3-bromo-4-(trifluoromethoxy)phenyl]-1-cyano-2-hydroxybut-2-e namide , C(12)H(8)BrF(3)N(2)O(3), PHI495, are potent inhibitors of Bruton's tyrosine kinase (BTK). The molecular structures of these compounds are similar and they display similar hydrogen-bonding networks and crystal packing. Examination of the crystal-packing interaction in the three compounds reveals an alternating direction of adjacent molecules in the crystalline lattice due to intermolecular cyano-amide hydrogen bonding. PHI492, a positional isomer of PHI493, does not form intermolecular O-H.O hydrogen bonds between molecules and crystallizes in a space group different from that of PHI493 and PHI495. The aromatic ring and the amide group of each molecule form a conjugated pi-system which ensures planarity, with further stabilization gained from intramolecular O-H.O hydrogen bonds.

Differentiation of intracardiac tumors and thrombi by echocardiographic tissue characterization: comparison of an artificial neural network and human observers.

The feasibility of classifying ultrasound images of intracardiac tumors and thrombi with a neural network-based algorithm was compared with the performance of experienced echocardiographers. The neural network used statistical descriptors of the apparent echocardiographic texture of the masses, and the blinded echocardiographers were given photographic prints of enlarged regions of interest without clinical data. The network classified 66% of the images correctly and the echocardiographers, 83%. The network and echocardiographers agreed in 88% of the images. Human observers usually base their classification of intracardiac masses on clinical data. The echocardiographic texture of tumors is quantitatively different from that of thrombi. This difference can be recognized by a neural network and potentially be useful in assisting with the diagnosis when clinical clues are insufficient.

Methysergide reduces nonnutritive blood flow in normal and scalded skin.

Methysergide is a serotonin antagonist and has been demonstrated to reduce wound blood flow and edema formation. We have determined the effect of methysergide on protein kinetics in normal and scalded skin of anesthetized rabbits. L-[ring-(13)C(6)]- or L-[ring-(2)H(5)]phenylalanine was used to reflect skin protein kinetics by use of an ear model, and L-[1-(13)C]leucine was used to reflect whole body protein kinetics. The results were that infusion of methysergide (2-3 mg. kg(-1). h(-1)) reduced the blood flow rate in normal skin by 50% without changing skin or whole body protein kinetics. After scald injury on the ear, administration of methysergide for 48 h reduced the weight of scalded ears (43 +/- 4 vs. 30 +/- 5 g, P < 0.01) and ear blood flow rate (42.6 +/- 4.9 vs. 5.8 +/- 1.0 ml. 100 g(-1). min(-1), P < 0.0001) and did not change wound protein kinetics. Methysergide reduced arteriovenous shunting and maintained inward phenylalanine transport from the blood to the skin pool. Using the microsphere technique, we found that the infusion of methysergide decreased blood perfusion by 33-36% in both normal and scalded ear skin. We conclude that methysergide administration reduces nonnutritive, as opposed to nutritive, blood flow in normal and scalded skin.

PKCepsilon modulates NF-kappaB and AP-1 via mitogen-activated protein kinases in adult rabbit cardiomyocytes.

We have previously shown that protein kinase C (PKC)-epsilon, nuclear factor (NF)-kappaB, and mitogen-activated protein kinases (MAPKs) are essential signaling elements in ischemic preconditioning. In the present study, we examined whether activation of PKCepsilon affects the activation of NF-kappaB in cardiac myocytes and whether MAPKs are mediators of this signaling event. Activation of PKCepsilon (+108% above control) in adult rabbit cardiomyocytes to a degree that has been previously shown to protect myocytes against hypoxic injury increased the DNA-binding activity of NF-kappaB (+164%) and activator protein (AP)-1 (+127%) but not that of Elk-1. Activation of PKCeta did not have an effect on these transcription factors. Activation of PKCepsilon also enhanced the phosphorylation activities of the p44/p42 MAPKs and the p54/p46 c-Jun NH(2)-terminal kinases (JNKs). PKCepsilon-induced activation of NF-kappaB and AP-1 was completely abolished by inhibition of the p44/p42 MAPK pathway with PD98059 and by inhibition of the p54/p46 JNK pathway with a dominant negative mutant of MAPK kinase-4, indicating that both signaling pathways are necessary. Taken together, these data identify NF-kappaB and AP-1 as downstream targets of PKCepsilon, thereby establishing a molecular link between activation of PKCepsilon and activation of NF-kappaB and AP-1 in cardiomyocytes. The results further demonstrate that both the p44/p42 MAPK and the p54/p46 JNK signaling pathways are essential mediators of this event.

Activation of PKCdelta by tyrosine phosphorylation in rat parotid acinar cells.

Receptor- and nonreceptor-mediated stimuli produce increases in both PKCdelta tyrosine phosphorylation and activity in rat parotid acinar cells and other cells. In vivo and in vitro increases and decreases in tyrosine phosphorylation resulted in increases and decreases, respectively, of PKCdelta activity. These studies demonstrated that increases in PKCdelta activity by G protein-coupled receptors and other stimuli were controlled by alterations in tyrosine phosphorylation.

Age and individual differences in visuospatial processing speed: testing the magnification hypothesis.

Forty young adults and 40 older adults performed seven visuospatial information processing tasks. Factor analyses of the response times (RTs) yielded a single principal component with a similar composition in both age samples. For both samples, regressing the mean RTs of fast and slow subgroups for the seven tasks (18 conditions) on the corresponding mean RTs for their age group accounted for 99% of the variance. Taken together, these findings suggest that individual differences in processing time were largely task independent. The magnification hypothesis, a simple mathematical model of the interaction between age and ability, is presented. This model correctly predicts the finding that in both the young and the older adult groups, individual differences increased systematically with task difficulty. The magnification hypothesis also explains the regression parameters describing individual differences among young adults and predicts correctly that equivalent parameters describe individual differences among older adults. According to the magnification hypothesis, the RTs of slower individuals are more affected by aging than those of faster individuals, and slower individuals may be more at risk with respect to other biological insults (e.g., changes in health status) as well.

[The association between A55V variant in UCP2 gene and body fat distribution, serum lipid profile in Chinese].

OBJECTIVE: Uncoupling protein 2(UCP2) could play an important role in energy metabolism and body weight regulation. The aim of this study was to investigate Ala55Val(A55V) variant in the UCP2 gene has effects on serum lipid profile, body fat and its distribution in Chinese. METHODS: The genotypes of A55V variant in the UCP2 gene were determined by a PCR-RFLP assay in 359 unrelated Chinese [including 193 normal glucose tolerance(NGT) and 166 type 2 diabetic subjects by ADA 97' criteria]. The parameters for regional adipose tissue distribution were measured by magnetic resonance imaging(MRI). RESULTS: In NGT group, an association between A55V variant in the UCP2 gene and body mass index(BMI) (P=0.0246), as well as femoral subcutaneous adipose tissue area (FA) (P=0.0017), was noted in females. A55V variant in the UCP2 gene was also associated with serum triglyceride (TG) level (P=0.0072) in males. However, in type 2 diabetes group, an association between A55V variant in the UCP2 gene and FA (P=0.0150) was replicated in females too. Those females who were homozygotes of AA in the UCP2 gene had decreased FA not only in NGT group but also in type 2 diabetes group. Furthermore, logistic regression analysis indicated that FA (P=0.0098 in NGT females, P=0.0071 in type 2 diabetic females) and BMI (P=0.0016 in NGT females), as well as TG level (P=0.0040 in NGT males) were associated with this variant in the UCP2 gene. CONCLUSION: A55V variant in the UCP2 gene is associated not only with FA (in NGT females and type 2 diabetic females) but also with BMI (in NGT females). Therefore A55V variant in the UCP2 gene appears to play a role in body fat and its distribution in Chinese females.

[3-D direct matching interpolation for rotary scanning echo-cardial images].

In the present paper, a method of three dimension direct matching interpolation for a series of 2-D echo-cardial images with intersecting each other and arranging at an angle in space has been developed. It is different from the conventional interpolation. During the interpolation, sectional images are acquired and the 3-D spatial position of "empty voxel" in sectional images is located; secondly, the cubic volume is constructed, and the motion orbit of images between two windows where the nearest original image intersects to the volume is found up, passing through the "empty voxel"; In the end, based on the matching pixels, the "empty voxel" is interpolated. This method simplifies the two interopolating procedures of the conventional methods into one procedure, and overcomes the shortcoming of the overlap of image data. Experiments show that the accuracy and reasonability of interpolation can be improved by the proposed method.

[The influence of epidermal growth factor, neurotensin on cytosolic calcium and membrane fluidity in carbon tetrachloride-injured primary cultured hepatocytes].

OBJECTIVE: To study the influence of epidermal growth factor (EGF), neurotensin (NT) on cytosolic calcium and membrane fluidity in carbon tetrachloride-injured primary cultured hepatocytes. METHODS: A micro-model of carbon tetrachloride -induced primary cultured hepatocytes injury was established. EGF and NT were added to the cells 1 h ahead of carbon tetrachloride given. Twenty-four hours later, cytosolic calcium and membrane fluidity in hepatocytes were detected with fluorescence probe of Fura-2/AM and 1, 6-dihenyl-1, 3, 5-hexatriene (DPH). RESULTS: EGF and NT could significantly antagonize the increase of cytosolic Ca(2+) and decrease of membrane fluidity in carbon tetrachloride-injured hepatocytes. CONCLUSION: EGF and NT have a function of maintaining membrane fluidity and steady-status of cytosolic calcium of hepatocytes.

[Analyses on the association of CA-repeat polymorphism and A1166-->C variant in the 3'-flanking region of AT(1)R gene with essential hypertension in Tibetans].

OBJECTIVE: To investigate whether CA-repeat polymorphism and A1166 --> C variant in the 3n-flanking region of AT(1)R gene are in association with the genetic susceptibily to essential hypertension (EH) in Tibetans. METHODS: A case-control study was carried out. Sibpair analysis and family linkage analysis were conducted. The CA-repeat polymorphism of AT &(1) R gene was identified by polymerase chain reaction(PCR) with fluorescence labeled dCTP as substrate and by semi-automatic sequence technology. The A1166 -->C variant was detected by PCR-RFLP. RESULTS: Association of AT&(1)R gene locus with EH was confirmed through the case-control study in well-characterized group of 113 Tibetan EH patients and 131 normotensives(chi(2)=26.44, P<0.001). A closer examination of this gene locus found 11 alleles from Tibetan population; allele A7 (138 bp) was more frequent in both the patients and the controls. Allele A8(140 bp) was in strong positive association with genetic susceptibility to EH in Tibetans. Frequency of allele A8 was 20.5% in EH and 7.3% in normotensives. The difference of allele frequencies between the groups was significant (chi(2)=9.64, P=0.002, OR=3.46, 95% CI 1.44-8.51). Affected sibpair analysis showed chi(2)=3.85, P=0.025; family linkage analysis gave Lod score of 0.80. No association between A1166 --> C variant in AT(1)R gene and EH in Tibetans was observed (P>0.05). CONCLUSION: The result suggests that CA-repeat polymorphism of AT(1)R gene be in association with EH in Tibetans, which implicates that AT(1)R gene may be in linkage disequilibrium with the causative genes of EH.

[Study on statistical method of distribution for erythrocyte morphological features by computerized image processing].

This study sought to develop a new statistic method for the semiautomatic analysis and classification of erythrocyte morphology based on the morphological features and shape analysis of erythrocytes by computer image processing. Shape factor as the description of the erythrocyte morphological features was used for the shape classification of erythrocytes. And the models and algorithms of erythrocytes image segmentation, cell body shape recognition and measure can be implemented through the VC++. The present method can efficiently and semi-automatically provide the statistical analysis of erythrocyte morphology, and can give the distribution of erythrocyte morphological features. The result showed that there was a significant difference between the distribution curves of the normal erythrocyte morphology (one apex) and hemolytic anemia's (two apices). By this way, it can be obtained the proportion data of different cell bodies' shapes. This method could provide some information for the study and diagnosis of the diseases (e.g. hemolytic anemia, pre-leukemia) related to erythrocyte morphology.

Therapeutic strategy in the management of stage II-IV epithelial ovarian carcinoma.

OBJECTIVE: To investigate the optimal time of debulking in stage/II to stage IV epithelial ovarian carcinoma, considering corresponding advantages of both surgery and chemotherapy. METHODS: From January 1989 to December 1996, ninety-five stage II to stage IV ovarian cancer patients were treated under two different regimens. Group A-76 cases (2 cases in IIa stage, 4 cases in IIb stage, 6 cases in IIc stage, 58 cases in IIIc stage and 7 cases in IV stage) was managed according to a traditional surgery-chemotherapy regimen; and group B-19 cases (17 cases in IIIc stage and 2 cases in IV stage) was managed with a chemotherapy-surgery-chemotherapy regimen. RESULTS: The optimal debulking rate (no macroscopic residual or residual < 2 cm) in group A was significantly lower than in group B, being 32.9% (25/76) and 68.4% (13/19), respectively (P < 0.001). The average survival time of those with a residual focus > 2 cm was shorter than those with a residual focus < 2 cm, in both groups. Sixteen out of the 51 patients with a residual focus > 2 cm had a second debulking operation, among whom 7 had preoperative chemotherapy. All of these 7 patients had either no residuals or residual < 2 cm. In 9 cases without preoperative chemotherapy, the residuals were all > 2 cm. The average survival time among these two groups were significantly different (P < 0.01). CONCLUSION: (1) For those patients in whom optimal debulking was clinically assessed to be possible, timely operation is mandatory. (2) For those inoperable advanced cases, chemotherapy-surgery-chemotherapy regimen is recommended. (3) For those with residuals > 2 cm and were assessed to be difficult to eradicate during second-look operation, multi-route chemotherapy (intro-arterial, intraperitoneal, and systematic) should be given before going on the second debulking operation. Positive attitude and proper regimen would offer better results. (4) A multicenter prospective study would give more decisive conclusion.

[The expression of transforming growth factor-alpha and epidermal growth factor receptor in human fetal ovary].

OBJECTIVE: To investigate the expression of transforming growth factor-alpha (TGF-alpha) and epidermal growth factor receptor (EGFR) in cultured and uncultured human fetal ovary. METHODS: The ovarian tissues were obtained from 20 human female fetuses from 16 to 40 weeks of gestational age and were cultured for 3-9 days. Expression of TGF-alpha and EGFR protein in the fetal ovarian tissues was examined by immunohistochemical technique. RESULTS: TGF-alpha and EGFR were expressed in the oocytes of 17 fetal ovaries, in part of follicular cells and stromal cells. There was no correlation between gestational age and TGF-alpha, EGFR expression (P > 0.05). The immunostaining intensity of oocytes in the cultured fetal ovary was weaker than those of the uncultured ones (P < 0.05). CONCLUSION: TGF-alpha and EGFR may play a role as intraovarian regulators through autocrine and paracrine mechanism.
