Effects of tadenan pretreatment on bladder physiology and biochemistry following partial outlet obstruction.

PURPOSE: Tadenan is a pharmaceutical agent used in the treatment of BPH. Prior studies demonstrated that pretreatment of rabbits with Tadenan significantly reduced the contractile dysfunction following two weeks of partial outlet obstruction. The specific aim of the present study was to determine the effect of Tadenan pretreatment on the time course of the response to partial outlet obstruction and correlate the effect of Tadenan on the contractile responses to field stimulation, bethanechol, and KCl with both mitochondrial enzyme activity (citrate synthase) and sarcoplasmic reticular function (calcium-ATP'ase activity). MATERIALS AND METHODS: Sixty male New Zealands white rabbit (3 to 5 kg.) were separated into 12 groups of 5 rabbits each. Each rabbit in groups 1-6 received Tadenan orally at 100 mg./kg./day for three weeks; each rabbit in groups 7-12 received vehicle (peanut oil). Each rabbit in groups 2-6 and 8-12 received a partial outlet obstruction as described below. One group of Tadenan treated and one group of vehicle-treated rabbits were euthanized at 1, 3, 5, 7, and 14 days following partial outlet obstruction. The non-obstructed groups were studied after 4 weeks of drug or vehicle treatment. Each bladder was rapidly removed and weighed, and 3 longitudinal strips prepared and mounted in individual baths for contractile studies. The remainder of the bladder was frozen for biochemical analysis. The contractile responses to field stimulation, bethanechol, and KCl were determined; and the enzyme activities of citrate synthase (marker for mitochondrial function) and calcium-ATP'ase (marker for sarcoplasmic reticulum) were determined. RESULTS: 1) Tadenan did not reduce the effect of partial outlet obstruction on bladder mass. 2) Although the contractile responses to forms of stimulation were reduced at 1 day following partial outlet obstruction, Tadenan pretreatment resulted in a significant protective effect on the contractile responses to field stimulation, bethanechol, and KCl at 3, 5, 7, and 14 days of obstruction. 3) The activities of both citrate synthase and calcium ATP'ase were reduced significantly at 1 day following obstruction for both Tadenan treated and vehicle treated groups. The activities of both enzymes returned to near normal levels at 7 and 14 days for the Tadenan groups whereas the activities of both enzymes remained significantly reduced in the vehicle treated groups. CONCLUSIONS: These results clearly demonstrate that Tadenan pretreatment protected the bladder from both the contractile and metabolic dysfunctions induced by partial outlet obstruction.

The role of human leukocyte antigen in susceptibility and clinical manifestations of sarcoidosis.

OBJECTIVE: To investigate the correlation of human leukocyte antigen (HLA) with susceptibility and clinical manifestations of sarcoidosis. MATERIAL AND METHODS: Fifty-five patients with sarcoidosis and one hundred and six normal subjects were investigated. Genomic DNAs were purified using the proteinase K-phenol extraction method. DNA samples were amplified by the polymerase chain reaction (PCR) procedure using the DRB1 group specific primer pairs. RESULTS: The gene frequency of HLA-DR5 increased significantly in patients with sarcoidosis (P < 0.01), and HLA-DR7 decreased (P < 0.05). Gene frequencies of HLA-DR9 and HLA-DR5 increased remarkably in male patients and in patients with stage I and stage IIa, respectively (both P < 0.05). CONCLUSIONS: It is suggested that HLA-DR gene might contribute to the susceptibility as well as various clinical manifestations of sarcoidosis.

[Studies on strategies for changing food composition of residents based on their nutritional status].

Investigations of resident food consumption and evaluations of their dietary nutrition were carried out to correctly lead to the adjustment of food composition, promotion of coordinating development of food consumption and production, and continuous improvement of people's nutritional level and health status. Results showed that residents consumed less protein of high quality, more fat in the urban area, insufficient some trace element and more salt in Hebei province. Based on these facts, it is recommended that to consume vegetable food as major dietary composition with a variety of grains and vegetables, to increase adequately animal food and legumes, to decrease salt intake, and to lower fat intake in the urban areas be the strategic goals for the food composition changes. Measures to implement these goals are discussed in the paper, also.

[The bonding strength between plat castable ceramics and mild fusing alloy].

Plat castable ceramics (PCC) was bonded to SDA-I mild fusing alloy using three bonding agents, and the tensile bond strengths were tested respectively in order to select the best bonding agent and the best way of surface treatment. The results obtained with scanning electron microscope and X-ray energy spectrum showed that the bonding behavior of TF agent was the best one. The greatest tensille bond strength (24.37 MPa) was obtained with TF agent when the surface of PCC was etched by hydrofluoric acid and the alloy was painted with KH-570. Remarkable tensile bond strengths were noted in the group where EM agent was used with PCC etched by hydrofluoric acid and alloy treated with sandblasting (15.20 MPa), and in the group where Porcelite dual cure cement was applied with PCC painted by KH-570 and alloy treated with sandblasting (15.25 MPa).

Role of intracardiac angiotensin II in cardiac dysfunction of rat during septic shock.

OBJECTIVE: To observe changes in the content of myocardial angiotensin II and its role in cardiac dysfunction of rat during septic shock. METHODS: Septic shock model was produced by cecal ligation and puncture (CLP) operation on rats. Experimental rats were given captopril 15 mg.kg-1/d per os for 3 days before CLP operation. Mean blood pressure and left ventricular pressure were recorded. Myocardial angiotensin converting enzyme (ACE) activities were determined by a fluorometric assay and myocardial angiotensin II content was measured by radioimmunoassay. Highly purified membrane of sarcoplasmic reticulum (SR) was prepared from rat hearts. Assays were made of ATP-dependent Ca2+ uptake by cardiac SR and (3H) ryanodine binding to SR. RESULTS: Myocardial angiotensin II content increased by 51.5% (P < 0.01) at the 18th hour post CLP, meanwhile there was a decrease in left ventricular +/- dp/dtmax value and the impairment in Ca2+ uptake and (3H) ryanodine binding to cardiac SR. Preliminary administration of captopril reduced myocardial ACE activity and angiotensin II content, but increased left ventricular +/- dp/dtmax value. In comparison to shock group, the initial rate and the capacity of SR Ca2+ uptake were increased by 120% (P < 0.01) and 33.9% (P < 0.05), the Bmax value of (3H) ryanodine binding to SR was also elevated, while the Kd value remained unchanged. CONCLUSIONS: The elevated intracardiac angiotensin II, resulting from the activation of myocardial ACE during sepsis, probably serves as one of the important mediators participating in the pathogenesis of heart failure: the effects of angiotensin II may be associated with the disturbance of Ca2+ transport function of cardiac SR.

[The morphometric study of endometrium during long-term use of TCu220C IUD].

OBJECTIVE: To observe the pathological changes of endometrium in women using TCu220C intrauterine devices (IUD) for 5-12 years, and to judge its long term safety. METHODS: A hundred women using TCu220C IUD for 5-12 years were randomly selected as IUD group. Samples of endometrium from the sites compressed by IUD and 1 cm apart were taken under hysteroscopy during proliferative phase. Another 100 women matched in age but not IUD users served as control group, and endometriums were taken by curettage. All the samples were studied for morphometry. RESULTS: All pathological changes were localized in the areas of endometrium in direct contact with IUD but not the adjacent areas. Inflammatory reactions mainly lymphocytes infiltration were seen in all IUD samples with depth of 0.91-1.20 mm. Atrophy was seen in 25% of the cases using IUD for 5-10 years with an average area of 0.81 mm2, while in 71% of those using IUD for 10-12 years with an area of 1.50 mm2. Fibrosis was shown in 18% of the cases using IUD for 5-10 years with area of 1.87 mm2, and 100% in 10-12 years with area of 2.04 mm2. There was no hyper-proliferation and malignant changes in IUD group. Hemorrhage and necrosis existed in all cases using IUD for 10-12 years. In the IUD group all the cases showed asynchronization of endometrium. CONCLUSION: It is safe for women using TCu220C IUD for 5-12 years. Since endometrial hemorrhage and necrosis were shown in women using IUD over 10 years, it is better to remove the IUD after 10 years if they have clinical symptoms.

[Preliminary diagnostic criteria for primary Sjogren's syndrome in China].

Primary Sjogren's syndrome (pSS) is a common autoimmune connective tissue disease in China yet without a universally accepted diagnostic criteria. In this study a new criteria was proposed and compared with other six sets of criteria. Fifty-five items in 112 pSS and 185 controls were evaluated. Results show the criteria we proposed contained one major and nine minor items. For the purpose of identifying patients in clinical studies, a major with at least three of the nine minor items or at least five of the minor items should be presented. The major item is anti-SSA/SSB(+) and the minors are, (1) dry eyes or dry mouth (> 3 months, persistently), (2) swollen salivary glands (recurrently or persistently), (3) rampant dental caries, (4) Schirmer test (< 5 mm in 5 min.) or corneal staining(+), (5) unstimulated salivary flow (< 0.03 ml/min) or abornal parotid sialography, (6) minor salivary gland biopsy (> or = 1 focus), (7) renal tubular acidosis, (8) hypergammaglobuminemia (gamma globulin > or = 30%) or hypergammaglobuminemic purpura, (9) RF > 1 : 20 or ANA > 1 : 20. Other connective tissure diseases, pre-existing lymphoma, AIDS, sarcoidosis, graft vs host disease must be excluded. The criteria we proposed had a high specificity of 98.2% and sensitivity of 94.1%.

HLA-DRB1 alleles genotyping in patients with rheumatoid arthritis in Chinese.

OBJECTIVE: To explore the role of HLA-DRB1 genes in the development of rheumatoid arthritis (RA) and the correlations between HLA-DR alleles and clinical manifestations of patients with RA. METHODS: 86 patients with rheumatoid arthritis and 106 race matched controls were studied in whom HLA-DR typing was performed by the method of DNA amplification with sequence-specific primers (PCR-SSP). The subtypes of HLA-DR4 were determined by the method of hybridization of PCR products with sequence-specific oligonucleotides (PCR-SSO). The absence or presence of HLA-DR4 and its subtypes was correlated with the clinical and serological characteristics of the patients. RESULTS: Compared with controls, an increased gene frequency of HLA-DR4 (48.8% vs 17.9%, P < 0.001) and a decreased frequency of HLA-DR7 (16.3% vs 27.4%, P = 0.06) were found. The DRB1* 0405 account for 61.9% of DR4+RA patients and 21.1% of DR4+ controls (P < 0.01). There was no difference between the DR4+ and DR4- patients with respect to age, sex, duration of disease, rheumatoid factor (RF), extra-articular manifestations including secondary Sjogren's syndrome. According to the wrist X-ray stage, the patients of DR4+ were more severe than that of DR4- (P < 0.05). CONCLUSION: HLA-DR4 and DR4 subtype of DRB1*0405 are related to the development of RA in Chinese. HLA-DR4 can be a useful prognostic marker in the patients with RA.

[Review of applying implant-magnetic attachments to oral and maxillofacial prostheses].

In order to evaluate the applying effect of MDIC implant-magnetic attachments on oral and maxillofacial prostheses, 38 patients, whose oral and maxillofacial defects had been restored with implant-magnetic attachments prostheses, were followed up for 1-4 years. The effects were evaluated by the measurement of the retentive force of the prostheses, X-ray and clinical examination. The results show: the successful rate of the restoration was 94.7%; two sets implant-magnetic attachments could offer an average of 14.7 N retentive force for lower complete dentures, which could meet various functional needs of the prostheses, but magnetic retentive force decreased as the applying time goes on the magnetic retainers should be changed after two years; negative rate of bone absorption on implant-bone surface was 92.6%; negative rate of gingival inflammation was 91.3%. The implant-magnetic attachments offer realize reliable retention, don't transmit lateral force to implants. Patients can insert and remove the prostheses and clean the implants and defect cavities conveniently. The above advantages are benefit to tissue health and functions of the prostheses. So, implant-magnetic attachments can be used in the restoration of various oral and maxillofacial defects.

[Clinical study and application of 3DCT in oral and maxillofacial regions].

3DCT reconstruction as a new roentgenographic technique has not been used so much in oral and maxillofacial regions in our country. From May 1993 to May 1995, 72 cases of 3DCT reconstruction have been performed by a Siemens Plus-s CT scanner for the maxillofacial injuries and jaw bone diseases. The 3DCT reconstruction of soft tissuses has been used too. It is showed that 3DCT reconstruction is a useful technique for the diagnosis and surgical planning of diseases in oral and maxillofacial regions.

[The diagnosis and surgical management of ectopic thyroid tumour].

The diagnosis and treatment of seven patients with ectopic thyroid tumour in different position suggest that fine needle aspiration biopsy cytology be used before operation and fast freezing section be made during operation. Isotope scanning should be used as a routine method to determine whether there is normal thyroid in neck, so that we can treat it appropriately and prevent serious complications after operation.

[Regulation of epinephrine on melatonin receptor of the young mouse thymus].

UNLABELLED: The melatonin binding sites in membrane preparations of the young mouse thymus were studies using [125I] iodomelatonin as the radioligand. Effect of epinephrine (E) on melatonin receptor (MR) of young mouse thymus were investigated. RESULTS: (1) E had inhibitory effect on MR of mouse thymus; (2) Propranolol could reverse the inhibitory effect of E but phentolamin could not; (3) cAMP had inhibitory effect on MR. These results indicate that E has the inhibitory effect on MR of the mouse thymus and the effect was mediated by beta-adrenergic receptor.

[Analgesic action of microinjection of neurokinin A into the lateral reticular nucleus and nucleus raphe magnus in rats].

Using the microinjection technique, the analgesic effect of neurokinin A (NKA) microinjected into the lateral reticular nucleus (LRN) and nucleus raphe magnus (NRM) was investigated in lightly pentobarbital-anesthetized rats using tail flick latency (TFL) as an index. Microinjection of NKA (0.5 microgram/0.5 microliter) into LRN significantly increased TFL lasting for 10 min (n = 12, P < 0.001). Microinjection of the same amount of NKA into NRM also produced evident increase in TFL for 5 min (n = 13, P < 0.001). The results indicate that NKA modulates pain reaction in both LRN and NRM in rats.

[The effect of electric acupuncture on SEP of local cerebral ischemic-reperfusion rats].

The results suggest that 3 hours after cerebral ischemia, the P1-N1 amplitudes of the somatosensory evoked potential (SEP) of the cerebral cortex decrease significantly by the stimulation of cerebrum, while the latency period of both wave P1 and P2 prolonged; 2 hours later, the abnormal data of SEP have not any improvement. These suggest that the post-ischemic cortex and subcortical neurons were injured permanently by a substantial lesions from reperfusional injuries. Cerebral tissue histologic studies show that the infarcted focus locates in caudate nucleus and lateral cortex of cerebral hemisphere. But after electric acupuncture at Huantiao at the late stage of cerebral ischemia of rats, the abnormal data of SEP show better recoveries and manintains till 2 hours after reperfusion, and even near to that of the normal rat. All of them suggest acupuncture is effective obviously for ischemic injury of cerebral tissue, and possesses a presetting protection for reperfusional injury of cerebral tissue. So it is concluded that the efficacy of acupuncture results from its comprehensive and optimal regulation function to multisystem of the whole body.
