FPL-64176 modifies pore properties of L-type Ca(2+) channels.

In addition to its known effects on Ca(2+) and Ba(2+) currents, the L-type Ca(2+) channel agonist FPL-64176 was found to affect channel function in isolated rat ventricular myocytes in the absence of Ca(2+), with other ions as current carriers through the channel. FPL-64176 induced Cd(2+) current through the L-type Ca(2+) channel, suggesting that certain selectivity properties had changed, perhaps indicative of a small change in pore structure. FPL-64176 slightly but significantly decreased the effectiveness of Co(2+) as a blocker of the channel. FPL-64176 also increased conductance through single L-type Ca(2+) channels recorded in the cell-attached configuration, from 71.9 +/- 11.6 to 94.1 +/- 8.3 pS, with Na(+) carrying the current at pH 9.0. At present it is uncertain whether FPL-64176 produces small alterations of a sole open state of the channel or whether it increases the prevalence of a second, higher conductance open state. These changes, particularly the conversion of Cd(2+) from a pure blocker to a permeant ion, may be of eventual help in discriminating among different models for Ca(2+) channel selectivity.

Apoptosis, proliferation and p53 gene expression of H. pylori associated gastric epithelial lesions.

AIM: To study the relationship between Helicobacter pylori (H. pylori) and gastric carcinoma and its possible pathogenesis by H. pylori. METHODS: DNEL technique and immunohistochemical technique were used to study the state of apoptosis, proliferation and p53 gene expression. A total of 100 gastric mucosal biopsy specimens, including 20 normal mucosa, 30 H. pylori-negative and 30 H. pylori-positive gastric precancerous lesions along with 20 gastric carcinomas were studied. RESULTS: There were several apoptotic cells in the superficial epithelium and a few proliferative cells within the neck of gastric glands, and no p53 protein expression in normal mucosa. In gastric carcinoma, there were few apoptotic cells, while there were a large number of proliferative cells, and expression of p53 protein significantly was increased. In the phase of metaplasia, the apoptotic index (AI, 4.36%+/-1.95%), proliferative index (PI, 19.11%+/-6.79%) and positivity of p53 expression (46.7%) in H. pylori-positive group were higher than those in normal mucosa (P<0.01). AI in H. pylori-positive group was higher than that in H. pylori-negative group (3.81%+/-1.76%), PI in H. pylori-positive group was higher than that in H. pylori-negative group (12.25%+/-5.63%, P<0.01). In the phase of dysplasia, AI (2.31%+/-1.10%) in H. pylori-positive group was lower (3.05%+/-1.29%) than that in H. pylori-negative group, but PI (33.89%+/-11.65%) was significantly higher (22.09+/-8018%, P<0.01). In phases of metaplasia, dysplasia and gastric cancer in the H. pylori-positive group, AIs had an evidently graduall decreasing trend (P<0.01), while PIs had an evidently gradual increasing trend (P<0.05 or P<0.01), and there was also a trend of gradual increase in the expression of p53 gene. CONCLUSION: In the course of the formation of gastric carcinoma, proliferation of gastric mucosa can be greatly increased by H. pylori, and H. pylori can induce apoptosis in the phase of metaplasia, but in the phase of dysplasia H. pylori can inhibit cellular apoptosis. And H. pylori infection can strengthen the expression of mutated p53 gene.

[The association between SA gene and essential hypertension in Han Chinese].

OBJECTIVE: To evaluate the linkage of SA gene locus to essential hypertension(EH) in affected Han Chinese sib pairs and to ascertain the association of SA gene CT79 polymorphism with essential hypertension in Han Chinese hypertensives. METHODS: In Han Chinese at Shanghai, 96 random individuals, 80 essentially hypertensive sib pairs with hypertensive family history, 200 essential hypertensives with hypertensive family history and 200 normotensive control(NC) subjects without such family history were enrolled in these serial studies. MF-PCR-SSCP technique was applied to detect the frequency distributions of SA gene microsatellite D16S3046, D16S3136 and D16S3068. The linkage of SA gene locus to essential hypertension was analyzed by Green's IBS for affected siblings. SA gene variant was screened by PCR-SSCP- silver staining and confirmed by DNA sequencing, then an association study was performed to determine whether such variant was associated with essential hypertension. RESULTS: The frequency distributions of SA gene microsatellites D16S3046, D16S3136 and D16S3068 were of high polymorphism. The polymorphism information contents of D16S3046, D16S3136 and D16S3068 were 0.86, 0.82 and 0.80, and the heterozygosities 0.88, 0.71 and 0.77. The linkage of SA gene locus to essential hypertension was not observed; the linkage analysis t values of D16S3046,D16S3136 and D16S3068 were 0.972, 0.622 and 0.236, and the P values 0.384, 0.543 and 0.871, respectively. The C two head right arrow T substitution was confirmed, but there was no association of such variant with essential hypertension. The frequency distributions of genotypes and alleles in the hypertensive group were not significantly different from those in the normotensive group (P>0.05). CONCLUSION: SA gene is not linked to or associated with essential hypertension in Han Chinese. SA gene may not be a susceptible gene contributing to the development of essential hypertension in Han Chinese.

In situ prostate cancer gene therapy using a novel adenoviral vector regulated by the caveolin-1 promoter.

Caveolin-1, a structural component of caveolae, is overexpressed in metastatic and androgen-resistant prostate cancer and highly expressed in tumor-associated endothelial cells. The mouse cav-1 promoter was cloned and placed upstream of the HSV-tk gene in an adenoviral vector (Adcav-1tk) and compared with a cytomegalovirus (CMV) or Rous sarcoma virus (RSV) promoter-driven HSV-tk, AdCMVtk and AdRSVtk vectors, respectively. Mouse and human prostate cancer cells and mouse endothelial cells were infected with Adcav-1tk, AdCMVtk or control vectors without the HSV-tk gene (Adcav-1 and AdCMV) and subsequently treated with ganciclovir (GCV). GCV-mediated in vitro cytotoxicity induced by the Adcav-1tk vector was comparable to that for AdCMVtk in multiple mouse and human prostate cancer cell lines. To evaluate the activity of Adcav-1tk in vivo, orthotopic mouse prostate cancer tumors were generated with RM-9 cells and injected in situ with Adcav-1tk, AdCMVtk, AdRSVtk, or AdCMVbetagal (control) and treated with GCV. All three HSV-tk transducing vectors produced statistically significant reductions in wet weight and increased apoptotic indices compared with the control vector. However, only Adcav-1tk produced significant necrosis, and only Adcav-1tk and AdRSVtk caused significant decreases in microvessel density. In conclusion, Adcav-1tk demonstrated efficacy in vitro and in vivo in preclinical models of prostate cancer. Our results suggest that the cav-1 promoter may have unique benefits in targeting gene therapy to prostate cancer and its associated vasculature.

[Modulation of multidrug resistance by 1-phenyl-2-palmitoylamino-3-morpholino-1-propanol in SKOV3-adriamycin-resistant cell line].

OBJECTIVE: To study the modulation of mdr1 and P-glycoprotein (P-gp) by 1-phenyl-2-palmitoylamino-3-morpholino-1-propanol (PPMP) in SKOV3-adriamycin-resistant (SKOV3/AdrR) cell line. METHODS: SKOV3/AdrR cells were treated with PPMP, mRNA expression of multidrug resistant (mdr1) gene was analyzed by reverse transcriptase-polymerase chain reaction. Intracellular rhodamine (Rh123) concentration was measured by flow cytometry. RESULTS: PPMP was found to inhibit mdr1 expression of SKOV3/AdrR at the mRNA level. This modulation of gene expression was content dependent and complete inhibition appeared at 25 mumol/L PPMP treatment. PPMP could increase intracellular Rh123 accumulation in SKOV3/AdrR cells. After 15, 25 mumol/L PPMP treatment, Rh123 accumulation in SKOV3/AdrR was markedly enhanced Rh123 fluorescence intensity were 389.98, 426.08 respectively (P < 0.05). CONCLUSIONS: PPMP could modulate mdr1 expression at the mRNA level in a content dependent manner. P-gp mediated outward efflux of cytotoxic agents can be blocked by PPMP in SKOV3/AdrR cell. PPMP possesses multi-drug resistance activity in ovarian cancer.

[Gap formation features of humid evergreen broad-leaved forest in central subtropical Wawushan Mountain, Sichuan Province, China].

The primary and secondary subtropical evergreen broad-leaved forests at an altitude of 1720-1750 m in Mt. Wawushan National Forest Park, southwestern China investigated to analyze their canopy gap formation characteristics. The sampling method and canopy formation causes were also discussed. In secondary evergreen broad-leaved forest, the gap density was surprisingly only 9 per hm2 and the size of all gaps was, no more than 10 m2. Almost every gap had only one gap maker, and the gap makers often died standing by suppression. Accordingly successful natural regeneration is difficult in small gap, due to the rapidly lateral growth of canopy trees. In primary evergreen broad-leaved forest, the gap density was 15 per hm2, and 56% of the gaps had an area of no more than 40 m2. The largest gap had an area of 256 m2. The canopy gap accounted for 11.1% and expended gap occupied 19.8% of land area in the forest. Median area of canopy gaps and expended gaps was 59 and 105 m2, respectively. Most gaps had more than one gap maker, and gap makers died falling and were often from mortality events separated in time. Most gaps aged over 10 yr and a few were formed recently and the estimated gap formation rate was 0.01 per year. According to falling direction of trees and the relations with growth process, slope aspect and wind, we deduced gap-maker death resulted from integrated influence among topography, climate particular in wind, tree growth characteristics, and interactions of population under competition. Adapting method of plot sampling and projection drawing used in the paper can improve investigation accuracy and help improving comparison of results in different investigation area.

[Investigation on the relationship between MDR and expression of N-glycosphingolipids in vincristine resistant KBv200 cell line].

OBJECTIVE: To study the relationship between MDR and expression of N-glycosphingolipids (H-GSLs) in tumor cells. METHODS: Ribozyme with specific catalytic activity, capable of cleaving mdr1 mRNA, was transfected into KBv200 cells. RNA dot blot was used to detect the expression of ribozyme in the transfected cells. Northern blot and immunocytochemistry were employed to examine the expression of mdr1 mRNA and P-glycoprotein. Cellular N-GSLs was isolated and purified with a modified Hakamori's method and analysed by high performance TLC. The effect of DL-PPMP, a glycolipids synthase inhibitor, on the reversion of MDR was assayed by MTT method. RESULTS: The ribozyme stably expressed in KBv200/5mR3 cells decreased the level of mdr1 mRNA expression by 81.9% and inhibited the formation of P-glycoprotein. The levels of monohexosylceramide (CMH) and dihexosylceramide (CDH) in KBv200 cells were increased as compared to those in KB cells. When MDR was reversed by the ribozyme, the KBv200/5mR3 showed a sharply decreased level of CMH. PPMP could reverse MDR by inhibiting synthesis of CMH. CONCLUSION: CMH is a kind of MDR-related glycolipid. Inhibition of its expression in tumor cells may be a new approach to reverse MDR.

[The value of number connection test in the diagnosis of subclinical hepatic encephalopathy].

OBJECTIVE: To explore the influencing factors and diagnostic values of number connection test (NCT) in detection of subclinical hepatic encephalopathy (SHE). METHODS: NCT was carried out in four hundred and fifty healthy volunteers to explore the influencing factors and to work out a reference value. Then forty patients with liver cirrhosis were studied with NCT, intelligence quotient (IQ) and evoked potentials(EP) to determine the value of NCT in the diagnosis of SHE. RESULTS: (1) NCT results were influenced by age and educational background. The older the age and the lower the educational level, the longer the NCT time (P < 0.001); (2) The abnormal rate of NCT was 30% (12/40), being lower than the IQ abnormal rate of 58% (23/40) (P < 0.01). (3) IQ or/and EP were abnormal in 85%. (4) The combination of four tests i.e. block design, picture arrangement, object assembly and NCT were abnormal in 53% of the patients. Comparing with the abnormal rate (58%) of full set of IQ test, the sensitivity and specificity were 87% and 94%, respectively. CONCLUSIONS: NCT result was influenced by age and educational background and the sensitivity was low for the scanning of SHE. Combination of four psychometric tests: block design test, picture arrangement, object assembly and NCT was recommended to substitute full set of IQ test in the diagnosis of SHE.

[Effects of DDPH on proliferation and immunophenotypes of the pulmonary vascular pericytes].

OBJECTIVE: To study the effects of 1-(2, 6-dimethylphenoxy)-2-(3, 4-dimethoxyphenylethylamino) propane hydrochloride (DDPH) on proliferation and immunophenotypes of newborn rat pulmonary vascular pericytes (PCs) induced by hypoxic endothelial cell conditioned medium (HECCM) from porcine pulmonary arteries. METHODS: The PCs were divided into 4 groups according to the endothelial cell conditioned medium (ECCM): normoxic ECCM (NECCM) group, NECCM + DDPH group, HECCM group and HECCM + DDPH group. Cell culture, immunocytochemical staining, image analysis and flow cytometry were used to investigate the effects of HECCM and DDPH on the expression of alpha-smooth muscle actin (alpha-SM-actin) and proliferating cell nuclear antigen (PCNA) and cell cycle in PCs. RESULTS: The alpha-SM-actin antigen in the PCs in HECCM group was more positively expressed than that in the other three groups, but CD(34) antigen and S-100 antigen were negatively expressed. The expression of alpha-SM-actin antigen, CD(34) and S-100 was positive in the groups of NECCM, NECCM + DDPH and HECCM + DDPH. The expression of alpha-SM-actin and PCNA in HECCM group was 1.32 times (F = 11.09, P = 0.000 1) and 1.24 times (F = 2.89, P = 0.025 7) that in NECCM group, 1.30 times (F = 3.65, P = 0.007 0) and 1.21 times (F = 2.63, P = 0.041 4) that in HECCM + DDPH group, respectively; The percentage of the cells in the G(0)-G(1) phase in the HECCM group was lower by 11.7% and 9.1%, in S phase higher by 5.6% and 4.2%, in G(2)-M phase higher by 6.1% and 4.9% than that in the groups of NECCM, HECCM + DDPH, respectively. The inhibitory rates of DDPH on the increased alpha-SM-actin and PCNA syntheses in PCs induced by HECCM were 23.4% and 17.1% respectively. The inhibitory rate on the increased PCs from G(0)-G(1) phase to S phase was 8.3%. CONCLUSION: DDPH might directly inhibit PCs from proliferation and immunophenotypical transformation of smooth muscle-like cells induced by HECCM.

[Study of telomerase activity and its related genes expression in leukemia cells].

OBJECTIVE: To investigate the changes of telomerase activity and its related genes expression in leukemia cells and their roles in leukemogenesis. METHODS: The telomerase activity in bone marrow cells of 47 leukemia patients were detected by a semi-quantitative telomeric repeat amplification protocol (TRAP)-silver staining, and the expression of telomerase related genes hEST2, TP1 and hTR by semi-quantitative reverse transcriptase polymerase chain reaction (RT-PCR). RESULTS: The telomerase activity and expression of telomerase related genes were significantly higher in 42 leukemia patients in progress stage than that in complete remission/chronic stage and in normal/anemia cases (P < 0.01). The telomerase activity and expression of TP1, hTR gene in 14 cases of acute leukemia complete remission (AL-CR)/chronic myelogenous (CML)-chronic stage were higher than that in normal/anemia cases (P < 0.05). Low levels of telomerase activity and telomerase related genes expression could be detected in part of normal bone marrow cells. The levels of telomerase activity and telomerase related genes expression in 3 cases of anemia increased after short-term culture. CONCLUSIONS: High level of telomerase activity and its related genes expression were specific markers of malignant hematopoietic cells and their changes might be useful for observation of treatment effectiveness in leukemia. Low level of telomerase activity was associated with hematopoietic stem cells with the ability of proliferation. hEST2 gene might be a up-regulation gene of telomerase; hTR might be related to feedback regulation of telomerase.

[The effect of leukotriene antagonist on the parameters of dose-response curve obtained after methacholine challenge test].

OBJECTIVE: To investigate the effect of leukotriene antagonist (zafirlucast) on the parameters of dose-response curve (DRC) obtained after methacholine challenge test. METHODS: A therapy trial was conducted with the use of zafirlucast in 20 cases of cough variant asthma (CVA) and 20 cases of asthma. Placebo was used in 4 cases of CVA and 10 cases of asthma for comparison with the two therapy groups respectively. Before and after the zafirlucast treatment a series of parameters of DRC were obtained after the methacholine challenge test and were calculated. These included the response threshold (PC35sGaw), the dose-response slope (DRS), the ratio of the area under the curve to the logarithm of the maximal concentration (AUC/1 g[Cmax]), and the maximal response. RESULTS: Zafirlucast significantly increased the PC35sGaw in the CVA and asthma groups. It conspicuously decreased all the parameters of DRS and AUC/1 g[Cmax] except for maximal response in the cases of CVA, and it had no significant effects on all the parameters of DRS and AUC/1 g[Cmax] except for PC35sGaw in the cases of asthma. CONCLUSION: Short-term use of Zafirlucast in asthmatic patients could improve the parameters of dose-response curve obtained after the methacholine challenge test.

[A comparison between the effects of albuterol and isoproterenol in bronchodilation test].

OBJECTIVE: To compare albuterol with isoproterenol as a bronchodilator in pulmonary function test and their value in the diagnosis and differential diagnosis of asthma. METHODS: Two groups (59 patients with asthma and 37 patients with COPD) were included in this study, and the patients of each group were randomly divided into 2 subgroups. The patients in the 2 subgroups inspired the two drugs in a cross-over by odd or even days. The pulmonary functions were evaluated. RESULTS: The changing rates of pulmonary functions induced by the two drugs in the two groups were positively correlated (P < 0.005), The changing rates of FEV1, MMEF, Raw, sGaw induced by albuterol were higher than the ones induced by isoproterenol in patients with asthma(P < 0.02), however the changing rates of FVC and PEF in asthma cases and all the above parameters in COPD cases were not significantly different between the two drugs(P > 0.10). The comparison of positive case number between asthma and COPD cases induced by both drugs showed significant difference(P < 0.005); especially, when the positive rate of sGaw induced by albuterol in asthma cases reached 98%, the positive rates in COPD cases was generally much lower. CONCLUSION: Bronchodilation test is a valuable diagnostic method for asthma. In this study, albuterol was superior to isoproterenol. The most sensitive index in bronchodilation test was sGaw, then in order, FEV1(92%), MMEF and Raw; the FVC and PEF were relatively insensitive.

[Measurement of central respiratory drive and inspiratory muscles strength in patients with uremia].

OBJECTIVE: To investigate the changes of central respiratory drive and inspiratory muscles function in patients with uremia. METHODS: We performed the measurement of forced vital capacity (FVC), maximal voluntary ventilation (MBC), forced expiratory volume in one second (FEV1), peak expiratory flow (PEF), maximal midexpiratory flow (MMEF), expiratory flow of 25 per cent of FVC (V25), lung carbon monoxide diffusing capacity (DLco), maximal inspiratory mouth pressure (MIP) and airway occlusion pressure (P0.1) in 25 patients with uremia and 20 normal subjects. RESULTS: In patients with uremia, the FVC, MBC, FEV1, PEF, MMEF and V25 which reflect the lung ventilatory function and the DLco which reflects the lung diffusing function were significantly lower than those in normal subjects. The patients' MIP which reflects inspiratory muscles strength was significantly lower and their P0.1 which reflects the central respiratory drive was significantly higher, compared with the normal subjects'. CONCLUSION: Our findings suggest that on the basis of the disorder of ventilation and diffusing function of the lungs, the inspiratory muscles function of the patients with uremia is significantly decreased and the central respiratory drive of the patients is increased.

[Measurement of the parameters of dose-response curve obtained after the methacholine challenge test].

OBJECTIVE: To investigate the clinical value in the measurement of parameters of the dose-response curve (DRC) obtained after the methacholine challenge test. METHODS: Twenty-seven cases of cough variant asthma (CVA), 29 of mild asthma, 19 of moderate asthma, and 15 healthy volunteers underwent the methacholine challenge test. The dose-response curves were constructed. The following parameters were calculated: position (PC35sGaw), dose-response curve (DRS), ratio of the area under the curve to the logarithm of the maximal concentration (AUC/lg[Cmax]) and maximal response. RESULTS: A plateau appeared in 11 cases among the healthy subjects, while a plateau appeared only in 2 cases of CVA and 2 of asthma. There were no significant differences among the asthmatic groups in the measurements of PC35sGaw and AUC/lg[Cmax] (geometric mean PC35sGaw: mild asthmatics = 0.369, moderate asthmatics = 0.251, cough variant asthmatics = 0.547), AUC/lg[Cmax] was significantly greater in the asthmatic groups than in the normal group [AUC/lg [Cmax](mean +/- s): mild asthmatics = 24.7 +/- 4.7, moderate asthmatics = 26.6 +/- 4.3, cough variant asthmatics = 25.6 +/- 3.7, normal subjects = 15.5 +/- 4.3, P < 0.01]. DRS of moderate asthmatics was significantly greater as compared with cough variant asthmatics (geometric mean DRS: mild asthmatics = 30.761, moderate asthma-tics = 59.020, cough variant asthmatics = 19.231, P < 0.05), but there was no difference between the other groups. PC35sGaw was negatively correlated with DRS (r = 0.866, P < 0.001) and with AUC/lg[Cmax] (r = 0.502, P < 0.001). CONCLUSION: Four parameters of DRC obtained after the methacholine challenge represent airway sensitivity and reactivity respectively. The parameters of DRC of asthmatic patients are different from those of normal subjects. The sensitivity is related to the reactivity, but they are not completely parallel with each other.
