Glial cell line-derived neurotrophic factor differentially stimulates ret mutants associated with the multiple endocrine neoplasia type 2 syndromes and Hirschsprung's disease.

Ret is a receptor tyrosine kinase involved in several neoplastic and developmental diseases affecting the thyroid gland and tissues of neuroectodermal origin. Different ret mutations are associated with different disease phenotypes. Gain-of-function of ret is caused by gene rearrangements in thyroid papillary carcinomas and by point mutations in multiple endocrine neoplasia (MEN) type 2A syndrome (MEN2A), in familial medullary thyroid carcinoma (FMTC), and in the more severe MEN2B syndrome. Conversely, Hirschsprung's disease (HSCR) is associated with loss of function of ret. Recently, it has been shown that glial cell line-derived neurotrophic factor (GDNF), by binding to the accessory molecule GDNFR-alpha, acts as a functional ligand of Ret and stimulates its tyrosine kinase and biological activity. To ascertain whether the biological effects of ret mutations are modulated by GDNF, we have investigated the responsiveness to GDNF of ret mutants in cell lines coexpressing GDNFR-alpha and MEN2A-, MEN2B-, FMTC-, or HSCR-associated ret mutants. Here, we show that triggering of GDNF affected only ret/MEN2B, i.e. it stimulated ret/MEN2B mitogenic and kinase activities, as well as its ability to phosphorylate Shc, a bona fide Ret substrate. In contrast, ret mutants associated with MEN2A or FMTC (carrying Cys634 or Cys620 mutations) were unresponsive to GDNF. HSCR mutations, by affecting either the extracellular or the intracellular Ret domain, impaired responsiveness to GDNF. These data suggest that the phenotype of human diseases caused by ret mutations can be differentially influenced by GDNF.

Immobilization of microalgae for biosorption and degradation of butyltin chlorides.

Since the discovery of their biocidal properties in the 1950s, organotin compounds have found a large spectrum of industrial applications such as wood and textile preservatives, fungicides and pesticides, and antifouling paint on ships and fishing equipment. The fate and environmental impact of butyltins have been the subjects of a large body of research in the last decades. Biosorption and degradation of butyltin compounds by immobilized microalgae chlorella were studied in this paper, aiming to find an alternative way to solve organotin pollution problem. Chlorella emersonii cells were entrapped in a calcium aginate matrix. The cell growth rates, respiratory rate and chlorophyll a content were studied and compared. Results showed that immobilized chlorella had increased respiratory and growth rates, and almost equal chlorophyll a content when compared with free cells. Cell leakage was slight during the 20-day experimental period Cell leakage from the matrix was unrelated to cell growth within the matrix. Immobilized chlorella was applied to deal with butytin contaminated aquatic solutions. Immobilized chlorella had increased degradation rates of tri-, di-, and mono-butyltin chlorides in aquatic solutions, and lower biological accumulation factors on cells, than free cells, which indicates a potential use for tackleing organotin polluted water body.

[Establishment of transgenic cell line CHL-3A4 and its metabolic activation].

OBJECTIVE: To establish a model cell line CHL-3A4 to study metabolic pathways for some genotoxic chemicals. METHODS: Complimentary DNA (cDNA) of cloned cytochrome P450-3A4 gene in human liver tissue was transferred to Bluescript M13 vector with reverse transcription polymerase chain reaction (RT-PCR) and DNA recombinant techniques. Restriction endonuclease map analysis and sequencing of partial cloned fragment proved that CYP3A4 cDNA was cloned into Bluescript M13 vector. Then, recombinant expression plasmid pREP9-3A4 was constructed in eukaryotic cell and transfected into Chinese hamster CHL cells. RESULTS: It was proved that a CHL-3A4 transgenic cell line was established, which could lead metabolic activation for aflatoxin B1 (AFB1), sterigmatocystin (STC) and cyclophosphamide (CPA). CONCLUSION: The CHL-3A4 cell line established did express human cytochrome P450 3A4 and could lead metabolic activation for three genotoxic chemicals AFB1, STC and CPA.

[Antitumor effect of M-CSF and/or IFN-gamma gene cotransfected-macrophages by intratumoral injection].

OBJECTIVE: To investigate the in vivo antitumor effects of M-CSF and/or IFN-gamma gene transfected-macrophages and their related immunological mechanisms. METHODS: Survival time and antitumor effects in vivo of tumor-bearing mice treated with gene-transfected macrophages were observed. NK cell and CTL activity was detected with 4 hr 51Cr release assay, cytotoxicity of macrophages was detected with indirect MTT methods, activity of IL-2, TNF, IFN-gamma were detected by routine methods. RESULTS: 25% of the tumor-bearing mice after treatment with IFN-gamma gene-transfected, M-CSF and IFN-gamma gene co-transfected macrophages survived more than 3 month. The CTL and macrophage activity of the tumor-bearing mice was enhanced. Levels of TNF, IL-2, IFN-gamma in the supernatants of splenocytes induced in vitro were also increased. Histologic analyses of the tumor tissue after treatment with M-CSF and/or IFN-gamma gene-transfected macrophages showed a large amount of lymphocyte infiltration. CONCLUSION: IFN-gamma and M-CSF gene co-transfection into macrophages are proved to enhance their antitumor potential efficiently through direct augmentation of their tumoricidal cytotoxicity and induction of host antitumor immunity.

Pathologic changes, mechanisms and diagnosis in renal bone disease.

OBJECTIVE: To investigate the incidence rate, pathologic changes, mechanisms and diagnostic methods in renal bone disease. METHODS: The blood levels of carboxyterminal parpthyriod hormone (C-PTH), 1,25(OH)2D3, calcium and phosphate, aluminum in serum and bone tissue were measured. The bone biopsy and bone scan with 99m technetium methylene diphosphonate (99mTC-MDP) were performed in 51 uremic patients. RESULTS: One hundred per cent of the patients had varying degree of pathologic changes in bone, in which 50.9% of the patients presented high-turnover bone disease, 7.8% of the patients presented low-turnover bone disease and 41.8% of the patients had mixed-type bone disease. The levels of serum C-PTH were predominently high in high-turnover bone disease while the levels of serum 1,25(OH)2D3 were significantly decreased in low-turnover bone disease. There was a high positive rate for the diagnosis of renal bone disease by bone scan with 99mTC-MDP. CONCLUSIONS: The examination of bone pathology is the most valuable method for the diagnosis of renal bone disease. Bone scan with 99mTC-MDP has reference value when clinical conditions do not allow to make bone biopsy.

The role of gut as a cytokine-generating organ in remote organ dysfunction after intestinal ischemia and reperfusion.

OBJECTIVE: To test the hypothesis that in a rat model of acute intestinal ischemia/reperfusion injury, endotoxemia from the gastrointestinal tract would play a role in mediating tumor necrosis factor (TNF) response and remote organ dysfunction. METHODS: Wistar rats underwent 45 minutes of superior mesenteric artery occlusion followed by 6 hours of reperfusion. The rats were treated intravenously with either TNF monoclonal antibody (MoAb) or saline solution 90 minutes prior to the onset of ischemia. RESULTS: Significant elevation of plasma TNF level in both portal and systemic circulation was detected immediately after the onset of reperfusion. The level peaked at two hours after reperfusion (P < 0.01). Similarly, a remarkable TNF mRNA expression in the intestine in controls was detected at 0.5 hour post-reperfusion, and sustained a marked elevation throughout the observation period (P < 0.05-0.01). TNF elevation was found associated with gut-origin endotoxemia, and the maximal TNF response occurred approximately 0.5-2 hours after the initial appearance of endotoxin in the portal vein. Concomitantly, multiple organ dysfunction in response to local ischemic insult was also observed in untreated controls upon reperfusion, but it was significantly attenuated by pretreatment with MoAb against TNF. CONCLUSIONS: Intestinal injury can result in the gut becoming a cytokine-liberating organ. The escape of endotoxin and bacteria from the gut may be responsible for the TNF expression and release, which would be an important mechanism underlying pathophysiological alterations associated with intestinal ischemia/reperfusion injury.

[ABR feature of medial and old patients with hyperlipidemia].

Sixteen medial and old patients with essential hyperlipidemia who were tested auditory brain-stem response (ABR) with click repetition rate 20/s and 40/s, respectively. In comparison with 12 medial and old people with normallipidemia and 10 health people, the IL and IPL of ABR of the hyperlipidemia group are longer than that of other two groups. There is statistical significant difference among the 3 groups of ABR date. The result showed that the hearing function of the medial and old patients with hyperlipidemia dropped potentially in early phase. Increasing the click repetition rate can improve the detection probability of positive reaction of ABR.

[Changes of prostaglandins in kidney tissue in gentamicin-induced acute renal tubular injury in rats].

OBJECTIVE: To examine the relationship between epidermal growth factor (EGF) and prostaglandins (PGs) in recovery of acute tubular necrosis (ATN) induced by gentamicin in rats and the changes of renal tissue PGs with EGF treatment. METHODS: Female wistar rats were divided into three groups; normal(NL, n = 7); GM-treated only (Group G, n = 20); GM 200 mg/kg, i.p. x 3 d; GM and EGF-treated (Group G + E, n = 19): EGF(20 micrograms) was given after last GM injection. [3H]thymidine incorporation rate (3HTdR) of renal tissue, serum creatinine concentration (Scr), renal levels of PGE2, 6-keto-PGF1 alpha, TXB2 were measured at day 1,4,8,12 after GM administration. RESULTS: [3H]thymidine incorporation rate of renal tissue in group G + E was significantly higher than that in group G after toxic injury. The histological lesions of group G + E was less severe than that in group G. 6-keto-PGF1 alpha in group G + E was increased significantly than that in group G, and renal TXB2 in group G + E was lower than that in group G. PGE2 and 6-keto-PGF1 alpha in group G + E was positively correlated with 3HTdR, respectively. CONCLUSIONS: (1) changes of renal prostaglandins may be related to the injury/proliferation of renal tubular epithelial cells in ATN. (2) Administration of exogenous EGF may enhance the release of PGE2 and 6-keto-PGF1 alpha of renal tissue and inhibit the synthesis of renal TXB2. The results indicate that effect of ameliorating recovery of renal tubular epithelial cells of EGF could be partly related to the changes of renal PGs.

[Application of the sphygmogram in evaluation of pilots' cardiovascular function in resting condition].

With the purpose of exploring a new method for aerospace medical monitoring, a newly developed method--sphygmogram was used in 169 pilots. The results showed that these pilots could be divided into three types by the features of their resting sphygmogram. Among them, with respect to their cardiovascular function, 71 (about 42%) were "completely normal", 79 were "essentially normal", and 19 had some changes in cardiovascular function. It is considered that the status of cardiovascular function of the pilots can be distinguished with the newly developed method of sphygmogram. It suggests that it is important to make dynamic observations of pilots' cardiovascular function and strengthen the medical care for those who show certain changes in their cardiovascular function as revealed by the sphygmogram.

[Observation of electronystagmogram in pilots under vestibular stimulation].

The relationship between electronystagmogram(ENG) and intensity of vestibular stimulation was observed in 62 pilots. 31 pilots received low intensity vestibular stimulation and the other 31 pilots received higher intensity vestibular stimulation. Electronystagmogram were recorded and analysed. The results showed that there was no relationship between the ENG parameters and stimulation intensity, but both slow phase and fast phase velocities and amplitudes were lower under Coriolis acceleration stimulus, while all of them were higher under angular acceleration and angular stimulations. It was also found that the fast phase amplitude of ENG was related to motion sickness symptoms of the subjects.

[Effects of vestibular stimulus on heart rate variability].

Possible effects of vestibular stimulation autonomic nerve function in pilots were studied by observing changes of Heart Rate Variability (HRV). Time domain and frequency domain analyses demonstrated that the primarily averages, standard deviation of the HRV averages(SD), least or greatest value, deviation of greatest minus least value, interrelated averages, square root of the mean of the sum of squares of differences between adjacent R-R intervals (RMSSD), total power(TP), low frequency(LF), high frequency(HF), LF/TP, HF/TP, and LF/HF all had significant increase or decrease (P <0.05 - <0.01) and the degrees of changes were related to the intensity of the stimulations.

Erosive-throat with rash (scarlet fever), an investigation on its brief history.

Being a kind of epidemic contagious disease in modern time, erosive-throat with rash was a fatal disease causing numerous victims. High attention was paid to by traditional medical professionals through various fields, theoretical and clinical. A correct conclusion was reached claiming that TCM did make contributions to its prevention and treatment before effective treatment in western medicine appeared in China.

[The effect of the macrophages conditioned media(MOCM) on NGF secretion in newborn rat schwann cells in vitro].

OBJECTIVE: To study the influence of macrophages on nerve growth factor (NGF) secretion in newborn rat schwann cells. METHODS: LPS-activated and not-activated adult rat peritoneal macrophages conditioned media (MOCM) were treated on purified cultured Schwann cells for 24, 48 and 72 hours in three different concentrations the NGF contents in the cultured Schwann cells suspension media were measured by a two-site enzyme-linked immunoserbent assay. RESULTS: Both L-MOCM (LPS-activated MOCM) and N-MOCM(not-activated MOCM) obviously enhanced NGF secretion in chwann cells in each different concentration or testing time. CONCLUSION: MOCM contains some factors which can promote NGF secretion in rat schwann cells in vitro, suggesting that macrophages regulating NGF secretion by secreted certain cytokines.

[The relationship between nm23-H1 loss of heterozygosity and metastasis in hepatocellular carcinoma].

OBJECTIVE: To demonstrate the relationship between nm23-H1 loss of heterozygosity (LOH) and clinical pathological characteristics of hepatocellular carcinoma (HCC) from the DNA level. METHOD: Southern blot hybridization was used to analyze genomic DNA in liver cancer and its corresponding liver tissue. RESULT: nm23-H1 has diallelic bands at 7.6 Kb and 2.3 Kb. Allelic loss of heterozygosity in tumour tissues was detected in 31.25% patients (5 patients). LOHs were more common in the tumours with intrahepatic metastasis or portal vein tumour embolism and the tumours poorly differentiated (Edmondson's classification III, IV). CONCLUSION: nm23-H1 LOHs may induce the metastasis potential of HCC and help to predict recurrence and metastasis of HCC.

[Retrograde partial hepatectomy: report of 11 patients].

OBJECTIVE: To study a new approach to hepatectomy for liver tumors resected difficulty by typical method. METHOD: The operative order is retrograde as compared with typical hepatectomy, namely, transection of the liver is performed first, isolating adhesions between the tumor and the diaphragm or partial invaded phrenectomy is performed second, and then after cutting corresponding ligaments, liver tumor is removed. If the surrounding organs were invaded or adhered by tumor too tightly to be separated. They were resected with the tumor. RESULT: The approach was used in 11 patients with malignant tumors of liver from June 1994 to January 1996. Operation time was 192 min (150 - 250 min) and estimated blood loss during operation was 1460 ml (800 - 4, 200 ml). No operative mortality was found and all of the patients recovered uneventfully. CONCLUSION: The retrograde partial hepatectomy is a good method for resection of liver tumors resected difficulty by typical hepatectomy.

[Effect of TNF-alpha monoclonal antibody on tissue lipopolysaccharide-binding protein mRNA expression in rats after thermal injury].

OBJECTIVE: To explore the effects of tumor necrosis factor-alpha (TNF-alpha) monoclonal antibody (MAb) on tissue TNF-alpha and lipopolysaccharide-binding protein (LBP) mRNA expression, and multiple organ dysfunction in rats after thermal injury. METHOD: 24 male Wistar rats were subjected to a 35% total body surface area full-thickness thermal injury. RESULT: The tissue TNF-alpha and LBP mRNA expressions of liver, lung, intestine, and kidney were markedly increased (P < 0.05 approximately equal 0.01) after thermal injury. Treatment with TNF-alpha MAb could significantly lower TNF-alpha mRNA expression of lung, intestine and kidney, but had no effect on liver TNF-alpha mRNA expression (P > 0.05). Similarly, the tissue LBP mRNA expression also decreased significantly (36.0% - 72.9%), and returned to normal in lung, intestine and kidney. In addition, biochemical parameters including GPT, TBiL, BUN, CK-MB, and LDH were markedly elevated after thermal injury, and decreased significantly after treatment with TNF-alpha MAb. CONCLUSION: The increase of tissue TNF-alpha gene expression caused by thermal injury might be associated with a marked elevation of tissue LBP mRNA expression, which could contribute to the development of multiple organ dysfunction. The early use of TNF-alpha MAb seems to be effective in inhibiting significant LBP mRNA expression in various tissues and ameliorating multiple organ damage after major thermal injury.

[The significance of changes in serum soluble lipopolysaccharide receptor CD14 levels in burned patients with multiple organ dysfunction syndrome].

OBJECTIVE: To determine whether an increased soluble lipopolysaccharide receptor CD14 (sCD14) level is associated with the development of multiple organ dysfunction syndrome (MODS) in patients after major burns. METHOD: 22 patients with burns covering more than 70% of body surface area were studied. These patients were divided into two groups according to the occurrence of MODS (MODS group, n = 9; non-MODS group, n = 13). Blood samples were obtained from burned patients. Serum concentrations of sCD14 and tumor necrosis factor (TNF) were determined by sandwich ELISA. Plasma endotoxin was measured using a quantitative chromogenic LAL test. RESULT: Compared to the healthy volunteers, a significant increase in serum sCD14 levels was observed in 22 patients on day 7 postburn, which remained marked elevation up to 21 days after thermal insult (P < 0.01). The serum sCD14 levels of the MODS group were significantly higher than those of the non-MODS group on days 7, 14, and 21 postburn (P < 0.05). Additionally, the serum sCD14 and endotoxin levels were positively correlated in patients who developed MODs on days 7, 14 and 21 postburn (P < 0.05), whereas no correlation was found between sCD14 and TNF levels at any time point (P > 0.05). CONCLUSION: Our data suggest that sCD14 elevation is associated with the development of MODS after major burns, and endotoxin release may be involved in the activation of sCD14 under certain pathophysiological conditions.
