Migration of medial smooth muscle cells to the intima after balloon injury.

Migration to the intima and other responses of M-SMC in the rat carotid artery and abdominal aorta after balloon injury were investigated in vivo. Migration occurred intensively between the second and fifth days after injury. About 80% of the cells were in the G1 and S phases of the cell cycle. The majority of the migrating cells were therefore simultaneously proliferating. Positive values of 42.3%, 48.9%, 44.4%, and 32.8% of the migrating cells on the fifth day in the carotid artery for PDGF-B, elastase III B, MMP-I, and MMP-9, were observed, respectively. Many of the cells expressed messages of PDGF-A and elastases II and III B by in situ hybridization. Fine structures of the migrating cells were characterized as a synthetic phenotype of the smooth muscle cell with reduced attachment to their surrounding ECM. A biphasic proliferative response of the M-SMC appeared on the second and fifth days. Migration occurred correspondingly in the proliferative period. The populations of M-SMC positive in immunostainings for PDGFs, their receptors, elastase III B, and MMP-1 and MMP-9 also increased biphasically, around 12 h and five days after the injury. The results of these studies suggest that the migrating cells were proliferative and synthesizing PDGFs, elastases, and collagenases.

Mg2+- or Mn2+-dependent endonuclease activities of human myeloid leukemia cells capable of producing nucleosomal-size DNA fragmentation.

The presence of at least two distinct Mg2+- or Mn2+-dependent, Ca2+-independent endonuclease activities was shown in the myeloid leukemia cell line P39. One of them was recovered from nuclear extracts and the other from a cytoplasmic fraction. The molecular size of the former was 30 kDa in both gel filtration and activity gel and that of the latter approximately 130-140 kDa in gel filtration and 65-70 kDa in activity gel. These two activities were almost completely inhibited by 0.1 mM ZnCl2 or 0.1 mM aurintricarboxylic acid, common inhibitors of apoptosis. Both could produce nucleosomal-size DNA fragmentation when incubated with diethyl-pyrocarbonate-treated nuclei as substrates, and the pattern of cleavage was 3'-OH and 5'-P. Taken together, either or both of these activities may be associated with apoptosis of myeloid leukemia cells.

cGMP-kinase mediates cGMP- and cAMP-induced Ca2+ desensitization of skinned rat artery.

(Rp)-8-Bromo-guanosine 3',5'-cyclic monophosphorothioate (Rp-8-Br-cGMPS) inhibited competitively both isozymes of type I alpha and I beta cGMP-dependent protein kinase (cGMP-kinase) purified from porcine aorta with apparent Ki values (microM) of 3.7 for I alpha and 1.8 for I beta. The compound also inhibited bovine heart type II cAMP-dependent protein kinase (cAMP-kinase), but with a Ki of 25 microM. Thus, it is a selective inhibitor of cGMP-kinase. In alpha-toxin-skinned smooth muscle preparations from rat mesenteric artery, 8-Br-cGMP (10(-7) M) and 8-Br-cAMP (10(-6) M) produced a rightward shift of the concentration-contraction curves for Ca2+, denoting a decrease in Ca2+ sensitivity of the contractile elements. The shift by 8-Br-cAMP as well as by 8-Br-cGMP was completely reversed by Rp-8-Br-cGMPS, while a selective inhibitor of activation of cAMP-kinase, (Rp)-adenosine-3',5'-cyclic monophosphorothioate (Rp-cAMPS), was without effects on the shift produced by these two compounds. These findings indicate the pivotal role that the activation of cGMP-kinase plays in the production of a decrease in Ca2+ sensitivity of contractile elements.

VSV-G-pseudotyped retroviral packaging through adenovirus-mediated inducible gene expression.

It has been technically difficult to generate recombinant adenoviruses encoding genes for cytotoxic products such as vesicular stomatitis virus G-protein (VSV-G), which is too toxic for the host cells to allow adenoviral propagation. In our companion paper (Yoshida, Y., and Hamada, H., Biochem. Biophys. Res. Commun., 230, 426-430, 1997), a tetracycline-inducible adenovirus system is reported. The inducible expression system enabled us to generate recombinant adenoviruses encoding genes for the cytotoxic viral VSV-G product. In this study, we generated recombinant adenoviruses encoding VSV-G and MoMLV gag-pol genes, both under the tetracycline-controllable promoter, and attempted retroviral packaging. Simultaneous infection of these adenoviruses together with tetracycline-transactivator (NtTA) expression resulted in efficient VSVG-pseudotyped retroviral packaging. Adenovirus-mediated recombinant retrovirus generation will be useful in studies with various pseudotyped mutants, as well as in assays for retrovirus-related genes and their products.

Effects of n-tritriacontane-16,18-dione, curcumin, chlorphyllin, dihydroguaiaretic acid, tannic acid and phytic acid on the initiation stage in a rat multi-organ carcinogenesis model.

The modifying effects of the naturally occurring antioxidants n-tritriacontane-16,18-dione (TTAD), curcumin, dihydroguaiaretic acid (DHGA), chlorophyllin, tannic acid and phytic acid on the initiation stage in a rat multi-organ carcinogenesis model were examined in male F344 rats. Animals were initiated with two i.p. injections of 2,2'-dihydroxy-di-n-propylnitrosamine (DHPN), followed by two i.g. administrations of N-ethyl-N-hydroxyethylnitrosamine (EHEN), and then three s.c. injections of 3,2'-methyl-4-aminobiphenyl (DMAB) during the first 3 weeks. Starting 1 day before the first carcinogen application, groups of rats received diet containing one of the antioxidants (0.2% TTAD, the others at 1% each) until 1 week after the last carcinogen exposure. Surviving animals were killed and complete autopsies were performed at the end of week 36. Histological examination revealed no inhibitory effects in terms of the multiplicities and/or incidences of neoplastic lesions in any of the organs examined, other than a significant increase in seminal vesicle atypical hyperplasia observed in rats treated with tannic acid. Thus, the antioxidants, with the exception of tannic acid, did not show any modifying effects on the initiation stage in the present multi-organ carcinogenesis model and at the present dose levels applied.

Adenovirus-mediated gene therapy of gastric carcinoma using cancer-specific gene expression in vivo.

The carcinoembryonic antigen (CEA) is a glycoprotein which overexpressed in the majority of human gastric cancers. We demonstrated that recombinant adenoviral vector (AdCEAtk), containing the CEA promoter, could transfer the herpes simplex virus thymidine kinase (HSVtk) gene into CEA-producing gastric cancer cells to confer sensitivity to ganciclovir (GCV) in vivo. In an ex vivo experiment, the tumor growth was inhibited after GCV treatment when the tumor contained more than 20% of AdCEAtk infected cells, indicating an efficient bystander killing effect. With intra-tumoral injection of AdCEAtk, the HSVtk were selectively expressed in approximately 30% of CEA producing cancer cells. By AdCEAtk injection and GCV administration, the growth of tumors was significantly inhibited by 20% as compared to untreated tumors. It is hoped that these results provide a strategy of tumor specific gene transfer for CEA producing gastric cancers.

In vivo gene therapy for alpha-fetoprotein-producing hepatocellular carcinoma by adenovirus-mediated transfer of cytosine deaminase gene.

The alpha-fetoprotein (AFP) gene is normally expressed in fetal liver and is transcriptionally silent in adult liver but overexpressed in human hepatocellular carcinoma (HCC). Here, we demonstrate that replication defective recombinant adenoviral vectors, containing the human AFP promoter/enhancer, can be used to express the Escherichia coli cytosine deaminase (CD) gene (AdAFPCD) and the beta-galactosidase gene (AdAF-PlacZ) in AFP-producing HCC cell lines. Expression of the CD gene by adenovirus from the AFP promoter/enhancer (AdAFPCD) induced cells sensitive to 5-fluorocytosine (5FC) in the AFP-producing cells but not in the AFP-nonproducing cells. Transduction by an adenoviral vector harboring an ubiquitous strong promoter and CD gene showed enzymatic activity and 5FC killing in all cell lines. When AdAFPlacZ was injected into the s.c. established hepatoma in vivo, expression of the beta-galactosidase gene was confined to AFP-producing HCC xenografts. Moreover, HCC xenografts regressed by transduction with AdAFPCD and subsequently with 5FC treatment in vivo. These findings suggest that utilization of the AFP promoter/enhancer in an adenoviral vector can confer selective expression of a heterologous suicide gene in hepatocellular carcinoma cells in vitro and in vivo.

Adenovirus-mediated inducible gene expression through tetracycline-controllable transactivator with nuclear localization signal.

Tetracycline-controllable expression vectors are widely used for inducible expression in mammalian cells. The limitation of this system is the difficulty in expressing high levels of the chimeric transactivator tTA. In this study, we demonstrate a utility of recombinant adenoviruses for the tetracycline-controllable expression system. Unexpectedly, the original tTA transactivator did not show sufficient regulation of the reporter gene expression driven by the tetracycline-responsive promoter (Tet). By adding the nuclear localization signal on the tTA transactivator (NtTA), we achieved tight regulation and high-level induction of the reporter gene expression. The NtTA driven by various promoters demonstrated strict tetracycline controllability at 1 microg/ml of tetracycline and above. The methodology for adenovirus-mediated inducible gene expression has wide applicability. Controllable expression of cytotoxic viral proteins will be applicable for antiviral vaccine productions and pseudotype viral vector generations.

The gnd gene encoding a novel 6-phosphogluconate dehydrogenase and its adjacent region of Actinobacillus actinomycetemcomitans chromosomal DNA.

A 10-kb DNA fragment containing the gnd gene from Actinobacillus actinomy-cetemcomitans Y4 was isolated and sequenced. The structural gnd gene codes for 6-phosphogluconate dehydrogenase that consists of 484 amino acids. In contrast to the gnd gene in Escherichia coli, Salmonella typhimurium, or Klebsiella pneumoniae, the gnd gene of A. actinomycetemcomitans was not located in the rfb or cps operon. The zwf gene encoding glucose 6-phosphate dehydrogenase, which is another enzyme consisting of pentose-phosphate pathway, sided at 3.8-kb upstream from the gnd gene. A phylogenetic tree based on sequence analyses showed higher homology of 6-phospho-gluconate dehydrogenase of A. actinomycetemcomitans with the eucaryotic enzymes rather than with bacterial enzymes.

Evidence for N-methyl-D-aspartate receptor-mediated increase in norepinephrine utilization in the prefrontal cortex of unanesthetized rats.

Local injection of N-methyl-D-aspartate (NMDA; 10-20 nmol/rat) into the prefrontal cortex of conscious rats caused a dose-related and an NMDA antagonist-reversible facilitation of norepinephrine (NE) disappearance in the cortical region during 35 min after inhibition of tyrosine hydroxylase. Destruction of ascending NE neurons by bilateral application of 6-hydroxydopamine into the superior cerebellar peduncle failed to affect [3H]N-(1-[2-thienyl]cyclohexyl)piperidine binding to the NMDA receptor-associated ion channel in the prefrontal cortex. These results indicate that, under unanesthetized conditions, the prefrontal NE neurons may be under glutamatergic facilitatory control mediated by the NMDA receptors which are located on the non-NE systems in the frontal cortex.

Laboratory diagnosis of anemia and related diseases using multivariate analysis.

To establish a simple computer program for the laboratory diagnosis of anemia and related diseases, multivariate analyses were applied to the results of routine hematological laboratory tests obtained from 48 patients and 51 healthy volunteers. The patients studied were limited to those who had not been treated hematologically by the time of their first visit to our hospital, and their first data obtained in our laboratory were analyzed. Final diagnoses were aplastic anemia (AA) in 21, myelodysplastic syndrome (MDS) in 14, iron deficiency anemia (IDA) in 3, polycytemia vera (PV)in 3, and idiopathic thrombocytopenic purpura (ITP) in 7. Eight parameters, WBC, RBC, Hb, Ht, MCV, MCH, MCHC, and PLT, were transformed to normal distribution and then applied to principal component analysis to evaluate their independence. Very close relationships were observed between Ht and Hb, and between MCV and MCH. One each of these pairs was selected by discriminant analysis and two sets, RBC, MCH, Hb, PLT, and WBC, and RBC, MCV, Ht, PLT, and WBC, were obtained. Two canonical components gave good discrimination of these five diseases and also of normal subjects. When disease prediction was made using this analysis, 37 of 48 patients (77.1%) were predicted correctly, and furthermore, when two disease predictions were allowed, all patients were diagnosed properly. Some overlaps were observed in this two-dimensional coordinate system, especially of AA and MDS, and also with normal subjects. To improve the system further, the additional parameters of age and sex were added to construct a three-dimensional analysis which resulted in much clearer discrimination. The whole procedure described is being developed with subjects who are not taking medication. Subsequently, the general application of this analytical procedure should be limited to only those not on medications. In conclusion, this is in essence a demonstration project; however, this trial of laboratory diagnosis using routine hematological laboratory results appears to be promising. Further extension of the study by increasing numbers of patients and disorders studied, including secondary anemias, will allow the design of diagnostic software for use with personal computers at the sites of primary care.

Proliferative activity of esophageal carcinomas and their lymph node metastases: comparison using argyrophilic nucleolar organizer region staining.

BACKGROUND: Many reports have concluded that quantification of the argyrophilic nucleolar organizer regions (AgNORs) measures proliferative activity and is a prognostic indicator in malignant disease. This retrospective study set out to evaluate the relationship between the AgNORs of the primary tumors and those of lymph node metastases in esophageal carcinoma. METHODS: Using a one-step silver staining technique, AgNORs were counted in surgical specimens from 54 patients with squamous cell carcinomas. RESULTS: The AgNOR scores of the lymph nodes metastases were significantly lower than those of the primary tumor (P = 0.0001). In 53 of 54 cases (98%), the AgNOR scores in the nodal metastases were lower than those of the primary tumor. The survival of 22 patients with AgNOR scores > or =4.0 for the primary tumor was significantly less than that of 32 patients with AgNOR scores <4.0 for the primary tumor (P = 0.0014). CONCLUSIONS: The AgNOR score of the lymph node metastases had no prognostic significance. The AgNOR score of esophageal primary cancer reflects the prognosis of patients. Scores for lymph node metastases were lower and did not reflect prognosis. The lower score in the lymph node metastases may result from the antitumor activity of macrophages in the lymph nodes.

Primary malignant lymphoma of the rectum with distant metastasis to the stomach: report of a case.

Primary extranodal malignant lymphoma frequently occurs in the gastrointestinal tract: however, it is rarely encountered in the rectum. We report herein the case of an 85-year-old man who underwent abdominoperineal resection for primary malignant lymphoma of the rectum, and 1 year later, developed hematemesis caused by gastric metastasis. To our knowledge this is the first case report of such distant metastasis developing from malignant lymphoma of the rectum.

Surgical removal of cavernous angioma in the medulla oblongata. A case report.

Recent advances in neuroradiology have enabled us to approach cavernous angioma in the medulla oblongata, rather rare vascular lesion in the central nervous system. We describe a such surgical case without additional neurological symptoms and discuss surgical indications in this paper. A 61-year-old woman presented with vertigo and swallowing disturbance. T1-weighted magnetic resonance image (MRI) showed a low intensity mass in the dorsolateral portion of the medulla oblongata, and T2-weighted imaging revealed a hemosiderin rim surrounding the lesion. Angiography showed no abnormalities. Surgery using far lateral approach achieved complete removal of the mass and hematoma. Histological examination of the surgical specimen disclosed cavernous angioma. This case suggests that direct surgery can be recommended for cavernous angioma located in the dorsal or lateral medulla oblongata to remove the hematoma and angioma if bleeding clearly provokes neurological symptoms.

Hemangiopericytoma of the sigmoid mesentery: report of a case with immunohistochemical findings.

Hemangiopericytoma has been described in various sites in the body but only rarely in the mesocolon. This report describes the clinical course of an 83-year-old man whose mesosigmoidal tumor (hemangiopericytoma) was resected on 11 November 1994. Immunostaining was done with the following primary antibodies: alpha-actin, vimentin, factor VIII-related antigen, chromogranin, and S-100. Staining for factor VIII-related antigen was strongly positive in the endothelial cells of the capillaries, but negative in the tumor cells. The tumor cells contained immunoreactive vimentin, but demonstrated no alpha-actin, chromogranin, or S-100. Since the operation, the patient has been disease-free for 11 months.

Anorectal malignant melanoma: report of a case.

A 74-year-old woman presented with anal bleeding and a protuberant mass. A biopsy of the mass revealed the proliferation of spindle-shaped cells with melanin pigments, and an abdominoperineal resection was performed, the histology of which confirmed malignant melanoma. Surgery was absolutely noncurative because massive metastases were encountered in the pelvic and paraaortic lymph nodes. However, postoperative chemotherapy, composed of decarbazine (DTIC), vincristine (VCR), and nimustine hydrochloride (ACNU), achieved satisfactory results. The patient has been well without any evidence of recurrence for more than 3 years.

Relapse of colon cancer followed by polymyositis: report of a case and review of the literature.

The direct causal relationship between dermatomyositis-polymyositis (PM) and malignancy remains controversial. We describe herein the case of a patient who underwent surgical treatment for colon cancer, which had preceded the onset of PM with tumor relapse. The PM markedly improved following the initiation of steroid therapy, and has remained under control, probably as a result of chemotherapy. The current concepts of variable clinical courses and the possible mechanism for the association of PM with malignancy are discussed following this case report.

Serum boron concentration from inhabitants of an urban area in Japan. Reference value and interval for the health screening of boron exposure.

Boron (B) levels were determined in the serum of 980 healthy inhabitants living in an urban area of Japan by means of inductively coupled plasma emission spectrometry (ICPES). The results showed a log-normal distribution of serum B for both sexes, although there are age-related differences. In male subjects, serum B increases rapidly up to 49 yr of age, reaching a plateau between ages 50 and 69 yr old, followed by a gradual increase up to 70 yr or older. Female subjects exhibit a gradual increase up to the age of 70 yr old. The reference value for male and female subjects was 79.8 micrograms/L and 67.9 micrograms/L, and the reference interval was 33.3-191.2 micrograms/L and 29.5-154.9 micrograms/L, respectively. The obtained reference value and interval of the nonexposed group may be useful for health screening for B exposure, either for people living in regions with high levels of B in the environment, or for workers who are exposed to this element.
