Normalized natural killer (NK) cell activity in long-term remission of acute leukaemia.

NK cell activity was studied in 28 cases of acute leukaemia in remission. It was found that patients in long-term remission for 5 years or more had normal NK cell activity but those in remission for less than 5 years had significantly lowered activity. This suggests that patients with acute leukaemia had not been constitutionally deficient in NK cell activity. Low activity was augmented but not fully restored by interferon pretreatment in vitro. Therefore, the low NK cell activity observed during remission is not solely attributable to inadequate secretion of interferon in the host. It is suggested that the production of NK cells in bone marrow is impaired, or the function of NK cells is suppressed, by the leukaemia itself or by chemotherapeutic agents. Increased consumption of NK cells is also a possibility.

Relationship between fibrinopeptide a (FPA) level and fibrinogen kinetics in patients with malignant disease.

The relationship between FPA level and fibrinogen turnover rate as well as fibrinolytic activity was studied in 18 patients with malignant diseases. It was found that the FPA levels were significantly elevated and were correlated with fibrinogen turnover rate (r = 0.74, p less than 0.001) and FDP (r = 0.58, p less than 0.02). The estimated FPA turnover rate was also correlated with fibrinogen turnover rate (r = 0.70, p less than 0.001). These results suggest that fibrinogen catabolism in patients with malignant disease is related to thrombin proteolysis. However, ratios of 1/2 FPA turnover rate to fibrinogen turnover rate suggest that intravascular thrombin proteolysis is not the major determinant of fibrinogen catabolism. It is suspected that extravascular thrombin proteolysis is responsible for the elevation of plasma FPA level which is correlated with acceleration of fibrinogen catabolism.

Blood-group A and the related glycolipids of human lung.

Three glucosamine-containing sphingoglycolipids were isolated from human lung tissue of a blood group-A subject. They were hexaglycosyl, pentaglycosyl, and tetraglycosyl ceramides. The hexaglycosyl ceramide exhibited blood group-A antigenicity, and the following chemical structure was proposed on the basis of sequential glycosidase treatment and methylation analysis. (Formula: see text) The pentaglycosyl ceramide for human lung was a blood group H(O)-active glycolipid having a carbohydrate composition of Fuc : GlcNAc : Gal : Glc (1 : 1 : 2 : 1). The tetraglycosyl ceramide is probably paragloboside (lactoneotetraosyl ceramide).

[Clinical evaluation of effectiveness of cefmetazole in bone and joint infections in the orthopedic field].

1. Cefmetazole (CMZ) was administered to a total number of 12 patients, 10 having acute or chronic osteomyelitis and 2 pyarthrosis in the orthopedic field. The efficacy rate was 91.7%. 2. Patients received an intravenous instillation of 1 to 2 g CMZ daily for an average period of 29.8 days. 3. Causative organisms were identified in 8 of the cases examined. In 7 cases S. aureus was isolated. CMZ showed an extremely strong antibacterial activity against S. aureus in a disk test in support of its excellent clinical results. 4. No abnormality was found with respect to subjective and objective symptoms and in laboratory tests. 5. The above results and migration of CMZ into the bone tissue at a high concentration suggest that CMZ is an effective and safe drug for bone and joint infection in the orthopedic field.

Acute monocytic leukemia cell isoferritin.

Ferritin was purified from acute monocytic leukemia cells, and its biochemical and immunological properties were investigated. The iron content of acute monocytic leukemia cell ferritin was extremely low, and this ferritin migrated slightly faster than normal adult liver ferritin on immunoelectrophoresis and polyacrylamide gel disc electrophoresis. Isoelectric focusing in polyacrylamide gel showed predominantly acidic isoferritins. This was considered to characterize the nature of isoferritin from normal monocytes or to be the result of neoplastic change of monocytes. Immunologic studies failed to recognized any antigenic differences between isoferritin from acute monocytic leukemia cells and that from normal adult liver. Therefore, selective quantitation of acute monocytic leukemia cell isoferritin, if any in leukemic sera, by means of immunoassay was considered difficult.

Isolation, characterization, and biosynthesis of Forssman antigen in human lung and lung carcinoma.

A glycolipid was isolated from normal lung and lung carcinoma tissues. This glycolipid was identified as the Forssman antigen of pentaglycosyl ceramide by means of chemical and immunological methods. The presence of this antigenic glycolipid was observed in all the tissues examined of adult and embryo lungs, and of lung tumors irrespective of histological type. The extracts of human lung and lung tumors were capable of catalyzing the synthesis of Forssman antigen from globoside.
