Expression and secretion of the S2 subunit of pertussis toxin in Bacillus brevis.

We have been trying to develop a mass production system for each of the subunits (S2, S3, S4, S5) of the pertussis toxin B oligomer (PTB) by using a Bacillus brevis-pNU212 system. In consequence a moderately efficient expression-secretion system for S2 was constructed by fusing the mature S2 gene from Bordetella pertussis Tohama with the signal-peptide coding region of pNU212 and by introducing the plasmid pNU212-S2 into B. brevis HPD31 by electroporation. The clone producing S2 secreted about 70 mg of recombinant S2 (rS2) per liter of PY-erythromycin medium after 5 days incubation at 37 degrees C. The rS2 purified by an ammonium sulfate fractionation at 30-50% saturation and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was identical to the native S2 in respect to the molecular weight determined by SDS-PAGE and the amino terminal amino acid sequence. The nucleotide sequence of the S2 gene in B. pertussis Tohama inserted into pNU212 was identical with that of the S2 gene in other virulent B. pertussis strains except that an adenine at position 52 of the latter was replaced by a guanine in the former, causing an amino acid substitution (glycine in the former for serine in the latter) at position 18.

Determination of interferon-alpha-producing capacity in whole blood cultures from patients with various diseases and from healthy persons.

To assess the clinical value of determination of the interferon (IFN)-producing capacity of patients, IFN production induced by Sendai virus (HVJ) in vitro was measured in cell cultures of whole blood from patients with various diseases. IFN production in patients with lung cancer, myelodysplastic syndromes, noninsulin-dependent diabetes mellitus, pulmonary tuberculosis, and asymptomatic HIV-1 infection was lower than that in healthy persons. Furthermore, periodic measurements of IFN production revealed decreasing IFN producing capacities in patients with lung cancer with progression of the tumor stage. However, increased IFN-producing capacities were observed in patients with tuberculosis after standard therapy. Further experiments showed that the main type of IFN induced in whole blood cultures was IFN-alpha, and decreased IFN production in patients did not result from a decreased number of leukocytes but rather from an impairment of cellular IFN production. The evaluation of IFN production in whole blood cell cultures may be a feasible method of assessing the impaired immune status.

Optimal pedaling rate estimated from neuromuscular fatigue for cyclists.

This study was designed to examine the optimal pedaling rate for pedaling exercise at a given work intensity for cyclists. Six college-aged cyclists each performed six sessions of heavy pedaling exercise at individually selected work rates based on their aerobic capacity. The optimal pedaling rate was evaluated on the basis of minimal neuromuscular fatigue as evidenced by the integrated electromyogram (iEMG) slope defined by the changes in iEMG as a function of time. The means of the iEMG slope demonstrated a quadratic curve versus pedaling rate. The mean values at 80 rpm (0.53 (SD 0.20) microV.min-1) and 90 rpm (0.67 (SD 0.23) microV.min-1) were significantly smaller than those values at any other pedaling rate. On the other hand, the mean value of oxygen uptake (VO2) expressed as a percent of the subject's maximal VO2 (% VO2max) at each pedaling rate also showed a quadratic curve with minimal values at about 60 or 70 rpm. VO2 at 70 rpm (84.0 (SD 5.0) % VO2max) was significantly smaller than those values at 80 rpm (86.3 (SD 3.5) % VO2max), 90 rpm (87.4 (SD 3.8) % VO2max), and 100 rpm (90.1 (SD 3.8) % VO2max). These data strongly suggest that the optimal pedaling rate estimated from neuromuscular fatigue in working muscles is not coincident with the pedaling rate at which the smallest VO2 was obtained, but with the preferred pedaling rate of the subjects. Our findings also suggest that the reason that cyclists prefer a higher pedaling rate is closely related to the development of neuromuscular fatigue in the working muscles.

Bul1, a new protein that binds to the Rsp5 ubiquitin ligase in Saccharomyces cerevisiae.

We characterized a temperature-sensitive mutant of Saccharomyces cerevisiae in which a mini-chromosome was unstable at a high temperature and cloned a new gene which encodes a basic and hydrophilic protein (110 kDa). The disruption of this gene caused the same temperature-sensitive growth as the original mutation. By using the two-hybrid system, we further isolated RSP5 (reverses Spt- phenotype), which encodes a hect (homologous to E6-AP C terminus) domain, as a gene encoding a ubiquitin ligase. Thus, we named our gene BUL1 (for a protein that binds to the ubiquitin ligase). BUL1 seems to be involved in the ubiquitination pathway, since a high dose of UBI1, encoding a ubiquitin, partially suppressed the temperature sensitivity of the bul1 disruptant as well as that of a rsp5 mutant. Coexpression of RSP5 and BUL1 on a multicopy plasmid was toxic for mitotic growth of the wild-type cells. Pulse-chase experiments revealed that Bul1 in the wild-type cells remained stable, while the bands of Bul1 in the rsp5 cells were hardly detected. Since the steady-state levels of the protein were the same in the two strains as determined by immunoblotting analysis, Bul1 might be easily degraded during immunoprecipitation in the absence of intact Rsp5. Furthermore, both Bul1 and Rsp5 appeared to be associated with large complexes which were separated through a sucrose gradient centrifugation, and Rsp5 was coimmunoprecipitated with Bul1. We discuss the possibility that Bul1 functions together with Rsp5 in protein ubiquitination.

Preparation and antirheumatic activity of optically active 2-acetylthiomethyl-4-(4-methylphenyl)-4-oxobutanoic acid (KE-298).

2-Acetylthiomethyl-4-(4-methylphenyl)-4-oxobutanonic acid (KE-298) is an antirheumatic agent. To elucidate the effects of optically active KE-298, we resolved the racemic acid and obtained the two optical isomers. (+)-KE-298 was converted to the 4-bromobenzyl ester derivative and the absolute structure was confirmed as (S) by X-ray crystallographic analysis. The pharmacological activities of the optical isomers and racemic KE-298 were compared by using the characteristic tests for KE-298. Though (+)-KE-298 showed a stronger suppressive effect on rat adjuvant arthritis than (-)-KE-298, no difference between the two isomers was detected in in vitro tests (enhancing effect on lymphocyte transformation, IL-1 antagonistic effect).

Serial electroencephalograms and somatosensory evoked potentials in a patient with isolated adrenocorticotropic hormone deficiency.

Serial electroencephalograms (EEGs) and somatosensory evoked potentials (SEPs) were carried out in a 36-year-old man with isolated ACTH deficiency. Before treatment with glucocorticoids, the EEGs displayed bilateral diffuse 5-6 Hz theta waves, and SEPs demonstrated a decreased amplitude. Following treatment with glucocorticoids, the EEGs showed gradual improvement; a diffuse alpha pattern was demonstrated on the 11th day after treatment and an occipital dominant alpha pattern was found on the 83rd day after treatment. After treatment, SEPs also showed improvement. The amplitude of N20 increased and N32 became clearly recognizable. The influence of glucocorticoids on EEGs and SEPs was discussed in the context of these observations.

[Chest wall reconstruction using polyester mesh].

From January 1987 through December 1994, we performed chest wall reconstruction using the polyester mesh in 15 patients with lung cancer, 11 with empyema after open drainage, 8 with chest wall tumor and 1 with radiation dermatitis and costal chondritis. Twenty five patients were resected 3 or more ribs. Chest wall defects were reconstructed with the polyester mesh covered with Gore-Tex soft tissue patch. Twenty two cases passed more than a year without signs of infection and follow-up averaged 27.6 months. Polyester mesh was removed due to bronchial fistula (3 cases), deformities (3 cases) and abscess formation (1 case). In these cases, polyester mesh was well incorporated and had no foreign body change. In conclusion, the polyester mesh seems to be a dependable prosthetic material for chest wall reconstruction.

[Correlation between an enzyme (glutathione S-transferase-pi) and effects of chemotherapy (cisplatin + etoposide)].

OBJECTIVES: We studied the correlation between cisplatin and etoposide chemotherapy and GST-pi staining in patients with untreated primary non-small cell lung cancer. METHODS: Of patients diagnosed as having primary lung cancer, 27 patients (26 men, 1 woman; median age 74.9 years) with stage III b or over were studied. Histological types included squamous cell carcinoma, adenocarcinoma, and large cell carcinoma. A course of treatment consisted of cisplatin 100 mg/m2/day x one day and etoposide 100 mg/m2/day x 3 days during 28 days. After 2 courses of treatment, the therapeutic effects were judged and examined in relation to immunostaining levels obtained with anti-GST-pi antibody. RESULTS: Of patients with PR (14), 11 showed GST-pi (-) staining. CONCLUSION: There is a correlation between GST-pi staining and the effects of cisplatin and etoposide chemotherapy.

[Flow cytometric analysis of the DNA content of resected non-small cell lung cancer with reference to long-term follow-up].

We measured the cellular DNA content of paraffin-embedded tumor specimens by flow cytometry from 340 cases of resected non-small cell lung cancer, and investigated the correlation of DNA content and prognosis of these cases with long-term follow-up. These 340 cases were divided into some populations according to pathological stage, histologic type, surgical curativity and N factor, and we compared the prognosis of DNA diploidy cases and DNA aneuploidy cases in each population. DNA aneuploidy cases had a significantly less favorable prognosis than DNA diploidy cases in population of stage I adenocarcinoma, stage IIIA non-small cell lung cancer and N2 cases among stage IIIA non-small cell lung cancer, all after curative operation. But in other populations, there was no significant difference in prognosis between DNA diploidy cases and DNA aneuploidy cases. In conclusion, DNA ploidy pattern is a prognostic factor for survival in patients with stage I adenocarcinoma and N2 cases of stage IIIA non-small cell lung cancer.

[T cell lymphoma presenting as recurrent bilateral pulmonary infiltrates over five years].

A 52-year-old man presented with BOOP-like recurrent bilateral pulmonary infiltrates. In 1989 a chest X-ray film showed an infiltrative shadow in the right S6 region which disappeared after administration of prednisolone. Thereafter, bilateral patchy infiltrates recurred many times, and each time they resolved rapidly with steroid therapy. Skin eruptions on the face recurred. In October 1994 the patient underwent an open-lung biopsy of the infiltrate in the left S6 region. The pathological findings were consistent with BOOP, except for the moderate-to marked infiltration of lymphocytes. A lymphoproliferative disorder was suspected, and Southern blot analysis of the specimen revealed a rearrangement of the TCR-beta gene, which led to the diagnosis of T cell lymphoma. Ten months after the diagnosis, no recurrence of the lymphoma had been detected. In this case a gene analysis of the biopsy specimen was very useful for the diagnosis of T cell lymphoma.

Pharmacological profile of KE-758, the main metabolite of the antirheumatic agent KE-298.

KE-298(2-acetylthiomethyl-4-(4-methylphenyl)-4-oxobutanoic acid) is a newly developed antirheumatic drug which suppresses the development of arthritis in animal models and has immunomodulatory effects. KE-298 is converted into the deacetylated form, KE-758 (2-mercaptomethyl-4-(4-methylphenyl)-4-oxobutanoic acid), the main serum metabolite in humans, and circulates in peripheral blood following oral administration. The authors compared effects of KE-758 and KE-298 on inflammatory cytokine production from human peripheral blood mononuclear cells and human umbilical vein endothelial cells (HUVEC), interleukin-1 (IL-1)-induced thymocyte proliferation (LAF) activity, adjuvant-induced arthritis in rats and lymphocyte transformation test in mice. KE-758 exhibited almost the same effect as KE-298 both in vitro and in vivo. The authors consider that KE-758 is pharmacologically a metabolite form of KE-298, in vivo.

[A case of mucosa associated lymphoid tissue (MALT) lymphoma].

We examined a 63-year-old female, who was referred to our hospital for a thorough examination of abnormal findings on the X-ray which were first detected in May, 1992, and suggested a tumor. After hospital admission, a bracheobronchial lymph node biopsy was performed, and the results suggested a pseudo-lymphoma. However, because the possibility of a malignant tumor was not completely negated and the patient, as well as her family, hoped to receive surgical treatment, we performed the tumor resection. The tumor had a clear border. S6 was 3 x 3.5 x 4 cm, S8 was 2 x 2.5 x 3 cm, and evaluation according to the tumor node metastasis (TNM) classification was T1 N0 M0, i.e., Stage I. In addition to an ordinary histopathological examination, an immunohistochemical examination was conducted for CD19+, CD20+, CD22+, IgD-, CD5-, CD10-. Overall, the tumor was diagnosed as a MALT lymphoma. Investigated IgH, TCR beta, bcl-1 and bcl-2 genes did not show reconstitution. Because it is difficult to differentiate a malignant lymphoma, primarily developed in the lung, from a MALT lymphoma on a morphological examination of a tissue specimen, immunohistochemical and genetic examinations are helpful for making a diagnosis.

Long-term culture of avian embryonic cells in vitro.

The pH of the embryonic blood, one of the most important environmental factors for embryonic cells, was found to range from 8.1 to 8.5 in chick embryos until 108 h after incubation. Based on these results, the culture medium adjusted to pH 8.0 was used to culture embryonic chick and quail cells. They were easily subcultured for a long period of time at pH 8.0. This pH culture condition may have wide application for manipulating embryonic cells or tissues and establishing cell lines from avian embryos.

Filtering noncorrelated noise in impedance cardiography.

Impedance cardiography (ICG) may be altered by noises as respiration and movement artifacts, mainly during exercise. In this work, a scaled Fourier linear combiner (SFLC) event-related to the R-R interval of ECG is proposed. It estimates the deterministic component of the impedance cardiographic signal and removes the noises uncorrelated to this interval. The impedance cardiographic signal is modeled as Fourier series with the coefficients estimated by the least mean square (LMS) algorithm. Simulations have been carried out to evaluate the filter performance for different noise conditions. Moreover, the method capability to remove uncorrelated noises was also examined in physiological data obtained in rest and exercise, by synchronizing respiration and pedaling with a metronome. Analyzing the ICG power spectrum, it was concluded that the proposed filter could remove the noises that are not synchronized with heart rate.

Immunoelectron microscopic localization of one of the spore germination proteins, GerAB, in Bacillus subtilis spores.

Ultrastructural localization of GerAB, one of the proteins of Bacillus subtilis spores related to L-alanine-initiated germination, was investigated by immunoelectron microscopy with antipeptide (residues 61 to 80 of GerAB) antiserum and a colloidal gold-immunoglobulin G complex. Immunogold particles were visualized in the boundary region between the cortex and coat of dormant spores, and they were broadly dispersed into the cortex region after germination.

[Study on surfactin, a cyclic depsipeptide. I. Isolation and structure of eight surfuctin analogs produced by Bacillus natto KMD 2311].

Crude surfactin was simply prepared from the culture filtrate of Bacillus natto KMD 2311 twice by acidification of the filtrate and extraction of the precipitate with ethanol. Eight surfactin analogs were isolated from the crude surfactin by RP-HPLC and gel filtration. The structure of each analog was deduced by means of amino acid composition of the acid hydrolysate and FAB-MS measurement to be a cyclic depsipeptide containing a hydroxyfatty acid. The structure of the hydroxyfatty acid moieties was elucidated as n- or iso- or anteiso-3-hydroxyfatty acid composed of carbon number 13-16 by GC analysis and EI-MS after the methanolysis of the analogs. The amino acid sequence of the peptide portion was assigned as acyl-Glu-Leu-Leu-Val-Asp-Leu-Leu by EI-MS for eight analogs. The isolated four compounds were found to be identical with the known surfactin analogs, A1, B1, B2 and C1. Although surfactin A2 and C2 had not been isolated, their structures were deduced to be a surfactin analog. Surfactin A3 and D were novel analogs. The acyl groups of surfactin A2, A3, C2 and D were anteiso-3-hydroxytridecanoic acid, n-3-hydroxytridecanoic acid, anteiso-3-hydroxypentadecanoic acid and iso-3-hydroxyhexadecanoic acid, respectively.
