Inositol trisphosphate-mediated Ca2+ influx into Xenopus oocytes triggers Ca2+ liberation from intracellular stores.

1. Inositol 1,4,5-trisphosphate (InsP3) functions as a second messenger by liberating Ca2+ from intracellular stores and by promoting influx of extracellular Ca2+. We examined whether Ca2+ influx modulates intracellular Ca2+ liberation in Xenopus oocytes by fluorescence monitoring of cytosolic free Ca2+ together with voltage clamp recording of Ca(2+)-activated Cl- membrane currents. Sustained activation of membrane Ca2+ permeability was induced by intracellular injections of a non-metabolizable InsP3 analogue, 3-deoxy-3-fluoro-D-myo-inositol 1,4,5-trisphosphate (3-F-InsP3), and Ca2+ influx was controlled by applying step changes in membrane potential to alter the driving force for Ca2+ entry. 2. Negative-going potential steps evoked intracellular Ca2+ signals comprising two components; an initial transient peak followed by a slower rise. The initial transient grew steeply over a narrow (ca 40 mV) voltage range but then increased little with further polarization, whereas the second component showed a nearly linear voltage dependence. 3. The transient Ca2+ signal continued to rise almost unchanged when Ca2+ influx was interrupted by stepping the potential to more positive values after brief hyperpolarization. In contrast, Ca2+ levels declined monotonically when positive-going steps were applied after longer intervals during the second component of the Ca2+ signal. 4. Large Ca(2+)-dependent transient inward (T(in)) membrane currents were evoked during the rising phase of the initial Ca2+ signal, but little current was associated with the second component of the Ca2+ signal. 5. The T(in) currents evoked by hyperpolarization were mimicked at fixed clamp potential by re-admitting Ca2+ to the bathing solution, and by flash photolysis of caged Ca2+ loaded into the oocyte. 6. T(in) currents were strongly inhibited by prior release of Ca2+ from InsP3-sensitive intracellular stores, and vice versa. Experiments with paired hyperpolarizing pulses and paired photorelease of InsP3 showed that responses to both stimuli recovered with similar time courses. 7. We conclude that the transient Ca2+ signal and associated T(in) current evoked by hyperpolarization arise because Ca2+ entering the oocyte triggers regenerative release of Ca2+ from InsP3-sensitive intracellular stores. Since membrane currents evoked by liberated Ca2+ were much greater than those evoked by Ca2+ entry per se, a major function of InsP3-mediated Ca2+ entry may be to modulate the activity of intracellular Ca2+ stores.

[The changes in free radical and prostaglandin metabolism in newborn infants with asphyxia].

Blood plasma thromboxane-B2 (TXB2) and 6-keto-PGF1 alpha levels were determined by radioimmunoassay technique during the first six to sixteen hours of life in 16 newborn infants with severe asphyxia, 18 newborn infants with mild asphyxia and 27 normal term neonates. Plasma lipid peroxidation (LPO) content was measured by TBA-colour-contrast method in 15 infants with severe asphyxia, 17 infants with mild asphyxia and 24 healthy term newborn infants. The results showed that blood plasma LPO, TXB2 and 6-keto-PGF1 alpha levels in infants suffering from severe asphyxia were higher than those in infants with mild asphyxia and normal infants (P < 0.01), but no significant difference was noted between the mild asphyxia group and normal control group (P > 0.01). These suggest that production of free radicals is increased and prostaglandin metabolism is triggered in the infants with severe asphyxia, that cerebral ischemia and hypoxia caused by asphyxia contributes to the augmented production of prostaglandins and free radicals, and that accumulation of free radicals and prostaglandin enhances brain damage and the metabolism of arachidonic acid appears to be an important source of the free radicals in infants with intrauterine asphyxia.

[The influence of neonatal hyperbilirubinemia on neonatal behavior].

Two groups of neonates, 30 cases with hyperbilirubinemia (307.8 +/- 99.86 mumol/L) and 30 normal newborns (control group with bilirubin 80.37 +/- 35.74 mumol/L), were evaluated with Neonatal Behavioral Assessment Scale (NBAS) and followed up at the age of 2 months and 5 months. All cases in the two groups were evaluated with NBAS at ages of 2-3 days, 12-14 days and 26-28 days. The results showed that there were statistically significant differences between the two groups in the aspects of attention and social responsiveness, the ability to modulate state of consciousness, the motor integrity and the reflexes. We also found that while the level of bilirubin was below 205.2 mumol/L, there was no correlation between the level of bilirubin and NBAS (r = -0.095, P > 0.02), but when bilirubin was at or above the level of 205.2 mumol/L, there was significant negative correlation (r = -0.53, P < 0.001) between the two. The follow-up revealed that 9 of 23 jaundiced infants had abnormal auditory reactions or abnormal muscle tonus at the age of 2 months; 8 of the 9 infants recovered, but 1 still had abnormal muscle tonus by the age of 5 months. The study suggests that hyperbilirubinemia above the level of 205.2 mumol/L would affect neonatal behavior and the greater the severity of jaundice, the more significant the influence on neonatal behavior. The effects on some jaundiced infants may last 5 months or more. NBAS is a valuable scale for early detection of mild brain dysfunctions which are less obvious on clinical examination.

[A voltage-clamp study on voltage-gated calcium channels translated in Xenopus oocytes by rat brain mRNA].

Voltage-gated calcium channels, expressed in Xenopus oocytes after injection of rat brain mRNA, were studied by using voltage-clamp technique. The properties of the calcium channels were characterized by barium current (IBa) passed through the channels. All oocytes used in this study were taken from five identified donors. Endogenous voltage--activated barium current measured in most oocytes from these donors were not detectable, or smaller than 15 nA, mRNA was extracted from the whole brains of 10 day postnatal rats and microinjected into the oocytes. IBa increased gradually during five days after mRNA injection. The maximum amplitude of the expressed voltage-activated barium current was usually larger than one hundred of nA on the third day after mRNA injection. In comparison, the expression of voltage-activated barium current was hardly detectable in oocytes injected by mRNA extracted from brains of embryonic rats. The voltage-dependence of activation and inactivation pharmacology of IBa were studied. It was found that IBa was inhibited potently by lanthanide cations (La+3,Nd+3,Sm+3,Eu+3,Gd+3,Dy+3,Er+3) at mumol/L concentration level. L-type calcium channel ligands, nifedipine and Bay K 8644 inhibited IBa at 100 mumol/L, while another dihydropyridine ligand (+/-) nimodipine enhanced IBa at the same concentration.

[Construction of clone about major DNA fragment coding for 150kd phosphoprotein of human cytomegalovirus and its restriction site analysis].

Human cytomegalovirus (HCMV) DNA EcoRI-Y fragment, which is major DNA fragment coding for 150kd phosphoprotein (pp150) of HCMV, was separated and recovered from recombinant cosmid pCM1015. The recombinant plasmids pHCY1 and pHCY2, which contain major DNA fragment coding for pp150, were constructed successfully, using the plasmid pUC19 as vector. The recombinant plasmids were analysed by restriction endonucleases and determined by Southern hybridization. The restriction sites of DNA encoding region for immunodominant epitopes of HCMV pp150 were analysed by GENEPRO system. The region could be digested by 76 kinds of restriction endonucleases. The sizes of cleaved fragments were between 4-2667 bp.

Expression pattern of skeletal muscle troponin T isoforms is fixed in cell lineage.

The expression of fast-muscle-type troponin T isoforms in chicken skeletal muscles was studied by two-dimensional SDS-polyacrylamide gel electrophoresis and immunoblotting. According to the pattern of troponin T isoform expression, chicken fast muscle was classified into two groups: One group expressed breast-fast-muscle-type troponin T in addition to leg-fast-muscle-type troponin T, the other expressed only leg-fast-muscle-type troponin T. To the former group belong breast and wing fast muscles and some of the back fast muscles, and to the latter group belong the fast muscles in leg, abdomen, and neck. Transplantation of breast muscle into leg was performed in order to change the physical environment and to investigate the mechanism of isoform expression. Histological observation of the transplant revealed severe degeneration of muscle cells, followed by differentiation of myoblasts in which breast-muscle-type troponin T was eventually expressed. The results showed that the pattern of troponin T isoform expression is primarily fixed in the cell lineage, although nerves modulate it.

Counting knowledge and skill in cognitive addition: a comparison of normal and mathematically disabled children.

The relationship between counting knowledge and computational skills (i.e., skill at counting to solve addition problems) was assessed for groups of first-grade normal and mathematically disabled (MD) children. Twenty-four normal and 13 MD children were administered a series of counting tasks and solved 40 computer-administered addition problems. For the addition task, problem-solving strategies were recorded on a trial-by-trial basis. Performance on the counting tasks suggested that the MD children were developmentally delayed in the understanding of essential and unessential features of counting and were relatively unskilled in the detection of certain forms of counting error. On the addition task, the MD children committed many more computational errors and tended to use developmentally immature counting procedures. The immature counting knowledge of the MD children, combined with their relatively poor skills at detecting counting errors, appeared to underlie their poor computational skills on the addition task. Suggestions for future research are presented.

Potentiation of inositol trisphosphate-induced Ca2+ mobilization in Xenopus oocytes by cytosolic Ca2+.

1. The ability of cytosolic Ca2+ ions to modulate inositol 1,4,5-trisphosphate (Insp3)-induced Ca2+ liberation from intracellular stores was studied in Xenopus oocytes using light flash photolysis of caged InsP3. Changes in cytosolic free Ca2+ level were effected by inducing Ca2+ entry through ionophore and voltage-gated plasma membrane channels and by injection of Ca2+ through a micropipette. Their effects on Ca2+ liberation were monitored by video imaging of Fluo-3 fluorescence and by voltage clamp recording of Ca(2+)-activated membrane Cl- currents. 2. Treatment of oocytes with the Ca2+ ionophores A23187 and ionomycin caused a transient elevation of cytosolic Ca2+ level when cells were bathed in Ca(2+)-free solution, which probably arose because of release of Ca2+ from intracellular stores. 3. Membrane current and Fluo-3 Ca2+ signals evoked by photoreleased InsP3 in ionophore-treated oocytes were potentiated when the intracellular Ca2+ level was elevated by raising the Ca2+ level in the bathing solution. 4. Responses to photoreleased InsP3 were similarly potentiated following activation of Ca2+ entry through voltage-gated Ca2+ channels expressed in the plasma membrane. 5. Ca(2+)-activated membrane currents evoked by depolarization developed a delayed 'hump' component during sustained photorelease of InsP3, probably because Ca2+ ions entering through the membrane channels triggered liberation of Ca2+ from intracellular stores. 6. Ba2+ and Sr2+ ions were able to substitute for Ca2+ in potentiating InsP3-mediated Ca2+ liberation. 7. Gradual photorelease of InsP3 by weak photolysis light evoked Ca2+ liberation that began at particular foci and then propagated throughout, but not beyond that area of the oocyte exposed to the light. Local elevations of intracellular Ca2+ produced by microinjection of Ca2+ acted as new foci for the initiation of Ca2+ liberation by InsP3. 8. In resting oocytes, intracellular injections of Ca2+ resulted only in localized elevation of intracellular Ca2+, and did not evoke propagating waves. 9. The results show that cytosolic Ca2+ ions potentiate the ability of InsP3 to liberate Ca2+ from intracellular stores. This process may be important for the positive feedback mechanism underlying the generation of Ca2+ spikes and waves, and for interactions between the InsP3 pathway and Ca2+ ions entering cells through voltage- and ligand-gated channels.

The role of endotoxin in the pathogenesis of experimental multiple system organ failure: a preliminary report.

In this study, an animal model of multiple system organ failure (MSOF) in rabbits, engendered by feeding E. coli prior to severe hemorrhagic shock, was used for the purpose of investigating 1) the relationship between lipopolysaccharide (LPS) and MSOF, and 2) the effectiveness of Re-LPS antiserum in preventing MSOF. The results showed that endotoxemia occurred very early, and its degree correlated well with that of organ dysfunction. Re-LPS antiserum administration abated the toxic effects and lowered the incidence of MSOF. These results suggest that sequential analysis of circulating LPS levels may be useful for the early diagnosis of MSOF, and that gut-derived endotoxin might play an important role in the pathogenesis of experimental MSOF.

[A voltage-dependent potassium channel of outward rectifier type in plasma membrane of oocyte from toad, Bufo bufo gargarizans].

Membrane properties of the fully-grown oocytes from toad, Bufo bufo gargarizans, were studied by using voltage-clamp technique. It was found that a sustained outward current was elicited by membrane depolarization to -30 mV or more positive value. The increase of the current was nearly proportional to the degree of depolarization. The peak value of the current ranged 2-5 microA at a membrane potential of 20 mV in oocytes from different toads. The current was inhibited by antagonists of potassium channel, TEA and 4-AP. The concentration of TEA capable of inhibiting half of the current was 2.6 mmol/L. Chloride channel antagonist 9-AC (2.5 mmol/L) had no effect on the current. Triple the extracellular calcium concentration did not show any effect either. The reversal potential of the current varied with an increase of 47.3 mV per decade change of the extracellular potassium concentration. Changing extracellular concentration of sodium or chloride did not shift the reversal potential. It was concluded that the outward current was a voltage-activated potassium current. The voltage-dependent potassium current decreased after treatment of the oocytes with progesterone to a state of maturation. A large decrease of the current (to about 1/20 of the control) occurred to the oocytes obtained from hibernating toads while a less striking decrease of the current (to about 1/3 of the control) was observed in the oocytes from toads all year round reared at 25-30 degrees C.

Prevalence of vitamin B12 deficiency among geriatric outpatients.

BACKGROUND: Healthy people can have low levels of cobalamin (vitamin B12) without symptoms or signs of cobalamin deficiency. Early detection of deficiency is imperative for treatment to be effective. Development of radioimmunoassay tests has greatly improved accurate determination of cobalamin (Cbl) levels. Nevertheless, results of studies of Cbl deficiency vary widely because of the variety of populations studied. METHODS: In a prospective study, we tested 100 consecutive, unselected geriatric outpatients in a primary care setting to determine the prevalence of cobalamin deficiency. All patients, 65 years of age or older, who visited the office of one of the authors during a period of 11 consecutive working days, had their serum Cbl level checked. If the level was 299 pg/mL or lower, serum intrinsic factor and parietal cell antibodies, serum gastrin, part 1 Schilling test, serum methylmalonic acid, and total homocysteine were done, when possible, for the diagnosis of type A gastritis and intracellular Cbl deficiency. RESULTS: Sixteen percent of geriatric outpatients had serum Cbl levels of 200 pg/mL or below, and 21% had levels between 201 and 299 pg/mL. Among the 16 patients with levels < or = 200 pg/mL, 2 patients had macrocytic anemia, 3 patients had peripheral neuropathy, and 8 patients had type A gastritis. Among the 21 patients with levels of 201 to 299 pg/mL, 2 patients had peripheral neuropathy, 9 patients had type A gastritis, and none of the patients had macrocytic anemia. Among the patients whose methylmalonic acid and total homocysteine levels were determined, the results were high in 80% of those with Cbl levels < or = 200 pg/mL and in 33% of those with levels from 201 to 299 pg/mL. CONCLUSIONS: The prevalence of Cbl deficiency in geriatric outpatients was found to be higher than in any recent report. The lower limit of the normal range for Cbl level should be increased to 300 pg/mL.

[The expression of kainate and GABA receptors in oocytes of Bufo bufo gargarizans].

Oocytes of Bufo bufo gargarizans were used as an expression system for analyzing structure of exogenous membrane protein and their functions. Poly (A)+ mRNA isolated from rat brain was injected into Toad (Bufo bufo gargarizans) oocytes (50 ng/oocyte). Rat brain kainic acid and gamma-aminobutyric acid(GABA) receptors expressed by the injected mRNA were integrated in the oocyte membrane 48 h after the injection at 19 degrees C. Membrane currents induced by kainic acid (5 x 10(-5) mol/L) and GABA (10(-4) mol/L) were 294.0 +/- 6.4 nA (n = 5) and 309.5 +/- 4.9 nA (n = 4) respectively. The kainic acid induced current reached its maximum value at about 10(-3) mol/L. Moreover, it was observed that the 36Cl- influx of the oocytes microinjected with mRNA was one-fold more rapid than the control oocytes. These results indicate that the oocytes of Bufo bufo gargarizans as those of Xenopus laevis can be used to express membrane proteins (receptors & transports) to acquire their proper functions from exogenous mRNA.

Regenerative release of calcium from functionally discrete subcellular stores by inositol trisphosphate.

Fluorescence imaging was used to determine the spatial and temporal patterns of subcellular calcium (Ca2+) liberation induced in Xenopus oocytes by photorelease of inositol 1,4,5-trisphosphate (InsP3) from a caged precursor. Increasing levels of InsP3 evoked Ca2+ release that began in a graded manner but, at varying threshold levels of InsP3, localized sites then showed transient and asynchronous 'puffs' of Ca2+ release. With higher levels of InsP3, Ca2+ from adjacent sites formed a focus for initiation of a propagating Ca2+ wave. The results show that InsP3-sensitive Ca2+ stores are arranged as distinct and functionally independent units, and that Ca2+ is released in both graded and regenerative fashions.

[Fungal toxin dihydroxyaflavinine inhibits non-competitively GABAA receptor channel expressed in Xenopus oocytes].

Dihydroxyaflavinine is an indole-derived metabolite of Aspergillus flavus. Its action on GABA-induced response was quantitatively studied on the GABAA receptor expressed in Xenopus oocytes after injection of chick brain mRNA under voltage-clamp conditions. Dihydroxyaflavinine inhibits GABA-induced current non-competitively with KI = 12 mumol/L. This blockage is rapidly reversible. In comparison, the inhibitory effect of penicillin on GABAA receptor is enhanced by increasing GABA concentration. Ro 15-1788 (a benzodiazepine ligand with KD = 0.6--2 nmol/L) of concentration as high as 1 mumol/L, does not mask the action of 10 mumol/L dihydroxyaflavinine, indicating that dihydroxyaflavinine acts on a site different from benzodiazepines. Dihydroxyaflavinine appears to expedite desensitization of the receptor, which is similar to the action of picrotoxin and in contrast with that of penicillin and bicuculline.

[A case of ruptured distal arch aneurysm associated with postoperative unilateral respiratory disorders].

This paper describes a patient with ruptured distal arch aneurysm associated with postoperative unilateral respiratory disorders whose life was able to be saved by surgery. The patient was a 68-year old male who underwent emergency surgery due to sealed rupture of distal arch aneurysm while preparing for selective surgery. Artificial graft replacement of the distal arch was carried out under centrifugal pump bypass. Respiration was controlled using differential lung ventilation (DLV) techniques. This was done because weaning from the respirator was difficult due to decreases in lung-thorax compliance on the opened chest side after surgery. Weaning from the respirator was achieved on the 10th day of illness after surgery. Postoperative prognosis of ruptured thoracic aneurysm is extremely poor, with high mortality being attributable to the fact that most patients are elderly and the incidence of respiratory and cardiovascular complications is very high. In addition, problems are often encountered in postoperative respiration management in patients with thoracic aneurysm who need open chest surgery. DLV seems to be a useful support in such patients who have underlying respiratory disorders.
