Analysis of dystrophin in muscular diseases by two-dimensional gel electrophoresis using agarose gels in the first dimension.

We analyzed dystrophin in case of normal control, Duchenne muscular dystrophy (DMD), Becker muscular dystrophy (BMD) and infectious muscular disease using two-dimensional gel electrophoresis and immunoblotting with 3 monoclonal dystrophin antibodies: Dys 1, a mid-rod-domain antibody; Dys 2, a C-terminal-domain antibody; and Dys 3, an N-terminal-domain antibody. In cases of normal control, a clearly separated doublet of bands was observed for Dys 1 and 3 at molecular weights 400 and 420 kDa. The isoelectric point was between pH approximately 5.7-approximately 5.9, similar to that for the myosin heavy chain. In one DMD case, a single faint band was observed for Dys 2. BMD presented a single-band pattern for each antibody. Infectious diseases cases showed 3- to 5-band patterns for Dys 1 and single or no bands for Dys 2 and 3. The pI of the Dys 1 band was almost identical. These results suggest coexistence of normal dystrophin and its proteolytic products, both containing triple helical segment, and show that two-dimensional gel electrophoresis may be applicable in the analysis of dystrophin in muscular disease.

Scatter factor binds to thrombospondin and other extracellular matrix components.

Scatter factor (SF) is an angiogenic growth factor that stimulates motility and invasion of carcinoma cells. SF is present in the extracellular matrix (ECM) of breast cancers, where it might act to promote tumor cell invasion and angiogenesis. To investigate how SF is incorporated into the ECM, we studied the binding of SF to various ECM components using a solid-phase binding assay based on the SF enzyme-linked immunosorbent assay. We found that SF binds to a variety of ECM molecules, with different binding capacities. The highest SF binding capacities were observed for thrombospondin-1 (TSP-1), fibronectin (Fn), and heparan sulfate proteoglycan, although SF did not bind to albumin. Mature two-chain SF and precursor single-chain SF bound approximately equally well to TSP-1 and Fn. Moreover, two SF alpha-chain peptides (NK1 and NK2) both bound to TSP-1 and Fn, suggesting that the whole SF molecule is not required for binding. Based on binding competition assays, TSP-1 exhibited higher affinity for SF than did nine other ECM molecules, including Fn and heparan sulfate proteoglycan. Although heparin in solution potently inhibited the binding of SF to TSP-1-coated surfaces, even very high concentrations of heparin could not elute SF already bound to TSP-1. SF binding was modulated by binding interactions among ECM molecules (TSP-1-Fn, TSP-1-collagen I, and Fn-collagen I), suggesting that the matrix capacity to bind SF depends upon its exact composition. SF bound in a dose-dependent fashion to ECMs secreted by three human breast carcinoma cell lines. Binding of SF to matrices from all three cell lines was significantly inhibited by preincubation of the matrices with antibodies against TSP-1, whereas antibodies against several other ECM components were less effective or ineffective in inhibiting SF binding. In addition, TSP-1 markedly inhibited chemotaxis of microvascular endothelial cells toward SF and SF-induced angiogenesis in the rat cornea neovascularization assay. Our findings suggest that 1) SF interacts with a variety of ECM components, 2) high affinity SF-TSP-1 interactions may mediate the binding of SF to the breast cancer matrix, and 3) the SF-TSP-1 interaction may contribute to modulation of angiogenesis. Possible implications of these findings for tumor angiogenesis are discussed.

Scatter factor protein levels in human breast cancers: clinicopathological and biological correlations.

Scatter factor (SF) is an invasogenic and angiogenic cytokine the cellular receptor of which is encoded by a proto-oncogene (c-met). We measured the immunoreactive SF content (nanograms of SF per milligram of protein) in tissue extracts from 166 breast cancers and correlated the values with various known prognostic parameters. Invasive cancers had nearly four times greater SF content than did ductal carcinoma in situ, and the difference was statistically significant (P < 0.02, two-tailed t-test). However, there were no significant differences in SF content among different histological types of invasive cancer. Invasive cancers that had spread to axillary lymph nodes exhibited higher SF content than did invasive cancers without regional spread (P < 0.02), but the difference in SF content between node-positive and node-negative tumors was not as great as that between invasive and ductal carcinoma in situ tumors. There was a trend toward increased SF content in larger primary tumors as compared with smaller tumors, but statistical comparison revealed borderline significance (0.05 < P < 1.0). There was no significant correlation between SF content and other parameters, including estrogen receptor, progesterone receptor, DNA ploidy, S phase, or Scarff-Bloom-Richardson score. We also measured the content of von Willebrand factor (a marker of blood vessels) and interleukin-1 beta (a pro-inflammatory cytokine) in the same tumor extracts. SF content showed a strong positive correlation with von Willebrand factor content (P < 0.001) but did not appear to be correlated with interleukin-1 beta. These findings suggest that SF is correlated with several other clinicopathological indicators of aggressive tumor behavior, consistent with the hypothesis that SF is a biological factor that may play a role in breast cancer pathogenesis.

Distribution of vagal preganglionic neurons in the rat brain innervating thoracic and abdominal organs revealed by retrograde DiI tracing.

To investigate the distribution and number of preganglionic neurons which regulate motility and secretion in thoracic and abdominal organs in the vagal parasympathetic nervous system, the neuronal tracer DiI was injected into the organs and the distribution of retrogradely labeled neurons was examined in the rat brainstem. The stomach received the vast majority of efferent projections from the dorsal motor nucleus of the vagus nerve (DMV). The cecum and the duodenum also received projections from the DMV, but they originated from a smaller number of preganglionic neurons. Preganglionic neurons projecting to the stomach occupied the middle part of the DMV, those projecting to the cecum occupied the lateral part of the DMV, and those projecting to the duodenum were found in the medial edge of the DMV. The ventral and dorsal sides of the stomach wall were innervated by the left and right vagus nerves, respectively. However, immediately after passing the boundary between the stomach and duodenum, the left and right vagal nerve fibers mixed in the ventral and dorsal walls of the distal gastrointestinal tract. The nucleus ambiguous is a mixture of parasympathetic preganglionic neurons and motoneurons. In this study, we revealed that the major targets of these preganglionic neurons were the lungs and other thoracic organs.

[Immunohistochemical detection of antigen of human cytomegalovirus in hepatocyte].

The expression plasmid of human cytomegalovirus (HCMV) 52kd glycoprotein was induced and amplified, and the recombinant protein was extracted. The rabbit antiserum to the recombinant protein was prepared. The antiserum-reacted to the antigen of HCMV AD165, and the titer of antiserum was 1 : 3200 by using 125I-Protein A. Using Avidin-Biotinperoxidase complex technique (ABC method), we detected the HCMV 52 kd glycoprotein in the hapatic slides of 9 cases. Among them, one case (an infant died of congenital heart disease) was held as a negative control. The other 8 cases (patients with clinically diagnosed cytomegalic inclusion disease) had viral hepatitis histopathologically. Of these 8 cases, 7 had positive signals but found in the nucleus and cytomembrane of the hepatocytes, no positive signal was found in the hepatocytes of the control. The result showed that HCMV 52kd glycoprotein was located in the nucleus and on the membrane of the infected hepatocyte. The antibody prepared with the antigen of gene engineering could find the virus antigen and represent the virus gene state which was active or silent. ABC method is not only simple, sensitive and specific, but also effective in localization. It is of importance to clinical diagnosis, differential diagnosis and prognostication.

[Dexamethasone's effect on glutamate receptor in experimental hypoxic-ischemic cerebral injury].

In order to probe into dexamethasone's effect on glutamate receptor (Glu R) in hypoxic-ischemic encephalopathy (HIE) of newborn, we established newborn pigs' model of HIE to investigate Glu R in their forebrain crude synaptic membrane and the effect of dexamethasone in different HIE times and different dosages on the changes of Glu R. This study included five groups: HIE group (no medication); dexamethasone treatment group (10 mg/kg before HIE); dexamethasone treatment group (10 mg/kg after HIE); dexamethasone treatment group (5 mg/kg before HIE); dexamethasone treatment group (5 mg/kg after HIE). The results showed that the mean receptor density (Bmax) of dexamethasone treatment group (10 mg/kg before HIE) was significantly lower than that of the other groups (P < 0.05), but there was no statistic difference in the affinity of Glu R between all the experimental groups. These findings suggested that the preventive use of dexamethasone in large dosage would reduce the binding of Glu, and could possibly protect the brain tissues from damage in HIE.

[Restriction fragment length polymorphism assays of clinical isolated human cytomegalovirus strain genomic DNA].

Ten clinical isolated strains of Human cytomegalovirus (HCMV) were obtained from 73 urine specimens of different people. Isolations from urine were carried out in human embryolung fibroblasts. Viral isolates were passaged four times. HCMV DNAs of laboratory strain AD169 and 10 clinical isolated strains were extracted with Hirt method, digested with each of the restriction enzymes EcoRI, Hind II. Comparison of restriction fragment length polymorphism (RFLP) of AD169 and isolated strains were made by hybridizing digested DNA with 32P labeled with HCMV Hind II cloned subgenomic fragments as the probe (pCM1035, pCM1015). pCM1035 is located in the joining region between the long(L) and short (S) unique sequences of the virus (L-S junction) pCM1015 is located in the terminal sequences of the virus. The results showed the genomic high degree of homology existed among all strains and the variable restriction site was in the L-S junction and terminal portion. The RFLP patterns of the clinical isolates which did not have relation in epidemiology were different, but the patterns of clinical isolates related in epidemiology were quite similar. Polymorphism frequently occurred in this case of EcoRI digested fragment hybridized with the probe of pCM1035. Southern hybridization of HCMV isolations is useful to researches into the molecular epidemiology and pathogenesis of HCMV infection.

[The treated effects of nimodipine on calcium of erythrocyte in experimental hypoxic-ischemic encephalopathy].

An animal model of hypoxic-ischemic encephalopathy (HIE) in newborn pigs was set up to study the histopathological changes of brain injury by light- and electron-microscopy, the changes in total calcium content and calcium iron of RBC, and the effects of two different doses of Nimodipine on HIE. Sixty-one newborn pigs were randomly divided into seven groups, the normal control group (n = 9); the HIE 1-hour group (n = 8) and 24-hour group (n = 8); HIE+ Nimodipine (1 microgram/kg.min-1) 1-hour group (n = 8) and 24-hour group (n = 8). The results showed that these were apparent brain edema, hematencephalon, neuron necrosis, and intracellular body degeneration in the HIE 1-hour group. The levels of RBC TCa and RBC Cai2+ in the HIE 1-hour group were significantly higher than those in the normal control group (P < 0.01). The brain tissues in all groups treated with Nimodipine showed histopathological betterment Compared with the HIE groups. The levels of RBC TCa and RBC Cai2+ in the Nimodipine treatment groups were significantly lower than those in HIE groups (P < 0.01) and were close to the levels in the normal control group (P < 0.05). The authors suggested that early application of Nimodipine in case of postnatal asphyxia might protect the newborn from severe hypoxicischemic brain lesions.

[The postoperative effects of tapered terminal ileal continent urinary reservoir].

We report postoperative effects of 23 cases having tapered terminal ileal continent urinary reservoir is reported. The operative technique was characterized by tapering the terminal ileum with stapler to form a continent efferent tract, segments of intestine were cut open and reformed a pouch, and the ureter implantation was performed by means of inserting the end of ureters into the pouch. Ileocolonic pouch wes constructed for 14 patients and colonic pouch for 9 patients. 22 patients were followed up for 1 to 6 years except one died of cerebral hemorrhage 2 weeks after operation. Urine continence were achieved in all patients and self catheterization could be easily undertaken with 20F-22F catheter in 21. Difficulty in catheterization was happened in one early case. One year after operation, the maximum volume of the ileocolonic pouch was 900 ml-1000 ml. The basal and intermittent pressure of the filling pouch was 0.49 kPa-1.96 kPa respectively. The maximum volume of the colonic pouch was 400 ml-500 ml. The basal and intermittent pressure of the filling pouch of the colonic pouch was 0.98 kPa-1.9 6 kPa and 1.96 kPa-3.93 kPa respectively. The complication of pouch overdistention was found in 6 cases, pouch stone formation in 2 and pouch infection in 12 episodes. There was no evidence of ureteral obstruction or reflux and significant hyperchloric acidosis. It is recognized that tapered terminal ileum is a good way to construct a continent efferent tract, both ileocolonic pouch and colonic pouch can meet the need of low inner pouch pressure so long as the intestines were cut open and reformed, and efforts must be made to prevent the complications of pouch overdistension, stone formation and pouch infection.

Quantal puffs of intracellular Ca2+ evoked by inositol trisphosphate in Xenopus oocytes.

1. Ca2+ liberation induced in Xenopus oocytes by a poorly metabolized derivative of inositol 1,4,5-trisphosphate (3-deoxy-3-fluoro-D-myo-inositol 1,4,5-trisphosphate; 3-F-InsP3) was visualized using a video-rate confocal microscope to image fluorescence signals reported by the indicator dye calcium green-1. 2. Low (10-30 nM) intracellular concentrations of 3-F-InsP3 evoked Ca2+ release as localized transient 'puffs'. Progressively higher concentrations (30-60 nM) gave rise to abortive Ca2+ waves triggered by puffs, and then (> 60 nM) to a sustained elevation of Ca2+ followed by the appearance of propagating Ca2+ waves. At concentrations up to that giving waves, the frequency of puffs increased as about the third power of [InsP3], whereas their amplitudes increased only slightly. 3. The rise of cytosolic Ca2+ during a puff began abruptly, and peaked within about 50 ms. The peak free Ca2+ level was about 180 nM, and the total amount of Ca2+ liberated was several attomoles (10(-18) mol), too much to be accounted for by opening of a single InsP3-gated channel. The subsequent decline of Ca2+ occurred over a few hundred milliseconds, determined largely by diffusion of Ca2+ away from the release site, rather than by resequestration. Lateral spread of Ca2+ was restricted to a few micrometres, consistent with an effective diffusion coefficient for Ca2+ ions of about 27 microns2 s-1. 4. The peak amplitudes of puffs recorded at a given site were distributed in a roughly Gaussian manner, and a small proportion of sites consistently gave puffs much larger than the main population. Intervals between successive puffs at a single site were exponentially distributed, except for a progressive fall-off in puffs seen at intervals shorter than about 10 s. Thus, triggering of puffs appeared to be stochastically determined after recovery from a refractory period. 5. There was little correlation between the occurrence of puffs at sites more than a few micrometres apart, indicating that puff sites can function autonomously, but closely (ca 2 microns) adjacent sites showed highly correlated behaviour. 6. Puffs arose from sites-present at a density of about 1 per 30 microns2 in the animal hemisphere, located within a narrow band about 5-7 microns below the plasma membrane. 7. We conclude that Ca2+ puffs represent a 'quantal' unit of InsP3-evoked Ca2+ liberation, which may arise because local regenerative feedback by cytosolic Ca2+ ions causes the concerted opening of several closely clustered InsP3 receptor channels.

Calcium puffs in Xenopus oocytes.

The second messenger inositol 1,4,5-trisphosphate (InsP3) functions in large part by liberating calcium ions from intracellular stores. This release process is highly non-linear and shows a regenerative characteristic that allows production of all-or-none calcium spikes which propagate as waves. However, at low concentrations of InsP3 an additional mode of calcium liberation is seen in Xenopus oocytes, transient 'puffs' of cytosolic calcium that last for a few hundred milliseconds and are restricted to within a few micrometres. Puffs are generally of similar size and the amount of calcium released (about 3 x 10(-18) mol) suggests that they arise through the concerted opening of several InsP3-gated calcium release channels. Puff sites are present at a density of about one per 30 microns 2 in the animal hemisphere of the oocyte. Each site functions autonomously, producing puffs at largely random intervals. We conclude that calcium puffs represent 'quantal' units of InsP3-evoked calcium liberation, which may result from local regenerative feedback by cytosolic calcium ions at functionally discrete release sites.

Acute administration of alpha-methyl-para-tyrosine alters levels of norepinephrine transporter mRNA in the rat brainstem.

This study investigated whether rat norepinephrine transporter (NET) mRNA levels would be altered by alpha-methyl-p-tyrosine (alpha-MPT), a tyrosine hydroxylase inhibitor. While NE levels decreased at 1 and 3 days but recovered at 7 days after alpha-MPT, NET mRNA levels decreased at 3 and 7 days but not at 1 day after alpha-MPT. The results indicate that acute treatment with alpha-MPT led to a delayed time response in its effects on NET mRNA and NE levels in the rat brain.

Dioxin activates HIV-1 gene expression by an oxidative stress pathway requiring a functional cytochrome P450 CYP1A1 enzyme.

We have studied the effect of several environmental chemicals on the transient expression of a chloramphenicol acetyltransferase (cat) reporter gene linked to the promoter sequences in the long terminal repeat (LTR) of the human immunodeficiency virus type 1 (HIV-1). Aflatoxin B1, 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD; dioxin) and benzo[a]pyrene cause a significant increases in CAT expression in mouse hepatoma Hepa-1 cells. The induction of CAT after TCDD treatment is abolished by administration of N-acetyl-L-cysteine or 2-mercaptoethanol and does not take place in a mutant cell line that lacks CYP1A1 enzymatic activity. Linker-scanning mutational analysis of transcription factor binding sites in the promoter revealed that both the NF kappa B and an adjacent aromatic hydrocarbon response element (AhRE) are required for TCDD-dependent CAT expression. In addition, mutation of the NFAT/AP-1 binding sites in the negative regulatory region of the promoter increases the magnitude of the TCDD effect. We conclude that induction of a functional CYP1A1 monooxygenase by TCDD stimulates a pathway that generates thiol-sensitive reactive oxygen intermediates which, in turn, are responsible for the TCDD-dependent activation of genes linked to the LTR. These data might provide an explanation for findings that TCDD increases infectious HIV-1 titers in experimental systems and for epidemiologic reports suggesting that exposure to aromatic hydrocarbons, such as found in cigarette smoke, is associated with an acceleration in AIDS progression.

Therapeutic effects of the blood-activating and stasis-reducing method in 80 cases of coronary heart disease.

The pathogenesis and treatment of coronary heart disease (CHD) may vary, whereas the author believes that the disease is due to blood stasis on a fundamental basis of deficiency, and the principle of treatment is activation of blood. Since stasis may lead to deficiency and vice versa, CHD cannot be cured without removal of stasis, and stasis cannot be completely removed without proper correction of deficiency. The basic method for the treatment of CHD is therefore activation of blood for reduction of stasis. At the same time, according to the conditions of individual patients, the methods of replenishing Qi, warming Yang, nourishing Yin, reducing phlegm and subduing Yang should be applied simultaneously to resolve the problem of deficiency. The author treated 80 cases of CHD by the above rationale and found the results satisfactory, i.e., 28 cases (35%) showed marked symptomatic relief, 46 (57.5%) showed symptomatic improvement, while 6 cases (7.5%) failed. The total effective rate was 92.5%. ECG examination indicated 9 cases markedly effective, 27 improved, while 44 cases failed, for a total effective rate of 45%. Blood pressure and blood lipids also showed significant drops (P < 0.001).

Evaluation of a tubular mesh and a spiral stent woven from titanium nickel alloy for prostatic obstruction in patients at high risk for surgery. Report of 62 cases.

The use of two urethral stents woven from titanium nickel alloy (TiNi) in the form of a tubular mesh or a spiral stent (both made in China) is described. They were implanted in 62 patients with prostatic outflow obstruction. All patients were considered contraindicated for surgery. They were divided into a spiral stent group (group 1) treated between March 1992 and May 1993, comprising 35 cases, and a tubular mesh group (group 2) treated between October 1993 and December 1994, comprising 31 cases, including four failures in group 1. Thirty-three out of the 35 patients in group 1 were treated successfully. Good results were achieved in eight cases (22.8%) and significant improvements occurred in 24 (68.5%), giving a total effective rate of 91.3%, with a follow-up of 11 to 27 months. Fourteen stents were removed within 6 months after the insertion and six more were removed within 12 months. The mean effective time of the stent in situ was 10.8 months. All 31 cases in the mesh group were treated successfully. Dramatically good effects were obtained in 28 cases (over 90%) and distinct improvements were achieved in two, giving a total effective rate of over 96.5%, with a follow-up of 6 to 14 months (mean 10.5 months). Cystoscopy was carried out in 12 patients after 6 months following the insertion. The major part of the meshes became covered by urothelium. Compared with the spiral, the mesh makes it possible to insert a prosthesis with a larger diameter and anti-pressure. The spiral, however, can be used as a temporary alternative for the relief of prostatic obstruction. A tubular mesh can work well for the relief of prostatic obstruction and remain in situ without causing major problems for at least 1 year.
