[Study on invasibility and multiplication of Toxoplasma strains with different virulence toward Vero-cells].

AIM: To investigate the pathogenic mechanisms of Toxoplasma strains with different virulence. METHODS: Tachyzoites from 3 strains, Viz. RH, B36 and Fukaya strains, were challenged to in vitro cultivated Vero-cells. Systematic examinations on the earliest invasion time, the invasion rate and intracellular multiplication were performed under different cultivation conditions. RESULTS: The tachyzoites of all the 3 strains invaded the host cells within a short period after inoculation. Invasion rates were all increased along with the prolonged duration of infection. The intracellular multiplication was found to be most active in RH strain, moderate in B36, and comparatively slow in Fukaya strains. Using purified tachyzoites freed from host debris and proteins and adding sufficient FCS in the medium may facilitate the invasion and subsequent multiplication of the parasite. CONCLUSION: Strain differences in pathogenicity to the host may be correlated to the genetically predetermined multiplication capabilities of the parasites after being invaded to the host cell, and that environmental factors may give certain impact on the invasibility of the parasite.

[Correlation between the dose and the anti-Toxoplasma effect of activated mouse macrophages induced by IFN-gamma and the synergism between IFN-gamma and TNF-alpha].

AIM: To find out the dose dependence of the anti-Toxoplasma effect induced by IFN-gamma and to determine the possible synergistic activity between TNF-alpha and IFN-gamma. METHODS: The in vitro effect of cultivated mouse peritoneal macrophages activated by IFN-gamma alone or IFN-gamma combined with different doses of TNF-alpha on the intracellular tachyzoites of RH strains and the nitric oxide (NO) level in the culture medium supernatant were simultaneously determined. RESULTS: With the increase in the dose of IFN-gamma, the anti-Toxoplasma effect was augmented and the NO level was enhanced. At 24 hours after tachyzoite invasion, a significant reversed correlation was demonstrated between the NO level and the number of intracellular parasites. CONCLUSION: The anti-Toxoplasma effect of macrophages activated by IFN-gamma appears to be dose-dependent and TNF-alpha acts synergitically with IFN-gamma in the activation of macrophages. The production of reactive NO could be an important effector in the IFN-gamma primed anti-Toxoplasma action.

[Studies on characterization of glutamine synthetase from Streptomyces lincolnensis].

Glutamine synthetase from Streptomyces lincolnensis was purified to electrophoretic homogeneity. The effects of pH, temperature and metal ions on the purified enzyme were studied, and the substrate specificity of glutamine synthetase were reported. Under the standard biosynthetic assay system, the substrate saturation curves of glutamine synthetase from Streptomyces lincolnensis for ammonia did not agree with the Michaelis-Menten relationship, with either Mn2+ or Mg2+ as divalent cation, and Hill constants not equal to 1. The K(m) values of GS for Glu and ATP were different with different divalent cation.

[Effect of aqueous extract of Lepidium apetalum on dog's left ventricular function].

Left ventricular myocardial contractility, function of pump-blood and coronary flow in dog were increased by i.v. aqueous extract of Lepidium apetalum (AELA) 0.2 ml/kg (containing crude medicinal herb 2 g/ml). The effects were similar to i.v. isoperaline 10 micrograms/kg, but AELA did not obviously change heart rate, arterioveno-oxygen partial pressure difference and arteriovenooxygen solubility. The results indicated the AELA had advantages of cordial effect and increasing coronary flow instead of enhancing oxygen consumption of cardiac muscle.

[Photoacoustic spectra of La (III), Gd (III), Tb (III) 8-hydroxyquinoline complexes and relaxation process].

8-hydroxyquinoline complexes of La(III) Gd (III) and Tb (III) were synthesized and the photoacoustic (PA)spectra of these complexes were measured in the region of 300-800nm at room temperature. The PA spectra and excitation spectra are compared and interpreted. It is observed that the photoacoustic spectra and the excitation spectra behave complementarily. The different fluorescence properties of the complexes and the relaxation process models are studied by the photoacoustic amplitude and phase spectra. It is confirmed by the PA phase spectrum that the PA absorption of pi-pi* transition of Tb (Ox)3 complex is dominantly contributed by the slow radiationless relaxation which is due to inner-molecular energy transfer.

Inhibition of intraocular neovascularization caused by retinal ischemia in pigs by PKCbeta inhibition with LY333531.

OBJECTIVE: The authors tested the antiangiogenic properties of an orally administered protein kinase-Cbeta inhibitor, LY333531, in a pig model of preretinal neovascularization caused by retinal branch vein occlusion to determine the effectiveness of this therapy in preventing intraocular neovascularization from an ischemic stimulus. METHODS: In 20 eyes of 10 pigs, branch retinal vein occlusions were created in a standardized manner using photodynamic thrombosis with rose bengal dye and thermal burns from an argon laser with green light. Five animals received 1 mg/kg LY333531 daily in two oral doses, and five animals were untreated. The eyes were followed clinically for 12 weeks with ophthalmoscopy, fundus photography, and fluorescein angiography. A standardized grading system permitted masked assessment of disc proliferations using stereo fundus photographs. After the pigs were killed, all neovascularization was confirmed histopathologically in a masked fashion and a final grade was assigned to each eye. The Mann-Whitney test was used for statistical analysis of the median values of the unpaired data between the two eyes of each animal (data were rounded up). RESULTS: Significant inhibition of neovascularization was observed in eyes from animals treated with the study drug (P = 0.03). Although some of the treated eyes demonstrated no clinically evident new vessels, histopathologic and photographic analysis demonstrated fine new vessels on the optic disc in all eyes (mean grade 1.9). All the untreated eyes developed clinically evident neovascularization (mean grade 3.1). The oral drug was well tolerated, and no side effects were documented. CONCLUSIONS: A specific protein kinase-Cbeta inhibitor, LY333531, effectively inhibited preretinal and optic nerve head neovascularization in the pig model of branch retinal vein occlusion. This was consistent with the known pathways of signal transduction by growth factors in activated cells and suggested that inhibition of this key regulatory isozyme is effective in the treatment of ischemia-mediated neovascular diseases.

Bilateral choroidal metastases as the initial presentation of a small breast carcinoma: a case report.

Most ocular tumors metastasize from systemic origins in breast carcinoma in females, and bronchial carcinoma in males. Here, we report a case of choroidal carcinoma metastasis from the breast with visual problems being the only initial manifestations. In this case, both eyes were involved at almost the same time, with initial manifestation of blurred vision which progressed to complete visual loss. At first, the patient was diagnosed with malignant melanoma, and enucleation of the right eye was performed in another hospital. However, the tumor had already metastasized rapidly to numerous organs, including the lungs, brain and bone, although it had not affected the liver. Clinical presentations were, therefore, not compatible with those of malignant melanoma, which has usually been reported to metastasize to the liver. Persistent hypercalcemia and raised carcinoembryonic antigen (CEA) concentrations prompted investigations into the possibility of systemic malignancy. A very small breast nodule was finally located by thorough physical examination, and a lumpectomy was performed. A detailed review of the histopathology showed the tumors from the breast and the right eye to have the same origin. Simultaneous bilateral choroidal metastases from other malignancies is not uncommon; however, it is quite rare for breast carcinoma to present with visual problems as a first manifestation. Detailed history taking and physical examination are therefore essential when searching for a primary tumor, so that appropriate therapy can be given earlier.

Laminin fragment E4 inhibition studies: basement membrane assembly and embryonic lung epithelial cell polarization requires laminin polymerization.

Laminins (LMs), the main constituents of basement membranes (BMs), are heterotrimeric glycoproteins composed of alpha, beta, and gamma chains held together by disulfide bonds. In the presence of Ca2+, some laminins, such as laminin-1 self-assemble into a polymer through the interaction of their three NH2 termini. Here we exposed lung organotypic cultures to a proteolytic fragment of laminin-1 that blocks laminin polymerization. This fragment, referred as E4, comprises the outer globular region of laminin beta1 chain. Inhibition of laminin polymerization in lung organotypic cultures resulted in impaired basement membrane assembly and failure of epithelial cells to polarize. In addition, we found that in control organotypic cultures, the bronchial smooth muscle cells were arranged in concentric layers around the newly formed epithelium. However, in E4-treated cultures, the smooth muscle cells were in disarray. Exposure of organotypic cultures to laminin-1 proteolytic fragment P1', that comprises part of alpha1, beta1, and gamma1 chains, but does not overlap with fragment E4, had no effect in basement membrane assembly. Exposure to fragment E4 also caused an increased release of laminin-1 into the culture medium, suggesting a failure to retain laminin at the epithelial-mesenchymal interface. These studies provide the first direct evidence linking epithelial cell polarization to laminin polymerization at the epithelial-mesenchymal interface and assign a key role to the outer globular region of laminin beta1 chain.

Transmission and natural regulation of infection with Ascaris lumbricoides in a rural community in China.

A longitudinal study on a cohort of 215 people who remained untreated from June 1993 to June 1994 was carried out using fecal egg counts to examine the course of transmission and aspects of the natural regulation of the infection with Ascaris lumbricoides. The results indicate that the parasite population remained relatively stable during the years since overall prevalence remained steady above 60%. About 40% of the people in the cohort retained their infection intensity status throughout the year, in terms of light, medium, and heavy infections. The regulation of infection in the population could be attributed to the tendency for the loss of worms from people with initial light infections and the movement to medium infection intensity status of people with both light and heavy infections. Compared with adults, children showed a stronger tendency of retaining initial medium infections but weaker tendencies for keeping initial light infections and clearing themselves of infection. Predisposition to light and medium infections was detected in people who had experienced an uninfected period of 4-12 mo before reinfection was established.

In vivo three-dimensional MR microscopy of mice with chronic relapsing experimental autoimmune encephalomyelitis after treatment with insulin-like growth factor-I.

PURPOSE: The purpose of this study was to determine the ability of three-dimensional in vivo MR microscopy to depict the treatment effects of insulin-like growth factor-I (IGF-I) in SJL mice with chronic relapsing experimental autoimmune encephalomyelitis (crEAE). METHODS: The experiments were performed at 4.7-T on 10 crEAE mice and on one set of control animals. Five crEAE mice were treated with IGF-I and five were treated with a placebo. RESULTS: In the crEAE mice treated with the placebo, in vivo MR microscopy showed areas of abnormal signal throughout the cerebrum, brain stem, and cerebellum. These findings were not present in either the IGF-I-treated mice or the normal control animals. The diffuse alterations in signal intensity in the placebo-treated crEAE mice were not identified on histologic sections of the same areas. CONCLUSION: Differences between the IGF-I- and placebo-treated groups may reflect changes in stabilization or permeability of cell membranes and/or of the blood-brain barrier, although other alternative contrast mechanisms could be playing a role. In vivo MR microscopy depicted changes resulting from treatment of crEAE with IGF-I.
