[Effect of food composition of mixed food on glycemic index].

UNLABELLED: In order to study the effect of protein, fat and dietary fiber on glycemic index(GI) of mixed food, the response of blood glucose and plasma insulin to nine diets with different components of food were tested by using glucose oxidase and radioimmunoassay. Glycemic index of 9 mixed foods show as follows, rice: 83.2 +/- 3.1, rice + stir fry pork: 72.0 +/- 14.0, rice + stir fry pork and celery: 57.1 +/- 11.2, rice + stir fry garlic sprout: 57.9 +/- 7.8, rice + stir fry garlic sprout and eggs: 62.8 +/- 16.7, steamed bread: 80.1 +/- 22.5, steamed bread + butter: 68.0 +/- 16.3, and steamed bread + beef: 49.4 +/- 22.8. Protein(P, beta 1 = -0.696, P < 0.01) and dietary fiber(Fi, beta 2 = -7.364, P < 0.01) can reduce the blood glucose response and were significantly related to GI. Fat also can inhibit the increment of blood glucose, but there is no significant relation with GI. When the co-ingestion of protein with carbohydrate, the serum insulin response increased greatly and the glycemic response reduced. The addition of fat can reduce the glycemic response without change in serum insulin. Dietary fiber can reduce the serum insulin response and inhibit the glycemic response. CONCLUSION: Protein and dietary fiber of mixed food could markedly affect the glycemic index of foods and reduced the blood glucose response.

[Endogenous protective effects of superoxide dismutases on infectious brain injury in rats].

We studied the alterations of MDA and three forms of SOD activities such as T-SOD, CuZn-SOD, and Mn-SOD in rat cerebral tissues injected by bordetella pertussis (BP) to elucidate protective mechanism of SOD against the infectious brain injury. The results were that water content(WC), Evans blue content(EB), MDA, and Mn-SOD activities in 4 h and 24 h BP-treated groups increased and T-SOD and CuZn-SOD decreased compared to corresponding normal saline(NS)-treated groups, respectively(P < 0.01); MDA increased and had a positive correlation with WC and EB in 4 h BP treated group (r = 0.9650, r = 0.9441, P < 0.01, P < 0.01, respectively); Mn-SOD activities were elevated and had a negative correlation with WC, EB, and MDA (r = -0.8650, r = -0.9021, r = -0.9346, P < 0.01, P < 0.01, P < 0.01, respectively) in 24 h BP-treated group. The results suggest that the increase of component Mn-SOD activities may play an important role in vivo endogenous protective mechanism against delayed infectious brain injury.

[Relationship between brain edema induced by freezing injury and concentration of Evan blues and cytosolic free calcium in brain tissues].

The authors studied the changes of Evan blues(EB) of brain tissues and concentration of cytosolic free calcium ([Ca2+]i), and relationship between them and water content of brain tissues following freezing brain edema in rats, to elucidate mechanism and type of brain edema induced by freezing injury. The results indicated that there were close positive relationship between water content and EB of brain tissues, and between [Ca2+]i and water content. We found that not only cytotoxic brain edema(intracellular edema) occurred but also occurred vasogenic brain edema(extracellular edema) followed by blood brain barrier breakdown after brain edema. Freezing brain edema is mixed brain edema.

[Cerebral hemodynamic impairment and therapeutic effect of external counterpulsation on elderly patients with brain infarction].

Hemodynamic parameters of middle cerebral artery(MCA) were measured with Transcranial Doppler (TCD) in 31 elderly patients with brain infarction during convalescence and 28 healthy elderly controls. Twenty-two elderly patients with brain infarction and cerebral hemodynamic impaired were treated by External Counterpulsation(ECP). The results showed that the Vp and Vm of MCA in patients were significantly lower than that in the healthy controls(P < 0.001), the DVp, DVm and asymmetry of MCA in patients were greater than that in the healthy elderly(P < 0.001, P < 0.005). The Vp and Vm of MCA in patients after ECP were significantly higher than before(P < 0.001). The DVp, DVm and asymmetry of MCA in patients after ECP were significantly lower than before(P < 0.001, P < 0.005). The hemodynamic therapeutic effect is 81.8%.

[Effect of pertussis bacilli on the release of excitatory amino acids in brain slice of rats].

OBJECTIVE: To investigate the effect of pertussis bacilli on the release of excitatory amino acids (EAAs) in rat-brain slice and to know whether MK-801(dizocipine) protects brain tissue from excitotoxicity. METHODS: SD rats were randomly divided into 3 groups: 1. Normal slice cultured group(NC); 2. 10% pertussis bacilli(PB) slice cultured group(PBC); 3. 10% PB slice cultured group after pretreatment with MK-801. The release of EAAs from brain slices was measured by high performance liquid chromatography. RESULTS: The concentrations of glutamate(Glu 29.53 +/- 8.00), aspartate(Asp 23.74 +/- 5.79) and LDH(38.03 +/- 8.45) were significantly higher in PBC group than those(Glu 16.46 +/- 4.00, Asp 15.27 +/- 5.31, LDH 23.10 +/- 5.11) in MPBC group and (Glu 16.09 +/- 3.74, Asp 11.47 +/- 2.51, LDH 25.82 +/- 4.50) NC group. CONCLUSION: Pertussis bacilli may elicit a large increase of extracellular EAAs and LDH in vitro, while MK-801 protects brain tissue from excitotoxicity.

[Chemical studies on the polysaccharide in fructus Corni].

OBJECTIVE: To study the tonic constituents in Fructus Corni, the pulp of Cornus of ficinalis. METHOD: Hot-water extraction, ethanol precipitation and column chromatography on DEAE-cellulose and Sephadex G-200 were used to isolate and purify the polysaccharides in Fructus Corni. HPGPC was used to determine the purity and measure the mean molecular weight of the polysaccharides. TLC, PC, GC, GC-MS, 13CNMR, partial acid hydrolysis and permethylation to elucidate the structures. RESULT: A water-soluble polysaccharide with mean molecular weight of 2.47 x 10(14), named Co-4, was obtained. It's main chain was (1-->4) linked beta-D-glucan, with branching points at position 6. CONCLUSION: A thorough study on the polysaccharides of Fructus Corni has been made for the first time and the structure of Co-4 has been elucidated.

[Chemical constituents of Cynanchum chinense R. Br].

OBJECTIVE: To investigate the chemical constituents in the aerial part of Cynanchum chinense. METHOD: The chemical constituents were extracted with solvent and separated by column chromatography. The structures were identified by spectral methods: RESULT: The structures were identified as 7-O-alpha-L-rhamnopy-ranosyl-kaempferol-3-O-alpha-L-rhamnopyranoside, 7-O-alpha-L-rhamnopy-ranosyl-kaempferol-3-O-beta-D-glucopyranoside and 7-O-alpha-L-rhamnopyr-anosylkaempferol-3-O-beta-D-glucopyranosyl(1-->2)- beta-D-glucopyranoside. CONCLUSION: All the three compounds were separated from C. chinense for the first time.

[Determination of flavonoids in pollen typhae(Puhuang) by HPCE and HPLC].

OBJECTIVE: To determine two flavonoids isorhamnetin-3-O-neohesperidoside(I) and typheneoside(II) in Pollen Typhae by HPCE and HPLC. METHOD: HPCE conditions: buffer = 0.02 mol.L-1 borate, 0.05 mol.L-1 SDS and 10% acetonitrile; V = 20 kV; T = 30 degrees C; lambda = 270 nm; sample size = 30 kPa.s, HPLC conditions: C18 lumn, acetonitrile-water (15:85) as mobile phase detection wavelength at 254 nm. RESULT: The contents of flavonoids I and II in five different Typha species and eighteen commercial samples of traditional Chinese drug Puhuang were between 0.1%-0.6%. CONCLUSION: These two methods are rapid, simple, accurate and can be used for the quality control of this traditional Chinese drug.

[Study of the immunological tolerance induced by peripheral intravenous infusion of donor-specific antigens].

In the experiment, donor specific tolerance was induced by DST, DSSL and DSBM through peripheral intravenous transfusion in rat cervical cardiac transplantation model. The results showed there were significant differences in graft survival time between the blank control group and the other groups (P < 0.05). The mean graft survival time days of the DST, DSSL and DSBM groups were 28.9 +/- 13.7, 42.9 +/- 6.6 and 40.3 +/- 11.6 days, respectively. The effect of component infusion was obviously better than that of DST (P < 0.05), but no statistical difference was observed between DSSL and DSBM. CsA gave a noticeable synergistic effect to the induction of immunological tolerance. MLR and CD8+ T cell were remarkably reduced(P < 0.05) in the cases of component transfusion. This study suggested that components transfusion may be of clinical use for donor-specific tolerance inducing.

[Diagnosis and treatment of internal jugular phlebectasia (three cases report].

OBJECTIVE: To discuss the diagnosis and treatment of patients with rare abnormal veins: the internal jugular phlebectasia. METHODS: Three patients with swollen during straining or crying were diagnosed as internal jugular vein dilatation by ultra-sonography, DSA and MRI. Then an operation of envelopment of the right internal jugular vein with the sternocleidomastoid muscle was performed in two cases. In the third case, the bilateral internal jugular ectasia, the right internal jugular vein were resected and the left internal jugular vein was enveloped with the resected vein. RESULTS: These three patients were treated satisfactorily by operation, and no recurrence in the following two years. CONCLUSION: To those who want their internal jugular phlebectasia to be treated, the operation of envelopment of internal jugular vein with sternocleidomastoid muscle is a good method.

Effects of ginsenosides on myocardial reperfusion arrhythmia and lipid superoxidation in high cholesterol diet rats.

To explore the effects of GSL on myocardial reperfusion arrhythmia and lipid superoxidation in high cholesterol diet rats. Hyperlipidemia model was set up with administered high cholesterol emulsion 15 ml/kg to rats orally for 14 days. In GSL group, rats were given GSL i.p. 75 mg/kg simultaneously when administered high cholesterol emulsion. The experiment of myocardial ischemia reperfusion was performed on all rats. The results showed: (1) After administration of high cholesterol emulsion to rats orally for 14 days, hyperlipidemia model was set up successfully, simultaneously treatment with GSL. It lowered serum lipid; (2) In hyperlipidemia state, serum MDA increased (p < 0.01, SOD and NO decreased markedly (p < 0.01 and p < 0.05 respectively) after 2 h of myocardial reperfusion; the rate of reperfusion arrhythmia (RPAr) increased within 10 min of reperfusion, four out of nine rats died of ventricular fibrillation (VF); and (3) GSL decreased MDA, increased SOD and NO after 2 h of myocardial reperfusion. All changes were significant (p < 0.01); the rate of RPAr decreased, no VF occurred and all rats survived. Hyperlipidemia aggravated myocardial ischemia reperfusion injury and increased the incidence of RPAr. The results suggested that GSL reduced myocardial ischemia reperfusion injury and RPAr in high cholesterol diet state through antiperoxidating and inducing the production of NO.

[Gas chromatography/mass spectrometry analysis for determination of methyl alpha-(6-methoxyl-2-naphthyl) propionate from carbonylation].

alpha-(6-Methoxyl-2-naphthyl) ethanol can be carbonylated to form methyl-alpha-(6-methoxyl-2-naphthyl)propionate catalyzed by a catalyst in the pressnce of methanol. High efficiency capillary gas chromatography and GC/MS technique were used for determining products which were obtained from the carbonylation. An OV-101 fused silica capillary column (25 m x 0.2 mm i.d.) and a flame ionization detector (FID) were employed in the GC analysis. The column temperature was kept at 250 degrees C. Mass spectra were obtained by electron impact at 70 eV. The structures of four compounds were identified. The major product is methyl-alpha-(6-methoxy-2-naphthyl) propionate, and the two by-products are 2-vinyl-6-methoxynaphthalene and 1-methoxy-(6'-methoxy-2'-naphthyl)ethanol.

[The properties of new affinity adsorbent for adsorbing the cycle immune complex in blood].

This study was designed to select an affinity adsorbent from 5 kinds of adsorbents available. The adsorbent AA3 was found being specific to the adsorption of pathologic cycle immune complex (CIC). The physical properties such as specific surface area, proe volume and average pore diameter of adsorbents were determined. A discussion on the factors that affect the adsorption of CIC is also presented in this paper.

[Cloning of mitomycin C resistance gene from Streptoverticillium caespitosum ATCC27422 and studying its function].

Streptoverticillium caespitosum is the major producer of anti-cancer drug mitomycin. It demonstrated that there exists a strong restriction system for the transformation of foreign DNA in this strain. Using the blocked mutant S-6 which is sensitive to mitomycin C (MC) and does not produce mitomycin as a recepient and plasmid pIJ699 as vector, the gene library of the strain ATCC27422 was constructed. A clone of mitomycin C resistance gene(mcr) was obtained by screening from this library. The mitomycin C resistance gene cloned on the 6.2 kb fragment of plasmid pLX5 could be expressed in the Strptonmyces lividans TK54. It was the first time to introduce successfully the plasmid pLX5 into the wild type strain ATCC27422 by electroporation technique. And the transformant's resistance to mitomycin C has risen greatly: the MIC has gone up from 200 micrograms/ml to more than 1000 micrograms/ml. The result of fermentation showed that the yield of mitomycin of ATCC27422 harboring plasmid pLX5 was more than the wild strain ATCC27422's. It suggested that there is some relationship between the level of the resistance to mitomycin C and its biosynthesis.

[Establishment and identification of the monoepitopic monoclonal antibody of Plasmodium falciparum].

AIM: To prepare a monoepitopic monclonal antibody of Plasmoduim falciparum. METHODS: According to the theory of protein structure, a peptide with 6-9 residues representting the antigenicity of the original histidine-rich protein II (HRP-II) of P. falciparum was synthesized and used to immunize BALB/c mice after purity identification by capillary electrophoresis. RESULTS: Four hybridoma cell lines targeting a suitable peptide from pf HRP-II were obtained by using spleen embedment method and hybridoma technology. CONCLUSION: It is the first report to prepare a monoepitopic monoclonal antibody against original protein by selecting a suitable peptide from the primary sequence of protein.

[Anti-Toxoplasma effect of activated mouse macrophages induced by interferon-gamma combined with TNF-alpha].

AIM: To compare the inhibitory effect of the macrophages activated by IFN-gamma combined with TNF-alpha against RH strain and Fukaya strain. METHODS: The average parasite proliferation rates of the two strains within the cytokine-activated M phi s were calculated at different times post-challenge, the nitric oxide (NO) levels in the medium supernatant were simultaneously determined. RESULTS: In the macrophages activated by 100 U each of IFN-gamma and TNF-alpha, the invaded tachyzoites of RH strain were completely killed, while the invaded tachyzoites of Fukaya strain remained slow proliferation with significantly lower levels of NO detected at 24 h post challenge. CONCLUSION: The difference in the anti-Toxoplasma effect of the activated macrophages against RH and Fukaya strains might be attributed to the different amount of NO produced by the macrophages.
