Expression and characterization of the human YWK-II gene, encoding a sperm membrane protein related to the alzheimer betaA4-amyloid precursorprotein.

The YWK-II cDNA, RSD-2, encoding a sperm membrane protein was isolated from a rat testis cDNA expression library. Using the RSD-2 insert in combination with rapid amplification of cDNA ends (RACE), the corresponding human gene was isolated from a human testis cDNA expression library. The human testis cDNA, HSD-2, is 3654 bp in length and contains an open reading frame of 763 codons. Hydropathicity analysis showed that the deduced polypeptide is a single strand transmembrane protein. The deduced polypeptide has partial homology with the amyloid precursor protein (APP) and high homology with the amyloid precursor homologue, APLP2/APPH. The YWK-II gene was mapped and assigned to human chromosome locus: 11q24-25. Northern blotting of various human tissue RNAs using the HSD-2 cDNA as a probe showed that the gene is transcribed ubiquitously. The cytoplasmic domain of HSD-2 was expressed in Escherichia coli. In-vitro studies showed that the recombinant polypeptide bound to a GTP-binding protein (G(o)) and was phosphorylated by protein kinase C and cdc2 kinase. In mammalian F11 cells, the recombinant polypeptide was found to be coupled to G(o). Thus, the YWK-II component has the characteristics of a G(o)-coupled receptor and may be involved in G(o)-mediated signal transduction pathway. Protein kinase C and cdc2 kinase may regulate this pathway in spermatozoa by phosphorylating the cytoplasmic domain of the YWK-II component.

Cooperativity and resonances in periodically driven spin-boson systems.

We present an analytical recursive formulation for the quantum transport in symmetric two-level systems under the influence of both dissipation and periodic driving. The rate-matching condition for quantum stochastic resonance despite its different appearance is found to be physically the same as that in the classical case. Analyzed are also the Rabi resonance and its implication to quantum stochastic resonance. We demonstrate that no matter how weak the driving field is, transport can involve about 70% population in the vicinities of the third-harmonic as well as the fundamental-harmonic Rabi resonance. Recovered is also an adiabatic passage condition in which the transport carries a nearly 100% population in the low frequency and strong driving limit.

An alternate binding site for the P1-P3 group of a class of potent HIV-1 protease inhibitors as a result of concerted structural change in the 80s loop of the protease.

Structures of the complexes of HIV protease inhibitor L--756,423 with the HIV-1 wild-type protease and of the inhibitors Indinavir, L-739,622 and Saquinavir with the mutant protease (9X) containing nine point mutations (Leu10Val, Lys20Met, Leu24Ile, Ser37Asp, Met46Ile, Ile54Val, Leu63Pro, Ala71Val, Val82Thr) have been determined. Comparative analysis of these structures reveals an alternate binding pocket for the P1-P3 group of Indinavir and L--756, 423. The alternate binding pocket is a result of concerted structural change in the 80s loop (residues 79-82) of the protease. The 80s loop is pulled away from the active site in order to accommodate the P1-P3 group, which is sandwiched between the flap and the 80s loop. This structural change is observed for the complexes of the wild type as well as the 9X mutant protease. The study reveals that the 80s loop is an intrinsically flexible loop in the wild-type HIV-1 protease and that mutations in this loop are not necessary to result in conformational changes. Conformation of this loop in the complex depends primarily upon the nature of the bound inhibitor and may be influenced by mutations in the protease. The results underscore the need to understand the intrinsic structural plasticity of the protease for the design of effective inhibitors against the wild-type and drug-resistant enzyme forms. In addition, the alternate binding pocket for the P1-P3 group of Indinavir and L--756,423 may be exploited for the design of potent inhibitors.

Blunt cell growth potential in carcinogen resistant inbred DRH rats.

A carcinogen-resistant inbred strain DRH/Sea has been developed from the Crj:Donryu strain. The rats had a very low incidence of liver tumors when they were fed diets containing a hepatocarcinogen such as 3'-methyl-4-dimethylamino-azobenzene (3'-Me-DAB). Despite using 3'-Me-DAB during the stage of selection, the DRH/Sea rats developed normally, reproduced and did not have any spontaneous tumor in the lung, liver or uterus at over 1 year of age. Although their growth curves were similar to the Crj:Donryu rats, the progression of polyploidization in the liver was significantly delayed when compared with Crj:Donryu rats. Mitogenic changes that occurred in the liver caused by either 3'-Me-DAB or lead nitrate were less significant in the DRH rats than in Crj:Donryu rats. Furthermore, the growth rate of cultured fibroblasts derived from the DRH rats was slower than that of Crj:Donryu rats. These results, together with our previous results, suggest that slow growth potential is present under certain conditions in DRH rats. These findings may explain partly the meaning of the different susceptibility to hepatocarcinogens.

Squalamine improves retinal neovascularization.

PURPOSE: Modalities for inhibiting neovascularization may be one avenue to the development of effective therapies for retinopathy. The effect of squalamine, an antiangiogenic amino sterol, on oxygen-induced retinopathy (OIR) was assessed in a mouse model. METHODS: OIR was induced in C57BL6 mice by a 5-day exposure to 75% oxygen from postnatal day (P)7 through P12. Squalamine (25 mg/kg, subcutaneous)treated animals received either daily doses for five days from P12 to P16 or one dose just after removal from oxygen on P12. Each set of animals was killed at P17 to P21. Retinopathy was assessed with a retinopathy scoring system evaluation of retinal wholemounts and by quantification of neovascular nuclei on retinal sections. RESULTS: Animals receiving 5 days of squalamine after a 5-day exposure to oxygen had total retinopathy scores (expressed as median score with 25th and 75th quartiles in parentheses) of 4(3, 5) versus oxygen-only-reared animals with scores of 8(7, 9; P < 0.001). Animals reared in room air and animals exposed to squalamine only had similar retinopathy scores: 1(1, 2) and 1(0, 2). Oxygen-reared animals receiving single-dose squalamine also showed improvement, with a median retinopathy score of 4(4, 6.75) versus oxygen-only-reared animals with median retinopathy score of 9(7, 10; P < 0.001). There was a decreased number of neovascular nuclei extending beyond the inner limiting membrane on retinal sections in animals treated with 5 days (P < 0.01) and 1 day (P < 0.001) of squalamine. CONCLUSIONS: Squalamine significantly improved retinopathy and may be a novel agent for effective treatment of ocular neovascularization.

A preliminary study of single nucleotide polymorphisms of lipoprotein lipase gene in coronary atherosclerotic heart disease.

OBJECTIVE: This study inquired into the relationship between single nucleotide polymorphisms(SNPs) of lipoprotein lipase gene (LPL) and coronary atherosclerotic heart disease(CHD). METHODS: Exon 6 and fragment of intron 6 in LPL gene from 102 cases of CHD and 110 normal subjects were analysed by polymerase chain reaction amplification, temperature-modulated high-performance liquid chromatography (TmHPLC) and DNA sequencing. RESULTS: Three new SNPs (4212t/c, 4509t/c, 4576a/c) were detected in the intron 6 of LPL gene. There was significant difference in frequencies of 4212t/c, and 4576a/c between health group and CHD group(P<0.05). CONCLUSION: This study has accumulated additional data on SNPs in LPL gene and provided new data for exploring the mechanism of CHD.

Analysis of temperature-modulated high-performance liquid chromatography as a tool for detection of single nucleotide polymorphism.

OBJECTIVE: To establish and evaluate temperature-modulated high-performance liquid chromatography(TmHPLC) as a rapid and efficient technique of detecting single nucleotide polymorphism(SNP). METHODS: Several TmHPLC parameters, such as the eluting gradient ranges and column temperatures used for analysing the amplicons have been studied. The size of DNA fragments suitable for analysis of SNP was studied. Also the sensitivity of the TmHPLC was evaluated. RESULTS: The suitable chromatographic condition of TmHPLC was found out. The elution peaks of dNTP and primers could be separated from those of polymerase chain reaction products of the genomic DNA. The heterozygotes of SNP from the sequenced DNA fragment with 178bp, 269bp and 272bp could be distinguished from the homozygotes of SNP clearly. CONCLUSION: The authors have established and evaluated the TmHPLC method, which is an efficient, sensitive and rapid method for screening out SNP in the DNA fragments of larger size.

Dexamethasone and critical effect of timing on retinopathy.

PURPOSE: Administration of corticosteroids soon after birth has been reported to have deleterious, protective, and no effect on retinopathy of prematurity. Conflicting results may be due to timing of corticosteroid administration. The goal of this study was to determine effects of pretreatment and late dexamethasone on retinopathy in a mouse model. METHODS: The C57BL6 mouse model of oxygen-induced retinopathy (by placing animals in 75% oxygen from postnatal days 7 through 12) was used to create retinal neovascularization. Dexamethasone at 0.5 mg/kg per day was administered from day 1 through day 5 in the pretreatment group. The late-treatment group received 5 days of dexamethasone at the same dose beginning on day 12. Mice were killed at days 17 through 20, and retinal vasculature was assessed by a retinal scoring system of wholemount preparation after high-molecular-weight fluorescein-labeled dextran perfusion. In addition, retinal neovascularization was assessed by quantification of extraretinal neovascular nuclei in retinal sections. Statistical significance was defined as P: < 0.05 and was determined by the Kruskal-Wallis test, Mann-Whitney test, and Student's t-test. RESULTS: Oxygen-exposed animals that received treatment with dexamethasone before oxygen exposure had an improvement in retinopathy, with a median score of 6 (5,7; 25th,75th quartiles) compared with 10 (8,11) in the untreated oxygen-exposed (P: < 0.05). The group treated late (after oxygen exposure) with dexamethasone had a median score of 10 (9,11). Pretreatment reduced extraretinal vascularization, when assessed by quantification of neovascular nuclei, to a mean +/- SEM of 19 +/- 9, significantly less than in the untreated oxygen-exposed group (55 +/- 12; P: < 0.05). No difference was observed in the late-treatment group when compared with the untreated oxygen-exposed group. Significant growth retardation, indicated by body weight, was observed in the pretreatment (P: < 0.01) and late-treatment (P: < 0. 05) groups when compared with the control group. CONCLUSIONS: Timing of dexamethasone administration was critical to the inhibition of development of retinopathy in the mouse model. Degree of growth retardation, measured by body weight, also appeared to be time dependent. These data may explain the different results of clinical observations with respect to corticosteroid treatment, timing, and development of retinopathy.

Rat hepatocellular apoptosis induced by glycodeoxycholate.

OBJECTIVE: To explore the relationship between glycodeoxycholate (GDC) and rat hepatocellular apoptosis. METHODS: GDC was used to treat rat hepatocytes cultured in vitro and the apoptotic cells were observed and analyzed with light microscope, transmission electron microscope, TUNEL in situ hybridization, DNA agarose gel electrophoresis and flow cytometry. RESULTS: When rat hepatocytes were incubated with GDC (final concentration of 100 micromol/L) for 2h, apoptotic hepatocytes were observed with light and transmission electron microscope, and TUNEL in situ hybridization. When the hepatocytes were incubated with GDC (final concentration of 50, 100, 150 micromol/L, respectively) for 2h, or with GDC (final concentration of 100 micromol/L) for 2, 4, 6, 8h, respectively, agarose gel electrophoresis of the hepatocytes DNA demonstrated the typical ladder patterns. CONCLUSION: GDC with final concentration of 50, 100, 150 micromol/L could induce rat hepatocellular apoptosis.

Dissolution behavior of hydroxyapatite coating by hydrothermal method: an in vitro study.

Hydroxyapatite (HA) coated CaO-SiO2-B2O3-Na2O glass composites prepared by a hydrothermal coating method were soaked in four kinds of solutions to evaluate the dissolution behavior and the chemical stability. 0.5 mol/l HCl and 0.5 mol/l HNO3 were used as acidic solution, and 5% physiological saline and sodium lactate compound solution as the neutral physiological solution. It was found that HA coated glass composites had the better chemical stability and the corroding-resistance ability. The dissolution behavior of the HA coating was mainly correlated with the phase composition of the as-received HA coating and its microstructure as well as the pH value of the solution.

[Changes of T lymphocyte and CD4+ T helper subsets in peripheral blood from women with unexplained recurrent spontaneous abortion].

OBJECTIVE: To investigate the relationship between occurrence of unexplained recurrent spontaneous abortion(URSA) and immune function of T lymphocytes. METHODS: T lymphocyte subsets and CD4+ T helper subsets (including T helper 1 and T helper 2 cells) in peripheral blood from 30 cases with URSA and 24 healthy women (controls) were detected by enzyme-linked immunospot assay (ELISPOT). RESULTS: The percentage of CD4+ positive T lymphocytes in URSA group was higher than that in the controls (60% vs 45%, P < 0.05). Compared with the normal group, the percentage of Th1 cells increased (26% vs 14%, P < 0.01) but the percentage of Th2 cells decreased (7% vs 20%, P < 0.05), the ratio of Th1 to Th2 increased (3.80 vs 0.73, P < 0.01) in the URSA group. CONCLUSIONS: The occurrence of URSA might be associated with proliferation of the CD4+ positive T cells and enhancement of cellular immune function mediated by Th1 cells.

[A study on membrane fluidity of thyroid gland in rats with iodine deficiency or iodine excess].

OBJECTIVES: To explore the effect of iodine deficiency and excess on thyroid membrane fluidity and its pathogenesis. METHODS: Wistar rats were fed different levels of iodine in the diet (normal iodine as control, low iodine and excess iodine). Serum T4 and T3 levels, thyroid superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities and malondialdehyde (MDA) content were determined, and two parameters of polarization (Pr) and micro-viscosity (eta) using DPH as a florescence probe were tested. RESULTS: Serum T4 and T3 levels in the low iodine (LI) group were markedly decreased, and the thyroid SOD and GPx activities and MDA content were significantly increased compared with the other two groups. However no changes were found in the high iodine (HI) group. There were no differences in Pr and eta values among three groups at 12th week of experiment, but at 24th week, both Pr and eta values in LI group were increased significantly compared to the other two groups. But the effect of iodine excess as expected was not found in this study. CONCLUSIONS: Long-term iodine deficiency caused a marked increase of membrane micro-viscosity and decrease of membrane fluidity in rat thyroid gland. Oxidative damage of thyroid by free radicals and retardation of lipid metabolism are considered as two important pathogenesis factors.

[Tumor infiltrating dendritic cells in relation to prognosis of lung cancer].

OBJECTIVE: To study the degree of infiltration of dendritic cells in lung cancer in relation to prognosis. METHODS: S-100 protein as the marker of tumor-infiltraing dendritic cells (TIDC) was stained immunohistochemically in lung cancer specimens. The degree of infiltration was considered mild and marked according to the criteria reported by Furukawa. The survival rate was obtained by Log-rank test of the data. RESULTS: In 15 of 38 adenocarcinomas, whose TIDC infiltration was marked, the five-year survival rate was 60.0%, while in 23 with mild TIDC infiltration, the five-year survival rate was 21.7% (P < 0.01) Quite similar data were obtained from 44 squamous-cell carcinomas. Significantly higher five-year survival rate (61.1%) was observed in those with marked TIDC than those with mild infiltration (19.2%). CONCLUSION: The degree of TIDC infiltration is positively correlated with the survival rate of lung carcinoma patients.

[Dendritic cell infiltration in lung adenocarcinoma and its effect on prognosis].

OBJECTIVE: To study the infiltration degree of dendritic cells in lung adenocarcinoma and its effect on prognosis. METHODS: S-100 protein was used as the specific marker for dendritic cells, and immunohistochemistry was employed to detect the distribution of dendritic cells in lung adenocarcinoma. RESULTS: Among 46 cases of lung adenocarcinoma, 19 were markedly infiltrated by dendritic cells, the five-year survival rate being 61%; 27 showed slight infiltration, the five-year survival rate being 21%. Among the 46 cases of adenocarcinoma, 26 were low differentiated adenocarcinoma, in which 10 were infiltrated markedly by dendritic cells, five-year survival rate being 57%; and 16 were slightly infiltrated, five-year survival rate being 16%. These data were evaluated with the log-rank test, which showed a significant difference between the markedly infiltrated group and the slightly infiltrated group. CONCLUSION: The prognosis of lung adenocarcinoma which markedly infiltrated by dendritic cells is better than that of slightly infiltrated by dendritic cells.

[A study of chemo-hand instrument removal of caries in vitro].

OBJECTIVE: To study the efficiency of chemo-hand instrument removal material. METHODS: The effect on caries removal of N-monochlorochloraminobutyric acid and the electrical dental instrument was detected by the caries detection, microscope and SEM. RESULTS: The positive for the caries detection and residual caries were zero in both groups. CONCLUSION: The effect on caries removal of N-monochlorochloraminobutyric acid is almost the same as the control.

[Electron-immunostaining characteristics of platelet-derived growth factor, transforming growth factor beta1 and their receptors in epiretinal membranes].

OBJECTIVE: To investigate the ultrastructural distribution of platelet-derived growth factor (PDGF), transforming growth factor beta(1) (TGF-beta(1)) and their receptors in epiretinal membranes (ERM) and discuss the clinical significance of the distribution. METHODS: 25 membrane specimens were surgically removed by vitrectomy from 9 patients with rhegmatogenous retinal detachment. Double-staining techniques of immunoelectromicroscopy for eight antibodies (PDGF, TGF-beta(1), PDGF receptor subunits alpha and beta, TGF-beta subunits I and II, type I and III collagen) were used in these specimens as previously designed. RESULTS: PDGF and TGF-beta(1) staining appeared to be stronger on early (< 2 months) and late (> 6 months) stage membranes than that of middle (2 - 6 months) stage of membranes, and the immunostaining intensity inverted with the degree of proliferative vitreoretinopathy (PVR). We found that the gold particles of PDGF and TGF-beta(1) tended to get together with that of type I and III collagen. In addition, the stainings of four growth factor receptors were frequently positive in a type of epithelial-like cells with rich cytoplasm components, and also the gold particles of PDGF and TGF-beta(1) were found around these cells. CONCLUSIONS: A concentration change of PDGF and TGF-beta(1) exists in development of ERM throughout. The epithelial-like cells are of a type of active cells, and autocrine and paracrine activity may be involved in ERM pathogenesis. PDGF and TGF-beta(1) influence the contractile activity of ERM.

[Distribution of minimal residual leukemia in Brown Norway rats express LacZ gene].

OBJECTIVE: To investigate the distribution of minimal residual leukemia (MRL) in vivo. METHODS: The MRL model in BN rat was established by using LT12nl leukemia cell line carrying LacZ gene. The marker gene (LacZ) was detected by using reverse transcriptase polymerase chain reaction (RT-PCR) and nested PCR, X-gal staining, and cytomorphological and pathological examination. RESULTS: PCR assays showed positive result in humerus at day 4, femur at day 6, and spleen at day 9 after CTX treatment. The peak positive signal in humerus was stronger at day 9. X-gal staining showed negative in spleen at day 9. PCR showed negative results in peripheral blood and liver. CONCLUSION: PCR can be used as an effective tool to investigate the distribution pattern in experimental MRL.
