[Biocompatibility of self-designed absorbable hydroxyapatite/poly (DL-lactide) composites].

The biocompatibility of self-designed hydroxyapatite/poly (DL-lactide) rods was evaluated both in vitro and in vivo including Ames test, micronucleus test, acute and subacute systemic toxicity test, hemolysis test, hemopexis test and long-term muscle and bone implant test. The results indicate that the material has no toxicity, no stimulation and mutation, and it does not cause hemolysis and hemopexis. Consequently, the biocompatibility of the composite was good.

[Chemical constituents of Scolopendra negrocapitis].

The chemical constituent of Scolopendra negrocapitis Zhang et Wang were systematically analyzed and compared with S. subspinipes mutilans L. Koch. The results showed the lipid content was 18.7%, protein was 63.4% and total amino acid was 11.9%; S. negrocapitis contained the same twelve fat acids (the content of unsaturated fat acid was nearly 64% in its fat acids), twenty one amino acids and twelve trace elements as S. subspinipes mutilans had. The protein of S. negrocapitis showed forteen bands as determined by polyacrylamide gel electrophoresis. S. negrocapitis and S. subspinipes mutilans have the same main chemical composition. The contents of compositions are different. S. negrocapitis is worth as the nature resources of medicinal centepede.

[Local treatment with human laryngeal squamous carcinoma draining lymph node lymphocytes in nude mice bearing human laryngeal carcinoma xenografts].

OBJECTIVE: To increase the number of the tumor draining lymph nodes lymphocytes (TDLNLs) in tumor site and study its anti-tumor effect. METHODS: Interleukin 2-activated TDLNLs were administrated by local injection in 10 nude mice bearing human laryngeal cancer at 10(7) cells/mouse, once a week for two weeks. As a control, 10 nude mice bearing human laryngeal cancer were injected with D-Hanks buffer solution. RESULTS: The volume and weight of TDLNLs treated tumors were significantly reduced in comparison with those of the control (P < 0.05). The observed histopathologic characteristics of treated tumors were tumor necrotic foci and increased infiltration of lymphocytes in tumor and around tumor vessels. Proliferation of spleen nodules was also observed in the treatment group. Remote metastasis was not found in this group. In contrast with the control group, tumor necrosis and proliferation of spleen nodules were not observed and infiltration of lymphocytes in the tumor was not significant. There was a lung metastasis in the control group. CONCLUSION: Local injection of TDLNLs is effective against human laryngeal carcinoma xenografts in nude mice. It also could stimulate the immunoresponse of hosts and probably decrease the chance of tumor metastasis.

[A study of the mitogen effects of endothelin-1 on human dental pulp cell in vitro]

OBJECTIVE:To investigate the mitogen effects of endothelin-1 (ET-1)with different concentrations on human pulp cells.METHODS:Using in vitro pulp cell cultrue technique and thiazolyl blue tetrazolium bromide (MTT) staining and spectrophotometric assay.RESULTS:Endothelin-1 with different concentration had the mitogenesis on human pulp cells in different degree,which was dependent on ET-1 concentration.CONCLUSION:Endothelin-1 which was mitogen factor on human pulp cells may play a role in the regeneration of pulp tissue.

Molecular characterization of a novel tubby gene family member, TULP3, in mouse and humans.

Tubby and related proteins are derived from a small family of novel genes. The carboxytermini of this family are highly conserved across a number of species including humans, mice, Caenorhabditis elegans, Arabidopsis, rice, and maize. Splicing defects in both tub and another member of the gene family, TULP1 (tubby-like protein 1), lead to phenotypes of retinal degeneration in mice and humans, respectively. We describe here the isolation of the human and mouse homologs of a new family member, TULP3. The cDNAs code for proteins of 442 and 460 amino acids, respectively. The level of identity between the human TULP3 and the mouse homolog is 69%, lower than that observed for the homologs of the other family members (96% for human and mouse TUB), and is higher at the amino- and carboxytermini than in the central region of the protein. Phylogenetically, TULP3 is the family member most closely related to TUB. Also, like TUB, it has a wider pattern of tissue expression than either TULP1 or TULP2. TULP3 is detected at high levels in human RNA from testis, ovaries, thyroid, and spinal chord. Tulp3 is also highly expressed in mouse RNA from eyes and adipose depots, tissues not tested in the human Northern analysis. We also report that TULP3 maps to human chromosome 12p13. The murine homolog, Tulp3, maps to the telomere of mouse chromosome 6.

Remarkable difference in the incidence of liver tumors by N-nitrosodimethylamine in carcinogen-resistant DRH rat and its parental strain Donryu rat.

The carcinogen-resistant inbred DRH rat strain was developed from the carcinogen-sensitive Donryu rat and showed a remarkably lower incidence of liver tumors than the latter after administration of either 3'-methyl-4-dimethylaminoazobenzene (3'-Me-DAB) or 2-acetylaminofluorene (2-AAF). In the present study, we examined the tolerance of DRH rat to another type of hepatocarcinogen, that is, N-nitrosodimethylamine (DMN). Male DRH and Donryu rats, 3 weeks of age, were given a single i.p. injection of 10 mg/kg body weight DMN and were sacrificed 1 year later. Five of 11 Donryu rats (45%) had macroscopically detectable tumors: 9 liver tumors in 4 rats and 1 urinary bladder tumor in the other rat. On the contrary, no tumors were detectable in the livers or other organs of 10 DRH rats under the same conditions. These results and other circumstantial evidence indicate that the different susceptibility of chemical carcinogenesis between DRH and Donryu rats is independent of individual pathways of metabolic activation of carcinogens.

Efficacious, orally bioavailable thrombin inhibitors based on 3-aminopyridinone or 3-aminopyrazinone acetamide peptidomimetic templates.

We have addressed the key deficiency of noncovalent pyridinone acetamide thrombin inhibitor L-374,087 (1), namely, its modest half-lives in animals, by making a chemically stable 3-alkylaminopyrazinone bioisostere for its 3-sulfonylaminopyridinone core. Compound 3 (L-375,378), the closest aminopyrazinone analogue of 1, has comparable selectivity and slightly decreased efficacy but significantly improved pharmacokinetics in rats, dogs, and monkeys to 1. We have developed an efficient and versatile synthesis of 3, and this compound has been chosen for further preclinical and clinical development.

Genomic organization of four beta-1,4-endoglucanase genes in plant-parasitic cyst nematodes and its evolutionary implications.

The genomic organization of genes encoding beta-1,4-endoglucanases (cellulases) from the plant-parasitic cyst nematodes Heterodera glycines and Globodera rostochiensis (HG-eng1, Hg-eng2, GR-eng1, and GR-eng2) was investigated. HG-eng1 and GR-eng1 both contained eight introns and structural domains of 2151 and 2492bp, respectively. HG-eng2 and GR-eng2 both contained seven introns and structural domains of 2324 and 2388bp, respectively. No significant similarity in intron sequence or size was observed between HG-eng1 and HG-eng2, whereas the opposite was true between GR-eng1 and GR-eng2. Intron positions among all four cyst nematode cellulase genes were conserved identically in relation to the predicted amino acid sequence. HG-eng1, GR-eng1, and GR-eng2 had several introns demarcated by 5'-GCellipsisAG-3' in the splice sites, and all four nematode cellulase genes had the polyadenylation and cleavage signal sequence 5'-GAUAAA-3'-both rare occurences in eukaryotic genes. The 5'- flanking regions of each nematode cellulase gene, however, had signature sequences typical of eukaryotic promoter regions, including a TATA box, bHLH-type binding sites, and putative silencer, repressor, and enhancer elements. Database searches and subsequent phylogenetic comparison of the catalytic domain of the nematode cellulases placed the nematode genes in one group, with Family 5, subfamily 2, glycosyl hydrolases from Scotobacteria and Bacilliaceae as the most homologous groups. The overall amino acid sequence identity among the four nematode cellulases was from 71 to 83%, and the amino acid sequence identity to bacterial Family 5 cellulases ranged from 33 to 44%. The eukaryotic organization of the four cyst nematode cellulases suggests that they share a common ancestor, and their strong homology to prokaryotic glycosyl hydrolases may be indicative of an ancient horizontal gene transfer.

A photoactivated prodrug.

A photolabile derivative (1) of the anticancer drug, 5-fluorodeoxyuridine (2), was designed and synthesized as a model prodrug. Photolysis of 1 with long-wavelength UV light rapidly released 2 in solution. While compound 1 alone is nontoxic to cells, the presence of both 1 and UV irradiation (lambda = 350 nm) resulted in potent inhibition of cell growth.

Design and synthesis of a series of potent and orally bioavailable noncovalent thrombin inhibitors that utilize nonbasic groups in the P1 position.

As part of an ongoing effort to prepare therapeutically useful orally active thrombin inhibitors, we have synthesized a series of compounds that utilize nonbasic groups in the P1 position. The work is based on our previously reported lead structure, compound 1, which was discovered via a resin-based approach to varying P1. By minimizing the size and lipophilicity of the P3 group and by incorporating hydrogen-bonding groups on the N-terminus or on the 2-position of the P1 aromatic ring, we have prepared a number of derivatives in this series that exhibit subnanomolar enzyme potency combined with good in vivo antithrombotic and bioavailability profiles. The oxyacetic amide compound 14b exhibited the best overall profile of in vitro and in vivo activity, and crystallographic studies indicate a unique mode of binding in the thrombin active site.

New wiggler beamlines at BSRF.

A new multipole wiggler has been installed in the ring of the Beijing Electron Positron Collider (BEPC), and two new beamlines were extracted from it for experiments, including LIGA, microprobe fluorescence analysis, high-pressure diffraction and soft X-ray research. The beam profile and photon intensity were measured and compared with the calculated values. The main parameters and characteristics of the two beamlines are illustrated. Some experimental results are presented.

A compact vacuum-ultraviolet beamline for synchrotron radiation photoelectron spectroscopy combined with an ion-scattering spectrometer, SORIS.

Apparatus for high-resolution synchrotron radiation photoelectron spectroscopy combined with high-resolution medium-energy ion scattering, named 'SORIS', has been developed for simultaneous investigations of electronic states and atomic structures on surfaces. For this purpose, a compact vacuum-ultraviolet beamline of photon energy from 5 to 700 eV has been designed for the small storage ring 'Aurora' installed at Ritsumeikan University. Owing to the small electron-beam size in the storage ring, an energy resolution E/dE of >5000 can be obtained.

High-resolution spherical grating monochromator for the undulator beamline (BL-16B) at the Photon Factory.

The design and performance of a 24 m high-resolution spherical grating monochromator (H-SGM) for the undulator beamline (BL-16B) at the Photon Factory are described. With three interchangeable gratings of 400, 900 and 2000 lines mm(-1), the H-SGM is designed to cover the photon energy range 40-600 eV. With a resolving power of approximately 2000 in the photon energy range 40-530 eV, more than 10(11) photons s(-1) have been measured. The best resolutions obtained so far, at a substantially lower photon flux, are greater than 10 000 at 250 eV and 8000 at 400 eV

Endogenous cellulases in animals: isolation of beta-1, 4-endoglucanase genes from two species of plant-parasitic cyst nematodes.

beta-1,4-Endoglucanases (EGases, EC 3.2.1.4) degrade polysaccharides possessing beta-1,4-glucan backbones such as cellulose and xyloglucan and have been found among extremely variegated taxonomic groups. Although many animal species depend on cellulose as their main energy source, most omnivores and herbivores are unable to produce EGases endogenously. So far, all previously identified EGase genes involved in the digestive system of animals originate from symbiotic microorganisms. Here we report on the synthesis of EGases in the esophageal glands of the cyst nematodes Globodera rostochiensis and Heterodera glycines. From each of the nematode species, two cDNAs were characterized and hydrophobic cluster analysis revealed that the four catalytic domains belong to family 5 of the glycosyl hydrolases (EC 3.2.1, 3.2.2, and 3.2.3). These domains show 37-44% overall amino acid identity with EGases from the bacteria Erwinia chrysanthemi, Clostridium acetobutylicum, and Bacillus subtilis. One EGase with a bacterial type of cellulose-binding domain was identified for each nematode species. The leucine-rich hydrophobic core of the signal peptide and the presence of a polyadenylated 3' end precluded the EGases from being of bacterial origin. Cyst nematodes are obligatory plant parasites and the identified EGases presumably facilitate the intracellular migration through plant roots by partial cell wall degradation.
