Bulk magnetic susceptibility shifts in NMR studies of compartmentalized samples: use of paramagnetic reagents.

The bulk magnetic susceptibility (BMS) shift of a nuclear resonance frequency caused by a paramagnetic compound is of importance in vivo NMR, both magnetic resonance spectroscopy and magnetic resonance imaging. However, since it is a rather complicated phenomenon, it has been the source of many misinterpretations in the literature. We have reworked and organized the theory of the BMS shift. This includes accounting for the important effects of local susceptibility. We have conducted experiments on phantom samples in order to illustrate the principles involved. Our phantoms consist of capillaries and coaxial cylinders. They simulate the situations of blood vessels oriented parallel and perpendicular to the magnetic field and the interstitial spaces surrounding them. In most of our experiments, the paramagnetic compound was one of several different hyperfine shift reagents for cation resonances. These were chosen to cover a range of potencies, in both magnitude and sign, of the shifts they produce. However, we also used a reagent which was incapable of inducing a hyperfine shift and thus could cause only a BMS shift. Although we report only 23Na spectra in this paper, the latter samples simulate the cases where one observes the water 1H resonance in experiments employing hyperfine shift reagents for cations. There have been a number of such investigations recently reported in the literature. The principles considered in this paper allow us to offer new interpretations for the results of several experiments published in the last few years.

Magnetic susceptibility shift selected imaging: MESSI.

Paramagnetic compounds are often used to enhance contrast in MRI by virtue of their increase in the kinetics of the relaxation of water 1H magnetization. Here, we demonstrate a method for contrast enhancement which is based on the resonance frequency shifts caused by the bulk magnetic susceptibility (BMS) effects of such compounds. This involves the frequency selective excitation in the absence of field gradients, during the imaging sequence, of a portion of the water 1H resonance which is rendered inhomogeneous by BMS shifts only. The image which results is of that portion of the sample which gives rise to the portion of the spectrum excited. A phantom sample which simulates some aspects of tissue, particularly blood vessels with different orientations in the magnetic field, was prepared. The contrast enhancement exhibited here avoids some of the distortions attendant to the use of paramagnetic reagents. This new approach can, in principle, utilize the natural BMS differences found in all tissue.

Secondary structure prediction for the spectrin 106-amino acid segment, and a proposed model for tertiary structure.

A collective secondary structure prediction for the human erythrocyte spectrin 106-residue repeat segment is developed, based on the sequences of nine segments that have been reported in the literature, utilizing a consensus of several secondary structure prediction methods for locating turn regions. The analysis predicts a five-fold structure, with three alpha-helices and two beta-strand regions, and differs from previous models on the lengths of the helices and the existence of beta-strand structure. We also demonstrate that this structural motif can be folded into tertiary structures that satisfy the experimental spectrin data and several general principles of protein organization.

Transforming growth factor-beta. Effect on soft tissue repair.

Previous studies have demonstrated that TGF-beta possesses many of the biologic properties necessary for acceleration of the normal wound healing process. We report that recombinant human TGF-beta 2 (rhuTGF-beta 1) increases wound strength and accelerates wound closure when applied topically to experimental wounds. Doses of 5 to 1,000 ng/wound increased wound strength in a dose-response manner and wound strength increase as high as 161% above control in the rat incisional wound model. Increased wound strength was observed as early as 3 days following rhuTGF-beta 1 application and continued to Day 28. In the rabbit ear ulcer model, acceleration of wound closure was observed following doses of 5 to 100 ng/wound applied a single topical application. No adverse effects of rhuTGF-beta 1 were observed. The amount of fibrous tissue, scar formation, and mitotic figures were not significantly greater than control. Epithelialization of rhuTGF-beta 1-treated wounds was not impeded. rhuTGF-beta 1 induced bone formation in the rabbit ear ulcer model but not in the rat incisional model, suggesting that precursor cells, such as perichondrial cells, are required for the bone forming activities of TGF-beta 1.

The PRP4 (RNA4) protein of Saccharomyces cerevisiae is associated with the 5' portion of the U4 small nuclear RNA.

We have combined oligonucleotide-directed RNase H degradation and immunoprecipitation in a study of the association of the Saccharomyces cerevisiae PRP4 protein with the U4-U6 complex. We have found that three oligonucleotides were able to direct nearly to completion the RNase H-specific cleavage of the target RNA molecules as they exist in splicing extracts. Immunoprecipitation of the degradation products with PRP4 antibody showed that the 5' portion of U4 small nuclear RNA (snRNA) and the 3' portion of U6 snRNA coimmunoprecipitated with the PRP4 protein. Micrococcal nuclease protection experiments confirmed further that the 5' portion and 3' end of U4 snRNA were very resistant to nuclease digestion, whereas the 3' portion of U6 snRNA was protected to only a very small extent. We conclude that the PRP4 protein of S. cerevisiae is associated primarily with the 5' portion of U4 snRNA in the U4-U6 small nuclear ribonucleoprotein (snRNP).

Mechanisms underlying anoxic hyperpolarization of hippocampal neurons.

The outward current evoked in CA1 neurons by brief anoxia is strongly voltage dependent and is abolished by an atropine-sensitive action of carbachol (and also when recording with a GTP gamma S-containing microelectrode). In this respect, it closely resembles the M-type K current, but the involvement of other, voltage-independent, carbachol-sensitive K channels has not been excluded. When the anoxic outward current is eliminated, an anoxic inward current is revealed, which may be Cl- mediated. It is suggested that an early release of Ca2+ from a dantrolene (and perhaps GTP)-sensitive internal store activates Ca-sensitive Cl channels, as well as carbachol-sensitive (mainly M-type?) K channels. The opposing Cl- and K currents would account for the variable depolarizing and hyperpolarizing effects of anoxia.

Accelerated healing of ulcer wounds in the rabbit ear by recombinant human transforming growth factor-beta 1.

A dermal ulcer wound-healing model was established in rabbit ear to examine the effects of recombinant human transforming growth factor-beta 1 (rhTGF-beta 1) in wound healing. Histomorphometric examination of the wounds indicate a biphasic healing response 7 days after a single application of rhTGF-beta 1 at the time of wounding. Statistically significant healing occurred at 5-100 ng but not at higher doses of 500 or 1000 ng rhTGF-beta 1/wound. Enhanced collagen synthesis as determined by [3H]proline incorporation occurred at 15 and 25 ng and was significantly depressed at 500 ng rhTGF-beta 1/wound. Multiple doses of 100 ng rhTGF-beta 1 applied to the wound at the time of wounding and for 3 days after wounding provided results comparable to the single application of growth factor. Delaying treatment 24 hr after wounding did not enhance wound healing compared with vehicle. Our findings suggest that rhTGF-beta 1 can be a valuable growth factor to improve the healing of ulcer wounds.

[Short-term curative effect of cultured Cordyceps sinensis (Berk.) Sacc. Mycelia in chronic hepatitis B].

33 cases of chronic hepatitis B. patients treated with cultured Cordyceps sinensis mycelia have shown that the drug improves the liver function, promotes negative transfer HBsAg, and markedly helps to raise plasma albumin, resist high gamma globulin and to adjust body immunocompetence. It is therefore suggested that cultured Cordyceps sinensis mycelia may be used as a medicine for chronic hepatitis B. patients in adjusting protein metabolism and correcting inversion of albumin and globulin.

Heterogeneous enzyme immunoassay of alpha-fetoprotein in maternal serum by flow-injection amperometric detection of 4-aminophenol.

A sandwich-type heterogeneous enzyme immunoassay with flow-injection analysis for alpha-fetoprotein (AFP) in human serum has been developed. 4-Aminophenol, the product of enzymatic reaction, is detected amperometrically. The interassay CV for this electrochemical enzyme immunoassay was less than 8.2%, with a minimum detection limit for AFP of 0.163 micrograms/L. The calibration curve had a linear range of 0.316-100 micrograms/L. Studies with 48 human maternal serum samples, comparing results by this method with those by a commercial kit, showed a good correlation (r = 0.961). This procedure provides an alternative method for determining low concentrations of AFP in human maternal serum.

The effect of Chinese tea on the occurrence of esophageal tumors induced by N-nitrosomethylbenzylamine formed in vivo.

Five groups of rats (22 per group) were intubated with precursors of N-nitrosomethylbenzylamine (NMBzA), i.e., methylbenzylamine (1 mmol/kg body wt) and sodium nitrite (0.5 mmol/kg body wt), and with five different varieties of Chinese tea twice a week. One positive control group was intubated with precursors of NMBzA without tea, and a negative control group was given tap water and sodium nitrite only. After 12 weeks of treatment, significantly fewer lesions and papillomas in the esophageal mucosa were found in the tea-treated group than in the positive control group. The results confirmed the anticarcinogenic effects of Chinese tea in rats by blocking the in vivo formation of NMBzA.

[Analysis of 366 cases of neonatal asphyxia].

366 cases of neonatal asphyxia were studied during 1983-1988. Different management regimes were used for neonatal asphyxia of different causes. The occurrence of neonatal asphyxia decreased from 4.9% in 1983 to 3.0% in 1988 in this hospital. Conversely, the occurrence of high risk pregnancy increased from 76.2% to 80.9% during the same period. Umbilical cord problems, hypertonic and too frequent uterine contractions, placental insufficiency and prolonged labor were the main causes of neonatal asphyxia. Use of magnesium sulphate for hypertonic contraction, correct estimation of fetal weight, and early diagnosis and selection of indications of C.S. are important for the prevention of neonatal asphyxia. Antepartum diagnosis and management of cord problems have already been discussed.

[Isolation and mapping of arbitrary single copy DNA fragment located on human chromosome 11p11-q11].

A library of genomic DNA has been constructed in EMBL3 lambda phage, from a Chinese hamster/human lymphocytes somatic cell hybrid carrying human chromosome 11 and 20. Recombinants containing human genomic DNA origin can be isolated from the hybrid cell genomic library by using species-specific probe. 8 single copy fragments have been isolated from 13 recombinants. One of them designated as FD11-1 has been identified on chromosome 11 by hybridized it with hybrid cell clone panel and mapped on chromosome 11p11-q11 by chromosome in situ hybridization. On chromosome 11, 3 linkage groups were reported, which located on 11p15, 11p13 and 11q13 respectively. Therefore, the FD11-1 will supply a new locus on chromosome 11 for linkage analysis. Endonuclease recognizing sites and potential recognizing sites on FD11-1 will guide the further RFLP studies.

The effect of Chinese tea on occurrence of esophageal tumor induced by N-nitrosomethylbenzylamine in rats.

Five groups of rats (40 rats per group) were given five different varieties of Chinese tea (1 g/50 ml) and another two groups of rats (a positive control and a negative control) were given tap water as drinking water. Except for the negative control group, all the animals were intubated with N-nitrosomethylbenzylamine (NMBzA, 5 mg/kg body wt/week). Half of the animals in each group were sacrificed after 6 weeks and the remaining animals at the end of 12 weeks. The incidences of esophageal mucosa lesions (16-59%) at the end of 6 weeks were significantly lower in the tea-treated rats than in the positive control group (100%). The incidences of esophageal tumor at the end of 12 weeks (42-67%) were also significantly lower in the tea-treated groups than in the positive control group (90%). The same phenomenon was found with respect to the size and number of tumors in each tumor-bearing animal. The relative anticarcinogenic effects of the five varieties of Chinese tea were different. The Fujian oolong tea and jasmine tea exhibited the strongest effects. The results suggest that Chinese tea can effectively inhibit the carcinogenesis caused by an N-nitroso compound.

Observation on the frequency of sister chromatid exchanges in the peripheral blood lymphocytes of healthy old women with Taichiquan exercise.

The frequency of sister chromatid exchanges (SCE) in the peripheral blood lymphocytes of eighteen healthy old women with 88 style Taichiquan exercise was studied. When compared with eighteen old women without exercise, the training group showed a decreased frequency of SCE in cultured peripheral blood lymphocytes at resting conditions. It is assumed that long term Taichiquan exercise provokes beneficial effects on lymphocytic chromosomes in the elderly.
