Roles of opioid peptides of PAG in analogous electro-acupuncture inhibition of experimental arrhythmia: analyzed by specific antisera microinjection.

Using specific antisera microinjected into periaqueductal grey (PAG), we further studied the mechanism of the analogous electroacupuncture, deep peroneal nerve stimulation (DPNS), inhibition on the experimental arrhythmia, hypothalamic stimulation-induced ventricular extrasystoles (HVE). Neither normal serum nor anti-leu-enkephalin antiserum interfered with DPNS inhibition; anti-dynorphin A (1-8) tended to attenuate the inhibition; and anti-beta-endorphin antiserum blocked it. The results provided further evidence for the hypothesis that beta-endorphin in PAG plays an important role in the neural circuit of DPNS inhibition on HVE.

Electrophysiological studies on the termination sites of double projection spinal dorsal horn neurons in the cat.

Some projection neurons in laminae III, IV and V of the feline lumbar dorsal horn have been found to be antidromically driven from stimulation of both the ipsilateral cervical dorsal and dorsolateral funiculi that are well-dissected and insulated from one another. No detectable responses can be evoked from these neurons when stimulation is applied to sites rostral to the lateral cervical nucleus and the dorsal column nuclei, suggesting that the dorsal and dorsolateral funicular branches of these neuron's axons terminate in the lateral cervical nucleus and the dorsal column nuclei, respectively. The results indicate that these neurons are an intersection set of the spinal cervical tract (SCT) and dorsal column postsynaptic (DCPS) neurons and have been thus named spinal cervical tract-dorsal column postsynaptic (SCT-DCPS) neurons.

Inhibitory effect of analogous electro-acupuncture on experimental arrhythmia.

Rabbits were used to study the inhibitory effect of analogous electro-acupuncture on the experimental arrhythmias, i.e. hypothalamic stimulation-induced ventricular extrasystole (HVE). The result revealed that HVE, which was due to an increased cardiac sympathetic activity, could be inhibited by deep peroneal nerve stimulation with an electrical current of low frequency and low intensity. Such effect is related to endogenous opioid peptides and serotonin in the arcuatus area, the periaqueductal gray and and the medial medulla. The acupuncture correction of arrhythmias may have the same mechanism.

Structural proteins and lipids in a virus, PBCV-1, which replicates in a Chlorella-like alga.

PBCV-1, a large dsDNA-containing virus which replicates in a Chlorella-like green alga, is composed of approximately 64% protein, 25% DNA, and 5-10% lipid on a weight basis. Polyacrylamide gel electrophoresis of the dissociated virus particle resolves 50 to 60 proteins which range in apparent molecular weight from 10,000 to 135,000. Two of these proteins are glycoproteins and at least four are located on the viral surface. The major lipids are phosphatidyl choline, phosphatidyl ethanolamine, and an unidentified component. The effect of organic solvents, surfactants, and chelating and reducing agents on viral infectivity and ultrastructure are reported. Inhibitor studies established that PBCV-1 protein synthesis occurs on cytoplasmic ribosomes.

Origin of cartilage laminae in MRI.

To understand the origin of the laminated appearance of cartilage in MRI (the magic angle effect), microscopic MRI (mu MRI) experiments were performed at 14-microns pixel resolution on normal canine articular cartilage from the shoulder joints. Two-dimensional images of the spin-spin relaxation time (T2) of the cartilage-bone plug at two angles (0 degree and 55 degrees) were calculated quantitatively. A distinct T2 anisotropy was observed as a function of the cartilage tissue depth. The surface and the deep regions exhibit strong orientational dependence of T2, whereas the upper-middle region exhibits little orientational dependence of T2. These three mu MRI regions correspond approximately to the three histologic zones in cartilage tissue. The results from the bulk T2 measurements agreed with these mu MRI results. Our studies show that the laminated appearance of cartilage in MRI is caused by T2 anisotropy of the tissue. We further suggest that the molecular origin of the T2 anisotropy is the nuclear dipolar interaction. The structure of the cartilage tissue indicates that the collagen meshwork defines this T2 anisotropy. The results show that the T2 anisotropy provides an indirect but sensitive indicator for the orientation of macromolecular structures in cartilage. The clinical implications of this anisotropy are discussed.

Decreased high-density lipoprotein cholesterol and serum apolipoprotein AI concentrations are highly correlated with the severity of Alzheimer's disease.

Serum apolipoprotein (apo) AI concentration was studied in 98 Alzheimer's disease (AD) patients (77.56+/-8.83 years) and 59 healthy, elderly controls (75.37+/-5.27 years). ApoAI levels were significantly lower (p<10(-7)) in AD patients. An apoAI cutoff value of 1.50 g/L, could distinguish between the two groups with a sensitivity of 71% and a specificity of 69%. ApoAI levels were highly correlated with mini-mental state (MMSE) scores of patients (p<0.0001). These relationships remained significant after adjustment for multiple testing. Our findings raise the question of the potential implication of apoAI in the etiopathology of AD and bring serum apoAI concentration to the fore as an important biochemical marker.

The adenylosuccinate synthetase-1 gene is activated in the hypertrophied heart.

Adenylosuccinate synthetase 1 (ADSS1) functions as an important component in adenine nucleotide biosynthesis and is abundant in the heart. Here we report that the Adss1 gene is up-regulated in two in vivo rodent models of surgically induced cardiac hypertrophy. In addition, we examined an in vitro hypertrophy system of rat neonatal cardiomyocytes treated with angiotensin II to study Adss1 gene regulation. We show that this stimulus triggers a signaling cascade that results in the activation of the Adss1 gene. The induction of Adss1 gene expression was blocked by cyclosporin A in vitro, suggesting that calcineurin, a calmodulin activated phosphatase, is involved in this signaling pathway. Consistent with this view we provide evidence that the induction of Adss1 by angiotension II requires the presence of an NFAT binding site located 556 base pairs upstream of the Adss1 transcription start site. We propose that ADSS1 plays a role in the development of cardiac hypertrophy through its function in adenine nucleotide biosynthesis.

DNA damage in buccal epithelial cells from individuals chronically exposed to arsenic via drinking water in Inner Mongolia, China.

The purpose of this pilot study was to assess DNA damage in buccal cells from individuals chronically exposed to arsenic via drinking water in Ba Men, Inner Mongolia. Buccal cells were collected from 19 Ba Men residents exposed to arsenic at 527.5 +/- 23.7 micrograms/L (mean +/- SEM) and 13 controls exposed to arsenic at 4.4 +/- 1.0 micrograms/L. DNA fragmentation by the DNA ladder and TUNEL assay were used to detect DNA damage in buccal cells. In the DNA ladder assay, 89% (17/19) of the arsenic-exposed group showed < 100 bp DNA fragments, in contrast to 15% (2/13) of the controls (p < 0.0001). For the TUNEL assay, the mean frequencies of positive cells were higher in the exposed group (15.1%) than in the controls (2.0%) (p < 0.0001). This study showed that high arsenic exposure via drinking water resulted in DNA damage and DNA fragmentation in buccal cells thus may be an appropriate biomarker for assessing chronic effects of arsenic in humans. A study investigating DNA fragmentation from the individuals with low levels of arsenic exposure in this population is in progress.

Metabolism of subtoxic levels of selenium in animals and humans.

Since high levels of selenium are used as cancer chemopreventive agents in animals and humans, a better understanding of the metabolism of subtoxic levels is desirable. Absorption from rat small intestine using in situ double perfusion, ligated intestinal segments, and brush border membrane vesicles (BBMV) was used to study selenium absorption. A level of 1.2 mM intraluminal selenite was required to inhibit 50 percent of the transepithelial transport of 3-0-methylglucose, indicating a high tolerance of the intestinal tract to selenium. The relative efficiency patterns for uptake of different selenocompounds during in vitro perfusion and in vivo ligated segments were identical with selenomethionine (SeMet) > selenate > selenite. In contrast, selenite was taken up most rapidly by BBMV, followed by SeMet and selenate in decreasing order. Ligated segments, double perfusion experiments, and uptake by BBMV indicated that selenium as selenodiglutathione or selenodicysteine was taken up faster than when present as selenite. Selenate and SeMet appeared in the vascular effluent largely unchanged, but selenite was metabolized extensively during absorption. Most of the selenium in plasma from subjects living in a high selenium area of China was associated with albumin, which is likely a result of high dietary intake of SeMet. Cracked fingernails and extensive hair loss were the symptoms of selenium toxicity in these individuals. Low adverse effect level of dietary (mean LOAEL) selenium was calculated to be about 1540 +/- 653 micrograms per day (or 28 micrograms/kg body weight) and the maximum safe dietary (mean NOAEL) selenium was calculated to be 819 +/- 126 micrograms per day (or 15 micrograms/kg body weight).

Modulations of breast fibroblast and carcinoma cell interactions by a dextran derivative (CMDB7).

Modulation of interactions between human breast carcinoma cells (MCF-7ras) and fibroblasts from normal breast tissue (MG3) and from a post-radiation fibrosis with recurrent breast carcinoma (FPR7) by a dextran derivative (CMDB7) was investigated. In a coculture system, MCF-7ras proliferation was increased (50%) by fibroblasts, and fibroblasts showed a 84% growth increase with MCF-7ras. This co-stimulation did not depend on fibroblast origin. CMDB7 can inhibit the growth of MCF-7ras but not the growth of fibroblasts. The addition of CMDB7 blocks the MCF-7ras stimulation by fibroblasts but not the fibroblast stimulation by MCF-7ras. Fibroblast-Conditioned Medium stimulated over 2-3 fold MCF-7ras's DNA synthesis but CMDB7 did not influence this paracrine stimulation. IGFI, IGFII stimulated MCF-7ras's DNA synthesis whereas bFGF inhibited it. CMDB7 did not block IGFI and IGFII's stimulatory effects but increased bFGF's inhibitory effects. These results indicate that CMDB7 can inhibit fibroblast-breast cancer cell interactions possibly by interfering with paracrine growth factor loops.
