Changes in sarcolemmal proteins in subacute myocardial infarction in the dog.

Previous studies have shown degradation of cardiac structural proteins and disruption of the sarcolemma as a result of acute myocardial infarction. However, there is no evidence to date on changes in sarcolemmal membrane proteins induced by experimental subacute myocardial infarction. We studied subepicardial layers overlying myocardial infarct 4 days following ligation of the left anterior descending coronary artery in 12 dog hearts. We first demonstrated that this layer provides the anatomic-electrophysiologic substrate for reentrant arrhythmias using activation mapping techniques and histologic correlations. The makeup of membrane proteins was studied using SDS polyacrylamide gel electrophoresis, peptide mapping, and laser densitometry. Sarcolemmal membrane proteins were isolated by ultracentrifugation through a sucrose gradient. We found that a sarcolemmal polypeptide (MW 126,000; n = 12) in the normal tissues has a different mobility than the corresponding protein (MW 124,000; n = 12) of the ischemic tissues although their peptide analysis appeared similar, suggesting that the protein undergoes a post-translational modification. In addition, two proteins (MW 75,000; n = 12 and MW 88,000; n = 12) were present in greater amount in the ischemic than in the control tissues suggesting either acceleration in protein synthesis or slow down of degradation turnover. These results demonstrate that specific changes occur in membrane proteins subjected to ischemic insults which might be responsible for membrane alterations following ischemia and may contribute to the abnormal electrophysiologic properties and arrhythmia seen in vivo at this stage.

The ability of polymorphonuclear leukocyte priming agents to overcome influenza A virus-induced cell dysfunction.

The major mortality and morbidity resulting from influenza virus infections are due to secondary bacterial infections which occur in association with virus-induced inhibition of polymorphonuclear leukocyte (PMNL) function. The present study was undertaken to determine if compounds which prime PMNL function to subsequent stimulation with N-formylmethionyl-leucylphenylalanine (FMLP) or phorbol 12-myristate 13-acetate (PMA) can overcome influenza A virus (IAV)-induced inhibition of the PMNL chemiluminescence response to these stimuli. Granulocyte-macrophage colony stimulating factor (GM-CSF), guanosine triphosphate (GTP), and 1-oleoyl-2-acetylglycerol (OAG) were able to prime the PMNL response to FMLP and/or PMA and totally or partially overcome IAV-induced PMNL dysfunction in cells stimulated with FMLP or PMA. A direct correlation was found between the extent of PMNL priming due to GM-CSF, GTP, and OAG and the capacity of these compounds to overcome virus-induced PMNL dysfunction. The implications of these findings in regard to the mechanism by which priming agents overcome IAV-induced cell dysfunction and the potential of these compounds as therapeutic agents to treat secondary bacterial infections are discussed.

[Effect of acupuncture on neuronal loss induced by axotomy in the rat hypoglossal nuclei].

It was reported previously that acupuncture promotes regeneration of rat's peripheral nerve. In order to study the effect of acupuncture on recovery of motoneuronal lesion and regeneration of central nervous system, we studied the effect of acupuncture on neuronal loss induced by axotomy in the rat hypoglossal nuclei. A total of 6 pairs of rats (acupunctured and controls in pairs) were used, (each pair of) the rats were paired of the same litter and sex. After the right hypoglossal nerves were severed, rats were reared for 14 days. In this period, we gave acupuncture therapy to the acupunctured rats but nothing to the controls. After perfusion, serial paraffin sections of brain stems of rats were cut and stained with cresyl violet. Serial sections per rat were used for counting the perikaryon of the hypoglossal nuclei. The reduced percentage of neuronal population followed axotomy was calculated. It was found that neuronal loss was less severe in acupunctured rats. Difference in the reduced percentage of neuronal population between acupunctured rats and controls was significant statistically. It was shown that acupuncture increased survival of lesioned neurons, and thus acupuncture may be helpful to the recovery of neuronal lesion. The detailed mechanism is going to be explored.

Evolutionary conservation of target sequences for cis-acting regulation in c-myc exon 1 and its upstream region.

Transcriptional control of the c-myc proto-oncogene, an important factor in cellular growth, differentiation and in the genesis of various neoplasms, is mediated by multiple positive and negative regulators in the 5' end region of the gene. Here, we report the nucleotide sequence of the first c-myc exon and its upstream region from woodchuck, a rodent which can develop liver tumors associated with c-myc activation [Möröy et al., Nature 324 (1986) 276-279]. Alignment of these sequences with the corresponding human and murine regions shows a surprisingly high homology between woodchuck and human, and suggests the absence of species-specificity in the fundamental regulatory elements which govern c-myc expression.

Interleukin 1 induces ferritin heavy chain in human muscle cells.

Interleukin 1 alpha (IL-1) and tumor necrosis factor alpha (TNF) are two monokines which play a prominent role in the response to inflammation and injury. We recently observed that TNF leads to an increase in the synthesis of the heavy chain of ferritin, suggesting that TNF may be involved in iron homeostasis (Torti et al. (1988) J. Biol. Chem. 263, 12638-12644). The experiments reported here demonstrate that in cultured human muscle cells, IL-1 induces ferritin H mRNA and protein as effectively as TNF. TNF and IL-1 were additive in their effects on ferritin H expression, and IL-1 induction of ferritin H was not blocked by anti-TNF antibodies. Ferritin H induction was a specific response not observed with beta or gamma interferon, nor with transforming growth factor beta. Both differentiated myotubes as well as myoblasts responded to IL-1 with the induction of ferritin H. These results suggest that monokine-mediated alterations in the subunit composition of the ferritin molecule may be of biological relevance in the response to inflammation and injury.

Quantitative study of the structure-stability relationship of Tc complexes.

By studying structural parameters (van der Waals radius and bond length from x-ray crystallography) of more than 100 known Tc compounds and using the cone packing model, a stability indicator of the Tc compounds was derived. The indicator is based on the solid angle factor sum (SAS), which is calculated from van der Waals radii and bond lengths between all coordinating atoms and Tc. The average SAS value is 0.97 +/- 0.13. The SAS value reflects the limits of the packing of ligand around Tc. The quantitative calculation of the SAS is useful for predicting stability and designing new Tc radiopharmaceuticals.

Quantitative study of the structure-stability relationship of TcvO(III) complexes.

This paper describes the quantitative structure-stability relationship of TcvON2S2 compounds using a stability indicator based on the solid angle factor sum (SAS) for predicting the in vitro stability. The SAS values of six TcvON2S2 and one TcvON4 (D,L-HMPAO) complexes were calculated from their x-ray crystallography data. The rank order of in vitro stability of these compounds, as measured by ligand exchange reaction, is directly related to that of the SAS values. The SAS values are potentially useful for predicting stability and designing new 99mTc radiopharmaceuticals.

[Ultrastructural study of stromal elements of lung carcinoma].

We studied the fine features of stromal elements in 33 cases of human lung carcinoma by electron microscopy. Cancer cells showed the damages to the basement membrane and matrix. On the other hand, stromal elements showed changes responding to the invasion of cancer cells. Lymphocytes and macrophages showed the activity of killing cancer cells. Most cell directly contacts the myofibroblast and fibroblast. Matured myofibroblast, early myofibroblast, active fibroblast and resting fibroblast were present near the nest of carcinoma. We discussed the roles and significance of stromal reactions.

[Protective effect of 2-[p-(dimethylamino) styryl] pyridine methiodide (DSPM) on acute experimental myocardial ischemia].

ip DSPM 5 mg/kg 1 or 4 h before excision of the heart was found to prevent damage to the heart due to global myocardial ischemia. Compared with the controls, the heart rate, coronary resistance, contractile force and edema of the heart were all greatly improved during the 60 min reflow after 25 min of no-flow, especially the heart pretreated with DSPM 4 h prior to excision. In addition, DSPM 1.5, 6 mg/kg markedly reduced the myocardial Ca2+ content measured by isoproterenol in mice. DSPM at higher dosage also lowered the normal myocardial Ca2+ content. These results suggest that DSPM may have a protective effect on myocardial ischemia which might be associated with its calcium antagonistic action.

[Studies on the interrelationships between some acupoints on pericardial channel and heart].

The theory of acupoint-Zangfu correlation is one of the essential contents of the doctrine of Channel and Collateral. Being based on this theory, a study has been done from the experimental research point of view in our laboratory since 1979, to approach the specific interrelationships between some acupoints on the pericardial channel and heart, as well as their functional regularities, and has served for clinical practice, which provided the basis for the research on the connective pathways between the pericardial channel and heart. Coronary heart disease, one has been treated effectively by acupuncture, was selected and, by imitating the early stage of myocardial infarction, the reperfusion model of coronary artery in rabbits was designed to produce a reversible acute myocardial ischemic injury. As the indices, the ST segments of second standard lead and augmented unipolar limb leads of electrocardiogram were obviously improved when electroacupuncture (EA) was applied to the points "Jianshi" or "Neiguan", these could be observed repeatedly. Then the factors influencing the effects of acupuncture were analysed. The effects of EA at six points on "pericardial channel", "urinary bladder channel" and "liver channel" were compared. Results showed that both "Ximen" and "Weishu" were able to accelerate the restoration of acute myocardial ischemic injury. However, EA at "Zhongdu" and "Xiguan" could hardly affect, this indicates that "where the channel passes through, where it affects on".(ABSTRACT TRUNCATED AT 250 WORDS)

Influence of EA on the electrical conductance in the marginal area of the acute ischemic myocardium in the rabbit.

AMI was produced by occlusion of LVB for 30 min in the rabbit, lengthening the conductive time of myocardium in the ischemic area and resulting in asynchronous activation state of myocardium. EA could facilitate the conductance of electric activation in the ischemic marginal area and make the activation of myocardial cells in this area synchronous which is of importance in blocking the formation of reentrant circuit. The finding suggests that EA possesses an antiarrhythmic effect.

[Ultrastructural study of the parenchyma of lung carcinoma].

Thirty three cases of human lung carcinoma were studied by both light microscopy and electron microscopy. The diagnoses of 24 cases initially made by light microscopy were changed, extended, or supplemented by electron microscopic observation. Nineteen cases showed heterogeneity. Bidirectional or tripartite differentiation was frequently found in the same case, even in the same cell. The diagnosis and heterogeneity of lung carcinoma were discussed.

[Clinicopathologic and ultrastructural studies of olfactory neuroblastoma].

The clinicopathologic and ultrastructural studies of 12 cases of olfactory neuroblastoma were made. The ages of patients ranged from 2 to 58 years with a mean of 32. Eleven cases occurred in men and 1 case in woman. An acidophilic fibrillary intercytoplasmic background was present in every case. Its amount varied focally. Ultrastructurally, the acidophilic fibrillary background was composed of neuronal cell processes. Homer-Wright rosettes were found in 7 cases, Flexner type rosettes in 4 cases, 3 of which shared both Homer-Wright and Flexner type rosettes. Almost all cases with rosettes also had apparent acidophilic fibrillary intercytoplasmic background. So, we think that acidophilic fibrillary intercytoplasmic background was the most useful diagnostic feature by light microscopic observation. The diagnosis of olfactory neuroblastoma by electron microscopy was mainly based on neuronal processes, neurosecretory granules, microtubules, neurofilaments, synaptic-like junctions and occasionally both microvilli and olfactory vesicles at the luminal borders of Flexner type rosettes.

[Effects of fu-zheng jie-du decoction and cyclophosphamidum on the production of tumor necrosis factor in mice].

Fu-Zheng Jie-Du decoction (FZJD) was an adjuvant drug of anti-tumor. In the present paper, the authors studied the effects of FZJD and cyclophosphamidum (CTX) on tumor necrosis factor (TNF) production of peritoneal macrophages (PM) of normal and EMT6 solid tumor-bearing mice. The results showed that in vitro, lipopolysaccharide (LPS) could induce TNF production of PM of normal mice; in contrast; FZJD and CTX were unable to induce TNF and inhibited the LPS effect. In vivo, CTX could not influence the TNF production of PM of tumor-bearing mice but FZJD could enhance the TNF production of PM of tumor-bearing mice treated with CTX.

An immunoelectron microscopic study of peptide substances in the rat neurohypophysis.

The ultrastructural localization of peptide substances, such as vasopressin (VP), substance P (SP), enkephalin (ENK) and somatostatin (SRIF), and the distribution of peptidergic nerves in rat neurohypophysis were studied by immunoelectron microscopy. The results show that VP, SP, ENK and SRIF immunoreactive products are located in large granular vesicles (110-150 nm in diameter), at the periphery of microvesicles and on the outer membrane of mitochondria. The VP-, SP-, ENK- and SRIF- containing nerve fibers are distributed at the periphery of capillaries and in the vicinity of pituicytes. VP- and ENK-positive nerve terminals form axo-axonic synapses with negative terminals. VP- and ENK-terminals are pre- or postsynaptic elements. Axo-axonic synapses formed by two SRIF- positive terminals were also found. In addition, ENK- and SP-positive terminals with pituicytes form synaptoid structures respectively. The authors have discovered VP-positive pituicytes for the first time.
