Mitomycin C-induced acceleration of liver cell polyploidization in adult rat after partial hepatectomy.

Mitomycin C, a DNA cross-linking agent, was administered for a week intraperitoneally to a normal 9-week-old rat in which hepatocyte proliferative activity had almost ceased. In the rat treated with mitomycin C, the number of polyploid cells with the DNA amounts more than 4C was not increased in comparison with that in the control rat without any treatment. However, the partial hepatectomy in the rat pretreated with mitomycin C provoked prominent hepatocyte polyploidization much greater in degree than that found in the hepatectomized rat without mitomycin C treatment. These results seem to indicate that the existence of cross-linking DNA damages is a latent factor necessary for the induction of abortive mitosis of a cell after completion of DNA synthesis, which results in the production of a mononuclear polyploid cell in one-step higher DNA class. Cross-linking DNA damages and DNA synthesis are the essential factors for the manifestation of polyploidization.

A method of DNA histogram analysis by computer and its evaluation by simultaneous combination with 3H-thymidine autoradiography.

A method for determining the fractions of cells in the G1, S, and G2 + M phases of the cell life cycle, by quantifying DNA histograms derived from static fluorescence cytophotometry, was evaluated by simultaneous combination with 3H-thymidine autoradiography. DNA histograms were obtained by cytofluorometry on the Feulgen-stained autoradiographs of HeLa cells, and mouse and rat hepatocytes, after DNA labelling with 3H-thymidine. The synthetic histogram determined by "sum of discrete normal curves" technique was fitted to the experimental data according to a weighted least-squares method by a desk-top computer (HP 85F). The mean relative percent deviations of estimated cell cycle phase fractions from the actual phase fractions determined directly on an autoradiograph was 6.6 +/- 3.3%.

Studies on the binding of adenylyl-3', 5'-cytidine to ribonuclease.

The interaction of adenylyl-3',5'-cytidine (ApC) with ribonuclease-A (RNAase-A) was studied by steady-state kinetics and ultraviolet difference spectroscopy. X-ray difference Fourier synthesis at 4 A resolution was also used to study the binding of ApC to RNAase-S. Unlike well-studied compounds like uridylyl-3',5'-adenosine, ApC binds in an unique way: (1) the cytidine moiety is bound to the B1 and R1 sites, (2) the adenosine moiety protrudes to the solution and is not fixed spatially and (3) the phosphate group is bound to the non-specific site (the "Po site") previously postulated (Sawada, F. and Irie, M. (1969) J. Biochem. (Tokyo) 66, 415--418) as the binding site for the 5'-phosphate of uridine 2',5'-diphosphate or uridine 3',5'-diphosphate. This conclusion is consistent with that derived for adenylyl-3',5' -4-thiouridine based on CD difference spectroscopy (White, M.D., Keren-Zur, M. and Lapidot, Y. (1977) Nucleic Acid Res. 4, 843--851). The "Po site" is most likely the epsilon-amino group of Lys 66.

Crystal structure of monoclinic ribonuclease-S at 4 A resolution. The mode of binding of 4-thiouridylic acid and a fragment of folic acid, p-aminobenzoylglutamic acid.

A four-A electron density map was calculated for the monoclinic crystal of ribonuclease-S (RNase-S) based on two heavy-atom derivatives. Close geometrical similarity was found between the two crystallographically independent RNase-S molecules (called molecules ZA and ZB) in this crystal and that (called molecule Y) in the trigonal crystal. Using the rotational and translational parameters relating these three molecules, it was established that the crystallographic two-fold symmetry between the two molecules ZA in the monoclinic crystal was exactly identical to that between the two molecules Y in the trigonal crystal, suggesting the tendency of RNase-S molecules to associate in this way although the interaction is weak. The 4-A difference Fourier maps calculated for the monoclinic crystal established the following conclusions. (1) 4-Thiouridine-2'(3')-monophosphates binds to the B1 and R1 sites like other pyrimidine nucleoside-2'(3')-monophosphates as expected from previous spectrophotometric studies, but not to the B2 site even at the concentration of 20 mM. An attempt to visualize the photoproduct generated by irradiation of near-ultraviolet light in this complex failed. (2) p-Aminobenzoylglutamic acid, a fragment of folic acid, seems to bind to RNase-S with its benzene ring close to the B2 site and the alpha-carboxylate group close to the p1 site. The model is compatible with most of the chemical results obtained by Sawada et al. ((1977) Biochim. Biophys. Acta 479, 188-197).

Left vocal cord paralysis in cases of systemic lupus erythematosus.

Two cases, a 41-year-old female and a 53-year-old female presenting with systemic lupus erythematosus and left vocal cord paralysis, are described in this paper. In both cases, laryngeal lupus and pulmonary hypertension were not detected. The laryngeal symptoms and signs were not improved by treatment with steroids, despite the systemic lupus erythematosus being under control.

Malignant endocrine carcinoma of the stomach.

Small cell carcinoma and small portions of poorly differentiated adenocarcinoma of the stomach sometimes differentiate into secretory cells. We experienced a pure endocrine carcinoma of the stomach. The patient, who had a giant mass in the greater curvature of the body of the stomach and regional lymph node metastasis, underwent total gastrectomy. Immunohistologic staining of the resected specimens revealed that the tumor was an endocrine carcinoma. The tumor cell shape in the present case was polygonal or oval and the cell size was relatively large. The cell nucleus was coarse chromatic and nucleoli were seen, whereas, the nucleus of small cell carcinoma often shows a fine chromatic pattern and nucleoli are not seen. On the basis of these findings, the present case was considered to be a pure endocrine carcinoma which was not classified as a small cell carcinoma.

Computed tomography and angiogram of primary hepatic rhabdomyosarcoma: report of two adult cases.

Two adult cases of primary hepatic rhabdomyosarcoma, one of which was associated with hepatocellular carcinoma and had double malignant tumors, are described with emphasis on the radiologic features. On precontrast computed tomography, the lesion appeared as a well-defined hypodense mass like an abscess, while linear mottled enhancement was observed on the postcontrast scan. Angiography revealed displacement of the hepatic arteries and portal veins, irregular neovascularity in the peripheral portion, and scant tumor stain without dilatation of the feeding artery. Review of the literature revealed only 10 previous reports.
