The blood vascular architecture of the rat thyroid gland: a scanning electron microscope study of corrosion casts.

The blood vascular bed of the rat thyroid gland was reproduced by injection of a methacrylate casting medium and observed with a scanning electron microscope. The rat thyroid gland received the superior and inferior thyroid arteries and emitted the superior and inferior thyroid veins. Anastomosis between the interlobular arteries or between the interlobular veins was frequently observed in the thyroid gland. The arteriovenous anastomosis was rarely observed between the terminal branches of the lobular arteries and veins. The thyroid blood vascular bed was divided into lobular units which consisted of basket-like capillary networks surrounding the thyroid follicles; a small lobular unit consisted of a few networks, whereas a large one of fifty or more networks. Sizes and forms of the networks varied widely in each case. However, the networks in the superficial layers of the lateral parts of the thyroid gland were typically most developed. Regardless of its size and form, each network always received a proper efferent vessel continuous with the lobular vein, though it was sometimes provided with an accessory afferent or efferent vessel. Only occasionally were the adjacent networks fused with each other or connected by transfollicular capillaries. Thus, the present data suggest that each follicular capillary network is a fairly independent functional-unit in the thyroid microcirculation. The capillaries of the network were sinusoidal in nature and sometimes protruded fine projections which indicated the neogenesis of capillaries. The blood vascular bed of the newborn rat thyroid gland was not always differentiated into basket-like capillary networks.

The blood vascular architecture of the rat testis: a scanning electron microscopic study of corrosion casts followed by light microscopy of tissue sections.

The blood vascular bed of the rat testis was studied by scanning electron microscopy (SEM) of corrosion casts and by light microscopy of tissue sections. The testicular artery penetrates the pampiniform plexus and gives rise to the intertubular arterioles. Each of these arterioles courses in the intertubular connective tissue column, and gives off intertubular and peritubular capillaries. The intertubular capillaries pass the intertubular connective tissue column, whereas the peritubular capillaries reach the peritubular connective tissue sheet. The intertubular and peritubular capillaries anastomose with each other and converge into the intertubular venules in the intertubular connective tissue columns. Thus, the blood vascular bed of the rat testis consists of hexago- or pentago-columnar capillary networks which commonly surround the seminiferous tubules. The Leidig's cells are preferentially observed in the intertubular connective tissue columns. One of the intertubular capillaries is consistently thick, and directly continues into the intertubular venules (arteriolo-venular capillary channels), which finally drain into the pampiniform plexus. These findings suggest that the male sex hormone, testosterone, as secreted by the Leidig's cells, is discharged into the intertubular capillaries and then mainly carried by the arteriolo-venular capillary channels and intertubular venules into the pampiniform plexus. This specialized drainage may ensure the presence of highly concentrated testosterone in the pampiniform plexus and allow the testosterone-exchange from the pamipiniform plexus to the testicular artery. The arteriolo-venular capillary channels may also eliminate blood congestion in the testis to enhance the efficiency of the heat-exchange mechanism between the testicular artery and pampiniform plexus. Many arterio-arterial and arterio-venous anastomoses occur, which may regulate the blood flow within the testis.

Blood vascular beds of rat adrenal and accessory adrenal glands, with special reference to the corticomedullary portal system: a further scanning electron microscopic study of corrosion casts and tissue specimens.

Blood vascular casts of the rat adrenal glands were observed with a scanning electron microscope. The cortical capillary plexus drains, through the corticomedullary venous radicles, into the subcortical veins continuous with the medullary collecting veins. The medullary capillary plexus drains into the corticomedullary venous radicles, subcortical veins and medullary collecting veins. No portal vessel was noted between the cortical and medullary capillaries. These findings indicate that the cortical blood rich in glucocorticoids preferentially and continuously flows into the corticomedullary venous radicles, subcortical veins and medullary collecting veins all three of which are fenestrated in type, and also suggest that the vascular route from the cortical capillaries to the medullary collecting veins functions as a substitute for the portal system, controlling the biosynthesis of catecholamines in the adrenal medulla. The vascular bed of the accessory adrenal gland (extra-adrenal cortical or chromaffin body) is sometimes annexed to that of the adrenal gland. On rare occasions, the vascular beds of the extra-adrenal cortical and chromaffin bodies fuse with each other. Additional scanning of tissue samples confirmed the direct drainage of cortical capillaries into the medullary veins and also the endothelial fenestrations of these capillaries and veins.

Removal of apolipoprotein E-enriched high density lipoprotein by LDL-apheresis in familial hypercholesterolaemia: a possible activation of the reverse cholesterol transport system.

The presence of apo E-containing HDL in familial hypercholesterolaemia was investigated and its removal by LDL-apheresis using a dextran sulphate cellulose column was demonstrated by measurement of the apo E/apo A-I molar ratio of serum and by nondenaturing polyacrylamide gel electrophoresis followed by immunoblotting. The molar ratios of apo E/apo A-I in the density greater than 1.063 kg/l fraction of serum obtained from two homozygous patients with familial hypercholesterolaemia were higher (0.021 and 0.030) than that from normal subjects (mean +/- SE 0.011 +/- 0.002) (P less than 0.05). Polyacrylamide gel electrophoresis and immunoblotting showed an increase in apo E-containing HDL similar to HDL2, in the plasma obtained from the homozygous patient with familial hypercholesterolaemia. The increased amounts of apo E-enriched HDL were removed from plasma by adsorption with a dextran-sulphate cellulose column. These results suggested that LDL-apheresis using the dextran-sulphate cellulose column, may cause an increase in the turnover rate of the apo E-containing HDL and thus facilitate cholesterol removal from the peripheral tissues.

lon-sulA regulatory function affects the efficiency of transposition of Tn5 from lambda b221 cI857 Pam Oam to the chromosome.

In transposition of Tn5 from lambda b221 cI857 Pam Oam rex::Tn5 to the chromosome, lon mutants of Escherichia coli K12 are less efficient than lon+ parents. Reduced frequency of transposition of Tn5 is recovered to the wild type level by adding DL-pantoyl lactone, an agent which suppresses Lon- phenotypes. A suppressor mutation, sulA (or sfiA), suppresses the deficiency of transposition as well as other known lon-associated phenotypes, whereas a lon sulB (or sfiB) double mutant is phenotypically Lon+ but still transposition-deficient. An additive effect of the sulA genes on teh chromosome and on a high copy number plasmid is found in inhibiting transposition of Tn5. A hypothesis that the SulA product someway regulates transposition of Tn5 negatively is proposed.

Blood vascular architecture of the rat parathyroid glands: a scanning electron microscopic study of corrosion casts.

Blood vascular beds of the rat parathyroid glands were reproduced with a methacrylate casting medium and observed with a scanning electron microscope. The rat possesses only a single pair (one left and the other right) of parathyroid glands. Each gland was found to contain a rich capillary network which was completely isolated from the capillary plexus of the thyroid gland. Each parathyroid gland received some small afferent arteries from the superior thyroid artery and emitted a thick efferent vein continuous with the superior thyroid vein. The capillary network of the parathyroid gland consisted of freely anastomosing capillaries. The afferent arteries were divided in the superficial and deep layers of the gland. The thick efferent vein arose in the deep layer of the gland. Some small or accessory efferent veins arose in the superficial layers of the gland. These also drained into the superior thyroid vein.

[Aging phenomena of granulosa cells in the human ovary].

The human ovary ceases its function far earlier than that of other endocrine organs. For the study of the mechanism of decreasing ovarian function with aging, the functional changes in follicular granulosa cells (G-cells) which are indispensable to existing of oocytes were investigated. 1) The mitotic frequency which was calculated in G-cells of antral follicles in tissue preparation decreased markedly in over 40 year old women who are in climacterium. 2) Also the mitotic frequency of cultured G-cells which are removed factors of aging from a living body decreased after 40 years old. 3) In order to study DNA synthetic ability of cultured G-cells, 3H-thymidine up-take to the nuclei of G-cells was examined. It was also decreased in climacterium. 4) E2 production of cultured G-cells also decreased in women in 40's. 5) The binding activity of 125I-FSH and 125I-LH on the membranes of cultured G-cells were also decreased with aging. These data indicate that the decrease of DNA synthetic ability, multiplication, steroidogenesis, and gonadotropin binding activity of granulosa cells which are indispensable to existing oocyte is playing major role in aging and cessation of human ovary.

[Fundamental and clinical study on cefpiramide in obstetrics and gynecology. Obstetrics and Gynecology Study Group for Cefpiramide].

Fundamental and clinical studies on a new cephalosporin antibiotic, cefpiramide (CPM), was carried out under a joint study program, in order to evaluate the usefulness of the drug in treating infection of the female genital organs. The results obtained were as follows: CPM was readily transported to female genital organ tissues, and the concentrations of the drug exceeded 35 micrograms/g in various organ tissues in about 1 hour, following intravenous injection of 1 g. A level of more than 2 micrograms/g was maintained even 14 hours after the injection. The transport of CPM to various tissues was also studied following intravenous drip infusion of 1 g for 1 hour. The concentrations in tissues were slightly low but similar to those following intravenous injection. The peak concentration of the drug in the dead space exudate was 3.1-20.4 micrograms/ml, following intravenous injection and intravenous drip infusion of 1 g. The MIC80 of CPM were 3.13-12.5 micrograms/ml against S. aureus, Klebsiella sp., P. mirabilis and P. aeruginosa. Clinical effects of CPM were analyzed in 158 patients, including 56 cases with intrauterine infection, 37 cases with intrapelvic infection, 22 cases with external genital infection, 31 cases with adnexitis, 6 cases with postoperative wound infection and 6 cases with other infections. Excellent response was seen in 28 cases (17.7%), good response in 120 (75.9%) poor response in 10 (6.3%). The rate of response was calculated as 93.7%. Safety of the drug was analyzed in 258 patients, and side effects occurred in 4 (1.6%). Of these 4 patients, rash was in 1 patient, heat sensation in 1 patient, nausea in 1 patient and rash accompanying edema in 1 patient. Abnormal values in clinical laboratory findings were seen in 7 patients. Elevations of transaminase were seen in 5 patients and decrease of platelet was seen in a patient, and then elevations of transaminase with decrease of platelet was seen in a patient, and no other changes of particular note appeared.
