Translocation, activation and association of protein-tyrosine kinase (p72syk) with phosphatidylinositol 3-kinase are early events during platelet activation.

We have previously reported that a non-receptor-type protein-tyrosine kinase p72syk, exists in both membrane and cytosolic fractions in porcine platelets and is activated after thrombin stimulation. To facilitate the understanding of the function of p72syk, we have investigated the topological features, kinase activities and the interaction with another signal-transducing molecule, namely phosphatidylinositol 3-kinase, during platelet activation. Membrane and cytosolic fractions were separated from thrombin-treated porcine platelets, and the amount of p72syk was quantified by the immunoblot technique or the kinase activity of each fraction was determined by an immunoprecipitation kinase assay. After stimulation by thrombin, cytosolic p72syk rapidly translocated to the membrane fraction within 10 s and there was also a significant increase in the amount of p72syk in the cytoskeletal fraction. The autophosphorylation activity of membrane-associated p72syk significantly increased approximately tenfold and reached a maximum at 10 s; the activity subsequently decreased to almost the basal level within 120 s. For similar time courses, association of p72syk with phosphatidylinositol 3-kinase and tyrosine phosphorylation of p72syk were observed. These results suggest that translocation, activation, and association of p72syk with transducing molecules such as phosphatidylinositol 3-kinase, events which occur during platelet activation, may participate in early signal-transduction events.

Origin of brain 2',3'-cyclic-nucleotide 3'-phosphodiesterase doublet.

The present study established that 2',3'-cyclic-nucleotide 3'-phosphodiesterase doublet common to mammalian brain originates from an alternative splicing. Peptides specific to the predicted larger translation product were synthesized and antisera against these peptides were prepared. Immunostaining of SDS/PAGE blots showed that the antisera react with the larger protein, but not with the smaller protein, of 2',3'-cyclic-nucleotide 3'-phosphodiesterase doublet in all mammals studied.

Mechanisms of dramatic fluctuations of ornithine decarboxylase activity upon tonicity changes in primary cultured rat hepatocytes.

We investigated the mechanisms underlying the marked induction of ornithine decarboxylase (ODC) activity by hypotonic treatment and its rapid decay upon reversal to isotonicity in primary cultures of adult rat hepatocytes. Upon hypotonic treatment, ODC synthesis rate increased progressively whereas the amount of ODC mRNA increased only about twofold. In addition, ODC was stabilized severalfold. ODC activity rapidly decreased upon restoration of isotonicity, owing to immediate and nearly complete suppression of ODC synthesis and 3-6-fold stimulation of ODC decay. The stimulation of ODC decay caused by restoration of isotonicity was mostly independent of time and protein synthesis. ODC decay was also stimulated by putrescine, even under hypotonic conditions, depending on time and new protein synthesis. Restoration of isotonicity and putrescine treatment together caused a synergistic stimulation of ODC decay, confirming that these act by different mechanisms.

Electron microscopic observation of calcitonin gene-related peptide-like immunoreactivity in the organ of Corti of the rat.

Calcitonin gene-related peptide (CGRP)-like immunoreactive (CGRP-IR) nerve terminals in the organ of Corti of rats were studied by light and electron microscopy. Surface preparation of the organ of Corti were immunostained using anti-CGRP antiserum for avidin-biotin immunohistochemistry. Dense CGRP-IR fiber bundles were observed by light microscopy in the inner spiral bundles, tunnel spiral bundles and outer spiral bundles. Electron microscopic analysis indicated that CGRP-IR fibers belong to efferent nerves. In the inner spiral bundles, the CGRP-IR fibers showed a direct contact mainly with non-immunoreactive afferent fibers. Some CGRP-IR nerve endings in the inner spiral bundles formed contacts directly with inner hair cells. In the outer spiral bundles, CGRP-IR fibers formed synaptic contacts exclusively with the outer hair cells. It should be noted that the number of synapses of CGRP nerve endings with outer hair cells varied depending upon the sub-row: a falling gradient in number occurred along the inner-outer axis. Our results suggest that CGRP acts as an efferent neuromodulator in the organ of Corti.

[Fracture toughness of porcelain using indentation method].

Material resistance to brittle fracture was quantitatively evaluated in the commercial porcelains, CERA 8 (CE 8), VITA DUR (VITD), VITA VMK 68 (VIT), CERAMCO II (CE II), UNIBOND (UNB), NORITAKE SUPER PORCELAIN AAA (AAA), PENCRAFT (FEN), COSMOTECH (COM) and OPTEC HSP OPT) from values of fracture toughness (KIC) obtained in the crack on a mode I, determined upon insertion of Vickers indenter. The results are summarized as follows: 1) Mean maximum and minimum values of Vickers hardness degree of 9 kinds of commercial porcelains at 5kgf of load for 15s were 1348 (SD 98.1) for OPT abd 666 (SD 74.6) for CE 8, respectively. 2) The value of half of the diagonal of indentation (a) ranged from 42 (SD 1.5) to 58 (SD 3.3) microns, and that of half of the crack length (c) ranged from 101 (SD 4.0) to 175 (SD 17.2) microns. 3) The ratio of (c) to (a) (c/a ratio) was within the range of 2.3 to 3.3, and median cracks were present. 4) KIC in the commercial porcelains determined by the indentation method was within the range of 2.04 to 4.69MNm-3/2, showing a maximum for OPT and minimum for VITD. 5) KIC of OPT was significantly greater than that of any other material. 6) The porcelains were divided by fractography of the direction of crack course into 2 groups: a group of intra-granular fracture showing linear cracks (AAA, COM and OPT) and a group of Inter-granular fracture showing a range of non-linear cracks (CE 8, VITD, VIT, CE II, UNB ND PEN).

Ontogeny of calcitonin gene-related peptide in the organ of Corti of the rat.

The ontogeny of calcitonin gene-related peptide (CGRP)-like immunoreactive (CGRPI) fibers in the rat cochlea was examined using immunocytochemistry. It was found that the CGRPI cochlear system developed markedly in the postnatal period; CGRPI fibers first appeared in the inner spiral bundle at postnatal day 8, then at the base of the outer hair cells, tunnel spiral bundle and tunnel radial fibers at postnatal day 19. The ontogenetical profile elucidated by this study suggests that CGRP has a role as a neuromodulator or neurotransmitter in this system.

Is recombinant human granulocyte colony-stimulating factor (G-CSF) orally available in rats?

Oral availability of recombinant human granulocyte colony-stimulating factor (G-CSF) was investigated in rats by measuring the blood total leucocyte (BTL) counts. Oral test G-CSF solution was prepared with 10% HCO-60 (polyoxyethylated, 60 mumol, castor oil derivative), 1% DK ester (sugar ester) or 10% MYS-40 (polyethyleneglycol monostearate), in which the G-CSF concentration was 500 or 250 micrograms/ml. Each test solution was injected into the duodenum of three rats at the G-CSF dose level of 300 or 600 micrograms/kg, and BTL counts were monitored for 48 h. All of the test G-CSF solution raised the BTL levels within 24 h after injection. In particular, the HCO-60 solution increased the BTL levels over 2 times as compared to the predose level at 600 micrograms/kg dose and the effect was apparently dose-dependent. A short-term study suggested that the effect of G-CSF on the BTL level appeared at the fastest at about 5 h after administration of HCO-60 test solution, 300 micrograms/kg. In view of the pattern of BTL dynamics obtained after i.v. injection of HCO-60 solution at 25 and 50 micrograms/kg, the increase of BTL levels observed after oral administration of the HCO-60 solution is considered to be due to the orally supplied G-CSF.

Plasma fluoride concentration and urinary fluoride excretion in obese and non-obese patients following enflurane anesthesia.

Plasma fluoride concentrations and urinary fluoride excretions were measured following enflurane anesthesia (1.5%, 2 hours) in obese (8 cases) and non-obese (9 cases) patients. At the end of anesthesia, there was no significant difference in plasma fluoride concentrations between the two groups. In the several days following anesthesia, however, plasma fluoride concentrations in obese patients were higher than those in non-obese patients. Urinary fluoride excretions after anesthesia were greater in obese patients than those in non-obese patients, and the period of increased fluoride excretion was prolonged in obese patients. These results suggested that obese patients metabolized more enflurane than non-obese patients during the postanesthetic period. In obese patients, their excess fatty tissue may cause a greater and more prolonged elevation of blood enflurane concentrations after anesthesia.

Localization of calcitonin gene-related peptide in the organ of Corti of the rat: an immunohistochemical study.

The localization of calcitonin gene-related peptide (CGRP)-like immunoreactive (CGRPI) structures in the cochlea was examined in the rat using immunocytochemistry. Numerous CGRPI fibers entered the organ of Corti in the intraganglionic spiral bundle and formed a dense fiber patch at the base of the inner hair cells. Much fewer, but still a significant number of CGRPI fibers were seen at the synaptic region of the outer hair cells. Since no immunoreactive cells were seen in the organ of Corti and spinal ganglion, these fibers may be one of the major components of the olivocochlear bundles originated from the superior olivary complex.

PACS in Kochi Medical School Hospital.

We have evaluated clinically CRT-based diagnosis and the performance of a picture Archiving and Communication System (PACS) in the Department of Radiology, Kochi Medical School. The important requirements of PACS are a good human interface with the diagnostic workstation and a long-term image archiving device. Because the style of image management varies from hospital to hospital, an analysis of the image management policies of each hospital is also important for the configuration of PACS.

Individual variation in semen parameters of healthy young volunteers.

Individual variation in semen parameters was investigated in healthy young volunteers. Semen samples were collected approximately once a month over a one-year period for a total of 93 samples (5 to 10 samples per subject) from 12 volunteers in their twenties. Semen analysis was carried out according to the WHO Manual. The amount of variation in each semen variable was calculated for each subject by dividing the maximum value by the minimum value. The results showed that the semen volume varied by 1.9+/-0.8 fold (1.3 to 4.2 fold), the sperm concentration by 4.8+/-4.3 fold (1.5 to 17.2 fold), the percentage of sperm with forward progression by 2.8+/-1.4 fold (1.6 to 6.4 fold), the percentage of sperm with rapid linear progression by 3.4+/-2.6 fold (1.7 to 10.9 fold), the percentage of sperm with normal morphology by 1.9+/-0.4 fold (1.3 to 2.4 fold), and the percentage of live sperm by 1.5+/-0.4 fold (1.1 to 2.6 fold). A between-group comparison showed significant differences in all of the variables except the percentage of sperm with normal morphology. These results suggest multiple and considerable semen analyses are needed when evaluating semen parameters.

XX-male syndrome bearing the sex-determining region Y.

The case of a 25-year-old man who presented for evaluation of infertility is described. The physical examination revealed testicular atrophy without gynecomastia. Repeated seminal analyses showed azoospermia, and serum hormonal levels suggested a state of a hypergonadotropic hypogonadism. Chromosomal analysis demonstrated 46XX. Polymerase chain reaction revealed the existence of a sex-determining region Y. The etiology of this rare sex reversal syndrome is discussed and cases reported in Japan are reviewed.
